        곡물류의 형질전환 유도에 관한 연구 (4) - 벼 배발생 세포의 생화학적 특징

        Rice (Oryza sativa L.) calli containing both embryogenic callus (EC) and nonembryogenic callus (NEC) regions were initiated from the mature seed on MS medium supplemented with 2.0 mg/L 2,4-D, 0.5 mg/L kinetin. The calli were developed into two callus type which can be distinguished by visual examination depending on color and appearance. In order to illucidate the polypeptide composition between EC and NEC, the total protein extracted from two types of callus was analysed by electrophoresis. By one-dimesional analysis of SDS-PAGE and Isoelectric focusing, several protein bands showed quantitative and qualitative difference in each type of callus. The further analysis of the total protein with two-dimensional electrophoresis showed at least 20 EC specific protein and 10 NE specific protein. Also 3 specific protein spots showing microheterogeneity of 90, 65, 50 kD were detected in EC, while a series of acidic heterologous protein spots were visualized in NEC.

        벼 현탁배양을 통하여 분리된 원형질체로부터 식물체 재분화

        Plant regeneration was accomplished from protoplast culture of rice (Oryza sativa L., cv. Taebaeg). Embryogenic callus was induced from mature seed on MS medium containing 5 mM proline, 2.5 mg/L 2,4-D, 30 g/L sucrose in the dark at 28℃ and used to establish embryogenic cell suspension culture. Suspension cells were subcultured every one week in N6 medium supplemented with 5 mM proline, 200 mg/L casein hydrolysate, 2.5 mg/L 2,4-D and amino acids of AA medium. Suspension cultures were composed of cells that were densely cytoplasmic, potentially embryogenic and were at least maintained for more than 6 months in liquid medium. Protoplasts were isolated from fast-growing suspension culture cells and cultured in a slightly modified KpR medium by mixed nurse culture. Isolated protoplasts began to divide within 5∼7 days and thereafter, protoplast-derived calli were sequentially transferred to callus proliferating medium that soft agar MS medium contained 2㎎/L 2,4-D and produced distinct embryogenic cells. Microcolonies were then transferred to solid medium which consisted of MS medium containing 5 mg/L kinetin, 1 mg/L NAA, 1 mg/L ABA, 30 g/L sucrose and 10 g/L sorbitol under fluorescent light. Multiple shoots of 4∼5 per callus emerged and were transeferred to hormone-free MS medium for root initiation. Thereafter, The plantlets were transferred to pots of soil to mature in the culture room.

      • 배발생 세포 관련 유전자 분석

        Both embryogenic(EC) and nonembryogenic calli(NEC) derived from the matured seed embryo of rice (Oryza sativa L. cv Donggin) were analyzed for differences in their gene expression. Two partial cDNA of an embryogenic cell specific gene was cloned by differential display of amplified cDNAs using a 12-mers synthetic nucleotide as a single primer. The expression of these clones, designated as REC1 and REC2, respectively, was confirmed by northern blot analysis that high levels expression only in the embryogenic cells. The 250 base of the cDNA clone REC1 have no homology with known genes. The cDNA nucleotide sequenced of the clone REC2 which is 649 base in length, was identified as a partial sequence of genes in Alcaligenes eutrophus. The partial amino acid sequence deduced from the clone REC2 showed 75 to 82 % homology with cobalt-nickel resistant proteins of Alcaligenes eutrophus.

        챔버 내측에 스프링형상을 갖는 유수형 자외선 살균장치 시뮬레이션

        Interest in application of ultraviolet light technology for primary disinfection of potable water in drinking water treatment plants has increased significantly in recent years. The efficacy of disinfection processes in water purification systems is governed by several key factors, including reactor hydraulics, disinfectant chemistry, and microbial inactivation kinetics. The objective of this work was to develop a computational fluid dynamics(CFD) model to predict velocity fields, mass transport, chlorine decay, and microbial inactivation in a continuous flow reactor. In this paper, It describe the how to design optimal UV disinfection device for ground water, BWT and rainwater. Spring shape instrument silver coated located in inner side of disinfection chamber. It make lead the active flowing movement target water and maximize disinfection performance. To search the optimal design method, it was performed computer simulation with 3D-CFD discrete ordinates model and manufactured prototype. Using proposed design method, performed simulation and proved satisfied performance.

