      • Leptin Stimulates Endometriosis Development in Obesity

        Objective: In this study, we utilized obese mouse models to determine the effect of obesity on the development of endometriosis. Methods: Endometriosis is induced surgically in three distinct obesity mouse models : (1) high-fat diet-induced obesity; (2) obesity with leptin deficiency (ob/ob mice); and (3) leptin receptor deficiency (db/db mice). Ectopic endometriosis lesions are florescence-tagged, and the endometriosis lesions were visualized under stereo microscope and fluorescence. Fluorescence signaling from the mice as well as incubated small uterine fragments was detected by IVIS Spectrum. The signaling intensity was quantified using Living Image software, and the quantification of the number and weight of ectopic lesions were compared. Results: High-fat diet-induced obese mice revealed a significant increase in endometriosis development compared with regular-diet control mice. However, obese recipient mice with leptin deficiency and leptin receptor deficiency showed suppressed endometriosis development compared with control mice. Donor uterine tissues with leptin deficiency and leptin receptor deficiency suppressed endometriosis development compared with control donor in control recipient mice. We also demonstrated aberrant high levels of leptin concentration by treating mice with Intraperitoneal injections of leptin. This significantly increased endometriosis development compared with vehicle treatment group in control mice with normal body weight. Conclusion: The role of leptin and leptin signaling in the pathogenesis of endometriosis is demonstrated in an experimental mouse model. The disruption of leptin signaling hinders the induction of endometriosis, which implies leptin\'s potential role in the development and progression of endometriotic lesions and targets for possible therapeutic interventions.

      • Cesarean section and risk factors: Secular tends in South Korea, 2006-2015

        Objective: The aim of the study is to investigate the recent trend in the rate of cesarean section (CS) in South Korea from 2006 to 2015 and identify risk factors in association with these changes. Methods: Using the National Health Insurance Corporation data, we included every women who had delivery from 2006 to 2015. We investigated (1) mode of delivery, (2) the rate of complications during pregnancy (i.e. preeclampsia and placenta previa) and (3) pre-pregnancy factors (BMI, hypertension, DM and other pre-existing medical conditions) and its trends during study period. Results: During 10 years, the rate of CS increased from 36.3% in 2006 to 40.6% in 2015 (p <0.01). The rate of CS increased in primiparous women, women with multiple pregnancy and women with preeclampsia. Otherwise, the rate of vacuum delivery and VBAC decreased during study period. The rate of women with pre-pregnancy obesity increased but the rate of women with pre-pregnancy hypertension and DM decreased. Conclusion: The rate of CS in South Korea have been increasing from 2006 to 2015. This trend may reflect changes in the rate of risk factors. Identification of causes for increasing trend of CS observed in this study will allow the clinician to monitor for these factors and possibly reduce CS.

      • Incidence and 30-day mortality of peptic ulcer bleeding in Korea

        OBJECTIVES: To estimate the rates of peptic ulcer bleeding (PUB) incidence and the association of demographic factors with 30-day mortality after PUB. METHODS: Diagnostic algorithms for PUB were derived and validated on the basis of 115 true PUB patients at one tertiary hospital in 2005, followed by estimation of age-specific PUB incidence and 30-day mortality rates, using the Korean National Health Insurance claim database. A Cox proportional hazard model was used to determine the impact of demographic factors on the 30-day mortality rate ratio (MRR) after PUB. RESULTS: The diagnostic algorithm showed 89 and 88% positive predictive value and sensitivity, respectively. On the basis of this algorithm, the rate of PUB incidence was 22.1 per 100 000 during 2006–2007 and the age-specific incidence rate increased with advanced age. This incidence rate was more than three times higher among men than women. Among 21 107 PUB patients, the overall 30-day mortality rate was 2.15%, but it ranged from 0.83% for patients younger than 60 years to 7.65% for patients older than 80 years. The adjusted 30-day mortality rate ratio for patients older than 80 was 8.13 (95% confidence interval 6.10–10.8) compared with those younger than 60 and 7.09 (95% confidence interval 2.78–4.51) for patients with a high level of comorbidity compared with a low level of comorbidity. CONCLUSION: PUB incidence was higher among men and increased with advanced age. Increased 30-day mortality was observed in association with increasing age, after adjusting for comorbidity.

