      • Chitosan microspheres as delivery vesicle for effective mucosal immunity to Brucella abortus malate dehydrogenase (MDH)

        Introduction: Brucellosis is a world widespread zoonotic disease that is transmitted from domestic animals to humans, effective and safe vaccine development have been required. In previous study, stimulation of recombinant proteins in THP-1 cells revealed malate dehydrogenase (MDH) as a high pro-inflammatory cytokine inducer. Also, it has been reported to be immunogenic and to be related to the bacterial pathogenesis. Brucellosis can be transmitted via aerosol exposure; hence the induction of protective immunity at respiratory mucosal surfaces is usually an expected attribute in the field of development of new vaccines. Therefore, this study produced delivery vesicle with immunostimulatory activities using chitosan, which is known to enhance mucosal absorption for effective B.abortus MDH delivery to the respiratory tract. Materials and Methods: After protein expression and purification, size and specificity was confirmed by SDS-PAGE and Western blotting with an anti-His antibody. Chitosan microspheres (CMs) were prepared based on the ionic gelation of chitosan with sodium tripolyphosphate anions. Prepared CMs were dispersed in 1ml of protein solution (4mg/ml, pH 7.4 PBS) and loading efficiency was measured by quantifying the unloaded protein in the supernatant in each time. Transmissible electron microscope (TEM) and dynamic light scattering spectrophotometer (DLS) was using to characterize morphology and size distribution. Results: SDS-PAGE profiles indicated purified TF and MDH was 52kDa and 85.71kDa sized, respectively. The recombinant proteins expressed with pCold TF vector were confirmed by western blot using anti-histidine antibody. The loading efficiency of TF and MDH into CMs was 61.23±3.58 and 50.97±0.82 %. TEM results showing that morphologies of CMs were spherical shapes. Average microsphere sizes of CMs, TF-loaded CMs, and MDH-loaded PCMs measured by DLS were 0.32±0.09, 1.87±0.46 and 0.66±0.16Om, respectively, with the increased particle sizes of CMs after the loading of TF and MDH. Release of TF and MDH from loaded CMs was released at 27% and 39% up to 80 hours respectively. Conclusions: Chitosan microspheres were produced and characterized as delivery carriers for effective mucosal immunity. Immune stimulatory activities will be analyzed in THP-1 cells and mice intranasally immunized with the CMs in further studies.         Induction of Th2-related immune responses and production of systemic IgA in mice intranasally immunized with <i>Brucella abortus</i> malate dehydrogenase loaded chitosan nanoparticles

        <P><B>Abstract</B></P> <P>The aim of this study was to investigate the induction of mucosal immune responses by an important <I>Brucella abortus</I> antigen, malate dehydrogenase (Mdh), loaded in mucoadhesive chitosan nanoparticles (CNs) and immunized intranasally in a BALB/c mouse model. The production of cytokines was investigated in human leukemic monocyte cells (THP-1 cells) after stimulation with the nanoparticles. Mdh-loaded CNs (CNs-Mdh) induced higher interleukin (IL)-6 production than unloaded antigens and TF loaded CNs (CNs-TF). Using ELISpot to quantify cytokines and antibody-secreting cells in the intranasally immunized mice, IL-4 and IgG-secreting cells were found to be significantly increased at 4 weeks and 6 weeks post-immunization in the CNs-Mdh immunized group, respectively. Increases in Mdh-specific IgG, IgG1, and IgG2a antibodies were confirmed at 6 weeks after immunization, indicating a predominant IgG1 response. Analysis of the mucosal immune response in the intranasally immunized mice revealed, Mdh-specific IgA and total IgA in the nasal washes, genital secretions, fecal extracts and sera that were remarkably increased in the CNs-Mdh-immunized group compared to the CNs-TF-immunized group except total IgA of nasal wash. Therefore, the results indicated that the intranasal immunization of CNs-loaded <I>B. abortus</I> Mdh antigen effectively induced antigen-specific mucosal immune responses through the elicitation of Th2-related immune responses.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • 망막혈관종성증식에서 유리체강내 라니비주맙과 애플리버셉트 주입술 후 맥락막두께 변화의 비교

