        흰쥐 태아 구개융합부위 상피세포의 아포프토시스에 관한 연구

        악안면 부위에서 가장 빈번히 발생하는 선천적 기형의 하나인 구개열은 그 원인요소가 명확히 밝혀져 있지는 않지만 구개발생 과정 중 어느 한 단계에서의 이상으로 발생하며 양측 구개판의 융합 후 융합상피의 제거가 이루어지지 않는 것도 그 원인의 하나로 알려져 있다. 본 연구는 구개발생시 구개판 융합상피의 제거가 아포프토시스에 의한 것임을 확인하고 아포프토시스가 일어나는 시기와 위치를 규명하기 위해 시행되었다. 체중 200gm내외의 암컷흰쥐 12마리를 체중 200gm내외의 수컷흰쥐와 교배시켜 16.0일, 16.5일, 16.75일, 17.0일 태아를 얻었으며 각 태아에서 두부만을 파라핀 포매하여 전두절단 하였다. 조직절편에서 형태학적 소견을 관찰하기 위한 H-E염색과 아포프토시스를 인지하기 위한 TdT염색을 시행한 후 광학현미경을 통해 관찰하여 다음과 같은 결론을 얻었다. 1.16.0일과 16.5일 된 태아의 표본에서 접촉전의 양측 구개판의 내측단 상피에서는 아포프토시스를 관찰할 수 없었으며, 구개판의 접촉 직후 상피세포간의 융합이 이루어지는 부위에서는 극히 소수의 아포프토시스 세포들만이 관찰되었다. 2.16.75일 된 태아의 표본에서 연속성을 거의 잃지 않은 정중 상피융합에서는 아포프토시스를 거의 관찰할 수 없었으나, 비강측과 구강측의 상피삼각 그리고 비중격과 구개판이 접촉하는 부위, 특히 비중격의 외측연과 접촉하는 부위에서는 다수의 아포프토시스 세포들이 관찰되었다. 3.16.75일 된 태아의 표본에서 정중 상피융합이 연속성을 잃고 상피섬들로 나뉘어지는 시기에서는 상피섬과 상피삼각 부위에서 증가된 아포프토시스 세포들을 관찰할 수 있었으며 근접한 구강, 비강상피에서도 다수의 아포프토시스 세포들이 관찰되었다. 4.17.0일 된 태아의 표본에서 상피섬이 많이 소실되어 간엽조직에 의한 융합이 이루어지고 있는 부위에서는 아포프토시스 세포들이 거의 관찰되지 않았으나 상피삼각부위와 구강, 비강상피에서는 아직도 다수의 아포프토시스 세포들이 관찰되었다. 이상의 결과로 흰쥐 태아의 구개발생시 구개융합부위의 상피세포가 아포프토시스에 의해 제거된다는 사실을 알 수 있었으며 이러한 아포프토시스에 의한 융합상피세포의 제거는 주로 구개융합의 후반기에 상피섬과 상피삼각 부위에서 일어난다는 사실을 알 수 있었다. The purpose of this study was to prove that the medial edge epithelial cells covering the secondary palatal shelves were removed by apoptosis during palatal fusion. 12 mature female rats (Suprague-Dawley) were mated overnight with male rats and sacrificed on days 16.0, 16.5, 16.75, 17.0 of pregnancy. The embryos were removed from the uterus and the heads were embedded in paraffin. The paraffin blocks were sectioned and the sections were undergone H-E staining for general histologic feature and TdT staining for detection of apoptotic cells. The obtained results were as follows. 1.In the section of 16.0 and 16.5 day embryos, the palatal shelves were prior to contact and no apoptotic cells wereobserved in the medial edge epithelium. At the initial contact of palatal shelves, there was a few apoptotic cells in the fusing epithelium. 2.In the 16.75 day embryos, the samples that epithelial seams did not lost there continuity, apoptotic cells were rarely seen at the midline epithelial seam. In contrast, a lot of apoptotic cells were observed at epithelial triangles and the junction between palatal shelves and nasal septum. 3.In the 16,75 day embryos, the samples that epithelial seams lost their continuity and disrupted to epithelial islands, large number, of apoptotic cells were observed at epithelial islands and epithelial triangles. Some apoptotic cells were also observed at the oral, nasal epithelium near the midline. 4.In the 17.0 day embryos, most of epithelial islands were disappeared and mesenchymal confluence was achieved. Apoptotic cells were rarely observed in the mesenchymal tissue which replaced epithelial islands, but there were some apoptotic cells at the epithelial triangles, oral and nasal epithelium. From the results of the study, it was revealed that medial edge epithelial cells of fusing palate were removed by apoptosis. Apoptotic cells were found mainly in the disappearing midline epithelial seam and the oral and nasal epithelial triangles at some late stages of palatal fusion.

