        Upregulation and biological function of transmembrane protein 119 in osteosarcoma

        Osteosarcoma is suggested to be caused by genetic and molecular alterations that disrupt osteoblast differentiation. Recent studies have reported that transmembrane protein 119 (TMEM119) contributes to osteoblast differentiation and bone development. However, the level of TMEM119 expression and its roles in osteosarcoma have not yet been elucidated. In the present study, TMEM119 mRNA and protein expression was found to be up-regulated in osteosarcoma compared with normal bone cyst tissues. The level of TMEM119 protein expression was strongly associated with tumor size, clinical stage, distant metastasis and overall survival time. Moreover, gene set enrichment analysis (GSEA) of the Gene Expression Omnibus (GEO) GSE42352 dataset revealed TMEM119 expression in osteosarcoma tissues to be positively correlated with cell cycle, apoptosis, metastasis and TGF-β signaling. We then knocked down TMEM119 expression in U2OS and MG63 cells using small interfering RNA, which revealed that downregulation of TMEM119 could inhibit the proliferation of osteosarcoma cells by inducing cell cycle arrest in G0/G1 phase and apoptosis. We also found that TMEM119 knockdown significantly inhibited cell migration and invasion, and decreased the expression of TGF-β pathway-related factors (BMP2, BMP7 and TGF-β). TGF-β application rescued the inhibitory effects of TMEM119 knockdown on osteosarcoma cell migration and invasion. Further in vitro experiments with a TGF-β inhibitor (SB431542) or BMP inhibitor (dorsomorphin) suggested that TMEM119 significantly promotes cell migration and invasion, partly through TGF-β/BMP signaling. In conclusion, our data support the notion that TMEM119 contributes to the proliferation, migration and invasion of osteosarcoma cells, and functions as an oncogene in osteosarcoma.

        In this paper two so-called regularized Green's functions are introduced to derive the optimal maximum norm error estimates for the unknown function and the adjoint vector-valued function for mixed finite element methods of Laplacian operator. One contribution of the paper is a demonstration of how the boundedness of $L^1$-norm estimate for the second Green's function ${\lambda}_2$ and the optimal maximum norm error estimate for the adjoint vector-valued function are proved. These results are seemed to be to be new in the literature of the mixed finite element methods.

        Parametric pitch instability investigation of Deep Draft Semi-submersible platform in irregular waves

        Parametric pitch instability of a Deep Draft Semi-submersible platform (DDS) is investigated in irregular waves. Parametric pitch is a form of parametric instability, which occurs when parameters of a system vary with time and the variation satisfies a certain condition. In previous studies, analyzing of parametric instability is mainly limited to regular waves, whereas the realistic sea conditions are irregular waves. Besides, parametric instability also occurs in irregular waves in some experiments. This study predicts parametric pitch of a Deep Draft Semi-submersible platform in irregular waves. Heave motion of DDS is simulated by wave spectrum and response amplitude operator (RAO). Then Hill equation for DDS pitch motion in irregular waves is derived based on linear-wave theory. By using Bubnov-Galerkin approach to solve Hill equation, the corresponding stability chart is obtained. The differences between regular-waves stability chart and irregular-waves stability chart are compared. Then the sensitivity of wave parameters on DDS parametric pitch in irregular waves is discussed. Based on the discussion, some suggestions for the DDS design are proposed to avoid parametric pitch by choosing appropriate parameters. The results indicate that it's important and necessary to predict DDS parametric pitch in irregular waves during design process.

        Morphology and Molecular Characterization of Alternaria argyranthemi on Chrysanthemum coronarium in China

        Chrysanthemum coronarium is an economically important plant in Asia, and used medicinally, ornamentally and as a vegetable. In April 2017, leaf spot disease on C. coronarium was observed in Shiyan, Hubei, China. A single-spore isolate was obtained and identified based on morphology and sequence analysis using four regions (rDNA ITS, GAPDH, EF-1a, and RPB2). The results indicated that the fungus is Alternaria argyranthemi. The pathogenicity tests revealed that the species could cause severe leaf spot and blight disease on the host. This is the first report of leaf spot disease on C. coronarium caused by A. argyranthemi in the world, which is also a new record of Alternaria species in China.

        Highly heat-resistant NF membrane modified by quinoxaline diamines for Li+ extraction from the brine

        The Functional layers with excellent heat-resistence nanofiltration (HRN) property were designed by fixingthermal stabilized quinoxaline diamines (QHDA) on poly (m-phenyleneisophthalamide) (PMIA) substratethrough interfacial polymerization. QHDA was used as aqueous monomer while cinnamoylchloride (CNC), isophthaloyl chloride (IPC) and trimesoyl chloride (TMC) were served as organic monomersto realize the immobilization of thermally stable N heterocycles. The experimental results statedclearly that the compact package reaction of QHDA with TMC lead a raise of thermal stability and saltsrejection at high temperature stream. With the optimal TMC and 0.5 wt.% QHDA, the as-developed membraneachieved the excellent NF performance and thermal stability. The rejection of MgSO4 reached to94.6 % at 30 C and hardly decrease at 90 C. The reduction in rejection was satisfactory at high temperatures. The functional layer still showed highly stability after 10 h long-term operation at 90 C with only2.8 % reduction in MgSO4 rejection. Besides, the excellent Mg2+ and poor Li+ rejection made this membranehave huge potential in the application of extracting Li+ from brine. The separation factor ofMg2+-Li+ at high temperature was satisfactory. Over all, this study offered a prospect technology toexploit heat-resistant membranes for extraction lithium at high temperature.

