        Effects of endocrine disrupting chemicals on expression of phospholipidhydroperoxide glutathione peroxidase mRNA in rat testes

        Phospholipid hydroperoxide glutathione peroxidase (PHGPx), an antioxidative selenoprotein, is modulated by estrogen in the testis and oviduct. To examine whether potential endocrine disrupting chemicals (EDCs) affect the microenvironment of the testes, the expression patterns of PHGPx mRNA and histological changes were analyzed in 5-week-old Sprague-Dawley male rats exposed to several EDCs such as an androgenic compound [testosterone (50, 200, and 1,000 μg/kg)], anti-androgenic compounds [flutamide (1, 5, and 25 mg/kg), ketoconazole (0.2 and 1 mg/kg), and diethylhexyl phthalate (10, 50, and 250 mg/kg)], and estrogenic compounds [nonylphenol (10, 50, 100, and 250 mg/kg), octylphenol (10, 50, and 250 mg/kg), and diethylstilbestrol (10, 20, and 40 μg/kg)] daily for 3 weeks via oral administration. Mild proliferation of germ cells and hyperplasia of interstitial cells were observed in the testes of the flutamide-treated group and deletion of the germinal epithelium and sloughing of germ cells were observed in testes of the diethylstilbestrol-treated group. Treatment with testosterone was shown to slightly decrease PHGPx mRNA levels in testes by the reverse transcriptionpolymerase chain reaction. However, anti-androgenic compounds (flutamide, ketoconazole, and diethylhexyl phthalate) and estrogenic compounds (nonylphenol, octylphenol, and diethylstilbestrol) significantly upregulated PHGPx mRNA in the testes (p < 0.05). These findings indicate that the EDCs might have a detrimental effect on spermatogenesis via abnormal enhancement of PHGPx expression in testes and that PHGPx is useful as a biomarker for toxicity screening of estrogenic or antiandrogenic EDCs in testes.

        생활수준의 향상으로 일반가정에서 사용하는 전기용품의 종류도 무척 다양해졌으며 사용량도 급격히 증가되어 이로 인한 전기재해(화재, 감전)가 끊이지 않고 있다. 행정자치부 화재통계 연보에 따르면 ''99년도 총 화재발생건수 33,856건 중 전기로 인한 화재가 11,204건으로 33.1$\%$의 높은 점유율을 보이고 있어 일반가정에서의 전기안전 생활화가 절실히 요구되고 있는 실정이다. 일반 가정생활에서 편리하게 사용하고 있는 전기를 보다 더 안전하게 사용하기 위해서 우리 모두가 꼭 알아두어야 할 전기설비에 관한 기초상식, 전기사고의 종류와 예방대책, 정전 시 대처요령 등을 알아본다.

        C57BL6 생쥐에서 NDM 헤어토닉의 모발성장 촉진 효과

        NDM hair tonic is composed of several plant extracts which are known to be used in oriental medicine. This study was carried out to investigate the effect of NDM hair tonic on hair regrowth in an alopecia model of C57BL6 mice. Hair of six-weeks old mice were trimmed by electric clippers and electric shavers, so as not to damage the skin. The next day, mice without visible scraches were selected, randomized and separated in groups of 6 mice. There were four experimental groups including saline (negative control), 50% ethanol (vehicle control), 3% minoxidil (MXD), and NDM tonic. The test compounds were topically treated with 0.15 $m\ell$ per mouse per day for 2 weeks. The hair regrowth was determined photographically and histologically and the quantity of endocrine factors, IGF-1 and TGF-$\beta$, in the skin of mice was measured by PCR. No clinical signs were found in all animals. The topical treatment of NDM tonic for 2 weeks to dorsal skin accelerated hair regrowth faster than either the controls or MXD treatment. The NDM tonic treatment also promoted hair follicle development and elongation as compared with the controls or MXD treatment. Both NDM tonic and MXD treatment significantly increased the expression of IGF-1 and TGF-$\beta$ in the skin of C57BL6 mice as compared with the controls (p<0.05). These results suggest that NDM tonic has a hair growth activity and be useful for the treatment of baldness or alopecia.