        피로와 의료이용, 업무상 사고 및 질병결근

        Objectives: This study examined the relationship of fatigue to medical utilization, occupational accident and sickness absence. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project, 2002~2004). A total of 30,146 employees categorized within the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were recruited. A structured-questionnaire was used to assess the participants' sociodemo-graphics, job-related factors, health-related behaviors, personality trait, self=perceived fatigue (MFS) and the frequency of medical utilization, occupational accident and sickness absence. Data on medical utilization and occupational accident were confirmed from company health records. Chi-square test and logistic regression analysis were used to elucidate the relationship of self-perceived fatigue to medical utilization, occupational accident and sickness absence. Results: In logistic regression analyses, self-perceived fatigue was associated with medical utilization (outpatient), occupational accident and sickness absence although the strength of the associations was higher in the blue collar workers except for medical utilization. For medical utilization, the white collar worker's adjusted odds ratios of outpatient utilization for workers scoring in the third quartile and the highest quartile, compared to workers scoring in the lowest quartile were 1.50 (95% CI: 1.20-1.87) and 2.16 (95% CI: 1.69-2.75), respectively. The blue collar workers' adjusted odds ratios of outpatient utilization for workers scoring in the second (OR: 1.23, 95% CI: 1.06-1.43), third (OR: 1.42, 95% CI: 1.22-1.66) and highest quartile (OR:1.83, 95% CI: 1.55-2.17), respectively, compared to workers scoring in the lowest quartile were statistically significant. However, no associations were found between fatigue and inpatient utilization for either the white or blue collar workers. The blue collar workers' adjusted odds ratios of being injured for workers scoring in the second, third and highest quartile were 1.41 (95% CI: 1.00-2.00), 1.57 (95% CI: 1.10-2.22) and 2.41 (95% CI: 1.69-3.44), respectively, compared to those scoring in the lowest quartile, but, those of the white collar workers were not significant. There was more likely to an increased risk of sickness absence in workers scoring in the second, third and highest quartile compared to those scoring in the lowest quartile in both white and blue collar workers, but the magnitudes of risk were higher in the blue collar workers than the white collars workers. Conclusion: This result suggests that fatigue is a determinant predictor of medical utilization, occupational accident and sickness absence, and that the pattern of risks for the three outcomes differs according to the occupational type. Some limitations of this study and considerations for future study were also discussed.

