        Effects of Aralia cordata Thunb. on Proteoglycan Release, Type II Collagen Degradation and Matrix Metalloproteinase Activity in Rabbit Articular Cartilage Explants

        Background & Objective: Articular cartilage is a potential target for drugs designed to inhibit the activity of matrix metalloproteinases (MMPs) to stop or slow the destruction of the proteoglycan and collagen in the cartilage extracellular matrix. The purpose of this study was to investigate the effects of Aralia cordata Thunb. in inhibiting the release of glycosaminoglycan (GAG), the degradation of collagen, and MMP activity in rabbit articular cartilage explants. Methods : The cartilage-protective effects of Aralia cordata Thunb. were evaluated by using glycosaminoglycan degradation assay, collagen degradation assay, colorimetric analysis of MMP activity, measurement of lactate dehydrogenase activity and histological analysis in rabbit cartilage explants culture. Results : Interleukin-la (IL-1a) rapidly induced GAG, but collagen was much less readily released from cartilage explants. Aralia cordata Thunb. significantly inhibited GAG and collagen release in a concentration-dependent manner. Aralia cordata Thunb. dose-dependently inhibited MMP-3 and MMP-13 expression and activities from IL-1a-treated cartilage explants cultures when tested at concentrations ranging from 0.02 to 0.2 mg/ml. Aralia cordata Thunb. had no harmful effect on chondrocytes viability or cartilage morphology in cartilage explants. Histological analysis indicated that Aralia cordata Thunb. reduced the degradation of the cartilage matrix compared with that of IL -1a-treated cartilage explants.

