        In order to develop the curriculum of the fishery department in the fisheries Jr. college, We have collected and analyzed the domistic and foreign data adquated to the fishey department under the international trend, also we gathered the opinion from the inquiring paper investigation and consultation to the industrial field, concerned college and graduate during the 6 month from September 1982. The fishery department curriculum in fisheries Jr. college will be industrialized with the concerned technics and knowledge in relation to speciality of maritime affairs. Accordingly, we have developed the adquate curriculum which is practical and can be cultivated the technician to adopt in the industrial society.

      • PES를 이용한 Mg/Si(111)계의 초기 성장모드 연구

        LEED와 PES를 이용하여 Si(111)7×7 표면위에 흡착된 Mg의 초기 성장모드를 연구하였다. 상온∼200℃의 기판온도에서 흡착초기에서부터 Mg silicide의 형성을 관측할 수 있었고 이 구조는 최종적으로 2/3√3×2/3√3 R30° 구조의 LEED pattern을 가진 epitaxial silicide로 완성되었다. 계속된 Mg의 흡착은 이 silicide 위로 Mg bulk가 무질서하게 성장됨을 보여준다. 또한 300, 450℃의 기판온도에서는 각각 1×1 그리고 3×1 구조가 관측되었다. The initial growing mode of Mg adsorbed on Si(111)7×7 surface has been studied by low energy electron diffraction (LEED) and photoelectron spectroscopy(PES). With increasing deposition time, the LEED pattern was changed to diffuse 7×7, diffuse 1×1 and 2/3√3×2/3√3 R30° structures at room temperature, successively. Also, we observed the 1×1 and 3×1 patterns at 300 and 450℃, respectively. From the Leed and PES results, it has been found that a Mg silicide is formed at the initial adsorption stage and that the 2/3√3×2/3√3 R30° structure is responsible for the thin epitaxial Mg silicide at RT∼200℃.

        Cyanobacterium Synechocystis PCC6803 내에서 Bacillus thuringiensis subsp. morrisoni PG-14 cryIVD 유전자의 발현

        미생물 살충제로 사용되고 있는 B. thuringiensis를 모기유충방제에 적용하기 위한 시도는 B. thuringiensis가 수서생태계에서 부유하지 못하고 가라앉으며, 생활환을 이루지 못한다는 문제점으로 인해 적용에 어려움이 있다. 따라서 본 연구는 모기유충에 강한 독성을 갖는 B. thuringiensis subsp. morrisoni PG-14의 cryIVD 유전자를 모기유충의 먹이인 cyanobactrium에 도입하기 위하여 발현벡터 pCYASK 5-1을 제작하고, cyanobacterium Synechocystis PCC6803에 형질전환시켜, 세포내에서 cryIVD 유전자의 발현과 뇌염모기에 대한 독성을 조사하였다. 그 결과 형질전환체 내에서 cryIVD 발현은 immunoblot 분석을 통해 B. thuringiensis subsp. morrisoni PG-14에서 발현된 단백질과 같은 분자량으로 발현되는 것을 확인하였으며, 모기유충에 대한 형질전환체의 독성은 1.40×10^6 cells/ml 농도에서 약 80%의 치사율을 나타내었다. Bacillus thuringiensis subsp. morrisoni PG-14 is a gram-positive soil bacterium producing mosquitocidal parasporal inclusions composed of several crystal proteins. Among these crystal protein genes, cryIVD gene is one of major component which has 72 kDa in size. However, these parasporal inclusions sink quickly from the surface of water where mosquito larval feeding occurred. To develope mosquitocidal cyanobacterium, therefore, we constructed the expression vector, pCYASK 5-1 harboring cryIVD gene. The expression vector, pCYASk5-1 was transformed into the cyanobacterium Synechocystis PCC6803 reported as a natural mosquito larval food source and the transformants were selected with kanamycin. Expression of cryIVD gene in transformant was characterized by SDS-polyacrylamide gel electrophoresis (PAGE) and immunoblot analysis. The mosquitocidal activity of a transformant was determined electrophoresis (PAGE) and immunoblot analysis. The mosquitocidal activity of a transformant was determined with Culex tritaeniorhynchus. The results showed that the transformed cyanobacterium is toxic to mosquito larvae and will be expected as a potential agent that is used for mosquito control.

        The diagnostic requirements were suggested and explained regarding the systems of differentiation of symptoms and signs in the third year study of standardization and unification of the terms and conditions used for diagnosis in oriental medicine. The systems were as follows : - analyzing and differentiating of epidemic febrile disease - analyzing and differentiating in accordance with the Sasang constitution medicine based on four-type recognition - differentiation of disease according to pathological changes of Chong and Ren channels - standards for diagnosis of women's disease - standards for diagnosis of children's disease - standards for diagnosis of motor and sensor disturbance (-muscle. born, joint, etc.) - standards for diagnosis of neuropsychiatric disease - standards for diagnosis of five sense organ disease - standards for diagnosis of external disease The indivisual diagnosis pattern was arranged by the diagnostic requirements in the following order : another name, notion of diagnosis pattern, index of differentiation of symptoms and signs, the main point of diagnosis, analysis of diagnosis pattern, discrimination of diagnosis pattern, prognosis, a way of curing a disease, prescription, herbs in common use, disease appearing the diagnosis pattern, documents. The standards for diagnosis of each disease was arranged by the diagnostic requirements in the following order : another name, notion of disease, the main point of diagnosis, analyzing and differentiating of disease, analysis of disease, discrimination of disease, prognosis, a way of curing and prescription of disease, disease in western medicine appearing the disease in oriental medicine, documents.