        스쿼트운동장치의 풋플레이트 구조해석에 관한 연구

        스쿼트운동은 운동방법과 자세에 대한 이론적 근거 및 동작에 대한 기준 확립을 통해 안전하고 효과적으로 운동성과를 기대할 수 있는 중요한 프리웨이트운동 중의 하나이다. 그러나 바르지 못한 운동에 의한 부작용과 그에 대한 과학적 대응방안에 대한 연구와 운동모형의 개발 또한 필요하다. 스쿼트운동을 위한 풋플레이트를 설계하기 위한 구조해석의 주안점은 동적 거동을 단순화하여 구분 동작으로 모델링을 수행한다. 모델링을 기반으로 설계된 풋플레이트 바닥면에 위치한 로드셀의 한계 하중으로 인한 구조적 안전성 여부를 변형율과 응력의 크기를 산출함으로써 구조의 안정성과 이로 인한 운동기구의 설계방법을 제시한다. 이러한 스쿼트운동에 따른 세그먼트별 모델링과 지면반발력에 대응한 역학적 시뮬레이션 분석, 그리고 하중해석을 통한 운동역학분석의 결과를 통해 프리웨이트운동을 지원하는 운동보조장치, VR, 등의 기구에 적용하여 관련 시스템설계를 완성할 수 있다. 본 논문에서는 스쿼트운동 기구에 활용되는 풋플레이트의 설계 시에 인가되는 수직하중분포에 대한 설계방법을 제시하고 이러한 결과를 토대로 보다 안전하고 신뢰성 높은 운동보조기구시스템의 설계 및 제작기술을 제시하였다. Squat exercise is one of the important free weight exercises that can safely and effectively expect the athletic performance by establishing the rationale. Therefore, it is necessary to study the side effects caused by incorrect exercise, scientific countermeasures and to develop a exercise estimation model. It is effective and accurate to use a variety of assistive devices to calibrate athletic posture. The issues of the structural analysis for designing a foot plate for squat exercise is to model the behavior by the dynamic behavior. It should be consider that the center of gravity of each segmented body is different when the maximum load is applied. It is applied to complete system design through simulation method with kinematic dynamic, ground reaction force and load analysis for the free weight exercise equipment, VR device, and safety foot plate. In this paper, the authors propose the design method for the vertical load distribution applied in the design of the foot plate used for the squat exercise mechanism, and based on these results, design make the more safe and reliable free weight exercise equipment system.

        UV램프를 이용한 유수처리형 살균장치의 설계방법

        A number of factors combine to make ultraviolet radiation a superior means of water purification for ground water, rainwater harvesting systems and so on. Ultraviolet radiation is capable of destroying all types of bacteria. Additionally, ultraviolet radiation disinfects rapidly without the use of heat or chemical additives which may undesirably alter the composition of water. In a typical operation, water enters the inlet of a UV lamp and flows through the annular space between the quartz sleeve and the outside chamber wall. The irradiated water leaves through the outlet nozzle. Several design features are combined to determine the dosage delivered. The first is Wavelength output of the lamp, the Second is Length of the lamp - when the lamp is mounted parallel to the direction of water flow, the exposure time is proportional to the length of the lamp, the third is Design water flow rate - exposure time is inversely related to the linear flow rate, the forth is Diameter of the purification chamber - since the water itself absorbs UV energy, the delivered dosage diminishes logarithmically with the distance from the lamp. In this paper, It describe the how to design optimal UV disinfection device for ground water and rainwater. To search the optimal design method, it was performed computer simulation with 3D-CFD discrete ordinates model and manufactured prototype. Using proposed design method manufactured prototype applied to disinfection test and proved satisfied performance.

        영어 수동구문의 by-phrase와 한국어 수동구문의 -에/에게

        The main purpose of his paper is three-fold: First, we summarize the arguments that by-phrase in English passives is an adjunct, not an optional complement and analyse the passive constructions on the basis of hypotheses of Bouma, Malouf & Sag(1998). That is, the prepositional phrase, by-phrase, has been treated as an adjunct in previous studies so far: Kiparsky(1987), Grimshaw(1990),Wechsler(1995) and Jo & Chung(2000). Second, we point out that the dative particle -ey/eykey shows that it is a postposition rether than a morphological case-marker. We use the various tests to see if -ey/eykey is a case or a postposition. Third, on the basis of Urushibara(1991), O'Grady(1991) and Chung(1995), we argue the -ey/eykey in Korean passives should not be a complement but an adjunt. Empirically, we have also demostrated that our argument enable us to analyse further Korean passive types in Korean.(Chosun University)

      • 벼의 배발생 세포로부터 식물체 재분화

        벼 현탁배양 세포로 부터 체세포 배발생에 의해 재분화 개체가 이루어졌다. 배발생에 의해 재분화 개체가 이루어졌다.배발생 캘러 스는 2.5mg/ℓ2.4-D 가 첨가된 MS 배지에서 유도되었으며,현탁배양은 5mM proline, 2.5mg/ℓ2.4-D 그리고 200mg/ℓcasein hydrolysate 가 첨가된 N6 medium 에서 계대배양하였다. 이와같은 현탁배양세포는 세포질이 충만된 세포들로 구성되었으며,액체배지에서 10개월이상 유지시켰을때,체세포 배 발생 빈도는 지속적으로 유지되었으며 현탁배양세포를 5mg/ℓkinetin, 1mg/ℓNAA, 1mg/ℓABA, 3% sucrose 와 1% sorbitol이 첨가된 MS 고체배지로 옮겨졌을때 높은 빈도의 식물체 재분화가 이루어졌다. This study was carried out to obtain the high frequency of plant regeneration from cultured cells of rice(Oryza sativa L. ev Taebaeg). Embryogenic callus was induced from matured-seed on MS medium conatining 2.5mg/ ℓ 2.4-D and used to establish embryogenic cell suspension culture. Embryogenic cell suspension cultures were subcultured every one week in N6 medium supplemented with 5 mM proline,200mg/ℓcasein hydrolysate and 2,5mg/ℓ2.4-D. Embryogenic suspension cultures were composed of cells that were densely cytoplasmic, potentially embryogenic and maintained for more than 10 months. Somatic embryo formed when embryogenic cell suspentions were transferred to solid medium which consisted of MS medium containing 5mg/ℓkinetin, 1mg/ℓNAA, 1mg/ℓABA,3% sucrose and 1% sorbitol and subsequently the high frequency of plant regeneration were achived.