        멸균 방법에 따른 교정용 플라이어의 물성 변화에 대한 비교 연구

        간염이나 AIDS의 영향으로 멸균에 대한 관심이 높아지고 있으며, 교정 진료실에서도 교정용 플라이어의 멸균이 필요하게 되었다. 제조회사에서는 멸균과정의 열이 교정용 플라이어에서의 야금학적 변화를 야기하지 않는다고 하지만 교정치과의사들은 가압 증기 멸균기나 대류식 건열 멸균기를 이용하여 반복적으로 멸균을 시행한 경우에 ligatrue cutter의 날이 쉽게 무디어져서 ligature cutter의 수명이 짧아진다는 것을 임상적 경험으로 알고 있다. 본 연구는 멸균과정이 교정용 플라이어의 물성에 미치는 영향을 알아보기 위하여 시행되었다. 본 연구에 사용된 교정용 플라이어는 멸균과정을 견딜 수 있도록 개발했다고 주장하는 AEZ, Unitek과 Dentronix ligature cutter이었고, 멸균기는 대류식 건열 멸균기인 Bowmar RHT-1000과 Dentronix DDS-5000와 가압 증기 멸균기인 Eschmann SES-2000이었다. 멸균하기 전과 200회와 400회 멸균한 후의 ligature cutter의 날 부위의 표면과 파단면의 상태는 주사 전자 현미경으로, 미세 조직은 광학 현미경으로 관찰하고, micro-Vickers와 Rockwell 경도 시험기로 경도를 측정하고, SEM-EDX로 조성분석을 시행하여 다음과 같은 결론을 얻었다. 1. 주사 전자 현미경을 이용한 교정용 플라이어의 날 부위의 표면과 파단면의 관찰에 의하면 멸균 횟수의 증가에 따라서 표면 조직에서 검은 반점 형태의 부식 생성물의 수와 크기가 증가하는 경향이 있었으며, 파단면의 관찰에서도 멸균 횟수의 증가에 따라서 전형적인 취성 파면인 벽개면의 분포율이 증가하는 경향이 있었다. 이러한 부식 현상과 금속 조직의 취성화로 기계적인 특성이 저하될 것으로 사료되었다. 2. 광학 현미경을 이용한 미세 조직 사진의 관찰에서도 멸균횟수의 증가에 따라서 Cr계 탄화물의 수와 크기가 증가하는 경향이 있었다. 이러한 금속 조직의 조대화에 따라서 기계적 특성이 저하된 것으로 생각되었다. 3. 교정용 플라이어의 날 부위의 경도는 멸균 횟수의 증가에 따라서 감소되었는데 경도의 감소는 멸균에 의한 부식생성물과 표면조직의 변화에 따른 것으로 생각되었다. 4. SEM-EDX를 이용한 조성분석의 결과에 의하면 AEZ과 Unitek ligature cutter는 Fe-Cr계 stainless steel이었고, Dentronix ligature cutter는 Co-Cr계 합금이었는데, 제조회사에 따라서 금속의 조성은 달랐지만, 멸균 전후의 비교에서는 큰 차이가 없었다. 5. 반복적으로 멸균을 하면서 실제 임상에서 사용한 교정용 플라이어의 날의 표면 조직을 주사 전자현미경으로 관찰한 결과, 미세 균열을 발견할 수 있었다. Sterilization has received much attention in orthodontic practices over the past several years. The present study was undertaken to investigate the effects of sterilization on the physical properties of orthodontic pliers-AEZ, after 200 and 400 sterilization cycles using the Bowmar RHT-1000, the Dentronix DDS-5000, and the Eschmann SES-2000. The tip surface and the fracture and the fracture surface were observed with a scanning electron microscope. The microstructure was observed with an optical microscope. The hardness test was carried out with the micro-Vickers hardness tester and the Rockwell C Scale hardness tester. The chemical composition was analyzed by energy dispersive X-ray spectrometer. The results of this study were as follows: 1. The number and the size of corrosion products on the tip surface and the proportion of cleavage planes in fractured specimen increased, but the hardness of the tip decreased in proportion to sterilization cycles. From these observations, it was considered that mechanical properities decreased in proportion to sterilization cycles. 2. The number and the size of chromium carbides increased in proportion to sterilization cycles. Coarse microstructrue decreased mechanical properities. 3. The AEZ and Unitek ligature cutters were Fe-Cr stainless steel, but the Dentronix ligature cutter was Co-Cr alloy. There were differences among manufactures, but the chemical composition was not changed after sterilization cycles. 4. The tip edge of ligature cutter used in a clinic revealed microcracks with the SEM observation. Clinical experience confirmed that ligature cutters were gradually degraded by sterilization.

        In postmodern era, information technology modifies the concept of human beings and their relationships with machines. Most of postmodern theorists refuse to take humanism for granted from their philosophical viewpoints. This is the very ground of considering the subjectivites of posthuman. We try to think over the status of posthuman reading the fictions of William Gibson, Thomas Pynchon, and Kathy Acker. William Gibson is mapping the ways that new technologies are impacting on human life creating posthumanistic individuals and environments through the imagined future. Thomas Pynchon, who tried to show the fear of technological civilization in his early works, considers the possibility of oppositional subjectivity in virtual space in Gravity's Rainbow and Vineland. And Kathy Acker introduces the possible female subjectivities in the age when technology encroaches even human body itself. Searching these kind of possibility they imaging a collective or individual agency who secedes the modern binary oppositional system. Donna Haraway says that she wants to be a cyborg rather than a goddess. Her concept of cyborg can be related with the postmodern collective subjectivities in Pynchon's Vineland and the kind of self to be newly encoded in Acker's The Empire of the Senseless. This is not to say that cyborg imagery suggests some utopian landscape of the coming future, but it involved with the dream of heteroglossia. But the term, "posthuman" is not completely defined yet. Not all of the writers use this term to mean the same thing. As Hayles recognizes, the face and the future of posthuman is not clear. The problem, however, is that we are moving into the shadow of unknown. Accordingly, posthuman is less a term suggesting utopian or distopian view in itself than an open space which has the possibility of both.

