      • PKCδ-dependent Activation of the Ubiquitin Proteasome System is Responsible for High Glucose-induced Human Breast Cancer MCF-7 Cell Proliferation, Migration and Invasion

        Type 2 diabetes mellitus (T2DM) has contributed to advanced breast cancer development over the past decades. However, the mechanism underlying this contribution is poorly understood. In this study, we determined that high glucose enhanced proteasome activity was accompanied by enhanced proliferation, migration and invasion, as well as suppressed apoptosis, in human breast cancer MCF-7 cells. Proteasome inhibitor bortezomib (BZM) pretreatment mitigated high glucose-induced MCF-7 cell growth and invasion. Furthermore, high glucose increased protein kinase C delta ($PKC{\delta}$)-phosphorylation. Administration of the specific $PKC{\delta}$ inhibitor rottlerin attenuated high glucose-stimulated cancer cell growth and invasion. In addition, $PKC{\delta}$ inhibition by both rottlerin and $PKC{\delta}$ shRNA significantly suppressed high glucose-induced proteasome activity. Our results suggest that $PKC{\delta}$-dependent ubiquitin proteasome system activation plays an important role in high glucose-induced breast cancer cell growth and metastasis.

      • BCR/ABL mRNA Targeting Small Interfering RNA Effects on Proliferation and Apoptosis in Chronic Myeloid Leukemia

        Background: To investigate the effects of small interference RNA (siRNA) targeting BCR/ABL mRNA on proliferation and apoptosis in the K562 human chronic myeloid leukemia (CML) cell line and to provide a theoretical rationale and experimental evidence for its potential clinical application for anti-CML treatment. Materials and Methods: The gene sequence for BCR/ABL mRNA was found from the GeneBank. The target gene site on the BCR/ABL mRNA were selected according to Max-Planck-Institute (MPI) and rational siRNA design rules, the secondary structure of the candidate targeted mRNA was predicted, the relevant thermodynamic parameters were analyzed, and the targeted gene sequences were compared with BLAST to eliminate any sequences with significant homology. Inhibition of proliferation was evaluated by MTT assay and colony-formation inhibiting test. Apoptosis was determined by flow cytometry (FCM) and the morphology of apoptotic cells was identified by Giemsa-Wright staining. Western blotting was used to analyze the expression of BCR/ABL fusion protein in K562 cells after siRNA treatment. Results: The mRNA local secondary structure calculated by RNA structure software, and the optimal design of specific siRNA were contributed by bioinformatics rules. Five sequences of BCR/ABL siRNAs were designed and synthesized in vitro. Three sequences, siRNA1384, siRNA1276 and siRNA1786, which showed the most effective inhibition of K562 cell growth, were identified among the five candidate siRNAs, with a cell proliferative inhibitory rate nearly 50% after exposure to 12.5nmol/L~50nmol/L siRNA1384 for 24,48 and 72 hours. The 50% inhibitory concentrations ($IC_{50}$) of siRNA1384, siRNA1276 and siRNA1786 for 24hours were 46.6 nmol/L, 59.3 nmol/L and 62.6 nmol/L, respectively, and 65.668 nmol/L, 76.6 nmol/L, 74.4 nmol/L for 72 hours. The colony-formation inhibiting test also indicated that, compared with control, cell growth of siRNA treated group was inhibited. FCM results showed that the rate of cell apoptosis increased 24 hours after transfecting siRNA. The results of annexinV/PI staining indicated that the rate of apoptosis imcreased (1.53%, 15.3%, 64.5%, 57.5% and 21.5%) following treamtne with siRNAs (siRNA34, siRNA372, siRNA1384, siRNA1276 and siRNA1786). Morphological analysis showed td typical morphologic changes of apoptosis such as shrunken, fragmentation nucleus as well as "apoptotic bodies" after K562 cell exposure to siRNA. Western blot analysis showed that BCR/ABL protein was reduced sharply after a single dose of 50nmol/L siRNA transfection. Conclusions: Proliferation of K562 cells was remarkbly inhibited by siRNAs (siRNA1384, siRNA1276 and siRNA1786) in a concentration-dependent manner in vitro, with effective induction of apoptosis at a concentration of 50 nmol/L. One anti-leukemia mechanism in K562 cells appeared that BCR/ABL targeted protein was highly down-regulated. The siRNAs (siRNA1384, siRNA1276 and siRNA1786) may prove valuable in the treatment of CML.

