<P>Since T cells express diverse sex steroid hormone receptors, they might be a good model to evaluate the effects of sex steroid hormones on immune modulation. Porcine testicular extract contains several sex steroid hormones and may be useful to study the effects of sex steroid hormones during T cell activation. We have examined the effects of the porcine testicular extract on T cell activation: proliferation and secretion of cytokines (IL-2 and IFN-관) by activated T cells were severely decreased after treatment with porcine testicular extract. The extract produced an immunosuppressive effect and inhibited the proliferation of activated T cells by blocking the cell cycle transition from the G(1) phase to S phase. These effects were mediated by a decrease in the expression of cyclin D1 and cyclin E and constitutive expression of p27(KIP1) after T cell activation.</P>

<P>β-Amyloid (Aβ)-induced neuronal toxicity in Alzheimer's disease (AD) is associated with complex mechanisms. Thus, a multi-target approach might be suitable for AD treatment. Following our previous study on the neuroprotective effects of red ginseng oil extract, its major compounds, including linoleic acid (LA), β-sitosterol (BS), and stigmasterol (SS), were examined to elucidate the mechanism of anti-apoptosis and anti-inflammation in Aβ25-35-stimulated PC12 cells. The results showed that the three compounds mitigated Aβ25-35 toxicity by regulating oxidative stress, apoptotic responses, and pro-inflammatory mediators. LA and SS strongly regulated intrinsic apoptosis markers, such as mitochondrial membrane potential, intracellular Ca<SUP>2+</SUP>, Bax/Bcl-2 ratio, and caspases-9, -3, and -8. However, BS blocked only the intrinsic apoptotic pathway, particularly by suppressing Ca<SUP>2+</SUP> accumulation. Furthermore, all three compounds downregulated iNOS and phospho-nuclear factor-κB, but only LA and SS inhibited the expression of cyclooxygenase-2 and phospho-IκB. In assays to evaluate MAPK expression for confirming upstream signal pathways, BS decreased the phosphorylation of p38 and ERK, but not JNK, while SS markedly decreased the phosphorylation of all three MAPKs, and LA clearly decreased the phosphorylation of ERK and JNK, but not p38. These results indicate that LA, BS, and SS act as neuroprotectives against Aβ25-35-induced injury by distinct molecular mechanisms, indicating their preventive and/or therapeutic potential to treat AD.</P>

The Impact of View Histories on Edit Recommendations

<P>Recommendation systems are intended to increase developer productivity by recommending files to edit. These systems mine association rules in software revision histories. However, mining coarse-grained rules using only edit histories produces recommendations with low accuracy, and can only produce recommendations after a developer edits a file. In this work, we explore the use of finer-grained association rules, based on the insight that view histories help characterize the contexts of files to edit. To leverage this additional context and fine-grained association rules, we have developed MI, a recommendation system extending ROSE, an existing edit-based recommendation system. We then conducted a comparative simulation of ROSE and MI using the interaction histories stored in the Eclipse Bugzilla system. The simulation demonstrates that MI predicts the files to edit with significantly higher recommendation accuracy than ROSE (about 63 over 35 percent), and makes recommendations earlier, often before developers begin editing. Our results clearly demonstrate the value of considering both views and edits in systems to recommend files to edit, and results in more accurate, earlier, and more flexible recommendations.</P>
