        This study conducted the indoor slope model test which reproduces the rainfall after building up the artificial slope by considering the construction condition of the actual slope in order to understand the characteristics of the change of the quantitative value such as the pore water pressure, dangerous rainfall volume etc, in accordance with the superior seepage of the unsaturated banking slope in the time of rainfall. With this result. the change of the pore water pressure caused by the rainfall seepage, the estimation of time for the slope degradation and of the dangerous rainfall volume, the change of the seepage line, the characteristics of the change in safety rate by using the simplified Janbu's method (1968) have been analyzed and studied. The abstract for this study is as follows: 1. Since the soil within the slope is in the unsaturated state in the early period after the rainfall started, it is started at the pore water pressure of the negative value(-) in the measuring point around the outer layer and of the positive value(+) in the deep area. As the rainfall permeates into the slope, it become saturated, therefore, the pore water pressure also increases which lead to the degradation. After that, it tends to be gradually diminished. The pore water pressure had shown relatively considerable increase right before the degradation, and 50~150 minutes before the slope degradation happens, it had shown sharp increase. 2. The safety rate is sharply decreased when reached the slope layer after being started its activity at the slope line area; the time to the complete degradation of the slope layer by being the 1.0 for the safety rate after the degradation at the slope line area started is displayed short as the rainfall intensity strengthens, and longer as the slope inclination becomes gentler. Likewise, the time to the complete degradation from the slope line to the slope layer is relatively short for the slope built by the 8M than by the one built by the SW.

        CITIS(Contractor Integrated Technical Information Service) is a contractor's developed service to provide electronic access and/or delivery of contractually required information. CITIS satisfies one of the major CALS objectives to furnish a single-entry point for authorized access to contractor-generated Contract Data Requirement List(CDRL) data. A critical requirement for the practical implementation of the CITIS concept is the ability to access multiple existing heterogeneous databases in a fashion transparent to the CITIS user. In this paper, we will propose a prototype system model for CITIS, and discuss the system architecture of proposed CITIS system as a global information management system for distributed and heterogeneous local information systems. Furthermore, we will discuss about the required server module and using-scenario of proposed CITIS system.

      • 공복 시 Caffeine 투여 후 일회성 운동이 흰쥐의 인슐린 및 골격근의 GLUT-4 발현에 미치는 영향

        This study was done to investigate the effects of caffeine ingestion on the expression of GLUT-4 in rats' skeletal muscle(soleus m) and blood insulin level after 12 hr fasting condition and one hour treadmill running. Total 19 rats(Sprague-Dawley) who were 7week Did were used for experimental subjects and raised for 1week in the room controled the air temperature and relative humudity(22℃ and 50%). They were divided three groups ; 12hr fasting group(F group, n=5), fasting+exercise group(FE, n= 7) and fasting+caffeine+exercise group(FCE group, n=7). FG and FCE group performed treadmill running for one hour and the treadmill speed was 18m/min that was supposed to be moderate exercise to rats. Each Dr rats in FCE group was forced to he given the caffeine solution(5mg/kg, Sigma Chemical co.) through stainless steel tube directly to their stomachs one hour before treadmill exercise. Immediately after treadmill running, we anesthetized them with the mixed solution(Ketamin 80g/kg and Rompun 5ml/kg) to draw 3ml blood from heart four insulin analysis and then obtained the rats' soleus muscle for GLUT-4 expression. We found that FCE group showed significantly higher GLUT - expression then the other groups and FE group showed the lowest GLUT-4 expression. This rusults suggest that the endurance type of exercise in fasting condition inhibit the GLUT-4 expression in working muscle and the caffeine helped to facilitate the GLUT-4 expression during endurance type of exercise. The future studies needed to confirm the action of caffeine to increase the GLUT- 4 and the mechanism.

