      • Down-regulated MYH11 Expression Correlates with Poor Prognosis in Stage II and III Colorectal Cancer

        The MYH11 gene may be related to cell migration and adhesion, intracellular transport, and signal transduction. However, its relationship with prognosis is still uncertain. The aim of this study was to investigate correlations between MYH11 gene expression and prognosis in 58 patients with stage II and III colorectal cancer. Quantitative real-time polymerase chain reaction was performed in fresh CRC tissues to examine mRNA expression, and immunohistochemistry was performed with paraffin-embedded specimens for protein expression. On univariate analysis, MYH11 expression at both mRNA and protein levels, perineural invasion and lymphovascular invasion were related to disease-free survival (p<0.05; log-rank test). Cancers with lower MYH11 expression were more likely to have a poor prognosis. Otherwise, MYH11 expression was unrelated to patient clinicopathological features. On multivariate analysis, low MYH11 expression proved to be an independent adverse prognosticator (p<0.05). These findings show that MYH11 can contribute to predicting prognosis in stage II and III colorectal cancers.

      • Seeing the World: Photoreaction and Photosensory Mechanisms of Cordyceps militaris

        Light is a necessary environmental factor for stroma formation and development of Cordyceps militaris, a well-known edible and medicinal fungus. In this study, photo morphogenesis and the blue-light receptor gene were studied using five representative strains of C. militaris. The results suggest that light was essential for colony pigmentation and could promote conidia production. Cmwc-1 gene inactivation results in thicker aerial hyphae, disordered fruit body development, a significant reduction in conidial formation, and carotenoid and cordycepin production. These characteristics were restored when the ΔCmwc-1 strains were hybridized with wild-type strains of the opposite mating type. A genomewide expression analysis revealed that there were 1042 light responsive genes in the wild-type strain and only 458 in the ΔCmwc-1 strain. Among five putative photoreceptors identified, Vivid, cryptochrome-1, and cyclobutane pyrimidine dimer photolyase are strongly induced by light in a Cmwc-1- dependent manner, while phytochrome and cryptochrome-2 were not induced. The transcription factors involved in the fungal light reaction were mainly of the Zn2Cys6 type. CmWC-1 regulates adenylosuccinate synthase, an important enzyme for adenosine de novo synthesis, which could explain the reduction in cordycepin production. Some G proteincoupled receptors that control fungal fruit body formation and the sexual cycle were regulated by CmWC-1, and the cAMP pathway involved in light signal transduction in N. crassa was not critical for the photoreaction in the fungus here. A transcriptional analysis indicated that steroid biosynthesis was more active in the ΔCmwc-1 strain, suggesting that CmWC-1 might switch the vegetative growth state to primordia differentiation by suppressing the expression of related genes.

        Determination of Chromium(VI) and Copper(II) in Organic Solvent - Solution by Liquid Core Optical Fiber Spectrophotometry

        A new hollow fiber filled with the lower refractive index liquid as core constructs a liquid core optical fiber(LCOF). The LCOF have been used as colorimetric cell to determine elements Cr and Cu in the presence of 70%- 50% ethanol or 50% dioxane aqueous solution, based on colored complex of Chromium(VI) with Diphenycarbazid(DPC) and copper with Chromaurd S(CAS) respectively. The sensitivity (1/ng/ml) of calibration curve of Cr and Cu are 0.052, 0.017 over the range 0 - 25, 0 - 24(ng/ml) respectively. The Cr in the animal gum and Nation Standard of China are determined with recoveries of 94 - 102%.

        Privacy-Preserving Key-Updatable Public Key Encryption with Keyword Search Supporting Ciphertext Sharing Function

        Public key encryption with keyword search (PEKS) allows a user to make search on ciphertexts without disclosing the information of encrypted messages and keywords. In practice, cryptographic operations often occur on insecure devices or mobile devices. But, these devices face the risk of being lost or stolen. Therefore, the secret keys stored on these devices are likely to be exposed. To handle the key exposure problem in PEKS, the notion of key-updatable PEKS (KU-PEKS) was proposed recently. In KU-PEKS, the users’ keys can be updated as the system runs. Nevertheless, the existing KU-PEKS framework has some weaknesses. Firstly, it can’t update the keyword ciphertexts on the storage server without leaking keyword information. Secondly, it needs to send the search tokens to the storage server by secure channels. Thirdly, it does not consider the search token security. In this work, a new PEKS framework named key-updatable and ciphertext-sharable PEKS (KU-CS-PEKS) is devised. This novel framework effectively overcomes the weaknesses in KU-PEKS and has the ciphertext sharing function which is not supported by KU-PEKS. The security notions for KU-CS-PEKS are formally defined and then a concrete KU-CS-PEKS scheme is proposed. The security proofs demonstrate that the KU-CS-PEKS scheme guarantees both the keyword ciphertext privacy and the search token privacy. The experimental results and comparisons bear out that the proposed scheme is practicable.