        도재표면의 처리방법과 접착제의 중합방식에 따른 교정용 브라켓의 전단강도의 연구

        최근에는 교육수준의 향상, 소득증대, 대중매체의 발달 등으로 인하여 성인 교정환자의 수요가 상당히 증가하고 있다. 성인환자는 어린환자에 비해 상대적으로 구강내 도재수복물을 장착하고 있는 경우가 많아 도재치에 대한 교정용브라켓의 직접접착의 기회도 증가하고 있다. 자연치에서는 법랑질을 산부식하여 비교적 쉽게 접착력을 증진시킬 수 있으나 도재치에서는 임상에서 브라켓이 너무 쉽게 탈락하거나 오히려 도재파절의 위험성이 존재하여 도재치에 대한 효과적인 직접접착법에 대하여 관심을 갖게 되었다. 그러나, 이러한 접착효율에 대한 다양한 연구가 있었으나 아직 정설로 받아들여지는 것은 없는 실정이다. 본 연구는 임상에서 비교적 간단히 이용할 수 있는 몇 가지 방법을 in vitro상의 실험을 통하여 그 결과를 비교검토하여 효과적인 방법을 제시하고자 시행되었다. 우선 장석형 도재인 Ceramco II로 도재 시편을 제작하였다. 도재의 표면처리방법은 Glazing한 도재 자체를 사용하기 위해서는 무처리하였으며, 거칠기를 부여하기위해서는 불화수소산으로 산부식하거나 Microetcher II로 Sandblasting,또는 불화수소산과 Sandblasting을 복합처리하였다. 접착레진은 화학중합형인 Ortho-two와 광중합형인 Transbond를 사용하였다. 모든 시편에는 도재 프라이머를 도포하였다. 실험군은 7가지 군으로 나누어 브라켓의 전단강도를 측정하였다. 1. 도재의 표면처리에 대한 비교에서 Ortho-two군내에서는 불화수소산이나 andblasting으로 표면거칠기를 부여한군이 무처리군에 비해 높은 전단강도를 갖는다. Transbond군내에서는 각각의 표면처리에 따른 유의성있는 차이가 없었다. 2. 불화수소산이나 Sandblasting처리군에서는 화학중합형 레진인 Ortho-two가 광중합형레진인 Transbond보다 높은 전단강도를 갖는다. 3. 도재 파절율의 비교에서는 무칠군이 표면거칠기를 부여한 군에 비해 매우 낮은 파절율 갖는다. 4. E(Transbond + Intact) 군의 평균전단강도는 134.4kg/㎠ 로 본 실험군중 가장 낮은 것으로 나타났다. 이것은 선학들이 제안한 임상적용가능 전단강도보다 크므로 7가지군 모두 임상적으로 교정력을 가하기에 충분한 전단강도를 갖는다. 그러나, 도재의 파절율을 고려해볼 때 파절율이 상대적으로 낮은 무처리군이 임상적으로 더 효과적일 것으로 사료된다. As a result of increased education and communication, the field of orthodontics has recently been expanded to include a greater number of adult treatment procedures. With this increased demand for adult orthodontic treatment, a problem that frequently arises is the placement of appliances on teeth restored with porcelain. But conventional acid-etching is ineffective in the preparation of porcelain surface for mechanical retention of orthodontic attachments. Also, it is possible to damage on porcelain. The purpose of this study was to evaluate the effect of composite bonding materials and the porcelain surface treatment methods on shear bond strength, and to observe the porcelain fracture rates. To accomplish this purpose, this study was carried out with feldsphatic porcelain, Ceram II. Porcelain surface treatment methods were divided into intact glazed porcelain which had not treatment and surface roughening. Surface roughening by etching with Hydroluoric acid(HF), sandblasting with Microetcher II and compound treatment with etching and sandblasting. Bonding materials were Ortho-two and Transbond. All porcelain specimens were applicated with porcelain primer. 1. In comparision according to porcelain surface treatment, surface roughening groups by HF etching and sandblasting had higher shear bond than intact group. No significant difference was found in Transbond group. 2. Ortho-two group had the higher shear bond strength than that of Transbond group in HF etching and sandblasting. 3. E(Transbond+Intact)group had the lowest shear bond strength in all experimental group. The bond strength was higher than clinically successful bond strength. 4. Non-treated group had very higher porcelain rates than treated group. 5. This study indicates that porcelain surface-roughening may not be necessary to attachment of orthodontic brackets to porcelain surfaces.

      • 이동형 생체계측을 위한 임베디드 컴퓨팅

        연구목적: 초소형 정밀 기계 기술로 제작된 2차원 가속도 센서와 916 MH2 단거리 고주파 데이터 통신 기술을 응용하여 인체의 운동상태를 자동으로 감지하고 분류하는 기법을 제안 하고자 하였다. 재료 및 방법: 인체의 신호를 획득하기 위하여 미국 버클리대에서 제작한 무선 센서를 등 산용 조끼의 전면상단에 부착해서, 대상자의 흉골 부위에 위치하도록 하고, 앉은 상태에서 일어서거나 눕거나 하는 동작 및 그 역동작이 일어날 때, 가속도 센서에서 감지되는 4가지 종류의 파형 현화에 해당되는 데이터를 무선센서 네트웍을 통해 획득하였다. 결과: 획득한 신호를 주성분분석(Principal Component Analysis)과 지원벡터머신(Support Vector Machine)을 사용하여, 2차주성분까지를 도표화하고 클러스터링 함으로써, 4개의 영역으로 분할하는 것이 가능함을 말 수 있었다 결론: 소형 센서들이 작동되는 임베디드 컴퓨팅 환경에서 일상생활 중에 발생하는 인체의 4가지 동작을 자동으로 감지하고 분류하는 것이 가능함을 보였다. Purpose: New monitoring and classifying method of human motion contort was pro- posed by using 2-axial MEMS accelerometer and 916 MHz short-range data communica-tion technology. Materials and Methods: The wireless sensor of MICA mote was attached to a woman mountain vest and located close to the subject's sternum. During four types of sub-ject's motion, that is, sit-to-stand, stand-to-sit, sit-to-tying, and tying-to-sit, waveform changes of the accelerometer data were acquired by wireless sensor network. Results: The acquired signal was analyzed by Principal Component Analysis(PCA) and Support Vector Machine(SVM) technique. By clustering the first and second principal components, four regions corresponding to the four types of motion, were clearly segment-ed. Conclusion: Human motion context during daily life could be monitored and classified automatically by using small sensors in embedded computing environment

