Production of a Phage-displayed Mouse ScFv Antibody against Fumonisin B1 and Molecular Docking Analysis of Their Interactions

        Zu-Quan Hu,He-Ping Li,Jin-Long Liu,Sheng Xue,An-Dong Gong,Jing-Bo Zhang,Yu-Cai Liao 한국생물공학회 2016 Biotechnology and Bioprocess Engineering Vol.21 No.1

Fumonisins produced by Fusarium pathogens are mycotoxins present in maize and other grains in the field as well as during storage worldwide and pose a serious threat to humans and domestic animals. Fumonisin B consists of different chemotypes, and fumonisin B1 (FB1) is the most predominant fumonisin found in food/feed commodities. Recombinant antibody can be deployed to analyze the fumonisin toxicological mechanism and develop a simple and cost-effective method for the detection of fumonisins, which is vitally important for monitoring and preventing fumonisins from entering food/feed chains. In this study, FB1 conjugated to keyhole limpet hemocyanin was used to immunize mice, from which RNA was isolated to construct a recombinant antibody library. Successive panning of the library by phage display was used to select monoclonal phage clones reactive to FB1 conjugated to bovine serum albumin. Subsequent phage ELISA and sequencing analyses revealed four different reactive scFv antibodies specific to FB1. Soluble expression and ELISA analysis showed that one scFv antibody, FBMA1, had the highest reactivity and could be purified from bacterial cells in large quantities. Surface plasmon resonance measurements further revealed that the FBMA1 scFv antibody had a binding kinetics of KD = 1.89 × 10–7 M. Molecular modeling and docking analyses suggested that the FBMA1 antibody shaped a proper cavity to embed the whole FB1 molecule and that a steady-state complex was formed relying on intermolecular forces, including hydrogen bonding, electrostatic force and hydrophobic interactions. Thus, the scFv antibody can be applied for mechanistic studies of intermolecular interactions and fumonisin toxicity, and for the development of an immunoassay for fumonisin-contaminated food/feed samples.

Preparation of Robust Aramid/Epoxy Composites through Enhancing the Interface Performance by Nanocoating Solution

        Tingli Hu,Zengxiao Wang,Yutong Cao,Zu Ming Hu,Jun Rong Yu,Yan Wang 한국섬유공학회 2022 Fibers and polymers Vol.23 No.4

Aramid fiber reinforced composites (AFRC) have attracted considerable attention because of its high strength andmodulus. But the poor interface due to the smooth fiber surface and few active groups limits the application of the finalcomposites. In this study, robust AFRC was achieved via dip-coating approach. Instead of Kevlar fiber, sulfonefunctionalizedpoly(p-phthaloyl-p-phenylenediamine) (SPPTA) was served as precursor to prepare nanocoating solution. Byfurther grafting epichlorohydrin (ECH) and Shellac (SLC), the functional coating solution was obtained. After modification, the roughness and the polarity of the fiber were considerably improved. The modified AF/epoxy composites showedexcellent interfacial bonding and mechanical properties. For the sample grafted with SLC, the interfacial shear strength(IFSS) reached 43.7 MPa with a significant enhancement by 21.4 %; while the inter-laminar shear strength (ILSS) reached48.6 MPa with a reinforcement of 45.9 %. The flexural strength and tensile strength were increased to 283 MPa and831 MPa, indicating that the study provides an effective method to achieve high strength AFRC.

      • Overexpression of Cyclooxygenase-1 Correlates with Poor Prognosis in Renal Cell Carcinoma

        Yu, Zu-Hu,Zhang, Qiang,Wang, Ya-Dong,Chen, Jing,Jiang, Zhi-Mao,Shi, Min,Guo, Xin,Qin, Jie,Cui, Guang-Hui,Cai, Zhi-Ming,Gui, Yao-Ting,Lai, Yong-Qing Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.6

The aim of this study was to evaluate expression of COX-1 in renal cell carcinoma (RCC) and its prognostic value. mRNA of COX-1 was detected in 42 paired RCC and adjacent normal tissues with quantitative realtime polymerase chain reaction (qRT-PCR). Expression of COX-1 was also evaluated in 196 RCC sections and 91 adjacent normal tissues with immunohistochemistry. Statistical analysis was performed to assess COX-1 expression in RCC and its prognostic significance. The results of qRT-PCR showed mRNA levels of COX-1 in RCC tissues to be significantly higher than that in adjacent normal tissues (p < 0.001). Immunohistochemical assays also revealed COX-1 to be overexpressed in RCC tissues (p < 0.001). Statistical analysis demonstrated high expression of COX-1 was correlated with tumour size (p = 0.002), pathological stage (p = 0.003), TNM stage (p = 0.003, 0.007, 0.027, respectively), and tumour recurrence (p < 0.001). Survival analysis indicated patients with high expression of COX-1 had shorter survival time (p < 0.001), and COX-1 was an independent predictor. This is the first study to reveal overexpression of COX-1 in RRC and point to use as a prognostic marker in affected patients.

