      • Baicalin Induces Apoptosis in Leukemia HL-60/ADR Cells via Possible Down-regulation of the PI3K/Akt Signaling Pathway

        Background: The effect and possible mechanism of traditional Chinese medicine, baicalin, on the PI3K/Akt signaling pathway in drug-resistant human myeloid leukemia HL-60/ADR cells have been investigated in this current study. Methods: HL-60/ADR cells were treated by 20, 40, $80\;{\mu}mol/L$ baicalin followed by cell cycle analysis at 24h. The mRNA expression level of the apoptosis related gene, Bcl-2 and bad, were measured by RT-PCR on cells treated with $80\;{\mu}mol/L$ baicalin at 12, 24 and 48hr. Western blot was performed to detect the changes in the expression of the proteins related to HL-60/ADR cell apoptosis and the signaling pathway before and after baicalin treatment, including Bcl-2, PARP, Bad, Caspase 3, Akt, p-Akt, NF-${\kappa}B$, p-NF-${\kappa}B$, mTOR and p-mTOR. Results: Sub-G1 peak of HL-60/ADR cells appeared 24 h after $20\;{\mu}mol/L$ baicalin treatment, and the ratio increased as baicalin concentration increased. Cell cycle analysis showed 44.9% G0/G1 phase cells 24 h after baicalin treatment compared to 39.6% in the control group. Cells treated with $80\;{\mu}mol/L$ baicalin displayed a trend in decreasing of Bcl-2 mRNA expression over time. Expression level of the Bcl-2 and PARP proteins decreased significantly while that of the PARP, Caspase-3, and Bad proteins gradually increased. No significant difference in Akt expression was observed between treated and the control groups. However, the expression levels of p-Akt, NF-${\kappa}B$, p-NF-${\kappa}B$, mTOR and p-mTOR decreased significantly in a time-dependent manner. Conclusions: We conclude that baicalin may induce HL-60/ADR cell apoptosis through the PI3K/AKT signaling pathway.

        Intranasal administration of oxytocin attenuates stress responses following chronic complicated stress in rats.

        Background/Aims Gastrointestinal (GI) symptoms may develop when we fail to adapt to various stressors of our daily life. Central oxytocin (OXT) can counteract the biological actions of corticotropin-releasing factor (CRF), and in turn attenuates stress responses. Administration (intracerebroventricular) of OXT significantly antagonized the inhibitory effects of chronic complicated stress (CCS) on GI dysmotility in rats. However, intracerebroventricular administration is an invasive pathway. Intranasal administration can rapidly deliver peptides to the brain avoiding stress response. The effects of intranasal OXT on hypothalamus-pituitary-adrenal axis and GI motility in CCS conditions have not been investigated. Methods A CCS rat model was set up, OXT 5, 10, or 20 μg were intranasal administered, 30 minutes prior to stress loading. Central CRF and OXT expression levels were analyzed, serum corticosterone and OXT concentrations were measured, and gastric and colonic motor functions were evaluated by gastric emptying, fecal pellet output, and motility recording system. Results Rats in CCS condition showed significantly increased CRF expression and corticosterone concentration, which resulted in delayed gastric emptying and increased fecal pellet output, attenuated gastric motility and enhanced colonic motility were also recorded. OXT 10 μg or 20 μg significantly reduced CRF mRNA expression and the corticosterone concentration, OXT 20 μg also helped to restore GI motor dysfunction induced by CCS. Conclusion Intranasal administration of OXT has an anxiolytic effect and attenuates the hypothalamus-pituitary-adrenal axis in response to CCS, and gave effects which helped to restore GI dysmotility, and might be a new approach for the treatment of stress-induced GI motility disorders.         Nonlinear bending analysis of magnetoelectroelastic rectangular plates using higher order shear deformation theory

        Nonlinear bending analysis was performed of magnetoelectroelastic (MEE) composite plates under a mechanical and magnetoelectric (ME) load by using von Karman’s nonlinear geometric equation and the higher order shear deformation theory (HSDT). Nonlinear higher order partial differential equations for MEE plates were derived by using Hamiltonian equilibrium equation. The MEE plate is considered to have clamped boundary condition. The nonlinear high-order equations can turn into algebraic equations through Galerkin method. Then the effects of scale effect of MEE plate (for instance, the aspect ratio) and external load (for instance, mechanical) on the displacement of the considered MEE plate were investigated.

      • Graph-Optimized SLAM Algorithm for Real-time Dense Reconstruction of Greenhouse Crops

        Greenhouse agricultural technology is undergoing rapid advancements, confronting the challenges of digitization and automation. In this study, we introduce a novel system for real-time 3D reconstruction of greenhouse crops based on SLAM. Employing RGBD cameras, the algorithm captures real-time 3D point cloud frames, subsequently extracting features for pose estimation. Only keyframes are retained in memory, constituting a pose-graph to economize computational resources. This architecture undergoes backend loop closure detection and is subjected to global optimization, further fortified by marker constraints. The resultant optimized pose graph, combined with the keyframes, is synthesized into a holistic 3D point cloud model of the greenhouse environment. Our method highly correlates with manual measurements (R²=0.996, RMSE=6.26 mm). This research offers a robust 3D modeling approach for greenhouse crops, benefiting phenotypic analysis and digital agriculture.