      • 1990년대 fashion에 나타난 여성수트의 형식적 특성에 관한 연구

        Five pairs of coordinate concept for classifying the mode by H. Wolfflin who combines the aesthetics with the history of art are adopted to understand the characteristics of the modem suit which have been appeared with the various aspects due to the effects of the post- modernism since the latter half of the 20th century and as a result of applying them to the women's suit shown in A/W collection from 1990 to 1996, the modern suit could be divided into two; Quasi-Renaissance from and quasi-Baroque one. While the suit of Renaissance propensity showed the typical characteristics of 'suit' that the components of the jacket and skirt - collar, lapel, pocket, texture, hemlines etc.- emphasize the symmetry of human body itself, open and non-constructional concept of the Baroque propensity expressed the constructional elements of human body to the incidental thing. The rising designers avoided the existing idea through the effect of natural pleates of row cloth and the use of surface decoration and gave the design a change. Such a suit form has the meaning in that it gives the sense of flexible movement to the structure of the fixed suit and it pursues the aesthetic concept of a new design by escaping from the typical suit form. Kal Lagerfeld after Chanel, Gianfranco Ferre after Christian Dior and Claude Montana after Lanvin follow their masters in the concept of painiting and the trend of ambiguity and non-construction are found in the works of Jean-Paul Gaultier, Christian Lacroix, Chantal Thomas and Gianni Versace who are the experimentalist designers and the concept of openness and non-construction are in those of Issey Miyake, Vivienne Westwood, Oscar de la Lenta and Pacco Rabanne. It is found the design of the women's suit shows the quasi-Baroque propensity based on the traditional suit design depending on the characteristics of the rising designers' group succeeding to the existing designers' brands. And it can be confirm through this study that formalism of Wolfflin based on the thought of formal aesthetics is the useful standard for analyzing the forms of the modern suit.

        백서 척수후근절로부터 슈반세포의 효과적인 체외 배양법

        Schwann cells(SCs), an important component of the peripheral nervous system, intract with nerous to mutually support growth and replication for the peripheral nerve regentation. Recently, ading SCs to the lumen of guidance channel is widely tried to improve regeneration or to make regeneration possible over otherwise irreparable gaps. however, it is not easy to isolate and multiplicate SCs as much as enough to help the axonal regeneration. For the allogeneic SCs source for tubular nerve guidance, we developed a little bit improved technique of harvesting and multiplicating SCs. by culturing dispersed dorsal root ganglia in specially designed medium with growth factors and serial processing, we repeatedlly generate relatively homogenous SC cultures. Our technique was compared with other methods of literature using immunostaining methods such as GFAP, S100, BDNF and the total SC count assessment at different time interval after primary culture.

      • APX-115, a first-in-class pan-NADPH oxidase (Nox) inhibitor, protects db/db mice from renal injury

