      • Polyvinylpyrrolidone-Iodine 處理에 依한 數種菌類의 微細構造 變化

        It has been studied that fine structural changes of several microorganisms (Escherichia coli, Neisseria gonorrhoeae, Staphylococcus aureus, Candida albicans, Cryptococcus neoformans) treated with Polyvinylpyrrolidoneodine. The alterations observed as follows: 1. It was observed that change of nucleoid, aggregation of ribosomal particles and other cytoplasmic alterations in E. coli, N. gonorrhoeae, S. aureus which were accompanied by marked breakdown of cell membrane and cell wall. 2. Particularly capsules of S. aureus was collapsed by polyvinylpyrrolidoneIodine. 3. Ultrastructural changes of above mentioned was also appeared that the more marked changes as the exposure time are longer. 4. The intra-cytoplasmic membrane systems in the cytoplasm which can observed in the control group of C. albicans and C. neoformans were disappeared because they were effected extensively by drug. The electron transparent area of cells was increased in the longer exposure group and their cell membrane, cell wall and capsule were occurred marked changes. 5. In this experiment it was considered that treatment of polyvinylpyrrolidone Iodine on several microorganisms occurred responsible cellular changes.

      • 抗生劑( Siccanin 및 Azalomycin-F )處理에 依한 Candida albicans의 電子顯微鏡的 觀察 : Siccanin and Azalomycin-F

        The present study is an ultrafine structure of Candida albicans treated with Siccanin and Azalomycin F by means of electron microscopy. The results as follows: 1. In contrast to the bacteria, the normal Candida albicans contains nuclear envelop, mitochondria, endoplasmic reticulum, distinct cell wall, cell membrane and nucleus as observed in the eucaryotic cells. 2. Treated Candida albicans with Siccanin and Azalomycin-F was changed their morphology and cytoplasmic components, for examples, thicking of cell wall, partial delation of nuclear envelop, mictochondria, detachment of cell membrane from cytoplasm and increasing of lipid particles. 3. When the cells were treated with the tested antibiotics at 10 mcg per ml. for 60 minutes, the cellular damges were most severe.

      • Studies on Fine Structure and Enzyme Activity of Toxoplasma gondii in Macrophage

        마우스 腹腔內에 流動 파라핀을 注射하여 巨食細胞를 誘導하고 다시 Toxoplasma RH 株 接種시켜 Toxoplasma 營養型과 宿主細胞의 相互關係를 電子頭微鏡的으로 觀察하고 微細構造에 對한 酵素活性反應을 본 바 다음과 같은 結果를 얻었다. 1. 遊離 Toxoplasma 營養型이나 巨食細胞內의 體와 宿住細胞 사이에는 parasitophorous vacuolo 이 形成되었고 그 間隔에는 stereocilia 가 觀察되었으며 이는 Toxoplasma의 pellicle에서 由來됨을 알수있었다. 4. Parasitophorous vacuole은 Toxoplasma에서 生成되는 thread-like filament에 의해 形成됨을 보았다 5. 巨食細胞內에 散在해 있던 ribosome-like granular materials는 parasitophorous vacuole이 形成되면서 vacuole 內로 모여드는 경향을 볼 수 있었고 parasitophorous vacuole 주의로 宿主細胞의 mitochondria가 나열됨을 觀察하였다. 6. A챵 phosphatase 의 活性은 pellicle의 前半部 및 parasitophorous vacuole의 內容物, 그리고 phagosome-like vacuoles에서 强하게 나타났으며, peroxidase 의 活性은 rhoptries의 外膜과 parasitophorous vacuole 周圍에 나열된 巨食細胞 mitochondria 內의 cristae에서 强한 反應을 보였다. Micromorphology and enzyme activities of Toxoplasma gondii in macrophage were studied. One ml of sterile liquid paraffin was injected into peritoneal cavity of mouse five days prior to Toxoplasma inoculation to stimulate macrophage production in peritoneal fluid. Two or 3 days after the Toxoplasma inoculation, the peritoneal exudates were aspirated, and the isolated Toxoplasma or macrophages which contained parasites were subjected for micromorphological and enzymatic studies. The intra-and extracellular trophozoites of T. gondii had pellicle with an outer and inner membrane, a conoid, micronemes, elongated rhoptries with bulbous alveolar posterior ends, a nucleus, osmiophilic bodies and lipoid bodies. The lipoid bodies scattered in cytoplasm of Toxoplasma, but tended to get together after engulfed in macrophage. A parasitophorous vacuole developed in later stages of the parasite, and membranous folds and stereocilia were observed in it. Stereocila in the vacuole and the thread-like filaments seemed to be originated from Toxpolasma. Ribosome-like granular materials in macrophage gathered into the parasitophorous vacuole, and mitochondria of the host cell lined up around the periphery of the vacuole. Acid phosphatase activity was demonstrated in the two-layered anterior part of the pellicle, contents of the parasitophorous vacuole and phagosome-like vacuoles. Peroxidase activity was demonstrated in the outer membrane of rhoptries of the parasite and the cristae of the mitochondria which lined colse to the parasitophorous vacuole.

      • 數種 藥劑 處理에 依한 Candida albicans의 電子顯微鏡的 觀察

        This study is an ultra-microscopic structures of Candida treated with various chemicals such as gentian violet, nystatin, trichomycin and pimaricin by means of electron microscopy and reveals the followings. 1. The cells of Candida albicans have a complex organization resembling that of higher organisms. 2. In the cells seen after exposure to various chemicals (nystatin, trichomycin and pimaricin), thickening of cell walls and increasing of electron transparent area (ETA). 3. Gentian violet treated groups are seen change of cytoplasmic component and suggested that less actively metabolizing in the fungus cells. 4. Increase of electron transparent area was thought to be associated with autolysis.
