      • A Case Study Exploring TPACK Framework within the Context of Early Childhood Education

        The purpose of this exploratory single-case study was to investigate the affordances of iPads and how these affordances transpired within a technological, pedagogical content knowledge (TPACK) framework by four early childhood educators at a low-income family preschool in Honolulu, Hawaii. Early childhood education (ECE) will be defined as education from birth to age eight although this study focused primarily on educators for preschool age group that is from age three to five. The targeted participants included four preschool teachers with varying technological skills and knowledge. Methods of data collection were pre-survey, post-survey, follow-up survey, group interviews, follow-up interviews, classroom observations, and documentation of iPad workshops and other email correspondences. The data included existing data from a grant project completed at the end of 2013 and new data from follow-up interviews collected in May 2014. The data were analyzed using coding methods in two cycles. The three major findings were 1) the exploration of affordances provided by iPads showed common themes of efficiency and effectiveness in assessing, teaching, and learning. In addition, there were some affordances particular to teaching and learning; 2) the exploration in how participating teachers discovered the affordances of iPad over time indicated parallel progression in technological knowledge and change in their value system about the affordances of iPad; and 3) the exploration in the progression of technological knowledge and change in their value system about the affordances of iPad within the TPACK framework suggested that there was a close relationship between progression of technological knowledge (TK) towards TPACK and progression of affective-valuing (AV) towards affective-characterization by value system (AC).

        Pancreatic Islets Induce CD4+CD25−Foxp3+ T-Cell Regulated Tolerance to HY-Mismatched Skin Grafts

        BACKGROUND.: The ability to induce and maintain antigen-specific immunologic tolerance is the ultimate goal in allo-transplantation. Here, we report that the transplantation of unmanipulated murine pancreatic islets across the HY disparity induced transplantation tolerance that prevented HY-mismatched skin grafts being rejected. METHODS.: Three hundred islet equivalent of freshly isolated islets from male C57BL/6 donor mice was transplanted underneath the kidney capsule of syngeneic female recipients rendered diabetic by streptozotocin. Nephrectomy was carried out to remove the islet graft and retransplantation was performed using the contralateral kidney. For skin transplantation, donor tail skin was transplanted onto the lateral thorax. RESULTS.: Islets from male C57BL/6 donors transplanted to syngeneic female recipients cured diabetes and the mice survived indefinitely. The acceptance of second grafts and rejection of third party islet grafts indicated antigen-specific transplantation tolerance. However, flow cytometry and ELISPOT analysis demonstrated that the HY-specific T cells were not deleted or anergized. A 2-fold increase of CD4Foxp3 regulatory T cells (Tregs) was observed in spleen and lymph nodes. Notably, CD25 Tregs increased threefold over levels in naïve mice. Adoptive transfer of CD4 T cells to neonatal mice could transfer tolerance. At the graft site in long-term tolerant mice, CD4 T cells, 40% of which were CD4Foxp3 Tregs (43% CD25, 57% CD25) infiltrated the peri-islet spaces. CONCLUSIONS.: Unmanipulated pancreatic islets can induce immunologic tolerance associated with peripherally induced CD4Foxp3 Tregs, a significant proportion of which notably are CD25.

        Ion-Mediated Electron Transfer in a Supramolecular Donor-Acceptor Ensemble

        <P>Ion binding often mediates electron transfer in biological systems as a cofactor strategy, either as a promoter or as an inhibitor. However, it has rarely, if ever, been exploited for that purpose in synthetic host-guest assemblies. We report here that strong binding of specific anions (chloride, bromide, and methylsulfate but not tetrafluoroborate or hexafluorophosphate) to a tetrathiafulvalene calix[4]pyrrole (TTF-C4P) donor enforces a host conformation that favors electron transfer to a bisimidazolium quinone (BIQ<SUP>2+</SUP>) guest acceptor. In contrast, the addition of a tetraethylammonium cation, which binds more effectively than the BIQ<SUP>2+</SUP> guest in the TTF-C4P cavity, leads to back electron transfer, restoring the initial oxidation states of the donor and acceptor pair. The products of these processes were characterized via spectroscopy and x-ray crystallography.</P>

      • The effects of a 12-week jump rope exercise program on abdominal adiposity, vasoactive substances, inflammation, and vascular function in prehypertensive adolescent girls

        Intro/purpose: Childhood obesity is strongly associated with the pathophysiology of various diseases, including cardiovascular disease (CVD). It is necessary to combat the unfavorable outcomes of obesity at a young age by utilizing effective interventions, such as exercise programs. Therefore, we sought to examine the effects of a jump rope exercise program on CVD risk factors, including body composition, vasoactive substances, inflammation, and vascular function in prehypertensive adolescent girls. Methods: Forty girls (age 14-16) were recruited and randomly assigned to either a jump rope exercise group (EX, n = 20) or control group (CON, n = 20). The EX group exercised 5 days/week. Body composition, nitrate and nitrite levels, endothelin-1 (ET-1), c-reactive protein (CRP), blood pressure, and arterial stiffness were measured before and after 12 weeks. Results: There were significant group by time interactions following the 12-week jump rope exercise program. Body composition, central adiposity, blood pressure, and brachial-to-ankle pulse wave velocity, an indicator of arterial stiffness (baPWV), all significantly decreased (P < 0.05) after 12 weeks. Nitrate and nitrite levels increased significantly (P < 0.05) along with a significant reduction in CRP levels (P < 0.05). There were no significant changes in ET-1 (P = 0.22). Conclusion: These findings indicate that jump rope exercise may be an effective non-pharmacological intervention to improve body composition, vasoactive substances, inflammation, and vascular function in prehypertensive adolescent girls. Jumping rope is an easily accessible and enjoyable exercise modality that may have important health implications for the prevention of CVD and other metabolic diseases at an early age.