        <P>Recent studies have suggested that renal Nox is important in the progression of diabetic nephropathy. Therefore, we investigated the effect of a novel pan-NOX-inhibitor, APX-115, on diabetic nephropathy in type 2 diabetic mice. Eight-week-old db/m and db/db mice were treated with APX-115 for 12 weeks. APX-115 was administered by oral gavage at a dose of 60 mg/kg per day. To compare the effects of APX-115 with a dual Nox1/Nox4 inhibitor, db/db mice were treated with GKT137831 according to the same protocol. APX-115 significantly improved insulin resistance in diabetic mice, similar to GKT137831. Oxidative stress as measured by plasma 8-isoprostane level was decreased in the APX-115 group compared with diabetic controls. All lipid profiles, both in plasma and tissues improved with Nox inhibition:APX-115 treatment decreased Nox1, Nox2, and Nox4 protein expression in the kidney. APX-115 decreased urinary albumin excretion and preserved creatinine level. In diabetic kidneys, APX-115 significantly improved mesangial expansion, but GKT137831 did not. In addition, F4/80 infiltration in the adipose tissue and kidney decreased with APX-115 treatment. We also found that TGF-beta stimulated ROS generation in primary mouse mesangial cells (pMMCs) from wild-type, Nox1 KO, and Duox1 KO mice, but did not induce Nox activity in pMMCs from Nox2 knockout (KO), Nox4 KO, or Duox2 KO mice. These results indicate that activating Nox2, Nox4, or Duox2 in pMMCs is essential for TGF-beta-mediated ROS generation. Our findings suggest that APX-115 may be as effective or may provide better protection than the dual Nox1/Nox4 inhibitor, and pan-Nox inhibition with APX-115 might be a promising therapy for diabetic nephropathy.</P>

        Plasma leptin concentrations are greater in type II diabetic patients and stimulate monocyte chemotactic peptide-1 synthesis via the mitogen-activated protein kinase/extracellular signal-regulated kinase pathway

        Background: Leptin is an adipokine that is recently reported to be a biomarker of systemic inflammation. Although atherosclerosis causes cardiovascular diseases, it is not clear whether leptin contributes to the acceleration of this process. In this study, we investigated whether alterations of plasma leptin levels were related to diabetic nephropathy and systemic inflammation. In addition, we examined the physiologic action of leptin in cultured vascular smooth muscle cells (VSMCs). Methods: A total of 126 type 2 diabetic participants and 37 healthy controls were studied. The diabetic participants were divided into three groups according to stage of nephropathy. We investigated whether leptin induced monocyte chemo- tactic peptide-1 (MCP-1) synthesis through the mitogen-activated protein kinase (MAPK) pathway using cultured VSMCs. Results: Plasma leptin concentrations were significantly higher in the diabetic group than in the controls. Plasma leptin levels were positively correlated with body mass index, fasting and postprandial blood glucose, hemoglobin A1c, total cholesterol, urinary albumin excretion, high-sensitivity C-reactive protein (hsCRP), and MCP-1 plasma levels, and negatively correlated with creatinine clearance values. In cultured VSMCs, leptin increased MCP-1 production in a dose-dependent manner, and this stimulating effect of leptin on MCP-1 expression was reversed by the MAPK (MEK) inhibitor PD98059. In addition, leptin stimulated the phosphorylation of MEK, extracellular signal-regulated kinase, and E26-like transcription factor, which are components of the MAPK pathway. Conclusions: Overall, these findings suggest that activation of leptin synthesis may promote MCP-1 activation in a diabetic environment via the MAPK pathway in VSMCs and that it possibly contributes to the acceleration of atherosclerosis.

        Background: Anemia is commonly observed in elderly population, and the prevalence of anemia increases with advanced age. Anemia is a sign of poor health state and increase adverse outcomes in the elderly. Therefore, assessment of anemia is important in elderly patients. Objectives: The aim of this study was to classify the type of anemia and to assess the current state of anemia evaluation in the elderly population. Methods: Medical records-based retrospective study was performed for patients who were more than 80 years (>80) old and visited Dongguk University Ilsan Hospital from April 2005 to February 2014. Results: Total 548 patients who had anemia according to WHO criteria were identifi ed. The median age was 85 years old (range, 82-99) and median hemoglobin level was 11g/dL (range, 2. 7-12. 9). Twenty eight, 468, and 52 patients were classifi ed as microcytic anemia, nomocytic anemia, and macrocytic anemia, respectively. Among them, 397 patients (72. 4%) did not underwent proper the evaluation for the cause anemia. In detail: 8 (28. 5%) of microcytic anemia, 361 (77. 1%) of normocytic anemia, and the 28 (53. 84%) of 52 macrocytic anemia patients. The remaining 151 patients (27. 6%) had completed the evaluation, and 24 patients (15. 9%) were diagnosed as solid or hematologic malignancies. Conclusions: This fi nding showed physicians often neglected anemia in elderly over age 80. Though the patients have limited life expectancy, physicians need to discriminate the sub-population who will be benefi t from adequate evaluation and treatment.