        The purpose of this study was to evaluate the effects of diet condition and caffeine ingestion prior to long distance treadmill running on the GLUT-4 protein expression in Sprague-Dawley rats' soleus muscle. Total is rats were used For subjects and assigned to six groups: Fasting control(FaC group), feeding control(FeC), asting+exercise(FaE), feeding +exercise(FeE), Fasting +caffeine+exercise(FaCE), and Feeding+caffeine+exercise(FeCE). Each animal belongs to fasting group was forced to be in fasting state at least for 14 hours, The food was provided to each rat in feeding group 12minutes prior to treadmill running and the caffeine was ingested through the stainless steel tube 60minutes prior to exercise. All rats in exercise groups were forced to perform treadmill running for B9 minutes at the speed of 18m/min, and 0%grade, immediately after exercise, all animal was anesthetized by Chlorofolm (DC Chemical, Co, Ltd) and about 10ml of blood was drawn from each animal's heart and than soleus muscle was exposed and separated from the rat's leg. we found that the GLUT-4 in feeding groups increased much significantly compared to fasting groups(p<.05). The exercise in fasting state appeared to suppress the expression of GLUT-4 protein different from the exercise in feeding state. Cafreine ingestion prior to treadmill running of 60 minutes induced the significant increase or GLUT-4 protein expression both in fasting or feeding states.

      • ERK1/2-Mediated Schwann Cell Proliferation in the Regenerating Sciatic Nerve by Treadmill Training

        <P>Proliferation of Schwann cells in the injured peripheral nerve supports axonal regeneration, and physical training in experimental animals has been shown to promote nerve regeneration. Extracellular signal-regulated kinase 1/2 (ERK1/2) activity can mediate neuronal responses to lesion signals, but its role in non-neuronal cells in the injured area is largely unknown. Here we report that treadmill training (TMT) facilitates axonal regeneration via the upregulation of phospho-ERK1/2 protein levels in Schwann cells in the injured sciatic nerve. Low-intensity, but not high-intensity, TMT increased neurite outgrowth of dorsal root ganglion (DRG) sensory neurons and potentiated Schwann cell proliferation. TMT elevated levels of GAP-43 mRNA and protein, and phospho-ERK1/2 protein in the injured sciatic nerves. TMT also enhanced phospho-c-Jun protein levels in the injured nerve. In-vivo administration of the ERK1/2 inhibitor PD98059 eliminated phospho-c-Jun, suggesting ERK1/2 phosphorylation of the c-Jun protein. PD98059 treatment decreased levels of BrdU-labeled proliferating Schwann cells in the distal portion of the injured nerve, and delayed the axonal regrowth that was promoted by TMT. The present data suggest that increased ERK1/2 activity in Schwann cells may play an important role in TMT-mediated enhancement of axonal regeneration in the injured peripheral nerve.</P>

      • Beneficial Function of Cell Division Cycle 2 Activity in Astrocytes on Axonal Regeneration after Spinal Cord Injury

        <P>Migrating activity of reactive astrocytes induced after spinal cord injury (SCI) controls glial scar formation by limiting inflammatory responses around the injury area, and, therefore, can be beneficial for regenerative responses of spinal axons. Recently, we found that cell division cycle 2 (cdc2) activity in primary astrocytes facilitated neurite outgrowth of co-cultured neurons. Here, we investigated the effects of cdc2 activity on regenerative processes in vivo after SCI. Administration of the cdc2 inhibitor purvalanol A restricted compaction of the injury cavity and astrocyte infiltration into the cavity. After SCI, regenerative responses of anterogradely labeled corticospinal tract (CST) axons were attenuated by purvalanol A treatment. Using the polymeric tube that was implanted into the spinal cord as a nerve guide conduit, we found that purvalanol A treatments reduced astrocyte migration into the tube graft and, in parallel, retarded the extension of spinal axons into the tube. These results suggest that astrocytes with cdc2 activity may play a permissive role in mediating regrowth of spinal axons after lesion.</P>

      • Involvement of Cdc2 in Axonal Regeneration Enhanced by Exercise Training in Rats