      • 마우스 B 림프구에서 비특이적 자극에 의한 CD30 발현의 유도

        CD30 was originally identified on Hodgkin and Reed-Sternberg (H-RS) cells and subsequently defined as an activation antigen. CD30 expression is observed on activated, but not on resting, T and B lymphocytes. Despite of numericcal studies, the functions of CD30 in physiological condition remains open question. Moreover, CD30 expression of normal B lymphocytes has been poorly documented. In this study, CD30 expression of murine B lymphocytes and its regulation was analyzed. B cell-enriched (90%) spleen (SP-B) cells obtained by nylon-wool adherence were used for activation with LPS or plate-bound anti-mouse IgM. LPS stimulation induced minimal expression of CD30 and addition of cytokines (IL 2 or IL 4) resulted in higher number of activated cells, but not higher expression of CD30. Substantial CD30 expression of SP-B cells was induced by stimulation with plate-bound anti-mouse IgM in a dose dependent manner. IL 4 upregulated proliferation of and CD30 expression on activated SP-B cells. IL 2 enhancd B cell proliferation but not CD30 expression and also did not show any additional effect on IL 4-mediated upregulation of CD30 expression. These data suggest that the signaling pathway for B cell proliferation is different from that for induction of CD30 expression and T and B cells may show distinct response to cytokines in CD30 expression.

        To produce Sendai virus for induction of α-interferon(α-IFN, Sendai virus (Cantell's strain, 10,000 hemagglutinin units(HAU/ml) was injected into the allantoic cavity of 11 day-old fertile hens' eggs. After inoculating 100, 300, 1000 or 2000 HAU/0.2ml into the eggs, the hemagglutinin(HA) titre was checked daily. The highest titre was shown from the eggs inoculated with 2000 HAU/0.2ml, and cultured for 3days at 37℃. Difference in α-IFN productivity of Sendai virus made Flow laboratory(USA) and the virus made in our laboratory was not significant(P>0.1).

        It has been well documented that global suppression of the immune responses occurs after thermal injury and these changes resulted in decreased resistance to infection, which is a leading cause of high mortality of burn patients. Regarding to numerical changes of lymphocytes related to this immune defects, a lot of controversial results have been reported. In this study, the distribution of leukocytes was investigated in thermally injured mice. Mice (Balb/c) were anesthetized by intraperitoneal injection of 2,2,2-tribromoethanol and thermally injured by immersion of hair-removed dorsal skin (15% total body surface) in a boiling water bath (ca. 96℃). Mortality of mice injured for 7, 9 and 11 seconds was <5%, 25 and 100%, respectively. Almost all of mice injured for 7 seconds recovered from wounds after 40 days postinjury, which were used for all experiments. Significant decrease of peripheral blood leukocyte (PBL), lymph node cell (I,NC) and spleen cell (SPC), thymocyte(Thy) numbers was observed at day 2 and followed by gradual increase to (PBL) or over normal level (LNC and SPC) until day 24 of injury, whereas thymocytes sustained lowered numbers. Such increase of cell number coincided with higher proportion of PMN, particularly in spleen (5% vs 30%). Reduction of lymphocyte proportion was due to relative increase of PMN number, not the decrease of absolute number of lymphocytes. This increasing number of PMN may explain the diminished immune response, assessed with whole spleen cells in a murine model. These results also suggest that trafficking of PMN may be important for the recovery from thermal trauma. In addition, the Th/Ts ratio of injured mice was comparable to that of control group, which is not likely to be a critical factor leading to alteration of immunity in thermal injury.

        랫드의 화상모델에서 전복껍질과 참기름의 치료효과

        This study was performed to investigate effect of a mixture of ear shell ash and sesame oil on bum in a Sprague-Dawley rat model. Sixty male rats were assigned to 3 experimental groups including negative and positive control groups, and a test material group. The test compound was applied to formally once a day f3r 7 days. Distilled water was applied as a relative positive control. After dermal applications f3r 3, 5, and 7 days, blood hematology and skin histopathological examination were performed. Inorganic compounds in ear shell ash was mainlycalcium oxide(92%). The body weights of positive control rats were significantly low at day 3, 5, and 7 as compared with negative control and test material groups. There was no significant difference in body weight between negative control and test material groups throughout the experimental period. WBC counts in both positive control and test material groups were higher than those in negative control group, indicating progress of inflammatory reaction. In differential leucocyte count, panulocytes including neutrophils and eosinophils were higher in positive control and test material groups than negative control group. Hemoglobin and hematocrit values were lower in homing model rats, probably resulting from hemorrhage in dermis. Skin histopathology showed severe full thickness coagulative necrosis in epidermis and dermis, hemorrhage and infiltration of leucocytes in dermis in both positive control and test material groups throughout the experimental period. These results indicate that the test materials, ear shell ash plus sesame oil, moderately ameliorate homing probably concerning to process of skin injury repair including inflammation and fibrosis