      • 악성 혈액질환에서 성공적인 동종골수이식 후 숙주 기질 미세환경의 구축

        인체의 골수는 간엽모세포를 함유하고 있으며 이들은 골수미세환경의 주된 세포들로 분화가 가능하여 조혈기능을 지지한다. 본 연구에서는 성별이 다른 동종조혈모세포이식 환경에서 조혈모세포의 완전 생착을 보이고, 이식 후 1년에서 8년이 지난 11예의 재생불량성 빈혈 및 백혈병 환자들을 대상으로 하여 골수에서 MSC를 분리하고 체외 확장을 통해 배양된 MSC에서 X 염색체 탐식자를 이용한 FISH 및 microsatellite polymorphism PCR 기법으로 그 기원을 확인하였다. 그 결과 조혈모세포는 완전히 공여자 기원으로 대치되었음에도 불구하고 MSC는 모두 수여자 기원임을 알 수 있어, 동종조혈모세포이식에서 미세환경의 구축은 수여자의 자가 생산에 의한 골수 간질세포에 의한 것으로 생각된다. Background: Human bone marrow (BM) contains mesenchymal stem cells (MSC) that can differentiate into various cells of mesenchymal origin. It remains a matter of controversy whether donor-derived stromal cells are capable of engraftment following hematopoietic stem cell transplantation (HSCT) or not. Methods: To determine if donor-derived stromal cells are transferred to the recipients of allogeneic HSCT, we investigated the characterization of MSC in 11 patients 1 to 8 years after sex mis-matched allogeneic HSCT in severe aplastic anemia and leukemia. Results: All patients had complete engraftment with donor- derived stem cells as shown by detection of donor type DNA in peripheral blood mononuclear cells. Following culture, MSC showed the expression of SH2 and SH4, but none of the hematopoietic markers of CD14, CD34, or CD45. MSC which can be differentiated to osteogenic lineage showed the genotype of recipient completely using FISH or PCR analysis. Conclusion: This study confirmed that MSC isolated from recipients of allogeneic HSCT in severe aplastic anemia and leukemia are not of donor genotype despite of full hematopoietic engraftment with donor type. Donor cells did not contribute to reconstitute the marrow microenvironment.

        Cardiac tumors, especially the primary tumors involving any part of the heart are extremely rare and its relative incidence has been reported to be approximately 0.02%. We report a patient with huge cardiac hemangioma who complained of shortness of breath, general weakness, and dizziness, Imaging study by echocardiography and computed tomography of thorax revealed a huge lobulating mass like a bunch of grapes in the right chamber of heart. The surgical exploration of thorax was performed and a histological diagnosis of spindle cell hemangioma was obtained by microscopy. The patient was treated by surgical resection of the tumor and doing well after surgery. Our experience indicated that prompt diagnosis and treatment of cardiac hemangioma is imperative for patients' prognosis.

        Geotextiles have been extensively used in road constructions as effective and economical materials performing several functions such as separation, filtration, drainage, protection against frost break, and as reinforcement to improve the load bearing capacity and stability of road. In this study, a method for the analysis of embankment reinforced with geotextile is studied. Computer program based on the simplified Bishop method is modified in order to investigate quantitively the effect of gotextile on the slope stability of embankment on weak soils. Evaluation of the tensile strength required for the design safety factor is also possible utilizing the computer program in this study.

      • 비선형 캐리어 제어 부스트 정류기 특성에 관한 연구

        Average-current mode controlled boost rectifier is most universally used at present, While the method gives high power factor, low harmonics distortion, has drawbacks such as 3-feedback loop, complex component, difficulty of control-circuit design, and high cost. Research for improving these problem has been carried out, and Nonlinear Carrier(NLC) Control method is representative. This control method has easy algorithm for control, and its feedback loop and control components are reduced. In this paper, the control method was evaluated by simulation through Matlab/Simulink, and experimented with 550W boost rectifier. As the result, that can get high power factor above 0.95 and low harmonic distortion was confirmed. Also, a good dynamic response on line and load variation was confirmed.