        Mating triggers immediate upregulation of the heat shock response but downregulation of immune activity in the female reproductive tract of moths

        Mating induces female changes, and how females respond to male manipulation through ejaculates, especially at the molecular level, is currently a key topic of sexual conflict and coevolution in insects. Here, we show that mating induced significant transcriptional changes both in the reproductive tract (RT) and head of Spodoptera litura females, with all those genes that are directly related to fecundity, such as yolk protein and vitellogenin, being upregulated after mating, suggesting that mating promoted egg production. The most interesting changes are the upregulation of the heat shock response and the downregulation of immunity soon after mating in the RT. An increased heat shock response after mating may contribute to the formation of a favorable microenvironment for the interaction between gametes. The downregulation of immune activities may be due to males suppressing female immunity directly to promote sperm storage and fertilization in females. Another interesting finding is that mating results in the downregulation of five circadian regulation genes (including two clock core genes) in the RT. Moreover, mating also induced significant changes in other gene families, such as neuropeptides and cytochrome p450, suggesting that some of them may play important functions in mating responses and post mating physiological and behavioral switches. In addition, a prominent change in the head was found in which many olfactory-related genes were upregulated after mating. S. litura females usually start to lay eggs at night soon after mating, and thus, an increased olfactory function after mating is beneficial for host location by females during the night.

        The logistics service integrated network is the primary factor in the logistics integrated service. This paper analyzed both the connotation and structure of the logistics service integrated network. It elaborated on the governance logic of the logistics service integrated network in terms of relationship, interaction and collaboration. Since the governance model of a logistics service integrated network is dominated by contractual governance, this paper discussed its macro-governance mechanisms (e.g. trust mechanism, reputation mechanism, culture mechanism and joint sanctions mechanism) and micro-governance mechanism (e.g. entry barriers mechanism, incentive and restrictive mechanism, coordination mechanism and information feedback mechanism) so as to promote a coordinated effect on the logistics service integrated network.

        Functional Characterization of the Genes tauO, tauK, and tauI in the Biosynthesis of Tautomycetin

        Tautomycetin is a specific protein phosphatase I inhibitor. In an effort to elucidate the biosynthetic mechanism of tautomycetin,we inactivated genes of the tautomycetin biosynthetic gene cluster, tauI, tauO, and tauK, which encode for putative P450 oxidase, citryl-CoA lyase, and esterase enzymes,respectively. The mutant STQ0606 (ΔtauO) did not produce any detectable amount of tautomycetin intermediates but could convert dialkylmaleic anhydride to tautomycetin,strongly indicating that TauO was involved in dialkylmaleic anhydride biosynthesis. STQ1211 (ΔtauK) accumulated dialkylmaleic anhydride, whereas the cofermentation of STQ1211(ΔtauK) and STQ0606 (ΔtauO) restored the production of tautomycetin. Together, these results suggest that TauK was responsible for the conjugation of dialkylmaleic anhydride and the polyketide moiety in tautomycetin biosynthesis. The disruption of tauI resulted in the accumulation of 5-des-ketotautomycetin,revealing that TauI was responsible for the oxidation at C5 as the last step. Although the shunt pathways were involved in the biosynthesis of tautomycetin, the main post-polyketide synthase tailoring steps were dehydration,decarboxylation and oxidation, taking place consecutively. This study allowed us to predict the biosynthesis of tautomycetin more accurately and provided novel insights into the mechanism of the biosynthesis of tautomycetin.

        Molecular Characterization and Expression Patterns of Porcine Eukaryotic Elongation Factor 1 A

        The eukaryotic elongation factor 1 A (EEF1A) participates in protein synthesis by forming the eEF1A GTP tRNA complex to deliver aminoacyl-tRNA to the A site of ribosomes. This study described cDNA sequences and partial genomic structure of porcine EEF1A1. The porcine EEF1A1 gene encoded a protein with 462 amino acids, which shared complete homology with human, chimpanzee and dog. The temporal expression pattern showed the diversity of EEF1A1 level in mRNA was relatively minor in prenatal embryo skeletal muscle, however, the expression decreased during aging after birth in skeletal muscle of the Chinese Tongcheng pig. The spatial expression patterns indicated that the gene expressed in skeletal muscle, heart, lung, liver, kidney, fat and spleen. In addition, we assigned the gene to porcine chromosome 1 using a radiation hybrid panel.

        Experimental study and numerical simulation on springback of Ti-6Al-4V alloy under hot U-bending

        In this study, springback of Ti-6Al-4V sheet subjected to hot bending was investigated experimentally and by numerical simulation. The experiment determined the relative effects of forming temperature, forming speed and holding time on springback during hot bending of the alloy sheet. Distribution of temperature and equivalent stress fields experienced during hot bending were predicted by thermal-mechanical coupling numerical simulation. Analysis of the mutual influence of deformation and temperature was conducted using finite element modeling in DYNAFORM software. The results indicated that increased forming temperature, forming speed and holding time each reduced springback to a degree. Forming temperature had the greatest influence in limiting springback, followed by forming speed and holding time, the latter of which was of minor significance. The thermomechanical coupling numerical model is reliable and the predicted temperature field distribution was confirmed.
