Background/Aims: Nonalcoholic steatohepatitis (NASH) is prevalent in both economically developed and developing countries. Twenty percent of NASH progresses to cirrhosis with/without hepatocellular carcinoma, and there is an urgent need to find biomarkers for early diagnosis and monitoring progression of the disease. Using immunohistochemical and immunoelectron microscopic examination we previously reported that expression of matrix metalloproteinase-1 (MMP- 1) increased in monocytes, Kupffer cells and hepatic stellate cells in early stage NASH. The present study investigated whether serum MMP-1 levels reflect disease activity and pharmaceutical effects in NASH patients. Methods: We measured the serum levels of MMPs, tissue inhibitors of metalloproteinases (TIMPs), and several cytokines/ chemokines in patients with histologically proven early and advanced stages of NASH and compared them with those in healthy controls. Results: Serum MMP-1 levels in stage 1 fibrosis, but not in the more advanced fibrosis stages, were significantly higher than in healthy controls (P=0.019). There was no correlation between serum MMP-1 level and fibrosis stage. Serum MMP- 1 levels in NASH patients represented disease activity estimated by serum aminotransferase values during the followup period. In contrast, MMP-2, MMP-9 and TIMPs did not change with disease activity. Consistent with the finding that MMP-1 is expressed predominantly in monocytes and Kupffer cells, serum levels of monocyte chemotactic protein-1 and granulocyte-colony stimulating factor were significantly increased in NASH with stage 1 fibrosis. Conclusions: These results suggest that serum MMP-1 levels represent disease activity and may serve as a potential biomarker for monitoring the progression of NASH. (Clin Mol Hepatol 2018;24:61-76)

Objectives: Little is known regarding the associations between Borg’s rating of perceived exertion (RPE), cardiopulmonary responses, and carrying bags while level walking. We investigated the changes in RPE and cardiopulmonary responses, including energy cost (EC), oxygen consumption (VdotO₂), and heart rate (HR), when walking with a bag. Methods: This randomized crossover study included 10 healthy adults (aged 21–22 years). Four days of experiments were performed at intervals of 1–2 weeks. The participants walked on a level treadmill while carrying a bag with a strap weighting 5 kg, placed diagonally on their shoulders (DS), vertically on their shoulders (VS), vertically in one hand (VH), and on their backs using both shoulders (BS) for 20 min. Walking speed was stable among all experiments, and was set to the participants’ preferred speed. Results: No significant differences between bag carrying methods regarding EC, VdotO₂, and %HRreserve (based on HR) were observed, whereas RPE respiratory (RPE-resp) and RPE leg muscle (RPE-leg) scores varied based on bag carrying method used. RPE-resp score was significantly lower for DS and BS than VH (both p < 0.0083); however, no significant difference was observed when each RPE-leg score was compared. Conclusions: We found that RPE-resp score varied, whereas no changes in EC, VdotO₂, and HR were observed in healthy adults while walking with different bag carrying methods. Additionally, of all bag carrying methods assessed, the highest RPE-resp score was observed when individuals carried a bag with one-hand at their side.

To investigate the microcrack healing behavior of $Si_3N_4-20wt\%SiC-8wt\%Y_2O_3$ composite ceramics(SNCY8), we observe the crack length evolution a! the time of 20, 40, 60 minutes with in-situ optical microscopy by varying healing temperature of $800\~1200^{\circ}C$. Crack healing obviously occurred as heating temperature and time increased. We proposed a simple model of effective diffusion based on the crack length evolution with healing condition, and determined the effective diffusion coefficient as Our result implies that we may predict the healing ability quantitatively with temperature and time in structural ceramics through the effective diffusion coefficient model.
