        β-Lapachone suppresses radiation-induced activation of nuclear factor-κB

        Anticancer effects of β-lapachone (β-lap) are due to generation of ROS and metabolic catastrophes as a result of NAD(P)H:quinone oxidoreductase (NQO1)-mediated futile cycling between the oxidized and reduced forms of β-lap. It has been shown that NQO1 is also essential for the TNF-induced activation of NF-κB and that β-lap suppresses the TNF-induced NF-κB activation. We investigated whether or not NQO1 is involved and β-lap suppresses the radiation-induced NF-κB activation using A549 human lung cancer cells and NQO1-knock down A549 cells (shNQO1 A549cells). Irradiation with 4 Gy markedly increased the DNA binding activity of NF-κB in A549 cells, but not in the shNQO1 A549 cells, thus demonstrating that NQO1plays a pivotal role in irradiation-induced NF-κB activation. Treatment with 10 μM β-lap for 4 h almost completely abrogated the radiation-induced increase in NF-κB activation and the transcription of NF-κB target genes such as bcl2, gadd45β and cyclinD1. Moreover, β-lap markedly suppressed the activation of IκB kinase γ (IKKγ) and the subsequent phosphorylation of IκBα, thereby inhibiting NF-κB activation. It is concluded that β-lap suppresses the radiation-induced activation of NF-κB by interrupting the involvement of NQO1 in the activation of NF-κB, thereby inhibiting the transcription of survival signals. The radiosensitization caused by β-lap may, in part, be attributed to β-lap-induced suppression of NF-κB activation.

        Synthesis and Evaluation O-Benzyl Oxime-ether Derivatives Containing β-Methoxyacrylate Moiety for Insecticidal and Fungicidal Activities

        In attempt to lead compounds exhibiting both insecticidal and fungicidal activities, a series of O-benzyl oximeether derivatives were designed and synthesized by introducing β-methoxyacrylate pharmacophore into a scaffold. The insecticidal activity against Aphis fabae and the fungicidal activity against Erysiphe graminis were screened. The title compounds exhibited remarkable insecticidal and fungicidal activities. The most potent compound 6d was identified. Its insecticidal LC50 against A. fabae is 6.4 mg/L, which is lower than that of chlorfenapyr (19.4 mg/L) and even close to the level of imidacloprid (4.8 mg/L). Its fungicidal EC90 in preventive and curative treatment against E. graminis are 2.2 and 4.8 mg/L, respectively, which are lower than azoxystrobin (7.0 and 5.9 mg/L). These results indicate that compound 6d can be considered as a lead for further developing new O-benzyl oxime-ether typed candidates with both fungicidal and insecticidal activities.

        Diarylheptanoids suppress proliferation of pancreatic cancer PANC-1 cells through modulating shh-Gli-FoxM1 pathway

        Pancreatic cancer is one of the leading causes ofcancer, and it has the lowest 5-year survival rates. It isnecessary to develop more potent anti-pancreatic cancerdrugs to overcome the fast metastasis and resistance tosurgery, radiotherapy, chemotherapy, and combinations ofthese. We have identified several diarylheptanoids as antipancreaticcancer agents from Alpinia officinarum (lessergalangal) and Alnus japonica. These diarylheptanoidssuppressed cell proliferation and induced the cellcycle arrest of pancreatic cancer cells (PANC-1). Amongthem, the most potent compounds 1 and 7 inhibited theshh-Gli-FoxM1 pathway and their target gene expression inPANC-1 cells. Furthermore, they suppressed the expressionof the cell cycle associated genes that were rescued bythe overexpression of exogenous FoxM1. Taken together,(E)-7-(4-hydroxy-3-methoxyphenyl)-1-phenylhept-4-en-3-one (1) from Alpinia officinarum (lesser galangal) andplatyphyllenone (7) from Alnus japonica inhibit PANC-1cell proliferation by suppressing the shh-Gli-FoxM1 pathway,and they can be potential candidates for anti-pancreaticcancer drug development.