        In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria gonorrhoeae(PPNG), we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by menas of the chromogenic cephalosporin method. In 1998, 93 strians of N. genorrhoeae were isolated, among which 60(64.5%) were PPNG. The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 64.5% in 1998.

        Sexually Matured Female Mice showed Higher γ-Linolenic Acid (GLA) in the Muscle Tissue

        The sexual maturation occurred by the changes of steroid hormones was known to sex-dependently and/or age-dependently regulate the lipid metabolism in various animal species. Our current study demonstrates that lipid and its functional fatty acids can be changed depending on the status of sexual maturation. Of the functional fatty acids, ${\gamma}$-linolenic acid (GLA; 18:3n-6) is an important factor for maintaining human health. The purpose of our study was to investigate the level of GLA in mice with different stages of sexual maturation. To this end, the longissimus muscle (LM) of immature (3-week-old) and mature (7-week-old) female mice was analysed for the fatty acid composition by gas chromatography. Furthermore, both gene and protein level of ${\Delta}6$ desaturase (FADS2) which is involved in GLA metabolism by real time PCR and Western blotting, respectively. Mature females showed greater (P<0.05) serum $17{\beta}$-estradiol (E2) level and LM GLA contents than immature group. The mRNA and protein levels of FADS2, which converts precursor linoleic acid into GLA, were higher (P<0.05) in mature female mice than in immature mice. In conclusion, these results show that sexual maturation of female mice induces GLA and FADS2 contents in LM.

        Selective GSK-3β inhibitors attenuate the cisplatin-induced cytotoxicity of auditory cells

        <P><B>Abstract</B></P><P>Glycogen synthase kinase-3 (GSK-3) plays an important role in the regulation of apoptosis. However, the role of GSK-3 in the auditory system remains unknown. Here we examined whether the GSK-3-specific inhibitors, SB 216763 and LiCl, could protect against cisplatin-induced cytotoxicity of auditory cells. GSK-3 was activated by cisplatin treatment of HEI-OC1 cells. SB 216763 or LiCl treatments inhibited cisplatin-induced apoptosis in a dose-dependent manner and activated caspase-9, -8 and -3. In rat primary explants of the organ of Corti, SB 216763 or LiCl treatments completely abrogated the cisplatin-induced destruction of outer hair cell arrays. Administration of SB 216763 or LiCl inhibited cochlear destruction and the production of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and IL-6 in cisplatin-injected mice. Furthermore, administration of SB 216763 or LiCl reduced the thresholds of the auditory brainstem response (ABR) in cisplatin-injected mice. Collectively, these results suggest that cisplatin-induced ototoxicity might be associated with modulation of GSK-3 activation.</P>

        Activation-induced Cytidine Deaminase in B Cell Immunity and Cancers

        Activation-induced cytidine deaminase (AID) is an enzyme that is predominantly expressed in germinal center B cells and plays a pivotal role in immunoglobulin class switch recombination and somatic hypermutation for antibody (Ab) maturation. These two genetic processes endow Abs with protective functions against a multitude of antigens (pathogens) during humoral immune responses. In B cells, AID expression is regulated at the level of either transcriptional activation on AID gene loci or post-transcriptional suppression of AID mRNA. Furthermore, AID stabilization and targeting are determined by post-translational modifications and interactions with other cellular/nuclear factors. On the other hand, aberrant expression of AID causes B cell leukemias and lymphomas, including Burkitt's lymphoma caused by c-myc/IgH translocation. AID is also ectopically expressed in T cells and non-immune cells, and triggers point mutations in relevant DNA loci, resulting in tumorigenesis. Here, I review the recent literatures on the function of AID, regulation of AID expression, stability and targeting in B cells, and AID-related tumor formation.

        Gain of a New Exon by a Lineage-Specific Alu Element-Integration Event in the BCS1L Gene during Primate Evolution

        BCS1L gene encodes mitochondrial protein and is a member of conserved AAA protein family. This gene is involved in the incorporation of Rieske FeS and Qcr10p into complex III of respiratory chain. In our previous study, AluYRa2-derived alternative transcript in rhesus monkey genome was identified. However, this transcript has not been reported in human genome. In present study, we conducted evolutionary analysis of AluYRa2-exonized transcript with various primate genomic DNAs and cDNAs from humans, rhesus monkeys, and crabeating monkeys. Remarkably, our results show that AluYRa2 element has only been integrated into genomes of Macaca species. This Macaca lineage-specific integration of AluYRa2 element led to exonization event in the first intron region of BCS1L gene by producing a conserved 3' splice site. Intriguingly, in rhesus and crabeating monkeys, more diverse transcript variants by alternative splicing (AS) events, including exon skipping and different 5' splice sites from humans, were identified. Alignment of amino acid sequences revealed that AluYRa2-exonized transcript has short N-terminal peptides. Therefore, AS events play a major role in the generation of various transcripts and proteins during primate evolution. In particular, lineage-specific integration of Alu elements and species-specific Alu-derived exonization events could be important sources of gene diversification in primates.