      • The Effect of Shopping Orientation on Cosmetic Attribute Evaluation, Purchase Motivation, and Re-purchase Intention

        The Purpose of the current study is to investigate the differences in cosmetics attribute evaluation, purchase motivation, and brand re-purchase intention with respect to shopping orientations of Korean female university students. Questionnaires were distributed to 250 female students at Kyungpook National University and 220 completed questionnaires were analyzed. The results were as follows. First, shopping orientations and cosmetics attribute evaluation each generated four factor solutions, whereas cosmetics purchase motivations produced three factors. Second, three consumer groups - Efficiency Shopper Group, Indifference Shopper Group Ambivalence Shopper Group - with different shopping orientations were identified. Third, the study found significant differences in consumers' attribute evaluation such as function and price among the groups. The study also revealed a significant difference in contingent purchase motivations and brand re-purchase intention among the groups. From these results, we could identify that cosmetics re-purchase intention was significantly different among three different groups with different shopping orientations. Furthermore, consumer classification according to shopping orientations in cosmetics product purchase can be used by cosmetics marketers and managers to establish product plan and marketing strategy development. Additionally, the current study has originality and value that the relationship between shopping orientation and re-purchase intention has not been studied very much in the cosmetics product domain.

        Is Nonstandard Employment Hazardous to Workers' Health Status? A Focus on Special Employment in South Korea

        Purpose: Workers in special employment relationship (WSERs) are workers in nonstandard employment arrangements who lack worker protection accorded in standard employment arrangements. This study aimed to describe self-rated health (SRH) and depressive symptoms (DS) among Korean WSERs in comparison to regular wage workers (RWW) and identify associations between working conditions and those outcomes. Methods: In this study, secondary data analysis using the 5th Korean Working Conditions Survey was used. The sample totaled 29,120, including 1,538 WSERs and 27,564 RWWs. Sociodemographic and work-related characteristics were employed as explanatory variables and SRH and DS as dependent variables. Using multiple logistic regression, the determinants of fair/poor SRH and DS were identified. Results: The prevalence rates for fair/poor SRH and DS in WSERs were 25.2% and 28.3%, respectively, and 20.7% and 25.0% in RWWs, respectively. Compared to RWWs, WSERs had 31% (aOR=1.31, 95% CI=1.14~1.49) and 20% (aOR=1.20, 95% CI=1.06~1.36) higher odds of SRH and DS, respectively. Some factors, such as a lack of rest guarantee and sickness presenteeism, had a larger influence in the WSER than in the RWW group. Conclusion: Compared to RWWs, WSERs reported having poorer working conditions and were more likely to report poor general and mental health. Therefore, in Korea, public health policymakers should consider measures to protect the working conditions and health of WSERs, a growing segment of the working population. The study produced new epidemiological evidence regarding the relationships between employment arrangements and health.

        Stc1: A Critical Link between RNAi and Chromatin Modification Required for Heterochromatin Integrity

        <P><B>Summary</B></P><P>In fission yeast, RNAi directs heterochromatin formation at centromeres, telomeres, and the mating type locus. Noncoding RNAs transcribed from repeat elements generate siRNAs that are incorporated into the Argonaute-containing RITS complex and direct it to nascent homologous transcripts. This leads to recruitment of the CLRC complex, including the histone methyltransferase Clr4, promoting H3K9 methylation and heterochromatin formation. A key question is what mediates the recruitment of Clr4/CLRC to transcript-bound RITS. We have identified a LIM domain protein, Stc1, that is required for centromeric heterochromatin integrity. Our analyses show that Stc1 is specifically required to establish H3K9 methylation via RNAi, and interacts both with the RNAi effector Ago1, and with the chromatin-modifying CLRC complex. Moreover, tethering Stc1 to a euchromatic locus is sufficient to induce silencing and heterochromatin formation independently of RNAi. We conclude that Stc1 associates with RITS on centromeric transcripts and recruits CLRC, thereby coupling RNAi to chromatin modification.</P>

        <P><B>Summary</B></P><P>FLK1-expressing (FLK1<SUP>+</SUP>) mesoderm generates blood and vessels. Here, we show that combined BMP, Notch, and Wnt signaling is necessary for efficient FLK1<SUP>+</SUP> mesoderm formation from embryonic stem cells (ESCs). Inhibition of BMP, Notch, and Wnt signaling pathways greatly decreased the generation of FLK1<SUP>+</SUP> mesoderm and expression of the Ets transcription factor <I>Er71</I>. Enforced expression of ER71 in ESCs resulted in a robust induction of FLK1<SUP>+</SUP> mesoderm; rescued the generation of FLK1<SUP>+</SUP> mesoderm when blocked by BMP, Notch, and Wnt inhibition; and enhanced hematopoietic and endothelial cell generation. <I>Er71</I>-deficient mice had greatly reduced FLK1 expression, died early in gestation, and displayed severe blood and vessel defects that are highly reminiscent of the <I>Flk1</I> null mouse phenotype. Collectively, we provide compelling evidence that ER71 functions downstream of BMP, Notch, and Wnt signals and regulates FLK1<SUP>+</SUP> mesoderm, blood, and vessel development.</P>
