        A Hypoxia-Induced SCFFBXL1 E3 Ligase Ubiquitinates and Degrades the MEN1 Tumor Suppressor to Promote Colorectal Cancer Tumorigenesis

        Purpose Emerging evidence has shown that SKP1-cullin-1-F-box-protein (SCF) E3 ligases contribute to the pathogenesis of different cancers by mediating the ubiquitination and degradation of tumor suppressors. However, the functions of SCF E3 ligases in the pathogenesis of colorectal cancer (CRC) remain obscure. Materials and Methods The cancerous and adjacent noncancerous tissues from CRC patients were collected, and protein levels were analyzed. Lentiviral short hairpin RNA (shRNA) and plasmid transfection were used to knock down and overexpress gene expression in CRC cell lines. Immunoprecipitation (IP), mass spectrometry, and co-IP analyses were used to determine protein interactions and the assembly of the SCF complex. Cell proliferation, migration, and tumor xenograft assays were performed to examine the effects of SCF members on CRC cell growth in vitro and in vivo. Results Hypoxia activated the docking of hypoxia-inducible factor 1α (HIF1α) onto the CUL1 promoter and induced CUL1 expression in CRC cells. CUL1 coupled with RBX1, SKP1, and FBXL1 to assemble the SCFFBXL1 complex in CRC biopsies and cells. The SCFFBXL1 E3 ligase specifically ubiquitinated and degraded the MEN1 tumor suppressor. Knockdown of HIF1α or SCFFBXL1 members, or blockage of SCFFBXL1 by two inhibitors (DT204 and SZLP1-41) caused the accumulation of MEN1 protein and led to a significant decrease in cell proliferation and migration in vitro and tumor growth in vivo. Conclusion The SCFFBXL1 E3 ligase is required for the ubiquitination of MEN1, and disruption of this complex may represent a new therapeutic strategy for the treatment of CRC.         A Controllable LCL-T Resonant AC/DC Converter for High Frequency Power Distribution Systems

        High frequency alternating current (HFAC) has been widely used in a wide range of power distribution systems (PDS) due to its superior performance. A high frequency AC/DC converter plays the role of converting HFAC voltage to DC voltage. In this paper, a new LCL-T resonant AC/DC converter has been proposed, and an easier control method based on input voltage comparison is presented, without the complicated calculation of the zero-crossing point. Both a low distortion and near-to-unity power factor can be achieved by the proposed resonant converter and control strategy. The operational principle and steady-state analysis are given for the proposed resonant converter. A simulation model and experimental prototype are implemented with an operation frequency of 25kHz and a rated power of 20W. The simulation and experimental results verify the accuracy of the analysis and the excellent performance of the proposed topology.

      • Stable Tetrabenzo-Chichibabin’s Hydrocarbons: Tunable Ground State and Unusual Transition between Their Closed-Shell and Open-Shell Resonance Forms

        <P>Stable open-shell polycyclic aromatic hydrocarbons (PAHs) are of fundamental interest due to their unique electronic, optical, and magnetic properties and promising applications in materials sciences. Chichibabin’s hydrocarbon as a classical open-shell PAH has been investigated for a long time. However, most of the studies are complicated by their inherent high reactivity. In this work, two new stable benzannulated Chichibabin’s hydrocarbons <B>1-CS</B> and <B>2-OS</B> were prepared, and their electronic structure and geometry in the ground state were studied by various experiments (steady-state and transient absorption spectra, NMR, electron spin resonance (ESR), superconducting quantum interference device (SQUID), FT Raman, X-ray crystallographic etc.) and density function theory (DFT) calculations. <B>1-CS</B> and <B>2-OS</B> exhibited tunable ground states, with a closed-shell quinoidal structure for <B>1-CS</B> and an open-shell biradical form for <B>2-OS</B>. Their corresponding excited-state forms <B>1-OS</B> and <B>2-CS</B> were also chemically approached and showed different decay processes. The biradical <B>1-OS</B> displayed an unusually slow decay to the ground state (<B>1-CS</B>) due to a large energy barrier (95 ± 2.5 kJ/mol) arising from severe steric hindrance during the transition from an orthogonal biradical form to a butterfly-like quinoidal form. The quick transition from the quinoidal <B>2-CS</B> (excited state) to the orthogonal biradicaloid <B>2-OS</B> (ground state) happened during the attempted synthesis of <B>2-CS</B>. Compounds <B>1-CS</B> and <B>2-OS</B> can be oxidized into stable dications by FeCl<SUB>3</SUB> and/or concentrated H<SUB>2</SUB>SO<SUB>4</SUB>.         Structure and Properties of Low Dielectric Constant Polyetherimide Films Containing-CF3 and Cardo Groups

        Cardo containing phenoxy and -CF3 derivate diamine was synthesized, and further it was polymerized with 3,3',4,4'-biphenyl tetracarboxylic dianhydride to prepare polyetherimide containing Cardo group. The transmittance of the prepared polymer increased from 82% to 88% and the dielectric constant decreased from 2.91 to 2.83 with a slight decline in thermal and mechanical performance when -CF3 was introduced. The molecular simulation in terms of charge distribution, electric potential energy, chain stiffness and aggregation state revealed that -CF3 could restrain charge transfer and decrease the unit dipole moment and polarization for its larger volume. The optical and dielectric performance was significantly enhanced, but the thermal and mechanical performance decreased slightly when the -CF3 group was introduced. The glass transition temperature of the polymer declined from 329℃ to 311℃. The investigation conducted in this paper can offer valuable reference to the optimization and development of synthesis and comprehensive performance of polyimide.

        Excessive retinoic acid inhibit mouse embryonic palate mesenchymal cell growth through involvement of Smad signaling

        All-trans retinoic acid (atRA), the oxidative metabolite of retinoic acid (RA), is essential for palatogenesis. Overdose RA is capable of inducing cleft palate in mice and humans. Normal embryonic palatal mesenchymal (EPM) cell growth is crucial for shelf growth. Smad signaling is involved in many biological processes. However, it is not much clear if atRA could affect Smad signaling during EPM cells growth. In this study, the timed pregnant mice with maternal administration of 100 mg/kg body weight of RA by gastric intubation were cervical dislocation executed to evaluate growth changes of palatal shelves by hematoxylin and eosin (H&E) staining. At the same time, a primary mouse EPM (MEPM) cell culture model was also established. MEPM cells were treated with atRA (0.1, 0.5, 1, 5 and 10 μM) for 24, 48 and 72 h. The results indicated that the sizes of the shelves were smaller than those in control. AtRA inhibited MEPM cell growth with both increasing concentration and increasing incubation time, especially at 72 h in vitro. Moreover, atRA significantly increased the mRNA and protein expression levels of Smad7 (P < .05), but the mRNA and protein expression levels of PCNA were reduced (P < .05). We also found atRA inhibited phosphorylation of Smad2 compared with untreated group (P < .05). However, the protein and mRNA levels of Smad2 did not change both in atRA-treated and untreated group (P > .05). We demonstrated that RA induced inhibition of MEPM cell growth that could cause cleft palate partly by down-regulation of Smad pathway.