        Two new records of genus Nudaria Haworth, 1809 (Lepidoptera, Erebidae, Arctiinae, Lithosiini) from China

        Two species of genus Nudaria Haworth, N. fasciata Moore, 1878 and N. phallustortens Holloway, 2001, are reported for the first time from China. The adult and genital characters of the two species are briefly redescribed, with illustrations and distributional data given.

        Surface roughness is well known to affect cellular activity. This study presents a method to fabricate nano-scale surface roughness on the surfaces of polymer, metal, glass with calcium phosphate thin film. Metastable calcium and phosphate ion solution was slowly heated to obtain calcium phosphate thin fibn. Then fresh calcium and phosphate ion solution was added to fabricate nano-scale roughness over tlùs initially forrned thin film. Various surfaces of nano- scale roughness lower than 800 nm of Ra v외ue was obtained. Initial cell adhesion was characterized over the fabricated surfaces using MC3T3-El osteoblastic cellline. Spreading of osteoblastic cell decreased according to the increase of surface roughness. Less stress fibers developed in the cells attaching to the rougher surface. Less focal contacts visualized by staining vinculin was found in the cells of irútial adhesion. All these features of the initial cell adhesion represents that less adhesion signals are generated from the rougher surface. Further study is required to evaluate the effect of these low adhesion signals on the overall cellular activitv.

        Modification of C-doped a-SiO2 after Swift Heavy-Ion Irradiation

        Amorphous SiO2 (a-SiO2) thin films were thermally grown on single-crystalline silicon. These a-SiO2/Si samples were first implanted (C-doped) with 100-keV carbon ion at room temperature (RT) at a dose of 5.0 × 1017 C-ions/cm2 and were then irradiated at RT by using 853 MeV Pb ions at doses of 5.0 × 1011, 1.0 × 1012, 2.0 × 1012, and 5.0 × 1012 Pb-ions/cm2, respectively. The microstructures and the photoluminescence (PL) properties of these samples induced by Pb ions were investigated using fluorescence spectroscopy and transmission electron microscopy. We found that high-energy Pb-ion irradiation could induce the formation of a new phase and a change in the PL property of C-doped a-SiO2/Si samples.       • Tax is Involved in Up-regulation of HMGB1 Expression Levels by Interaction with C/EBP

        The high mobility group box 1 (HMGB1) protein is a multifunctional cytokine-like molecule that plays an important role in the pathogenesis of tumors. In this study, real-time polymerase chain reactions and Western blot assays indicated that HMGB1 transcriptional activity and protein level are increased in $Tax^+$-T cells (TaxP). To clarify the mechanisms, a series of HMGB1 deletion reporter plasmids (pHLuc1 to pHLuc6) were transfected into $Tax^-$-T cells (TaxN, Jurkat) and $Tax^+$-T cells (TaxP). We found that promoter activity in $Tax^+$-T cells to be higher than that in $Tax^-$-T cells, indicating a significant increase in pHLuc6. Bay11-7082 (NF-${\kappa}B$ inhibitor) treatment did not block the enhancing effect. Chromatin immunoprecipitation assays revealed that Tax was retained on a HMGB1 promoter fragment encompassing -1163 to -975. Bioinformatics analysis showed six characteristic cis-elements for CdxA, AP-1, AML-1a, USF, v-Myb, and C/EBP in the fragment in question. Mutation of cis-elements for C/EBP reduced significant HMGB1 promoter activity induced by Tax. These findings indicate that Tax enhances the expression of HMGB1 gene at the transcriptional level, possibly by interacting with C/EBP.

        In Sichuan, the number of different editions of the musical Xiaoyaoji amounts to nearly forty。It is a long narrative folklore among Sichuan Han people. It intactly recites a romatic tragical story caused by affair. This thesis firstly defines the musical edition as folklore xiaodiao, then introduces and analyzes the story srtucture、qudiao and content. Many people know moonset in the west but ignore Xiaoyaoji, so this thesis explains the propagation history of Xiaoyaoji in the last part. The reason why Xiaoyaoji was ineluctably questioned and opposed at that time is because of the affair it involves. That’s why the related parts in prelude can only be “forgotten” and also the social reason for why Xiaoyaoji could be “selected”. The affair subject in Xiaoyaoji is cleared up without a clue from that time on.