      • <i>ALU</i> and <i>LINE‐1</i> hypomethylations in multistep gastric carcinogenesis and their prognostic implications

        <P><B>Abstract</B></P><P>Focal CpG island hypermethylation and diffuse genomic hypomethylation signify the changes in the DNA methylation status in cancer cells. <I>ALU</I> and <I>LINE‐1</I> repetitive DNA elements comprise ∼28% of the human genome. PCR‐based measurements of these repetitive DNA elements can be used as a surrogate marker of the genomewide methylation content. Our study aimed to identify the timing of <I>ALU</I> and <I>LINE‐1</I> hypomethylations during multistep gastric carcinogenesis and their prognostic implications in gastric cancer (GC). In our study, we analyzed the methylation statuses of <I>ALU</I> and <I>LINE‐1</I> in 249 cases of gastric biopsy samples and another independent set of 198 cases of advanced GC by pyrosequencing. Regardless of the <I>Helicobacter pylori</I> infection status, a significant decrease in the <I>ALU</I> methylation levels was noted during the transitions from chronic gastritis to intestinal metaplasia and from gastric adenoma to GC. <I>LINE‐1</I> methylation decreased during the transition from intestinal metaplasia to gastric adenoma and no further decrease occurred during the transition from gastric adenoma to GC. A low <I>LINE‐1</I> methylation status was strongly associated with poor prognosis in GC. A multivariate analysis revealed that <I>LINE‐1</I> methylation status was an independent prognostic factor. Our findings suggest that <I>ALU</I> and <I>LINE‐1</I> hypomethylations are early events during multistep gastric carcinogenesis. Furthermore, the <I>LINE‐1</I> methylation status can be used as a molecular biomarker to define a subset of GC patients with poor prognosis.</P>

        Synthesis of Nanoporous Adsorbents Using Alum Sludge.

        <P>This study optimizes the synthesis process of pellet-type adsorbents using alum sludge. The effect of the binder and heat treatment temperature on the nanopore formation in the adsorbent is investigated. The pellet-type adsorbent prepared using the powder-type sludge from water treatment is determined to be a material that contains nanopores. The specific surface area is increased significantly after the calcination process in the range of 132-172 m2/g. With the calcination treatment, the breakthrough time in the formaldehyde adsorption increases remarkably with an optimum calcination temperature of 400 °C. The breakthrough capacity of the formaldehyde increases to a maximum 2.96 mg/g at this temperature.</P>

        알럼 슬러지를 이용한 입상흡착제 압출 및 황화수소 제거 성능

        본 연구의 목적은 알럼 슬러지를 사용하여 입상흡착제를 제조하기 위한 압출 공정을 최적화하는 것이다. 함수율과 바인더인 메틸 셀룰로스의 함량이 압출 가능성과 입상흡착제의 물리적 특성에 미치는 영향을 연구하였다. 입상흡착제의 물리적 특성은 질소 흡착실험과 압축 강도 측정을 통해서 분석하였다. 증류수와 알럼 슬러지 가공분말의 비가 63/100인 경우가 입상흡착제의 외형이 원통형으로 잘 성형되었고, 압축강도도 가장 높게 나타났다. 메틸 셀룰로스의 함량을 증가시키면 압축강도가 개선되었으나 비표면적이 감소하였다. 성형된 입상흡착제의 소성과정을 거치면 입상흡착제의 표면적이 크게 증가하여 황화수소의 파과 시간이 획기적으로 증가하였으며, 황화수소 파과 용량은 1,700 mg/g 이상을 얻을 수 있었다. The objective of this study is optimization of extrusion process for preparation of pellet-type adsorbents employed with alum sludge. Effects of water content and methyl cellulose as a binder on the possibility of extrusion and physical properties of pellet-type adsorbents were investigated. The physical characteristics of the pellet-type adsorbents were studied using nitrogen adsorption and compression strength. With a ratio of water to sludge, 63/100, the adsorbent was well extruded with a cylindrical form and the compressive strength was the highest. With increasing methyl cellulose content, the compressive strength of pellet-type adsorbent could be improved, but the specific surface area decreased. The breakthrough time of the hydrogen sulfide could be increased significantly through calcination and the breakthrough capacity reached to 1,700 mg/g, which seems to be due to increase of surface area during calcination.