      • 도시 뒤에 가려진 구석 귀퉁이 : 농촌에 남겨진 부녀자의 일상생활

        본 연구는 산둥성(山東省) 남부의 쑹좡(宋莊)을 조사 지역으로 삼아, 미시적인 접근으로 이 마을에 남겨진 부녀자들 일상생활의 한 측면과, 그 부녀자들이 생계에 쫓겨 일반인은 상상하기도 어려운 체력적인 고생을 겪으며 정신적으로도 큰 대가를 치르고 있음을 확인했다. 그와 동시에 현실 속에서 남겨진 부녀자들이 농촌의 ‘지킴이’면서 도시의 ‘선택’ 뒤에 남겨진 ‘잔여물’임을 발견했다. 그녀들은 주체의식을 가지고 번거롭고 힘든 육체노동, 가사노동, 노인 봉양과 아이 양육의 임무를 맡고 있었다. 그녀들은 마을에서 가장 중요한 역할을 맡고 있었지만, 주관적인 인식과 객관적인 조건의 제약 때문에, 마을이나 사회라는 틀 속에서 자기표현을 할 기회가 없었다. 그와 동시에 그녀들의 모습은 지금의 주류매체와 대중매체 속에서 정상적으로 전달되질 못하고, 현대문학 속에서도 제대로 표현되지 못한 채, 왜곡되거나 가려져 버렸다. 이러한 현상에 대한 원인을 분석하고 좀 더 고려해 볼 필요가 있다. Songzhuang(宋莊) Village, located at the southern part of the Shandong Province(山東省), was used as the investigation area for this study. Adopting a microscopic approach to examine the everyday life of women left in this village, the study reveals that in order to maintain livelihood, these village women often have to go through physical and mental hardships unimaginable to most people. At the same time, the study sheds light on the fact that these women who are left behind in reality, are the ‘remains’ of those who ‘choose’ the city and are the ones staying behind to ‘protect’ the village. To upkeep traditions and identity, they are entrusted with the onerous and difficult tasks of physical labor, housework, elderly care and child care. Despite playing the most important role in the village, these women have no opportunity for self-expression within the frameworks of the village and society, due to subjective perceptions and the constraints of objective conditions. In addition, they are not properly portrayed in the current mainstream media and mass media, and are often misrepresented in modern literature which distorts and disguise their image. There is thus a need to analyze the cause of such a phenomenon and think deeper into it.

      • Research on Fuzzy PD Current Controller for FDWRD

        A fuzzy control scheme is proposed for the system of filling and draining off water of the rubber dam(FDWRD) which is a typical nonlinear, multi- variables, time- varying and strong coupling control system base on traditional PID control or model-based control methods can not meet its constant flow sufficient drainage control requirements, during to it couldn‘t be established accurate mathematical model. In a practical project, a fuzzy PD controller is designed for the constant current control of filling and draining off water of the rubber dam, combining with the traditional PID algorithm which is widely used in industrial process control, and achieved good control effect. This paper elaborates the control system of filling and draining off water of the rubber dam and the implementation process of the fuzzy PD controller, and then giving the actual control curve of constant current. Practice proves that the proposed control scheme is efficient and the system possesses good static, dynamic performance and certain robustness.

        조선 세종시기의 북방정책은 4군6진의 개척, 즉 영토를 조선과 여진사이의 미확정 지역인 압록강지역과 도문강지역으로 확장하는 것이다. 이로서 압록강과 도문강이 중조양국의 국계로 되었다. The so-called northern policy in the period of shizongin Korea refers to the opening up of four prefectures and six towns. The purpose of it is to extend the territory to the areas of upper reaches of Yalujang River and Tumenjiang River which haven`s been clearly delimited by Korea and nvzhen, thus making these two rivers the national boundaries between China and Korea.

        First-Principles Study of the Elastic, Hardness and Electronic Properties of MoAlB under High Pressure

        The structure, elastic and electronic properties of MoAlB have been studied under high pressure by using first principles. The optimized lattice parameters, elastic constants, bulk moduli, and shear moduli agree well with experimental and other theoretical values. The relationships of the lattice constants with pressure indicated an anisotropic behavior. The calculated elastic constants satisfy the mechanical stability conditions. Our calculated Vickers hardness of MoAlB is 11.56~GPa, which agrees with the experimental data and confirms its having a relatively softer nature than the other borides. The Cauchy pressure, the Poisson's ratio and the $G/B$ value suggest that MoAlB is a brittle solid under ambient conditions, but the brittleness decreases with increasing pressure. The band structure and the density of states reveal the metallic feature of MoAlB, and the energy levels shift toward both sides of the Fermi level with increasing pressure. The Mulliken population and density of states suggest a mixture bonding nature of covalence and metallicity in MoAlB.

        Triterpenoid saponins from Clinopodium chinense (Benth.) O. Kuntze and their biological activity

        Four new ursane-type triterpenoid saponins, clinopoursaponins A–D (1–4), six new oleanane-type triterpenoid saponins, clinopodiside VII–XII (5–10), as well as eight known triterpene analogues (11–18), were isolated from the aerial parts of Clinopodium chinense (Benth.) O. Kuntze. The structures of the new compounds were determined based on extensive spectral analyses, including 1D (1H and 13C) and 2D NMR experiments (COSY, NOESY, HSQC, 2D TOCSY, HSQC-TOCSY and HMBC), HR-ESI-MS and chemical methods. Compounds 1–18 were evaluated for their protective effects against anoxia/reoxygenation-induced apoptosis in H9c2 cells and cytotoxicities against murine mammary carcinoma cell line 4T1. Compounds 8, 9 and 18 exhibited significant protective effects, while compound 1 exhibited cytotoxic activity with IC50 value of 7.4 μm compared to 7.6 μm for the positive control 10-hydroxycamptothecin.

        Stochastic Resonance Control by Coupling Two Fractional Bistable Systems

        In order to enhance or produce stochastic resonance (SR), to obtain the required output response with specific properties, two bistable systems based on fractional over-damped Langevin equation are coupled into one multi-steady-state system via nonlinear method, one fractional bistable system where inherent parameters can be controlled is fed into the other system, the kinetic characteristics of the new coupled system is found much richer than each single fractional system. Theoretical analysis and numerical simulation results show that the SR can be enhanced or produced by adjusting parameters as coupling coefficient, fractional system inherent parameters as well as the external periodical signal amplitude while another fractional system’s parameters fixed. The coupled system has high practical value, which provides a reliable theoretical basis for the control and its applications of artificially generated stochastic resonance.