Calculation of thermal neutron scattering data of MgF2 and its effect on beam shaping assembly for BNCT

        Hu Jiaqi,Qiao Zhaopeng,Fan Lunhe,Tang Yongqiang,Cao Liangzhi,Zu Tiejun,He Qingming,Li Zhifeng,Wang Sheng 한국원자력학회 2023 Nuclear Engineering and Technology Vol.55 No.4

MgF2 as a moderator material has been extensively used in the beam shaping assembly (BSA) that plays an important role in the boron neutron capture therapy (BNCT). Regarded as important for applications, the thermal neutron scattering data of MgF2 were calculated, based on the phonon expansion model. The structural properties of MgF2 were researched by the VASP code based on the ab-initio methods. The PHONOPY code was employed to calculate the phonon density of states. Furthermore, the NJOY code was used to calculate the thermal neutron scattering data of MgF2. The calculated inelastic cross sections plus absorption cross sections are in agreement with the available experimental data. The neutron transport in the BSA has been simulated by using a hybrid Monte-Carlo-Deterministic code NECP-MCX. The results indicated that compared with the calculation of the free gas model, the thermal neutron flux and epithermal neutron flux at the BSA exit port calculated by using the thermal neutron scattering data of MgF2 were reduced by 27.7% and 8.2%, respectively.

Genomic rearrangement between wheat and Thinopyrum elongatum revealed by mapped functional molecular markers

        Li-Jun Hu,Cheng Liu,Zi-Xian Zeng,Guang-Rong Li,Xiao-Jin Song,Zu-Jun Yang 한국유전학회 2012 Genes & Genomics Vol.34 No.1

Thinopyrum elongatum serves as an excellent gene pool for wheat improvement. Genes for resistance to many biotic and abiotic stresses have been transferred from Th. elongatum to wheat through chromosome manipulation. For breeding programs, molecular markers enable screening of a large number of genotypes for alien chromosome introgressions. The main objective of the present study was to develop and characterize EST (expressed sequence tags) and PLUG (PCR-based Landmark Unique Gene) markers that can distinguish Th. elongatum chromatin from the wheat genomes. A total of 258 mapped EST primer pairs and 46 PLUG primer pairs were tested on DNA from wheat Chinese Spring (CS) and CS-Th. elongatum addition lines. The results showed that 43 primer pairs could be effectively mapped to specific Th. elongatum chromosomes. Twenty-two of the 43 markers displayed similar homoeologous chromosome locations to hexaploid wheat. Nine markers mapped to different linkage groups between wheat and Th. elongatum, while 12 makers mapped on two or three different Th. elongatum chromosomes. A comparison of molecular marker locations indicated that Th. elongatum genome was closely related to the D genome of wheat, and chromosome rearrangements and duplication had occurred in Th. elongatum and the wheat genomes. The markers will be useful in comparative gene mapping, chromosome evolutionary analysis, and gene introgression for wheat improvement using Th. elongatum accessions as gene donors.

      • Pathogenesis, Early Diagnosis, and Therapeutic Management of Alcoholic Liver Disease

        Kong, Ling-Zu,Chandimali, Nisansala,Han, Ying-Hao,Lee, Dong-Ho,Kim, Ji-Su,Kim, Sun-Uk,Kim, Tae-Don,Jeong, Dong Kee,Sun, Hu-Nan,Lee, Dong Sun,Kwon, Taeho MDPI AG 2019 INTERNATIONAL JOURNAL OF MOLECULAR SCIENCES Vol.20 No.11

<P>Alcoholic liver disease (ALD) refers to the damages to the liver and its functions due to alcohol overconsumption. It consists of fatty liver/steatosis, alcoholic hepatitis, steatohepatitis, chronic hepatitis with liver fibrosis or cirrhosis, and hepatocellular carcinoma. However, the mechanisms behind the pathogenesis of alcoholic liver disease are extremely complicated due to the involvement of immune cells, adipose tissues, and genetic diversity. Clinically, the diagnosis of ALD is not yet well developed. Therefore, the number of patients in advanced stages has increased due to the failure of proper early detection and treatment. At present, abstinence and nutritional therapy remain the conventional therapeutic interventions for ALD. Moreover, the therapies which target the TNF receptor superfamily, hormones, antioxidant signals, and MicroRNAs are used as treatments for ALD. In particular, mesenchymal stem cells (MSCs) are gaining attention as a potential therapeutic target of ALD. Therefore, in this review, we have summarized the current understandings of the pathogenesis and diagnosis of ALD. Moreover, we also discuss the various existing treatment strategies while focusing on promising therapeutic approaches for ALD.</P>