        Microbial Community on Healthy and Diseased Leaves of an Invasive Plant Eupatorium adenophorum in Southwest China

        Invasive plants have caused great economic losses and environmental problems worldwide. Eupatorium adenophorum is one of the most invasive weeds in China. To better understand its invasive mechanisms, in the present paper, the microbial communities of healthy and diseased leaves of E. adenophorum were obtained using both culture-independent and -dependent methods and their diversities were compared. The bacteria obtained from culture-independent method belong to Proteobacteria (95.8%), Actinobacteria (2.1%), and Firmicutes (2.1%) and fungi belong to Ascomycota (65.2%) and Basidiomycota (34.8%). Very few overlapped microbial species were found by culture-dependent and -independent methods. Healthy leaves display higher bacterial diversity than diseased leaves. Phylogenetic structures are very different between healthy and diseased phyllosphere microbial communities. Bacteria close to Acinetobacter and Pseudomonas were dominant on healthy leaves, whereas those close to Shigella were dominant on diseased leaves. 52.9% of fungal clones from healthy leaves were Ustilaginomycetes, close to Rhodotorula phylloplana and uncultured basidomycete; by contrast, 60% of clones from diseased leaves were Lecanoromycetes, close to Umbilicaria muehlenbergii. No bacteria but four fungal strains phylogenetically close to Myrothecium sp. and Alternaria alternate were pathogenic to seedlings and detached leaves of the invasive plant. Therefore, this plant may be resistant to pathogens from bacteria but not fungi in its introduced range.

      • Networking issues in medical implant communications

        IEEE 802.15 working group established a new task group, body area network (TG-BAN), to develop short range wireless technology in and around human body recently. This paper investigates networking issues in implant communications at 2.4 GHz frequency band. The object is better understanding of medical implant networks and how and where to start TG-BAN work. We applied IEEE 802.15.4b and 802.15.4a-chirp spread spectrum (CSS) for implant communications. We derived a path loss model in the layered body and found two unique issues: clear channel assessment (CCA) of implant devices, interference from neighbor piconet in adjacent channel, and multihop long distance communication. All of them can be attributed to the rapid attenuation of electromagnetic wave through lossy tissue, which is about 30 dB more than the free space propagation. The carrier sense multiple access mechanism and transmit mask of 802.15.4b cannot be directly adopted. The modulation of 802.15.4a-CSS is robust to the interference from adjacent channel. A time division based MAC protocol should be considered by TG-BAN. Instead of routing among implant devices, a simple two-hop protocol which uses a body surface forwarder was presented and verified for long distance implant communications.

      • Benefit of Post-mastectomy Radiotherapy of the Supra-/infraclavicular Lymphatic Drainage Area in Breast Cancer Patients

        Background: This study investigated the survival benefit of radiotherapy (RT) of the supra- and infraclavicular lymphatic drainage area in Chinese women with T1-2N1M0 breast cancer receiving mastectomy. Methods: A total of 593 cases were retrospectively reviewed from 1998 to 2007. The relationship between supra- or infraclavicular fossa relapse (SCFR) and post-operative RT at the supra-/infraclavicular lymphatic drainage area was evaluated. Results: The majority of patients (532/593; 89. 8%) received no RT while 61 patients received RT. The median follow-up was 85 months. Among patients without RT, 54 (10. 2%) developed recurrence in the chest wall or ipsilateral SCFR. However, none of the 61 patients who underwent RT demonstrated SCFR. One patient who received RT (1. 6%) experienced recurrence in the chest wall. Univariate analysis revealed that age and molecular subtype (both P < 0. 05) were two prognostic factors related to supraclavicular and infraclavicular fossa relapse-free survival (SFRFS). Multivariate analysis revealed that only Her-2 positive status (P = 0. 011) was an independent predictor of SFRFS. RT had no influence on distant metastasis (P = 0. 328) or overall survival (P = 0. 541). SCFR significantly affected probability of distant metastasis (P < 0. 001) and overall survival (P < 0. 001). Conclusion: Although RT was not significantly associated with SFRFS, postoperative RT was significantly associated with a lower locoregional (i. e., supraclavicular/infraclavicular and chest wall) recurrence rate. SCFR significantly influenced distant metastasis-free survival, which significantly influenced the overall survival of T1-2N1M0 breast cancer patients after mastectomy. Thus, prophylactic RT is recommended in T1-2N1M0 breast cancer patients, especially those who have Her-2 positive lesions.

        Transcriptional regulation of Glis2 in hepatic fibrosis

        The role of Gli-similar 2 (Glis2) in hepatic fibrosis (HF) is controversial. In this study, we focused on the functional and molecular mechanisms involved in the Glis2-mediated activation of hepatic stellate cells (HSCs)—a milestone event leading to HF. The expression levels of Glis2 mRNA and protein were significantly decreased in the liver tissues of patients with severe HF and in mouse fibrotic liver tissues as well as HSCs activated by TGFβ1. Functional studies indicated that upregulated Glis2 significantly inhibited HSC activation and alleviated BDL-induced HF in mice. Downregulation of Glis2 was found to correlate significantly with DNA methylation of the Glis2 promoter mediated by methyltransferase 1 (DNMT1), which restricted the binding of hepatic nuclear factor 1-α (HNF1-α), a liver-specific transcription factor, to Glis2 promoters. In addition, the enrichment of DNMT1 in the Glis2 promoter region was mediated by metastasis-associated lung adenocarcinoma transcriptor-1 (MALAT1) lncRNA, leading to transcriptional silencing of Glis2 and activation of HSCs. In conclusion, our findings reveal that the upregulation of Glis2 can maintain the resting state of HSCs. The decreased expression of Glis2 under pathological conditions may lead to the occurrence and development of HF with the expression silencing of DNA methylation mediated by MALAT1 and DNMT1.