        Human lactoferrin efficiently targeted into caprine beta-lactoglobulin locus with transcription activator-like effector nucleases

        Objective: To create genetically modified goat as a biopharming source of recombinant human lacotoferrin (hLF) with transcription activator-like effector nucleases. Methods: TALENs and targeting vector were transferred into cultured fibroblasts to insert hLF cDNA in the goat beta-lactoglobulin (BLG) locus with homology-directed repair. The gene targeted efficiency was checked using sequencing and TE7I assay. The bi-allelic gene targeted colonies were isolated and confirmed with polymerase chain reaction, and used as donor cells for somatic cell nuclear transfer (SCNT). Results: The targeted efficiency for BLG gene was approximately 10%. Among 12 Bi-allelic gene targeted colonies, five were used in first round SCNT and 4 recipients (23%) were confirmed pregnant at 30 d. In second round SCNT, 7 (53%), 4 (31%), and 3 (23%) recipients were confirmed to be pregnant by ultrasound on 30 d, 60 d, and 90 d. Conclusion: This finding signifies the combined use of TALENs and SCNT can generate bi-allelic knock-in fibroblasts that can be cloned in a fetus. Therefore, it might lay the foundation for transgenic hLF goat generation and possible use of their mammary gland as a bioreactor for large-scale production of recombinant hLF.

        Comparing eight types of ginsenosides in ginseng of different plant ages and regions using RRLC-Q-TOF MS/MS

        Background: This article aims to compare and analyze the contents of ginsenosides in ginseng ofdifferent plant ages from different localities in China. Methods: In this study, 77 fresh ginseng samples aged 2e4 years were collected from 13 differentcultivation regions in China. The content of eight ginsenosides (Rg3, Rc, Rg1, Rf, Rb2, Rb1, Re, and Rd) wasdetermined using rapid resolution liquid chromatography coupled with quadrupoleetime-of-flighttandem mass spectrometry (RRLC-Q-TOF MS/MS) to comparatively evaluate the influences of cultivationregion and age. Results: Ginsenoside contents differed significantly depending on age and cultivation region. The contentsof ginsenosides Re, Rc, Rg1, Rg3, and Rf increased with cultivation age, whereas that of ginsenoside Rb1peaked in the third year of cultivation. Moreover, the highest ginsenoside content was obtained fromChangbai (19.36 mg/g) whereas the lowest content was obtained from Jidong (12.05 mg/g). Ginseng fromJilin Province contained greater total ginsenosides and was richer in ginsenoside Re than ginseng of thesame age group in Heilongjiang and Liaoning provinces, where Rb1 and Rg1 contents were relatively high. Conclusion: In this study, RRLC-Q-TOF MS/MS was used to analyze ginsenoside contents in 77 ginsengsamples aged 2e4 years from different cultivation regions. These patterns of variation in ginsenosidecontent, which depend on harvesting location and age, could be useful for interested parties to chooseginseng products according to their needs.

      • Facile synthesis of Nafion-supported Pt nanoparticles with ultra-low loading as a high-performance electrocatalyst for hydrogen evolution reaction

        <P><B>Abstract</B></P> <P>x%Pt-Naf-CV (Pt-Nafion-Cyclic Voltammetry) catalysts with homogeneously distributed platinum nanoparticles and ultra-low Pt loading are successfully synthesized by using a facile potential cycling approach. The as-synthesized 0.8%Pt-Naf-CV catalyst exhibits an enhanced electrocatalytic activity for hydrogen evolution reaction (HER) in 0.5 M H<SUB>2</SUB>SO<SUB>4</SUB> solution, which obtains a low overpotential of 34 mV at 10 mA cm<SUP>−2</SUP>. The linear sweep voltammetry (LSV) curve of 0.8%Pt-Naf-CV catalyst is almost consistent with that of commercial Pt/C. However, the 0.8%Pt-Naf-CV catalyst displays a more excellent stability and durability in comparison with commercial Pt/C. Besides, the Pt loading of Pt/C (Pt–10 wt%) is about 10 times that of 0.8%Pt-Naf-CV catalyst. The improved electrocatalytic performances are derived from the synergistic effects of Pt and Nafion. The Nafion plays a significant role as a dispersant, carrier and structure directing agent on the morphology and size of the Pt catalyst.         Complete ¹H-NMR and <SUP>13</SUP>C-NMR spectral assignment of five malonyl ginsenosides from the fresh flower buds of Panax ginseng

        Background: Ginsenosides are the major effective ingredients responsible for the pharmacological effects of ginseng. Malonyl ginsenosides are natural ginsenosides that contain a malonyl group attached to a glucose unit of the corresponding neutral ginsenosides. Methods: Medium-pressure liquid chromatography and semipreparative high-performance liquid chromatography were used to isolate purified compounds and their structures determined by extensive one-dimensional- and two-dimensional nuclear magnetic resonance (NMR) experiments. Results: A new saponin, namely malonyl-ginsenoside Re, was isolated from the fresh flower buds of Panax ginseng, along with malonyl-ginsenosides Rb1, Rb2, Rc, Rd. Some assignments for previously published ¹H- and <SUP>13</SUP>C-NMR spectra were found to be inaccurate. Conclusion: This study reports the complete NMR assignment of malonyl-ginsenoside Re, Rb₁, Rb₂, Rc, and Rd for the first time.