        PURPOSE:: Physical activity can improve sensorimotor recovery after peripheral nerve injury. We examined the effects of treadmill training (TMT) on axonal regeneration in the injured sciatic nerve of the rat and further investigated cellular and molecular events that underlie enhanced axonal regrowth by training. METHODS:: After crush injury of the sciatic nerves, rats were randomly assigned into either TMT or sedentary groups. Three to 14 d after injury, changes in protein levels in the regenerating nerve were analyzed by Western blotting and immunofluorescence staining. Axonal regeneration was assessed by anterograde and retrograde tracing techniques. The animals' functional recovery was determined by the sciatic functional index. RESULTS:: We identified enhanced axonal regrowth in the distal stump of the sciatic nerve 7-14 d after injury in the rats with TMT. Cell division cycle 2 (Cdc2) mRNA and protein levels were highly increased in the injured sciatic nerves 3 and 7 d after injury, and decreased to basal levels 14 d later. Daily TMT accelerated distal shift of Cdc2 mRNA and protein induced in the regenerating nerves, and Cdc2 kinase activity was similarly increased in the distal stump by TMT. Cdc2 protein induced by TMT was mainly colocalized with Schwann cell marker S100&bgr; protein, and correlated with axial distribution pattern of bromodeoxyuridine-labeled proliferating cell population in the regenerating nerve. We further demonstrate that axonal regeneration and motor function recovery after injury, both of which were promoted by TMT, were greatly suppressed by in vivo administration of Cdc2 inhibitor roscovitine. CONCLUSION:: The present data suggest that Cdc2 kinase activated in the regenerating sciatic nerve may play an important role in TMT-mediated enhancement of axonal regeneration.

      • Growth-promoting activity of Hominis Placenta extract on regenerating sciatic nerve1

        <P>AIM: Extract of Hominis Placenta (HP) has been used in oriental medicine as an agent for improving physiological function. The present study was conducted to investigate whether HP treatment in an experimental sciatic nerve injury animal model produces growth-promoting effects on regenerating peripheral nerve fibers after injury. METHODS: After HP was injected into a sciatic nerve injury site, changes in protein levels were analyzed in the regenerating nerve area by Western blotting and immunofluorescence staining analyses. For quantitative assessment of axonal regeneration, a retrograde tracing technique was used to identify the neuronal cell bodies corresponding to regenerating axons, and the extent of neurite outgrowth in cultured dorsal root ganglia (DRG) sensory neurons prepared from animals that had experienced a sciatic nerve crush injury 7 d before neuron collection was analyzed. RESULTS: Induction levels of axonal growth-associated protein (GAP-43) in the injured sciatic nerves were elevated by HP treatment. HP treatment also upregulated cell division cycle 2 (Cdc2) protein levels in the distal stump of the injured sciatic nerve. Induced Cdc2 protein was detected in Schwann cells, suggesting that Cdc2 kinase activity may be involved in the growth-promoting activity of regenerating axons via Schwann cell proliferation. Cell body measurement by retrograde tracing indicated that HP treatment produced significant increases in regenerating motor axons. Finally, HP treatment of cultured DRG sensory neurons significantly increased neurite arborization and elongation. CONCLUSION: HP promotes the regeneration of injured sciatic axons by upregulating the synthesis of regeneration-related protein factors such as GAP-43 and Cdc2.</P>

        Review on Molecular Biological Approaches to Investigate the Effect of Endurance Training

        The purpose of this study is to examine details of domestic and foreign papers that verified the effect of endurance training using molecular biological approaches and to investigate the directivity of molecular biological study, analytical technology and analytical factors for the analysis of training effects. Existing domestic and foreign papers on endurance training that investigated the effect of training based on molecular biological analysis of human beings were reviewed. In order to increase reliability of the selected papers and their compatibility with the purpose of this study, the papers were reclassified by a group of experts that consisted of physical education professors and doctors of exercise physiology. The final papers selected were studied for the change in hormones and enzymes through blood and muscular tissues based on study topic and trend analysis from a macroscopic perspective. Analytical methods used include western blot, RT-PCR, ELISA and immunohistochemistry of molecular biology. Analytical variables investigated / analyzed were ACE, ACTN, PPARs, UCP, mitochondrial DNA variant, creatine kinase, renin-angiotensin, AMPK for blood and COX, myosin binding protein C fast-type, glycogen phosphorylase, pyruvate kinase, GLUT, PPARs, and AMPK for muscle. Such molecular biological analytical techniques and variables should be widely used to provide 1) conditioning of professional athletes and life sports athletes, 2) analysis of training effect, and 3) scientific and future-oriented methods for health enhancement.
