Composition of the Extracellular Matrix of Lymphatic Novel Threadlike Structures: Is It Keratin?

<P><I>Background</I>. The lumen of novel threadlike structures (NTSs) is enclosed by a single layer of endothelial cells surrounded by extracellular matrix (ECM). We hypothesized that collagen may be a component of the ECM associated with lymphatic NTSs. <I>Methods</I>. Six female New Zealand white rabbits were anesthetized, and the NTS structures within lymphatic vessels were identified by contrast-enhanced stereomicroscopy or alcian blue staining. Isolated NTS specimens were stained with acridine orange, YOYO-1, and 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate (DiI). The structural and molecular composition of the ECM was investigated using transmission electron microscopy (TEM), electrospray ionization-mass spectrometry, and proteomic analysis. <I>Results</I>. The lymph vessel wall was stained red by DiI, and rod-shaped nuclei were stained green by YOYO-1. The area surrounding the NTS was also stained red and contained green rod-shaped nuclei. TEM images showed that the NTS consisted of many ECM fibers and the ECM fibers appeared to be ~100 nm in diameter and had narrowly spaced striated bands. Proteomic analysis of the lymphatic NTS-associated ECM identified 4 proteins: keratin 10, cytokeratin 3, cytokeratin 12, and soluble adenylyl cyclase. <I>Conclusion</I>. The TEM study suggested that the lymphatic NTS-associated ECM did not contain collagen. This was confirmed by proteomic analysis, which showed that keratin was the major component of the ECM. </P>

Anesthetic management of penetrating neck injury patient with embedded knife -A case report-

Penetrating neck injuries can be a fatal event and they are difficult to manage for both surgeons and anesthesiologists. So, adequate preoperative evaluation is important to improve the patients’ outcomes, but this can not be done for hemodynamically unstable or uncooperative patient. Here we present our clinical experience with a patient with a penetrating neck injury and who was hemodynamically stable, but she was uncooperative and the knife was still embedded in her neck. The surgical exploration and bronchoscopic examination were successfully done under monitored anesthesia care.
Therapeutic Effect of Rebamipide on Ammonia-induced Gastric Mucosal Hemorrhagic Lesion in Rats

Rebamipide, 2-(4-chlorobenzoylamino)-3-[2(1H)-quinolinone-4-yl]-propionic acid, a novel antipeptic ulcer agent, has been reported to prevent various acute experimental gastric mucosal lesions and to accelerate the healing of chronic ulcers. Therapeutic effect of rebamipide was investigated with regard to the inhibitory effect on xanthine oxidase activity and type conversion of the enzyme which play a profound role in oxygen radicals generation system. Intraperitoneal administration of rebamipide at 60 mg/kg body weight reduced the xanthine oxidase activity, lipid peroxide content in ammonia induced hemorrhagic lesion. These results suggest that the therapeutic effect of rebamipide on gastric mucosal lesion may be in part due to the inhibitory activity of xanthine oxidase and type conversion rate of the enzyme.

Gastrodia elata (GE) is a oriental medicinal herb which has been used traditionally for the treatment of various brain diseases including convulsion and epilepsy. In order to examine the mechanism of anticonvulsive effect, we treated the methanol extract of GE (500 mg/kg, P.O) to the pentylenetetrazole (PTZ)-induced convulsive rats. Methanol extracts of GE siginificantly inhibited (35%) the convulsion state as well as the level of lipid peroxidation (25%) in the brain. The ether fraction of methanol extracts among the others effectively inhibited in vitro lipid peroxidation dose dependently (5.0×10^(-6)~2.0×10^(-5) g/ml). The scavenging effect on hydroxy radicals was found in all the fractions of ether, butanol, and dichloromethane. These results suggest that the anticonvulsive effect of GE is possibly due to the antioxidative effects of the active components in GE.
Effect of Thyroid Hormones in Oxidative Stress in Rat Liver

The effect of thyroid hormones on the hepatic xanthine oxidase activity was studied in rats after the intraperitoneal injections of comthyroid (triiodotyronine:thyroxine=1:4) at 0.3 mg/kg for 3 consecutive days. The aim of this study was to understand the precise mechanism of hyperthyroidism induced by oxidative stress. The concentration of lipid peroxides determined indirectly by the measurement of thiobarbituric acid reactants was increased in comthyroid treated rats. The hepatic glutathione content was decreased in comthyroid injected rat compared to the euthyroid state. It was also observed that the increment of xanthine oxidase activity has a profound role in oxygen radicals generation system in comthyroid treated rat. These findings suggest that the enhanced xanthine oxidase activity and depleting glutathione content in comthyroid treated rats result in pathophysiological oxidative stress including an increment of hepatic lipid peroxidation.

<P>Fatigue crack propagation behavior under cyclic tensile or torsional loading with biaxial static loads has been investigated. Two different biaxial loading systems, i.e. cyclic tensile loading with static torsional load and cyclic torsional loading with static tensile load, were employed to thin-walled tubular specimens. The crack propagation was measured by two crack gages mounted near the notch and crack opening level was measured by unloading compliance method. The directions of the fatigue crack propagated under respective biaxial loading conditions were examined and the growth rates were evaluated by using several cyclic parameters, including equivalent stress intensity factor range, Keff, crack tip opening displacement range, CTD, minimum strain energy density factor range, Smin. Furthermore, the growth rates were evaluated by effective cyclic parameters considering crack closure. It was found that the biaxial static stress superimposed on the cyclic tensile or torsional loading tests has no influence on the propagation directions of the cracks. Furthermore, it was shown that the fatigue crack growth rates under biaixial faigue loading were well expressed by using the cyclic fatigue parameters, Keq,eff, CTDeff, Smin,eff considering crack closure effect.</P>
