Crude saponin(100g) were obtained by extracting the radix(5kg) of Scrophalariae koraiensis Nakai. Crude saponin were positive in the Liebermann-Burchard test. We observed 5 sports by TLC using BuOH saturated with H_2O : ethyl acetate : water(4:1:5, upper phase) as solvent and 1% Ce(SO_4)_2 in 10% H_2SO_4 as a color former. We divided into 3 fractions by common column chromatography using BuOH saturated with H_2O : ethylacetate : water(4:1:5, upper phase) as solvent.
An event of gm peppers tolerant to pepper mottle virus (PepMoV)

In Korea, CMV (cucumber mosaic virus) is the most frequently occurring virus with a single infection rate of 45%. However, a total occurrence of CMV by co-infection, either couple or multiple, with BBWV (broad bean wilt virus), PepMoV (pepper mottle virus) and PMMoV (pepper mild mottle virus) covers over 90% in the field cultivation of pepper. The PepMoV is transmitted by several aphid species, and it has been considered the most frequently detected potyvirus when it co-infects with CMV or PMMoV. Since F1 hybrid that resistant to PepMoV has not been developed, we have developed transgenic peppers using Agrobacterium-mediated transformation with a Hc-Pro gene of the PepMoV. A large number of T1 peppers were tested for resistance to the PepMoV, and T1 peppers tolerant of PepMoV were selected. After consequent self-crossing up to T4 generation, highly tolerant peppers to PepMoV were selected. So far, BC3F1 lines have been selected by back-crossing with 4 elite lines through a breeding program. The horticultural differences of the GM line comparing to inbred lines were investigated and no statistical significance between GM and non-GM lines was found. Based on molecular analysis, One of GM lines, 10-2, contained the transgene in the non-coding region indicating that this line would be a GM event.
Anti-inflammatory and anti-diabetic effect of hot-water extract of Gryllus bimaculatus (HW-GB)

In this study, the inhibitory effect of hot-water extract of GB (HW-GB) on inflammation of LPS-stimulated Raw 264.7 macrophages and alloxan-induced diabetes was investigated. In the anti-inflammatory activity, treatment with HW-GB significantly inhibited the production of inflammatory mediators such as nitric oxide (NO), tumor necrosis factor (TNF-α) and IL-6 from LPS-stimulated Raw 264.7 macrophages in a dose-dependent manner. Treatment of HW-GB also showed the expression of mRNAs of iNOS synthase, TNF-α and IL-6 in RT-PCR analysis. Furthermore, this extract of GB was shown to inhibit LPS-induced inflammation in RAW 264.7 macrophages via down-regulation of activation of MAPKs such as ERK, JNK and p38. For type 1 diabetes experiment, HW-GB was administered orally for 5 days before alloxan injection. Oral administration of HW-GB significantly decreases the increase of blood glucose level in alloxan-induced diabetic mice. Additionally, HW-GB-fed mice reduced the increase of food intake and water consumption observed in alloxan-induced diabetic mice. Results of an experiment in which the expression of genes associated with metabolic activity by qPCR method revealed that oral administration of HW-GB prominently increased GLUT2 and GS gene expression in the liver, and enhanced GLUT 4, SIRT4, SIRT6, CPT1α, and GK gene expression in the muscle. Collectively, these results suggest that HW-GB has potential activity to inhibit inflammation and diabetes, and this extract is a promising candidate for development of functional foods and drugs to improve inflammation-related diseases and diabetes.
Maternal body mass index at delivery and preterm birth

Objective: To evaluate the association of the body mass index (BMI) at delivery on perinatal outcomes in Korean pregnant women. Methods: The study subjects were 2,485 pregnant women that visited the obstetrics clinic at Ewha Womans University Hospital for antenatal care and delivered single newborns between 25 and 42 weeks of gestation during 2005, 2006, and 2007. We used a logistic regression analysis to determine the risk of preterm delivery according to the maternal BMI at delivery. The adjusted odds ratios and 95% confidence intervals were calculated using multiple logistic regression models, controlling for maternal age, parity, and the gender of the neonates. Women were categorized as lean (BMI<19.8), normal (19.8≤BMI<25.9), overweight (26.0<BMI<28.9), or obese (BMI≥29.0) using the BMI cutoff points. Results: The lean BMI at delivery group showed a significant correlation with preterm birth (P<0.0001), spontaneous preterm birth (P=0.005), premature rupture of membrane (P<0.0001), and low birth weight (P<0.0001). Conclusion: Our results indicate that women that were underweight at delivery had significantly higher risk for preterm birth.

Our previous study demonstrated that CopA3, a disulfide dimer of the coprisin peptide analogue (LLCIALRKK), has antibacterial activity. In this study, we assessed whether CopA3 caused cellular toxicity in various mammalian cell lines. CopA3 selectively caused a marked decrease in cell viability in Jurkat T, U937, and AML-2 cells (human leukemia cells), but was not cytotoxic to Caki or Hela cells. Fragmentation of DNA, a marker of apoptosis, was also confirmed in the leukemia cell lines, but not in the other cells. CopA3-induced apoptosis in leukemia cells was mediated by apoptosis inducing factor (AIF), indicating induction of a caspase-independent signaling pathway. [BMB reports 2012; 45(2): 85-90].