DFT Studies on Two Novel Explosives Based on the Guanidine-Fused Bicyclic Structure

        Jin, Xing-Hui,Hu, Bing-Cheng,Jia, Huan-Qing,Liu, Zu-Liang,Lu, Chun-Xu Korean Chemical Society 2014 Bulletin of the Korean Chemical Society Vol.35 No.4

Density functional theory (DFT) calculations at the B3LYP/6-31G(d,p) theoretical level were performed for two novel explosives (compounds B and C) based on the guanidine-fused bicyclic skeleton $C_4N_6H_8$ (A). The heats of formation (HOFs) were calculated via isodesmic reaction. The detonation properties were evaluated by using the Kamlet-Jacobs equations. The bond dissociation energies (BDEs) for the thermolysis initiation bond were also analyzed to investigate the thermal stability. The results show that the compounds have high positive HOF values (B, 1064.68 $kJ{\cdot}mol^{-1}$; C, 724.02 $kJ{\cdot}mol^{-1}$), high detonation properties (${\rho}$, D and P values of 2.04 $g{\cdot}cm^{-3}$ and 2.21 $g{\cdot}cm^{-3}$, 9.98 $km{\cdot}s^{-1}$ and 10.99 $km{\cdot}s^{-1}$, 46.44 GPa and 59.91 Gpa, respectively) and meet the basic stability requirement. Additionally, feasible synthetic routes of the these high energy density compounds (HEDCs) were also proposed via retrosynthetic analysis.

Mao, Ying-Min,Luo, Zu-Yan,Li, Bo,Hu, Ting-Yang Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.3

Aim: Tanscatheter arterial embolization irrespective of with or without an anticancer agent and lipiodol has been controversial with regard to survival benefit. Therefore, we conducted a prospective study to analyze the effect of transcatheter arterial lipiodol chemoembolization (TACE) on the survival of HCC. Methods: A prospective study was conducted, and a total of 326 patients with primary liver cancer who were newly diagnosed were collected from January 2004 to January 2005 in Zhejiang Provincial People's Hospital of China. A univariate Cox's regression analysis was used to assess the survival of the HCC cases receiving TACE. Results: The duration of follow-up for the HCC patients treated with TACE ranged from 3 months to 60 months. For the overall patients, survival rate at 5 years was 42%. Both HBV Ag and HCV Ab positive patients showed significantly low survival rate at 5 years. The multivariate analysis revealed The IV TNM stage was related to an heavy increased risk of death of HCC patients, and Child C grade group showed a significant moderate increased risk. Conclusion: Our study showed TACE is associated with a better prognosis of HCC patients, and the HBV infection, TNM stage, Child-Pugh grade and number of TACE may influence the survival probability. Further TACE studies should be assess the quality of life of HCC patients, so as to provide more information for treatment of HCC.

The infection frequencies and dynamics of three secondary endosymbionts in the laboratory environments on Sitobion avenae (Fabricius) as determined by long PCR

        Chen Luo,Kun Luo,Zu-Qing Hu,Yue-Yue Tao,Hui-Yan Zhao 한국응용곤충학회 2016 Journal of Asia-Pacific Entomology Vol.19 No.2

Stable infections of maternally transmitted secondary endosymbionts in aphids are frequently found in field populations; however, whether this phenomenon changes in artificial lab conditions has not been clear. To investigate, (1): we collected Sitobion avenae (Fabricius) fromsix different regions and detected the three common secondary endosymbionts (Regiella insecticola, Hamiltonella defensa and Serratia symbiotica); (2): we raised Sitobion avenae fromChuzhou, Anhui for 14 months in the laboratory, and analyzed these aphids' DNA using the long PCR protocol after the 1stmonth, 3rdmonth, 5th month, 7thmonth, 9thmonth, 11thmonth, 12thmonth, 13thmonth and 14th month. The results showed that S. symbiotica and R. insecticola could be detected in all aphid populations, while H. defense were detected in three locations. After reared in the lab, the infection frequencies of Serratia symbiotica did not change across the 14 study months, but the infection frequencies of Regiella insecticola after the 9th month and of Hamiltonella defensa after the 11th month decreased significantly. Additionally, the results show that facultative endosymbiont infections may decline in artificial lab conditions, suggesting that temporal changes of endosymbionts may occur within the host under different environmental pressures. This result may partially explain why different geographic populations harbor a diverse variety of endosymbionts. The results of this study can be helpful in understanding the stability of secondary endosymbionts in Sitobion avenae under long-termreproduction in controlled laboratory conditions.