        Purpose: To compare changes in choroidal thickness after intravitreal ranibizumab and aflibercept injections in patients with retinal angiomatous proliferation (RAP). Methods: This study involved 39 eyes (21 ranimizumab-treated eyes, 18 aflibercept-treated eyes) of 36 patients who received either intravitreal ranibizumab or aflibercept injection for the first time and followed up for more than 6 months. Subfoveal choroidal thickness was measured before intravitreal injection and at 3 and 6 months after intravitreal injection to determine any significant change in subfoveal chorioidal thickness over time in each treatment group. The relevant changes at each time point were measured to compare differences between the treatment groups. Central macular thickness and the best corrected visual acuity were measured and compared. Results: Choroidal thickness at 3 and 6 months after injection was significantly lower than that before injection in both the ranibizumab and aflibercept groups (p < 0.001, in all). The changes in choroidal thickness were 17.7 ± 10.9 μm and 13.0 ± 11.0 μm at 3 and 6 months after ranibizumab injection, respectively, and 24.5 ± 12.5 μm and 18.3 ± 11.1 μm at 3 and 6 months after aflibercept injection, respectively. These results show that although no significant differences were observed according to injection type at 3 months, the aflibercept group exhibited a significantly greater decrease at 6 months (p = 0.094, 0.043). The best corrected visual acuity in the two groups was significantly improved for up to 6 months after the injection compared with before the injection, and the central macular thickness also significantly decreased. There was no significant difference in visual improvement and central macular thickness decrease between the two groups. Conclusions: The results confirmed that both of these two drugs significantly decreased the subfoveal choroidal thickness in patients with RAP for up to 6 months after the injection, and the intravitreal injection of aflibercept tended to show a greater decrease in subfoveal choroidal thickness than the intravitreal injection of ranibizumab. 목적: 망막혈관종성증식 환자에서 유리체강내 라니비주맙과 애플리버셉트 주입술 후 중심하맥락막두께의 변화를 비교하고자 한다. 대상과 방법: 처음으로 망막혈관종성증식으로 진단된 후 라니비주맙 또는 애플리버셉트 주입술을 시행 받고 6개월 이상 경과관찰 가능했던 36명 39안(라니비주맙 21안, 애플리버셉트 18안)을 대상으로 하였다. 첫 주사 전, 주사 후 3개월, 6개월째 중심하맥락막두께를측정하여 약제군별로 시간에 따라 의미 있는 변화를 보이는지 확인하였고, 시기별 변화량을 측정하여 약제에 따른 차이를 비교하였다. 최대교정시력 및 중심망막두께도 함께 측정하여 비교하였다. 결과: 라니비주맙군과 애플리버셉트군 모두 주사 전 중심하맥락막두께와 비교하여 주사 후 3개월, 6개월째 통계적으로 유의한 감소를보였다(p<0.001, in all). 라니비주맙 주사 후 3개월, 6개월째 변화량은 17.7 ± 10.9 μm, 13.0 ± 11.0 μm, 애플리버셉트 주사 후변화량은 24.5 ± 12.5 μm, 18.3 ± 11.1 μm로 애플리버셉트군에서 보다 큰 변화량을 보였으며 3개월째는 통계학적으로 유의한 차이가 없었으나 6개월째는 유의한 차이를 보였다(p =0.094, 0.043). 두 군 모두에서 최대교정시력 및 중심망막두께는 주사 전과 비교하여 주사 후 6개월까지 유의하게 호전되었으며, 호전정도는 두 군 사이에 유의한 차이를 보이지 않았다. 결론: 망막혈관종성증식 환자의 치료에서 두 약제 모두 주사 후 6개월까지 유의하게 중심하맥락막두께를 감소시켰고, 애플리버셉트를주사하였을 때 라니비주맙에 비해 보다 큰 맥락막두께 감소를 보이는 경향을 확인하였다.

        Preparation of Partial Mesophase Pitch-based Carbon Fiber from FCC-DO

        Partial mesophase (PM) pitch precursor was prepared from fluidized catalytic cracking-decant oils (FCC-DO) by chemical reaction in the presence of Br2. The PM pitch heated-treatment at 420℃ for 9 h exhibited the softening point of 297℃ with 23% yield, and 55% anisotropic content. The PM pitch precursor was melt-spun through circular nozzle by pressurized N2, stabilized at 310℃, carbonized at 700℃, 1000℃, and 1200℃. The enough stabilization introduced 16.4% of the oxygen approximately. The stacking height (Lc002) and interlayer spacing (d002) of the as-spun fibers were 4.58 nm and 3.45a and the value became minimum and maximum at 700℃ respectively in the carbonization procedure. The tensile strength increased with an increase in the heat treatment temperature exhibiting highest value of 750 MPa at 1200℃ carbonization.

        Clinical Significance of Diffuse $^{18}F$-FDG Uptake in Residual Thyroid Gland after Unilateral Thyroid Lobectomy

        Purpose We investigated the clinical significance of diffuse uptake in remaining thyroid after unilateral lobectomy for thyroid cancer. Methods A total of 144 thyroid cancer patients who underwent $^{18}F$-FDG PET/CT after lobectomy were enrolled in the present study. The PET/CT images were evaluated for the presence of diffuse $^{18}F$-FDG uptake with maximum SUV (SUVmax) >2.0 in the residual thyroid and placed into one of two groups: with diffuse uptake and without diffuse uptake group. Clinical, laboratory, and PET/CT parameters in both groups were compared. Correlations between SUVmax of thyroid and available parameters were analyzed. Results Forty-two of 144 patients (29.2%) had diffuse thyroid uptake (mean SUVmax: $3.2{\pm}1.1$). All patients with diffuse uptake and 96 (94.1%) without diffuse uptake were receiving thyroxine therapy (P=0.09). Thyroid function tests showed that most patients were euthyroid status (78.6 vs. 85.3%, P=0.36). TgAb levels were significantly higher in patients with diffuse uptake ($338.0{\pm}664.6$ vs. $57.3{\pm}46.4$, P<0.0001). Mean attenuation values in the diffuse uptake group were significantly lower ($72.2{\pm}15$. vs. $97.0{\pm}16.0$, P<0.0001). An inverse correlation was found between SUVmax and mean attenuation values of residual thyroid in all patients (r=-0.57, P<0.0001) and subgroup with diffuse uptake (r=-0.31, P<0.05). Conclusion In this study, diffuse $^{18}F$-FDG uptake in the residual thyroid after unilateral lobectomy was a relatively frequent finding and may be associated with chronic thyroiditis. This uptake is not influenced by thyroid status or thyroxine therapy. The $^{18}F$-FDG uptake is inversely correlated with mean attenuation value of thyroid.