        Guillain–Barré Syndrome After COVID-19 Infection in Korea: A Case Series

        Guillain–Barré syndrome (GBS) is an autoimmune-driven condition characterized by acute polyneuropathy, often emerging as a sequel to prior infections or vaccinations. This study presents the first reported cases of GBS emerging after the full recovery from coronavirus disease 2019 (COVID-19) infection in Korea. Despite experiencing mild acute COVID-19 symptoms, these patients faced substantial weakness attributed to GBS, significantly affecting their daily lives. The timely administration of intravenous immunoglobulin treatment halted the progression of symptoms, underscoring the critical importance of early intervention. These cases highlight the potential for neurological complications associated with COVID-19 and underscore the necessity for continuous monitoring and timely medical care.

        Characteristics of pellet-type adsorbents prepared from water treatment sludge and their effect on trimethylamine removal

        We optimized the preparation method of pellet-type adsorbents based on alum sludge with the aim of developinga high-performance material for the adsorption of gaseous trimethylamine. Effects of calcination temperatureon physical and chemical properties of pellet-type adsorbents were investigated. The porous structure and surfacecharacteristics of the adsorbents were studied using N2 adsorption and desorption isotherms, scanning electron microscope,X-ray diffraction, temperature-programmed desorption of ammonia, and infrared spectroscopy of adsorbed pyridine. The adsorbents obtained from the water treatment sludge are microporous materials with well-developed mesoporosity. The pellet-type adsorbent calcined at 500 oC had the highest percentage of micropore volume and the smallestaverage pore diameter. The highest adsorption capacity in trimethylamine removal attained over the pellet-type adsorbentcalcined at 500 oC can be attributed to the highest number of acid sites as well as the well-developed microporosity.

        Time-Lapse Live-Cell Imaging Reveals Dual Function of Oseg4, Drosophila WDR35, in Ciliary Protein Trafficking

        Cilia are highly specialized antennae-like organelles that extend from the cell surface and act as cell signaling hubs. Intraflagellar transport (IFT) is a specialized form of intracellular protein trafficking that is required for the assembly and maintenance of cilia. Because cilia are so important, mutations in several IFT components lead to human disease. Thus, clarifying the molecular functions of the IFT proteins is a high priority in cilia biology. Live imaging in various species and cellular preparations has proven to be an important technique in both the discovery of IFT and the mechanisms by which it functions. Live imaging of Drosophila cilia, however, has not yet been reported. Here, we have visualized the movement of IFT in Drosophila cilia using time-lapse live imaging for the first time. We found that NOMPB-GFP (IFT88) moves according to distinct parameters depending on the ciliary segment. NOMPB-GFP moves at a similar speed in proximal and distal cilia toward the tip (${\sim}0.45{\mu}m/s$). As it returns to the ciliary base, however, NOMPB-GFP moves at ${\sim}0.12{\mu}m/s$ in distal cilia, accelerating to ${\sim}0.70{\mu}m/s$ in proximal cilia. Furthermore, while live imaging NOMPB-GFP, we observed one of the IFT proteins required for retrograde movement, Oseg4 (WDR35), is also required for anterograde movement in distal cilia. We anticipate our time-lapse live imaging analysis technique in Drosophila cilia will be a good starting point for a more sophisticated analysis of IFT and its molecular mechanisms.
