        Simultaneous delivery of DNA vaccine and hydrophobic adjuvant using reducible polyethylenimine-functionalized graphene oxide for activation of dendritic cells

        The activation and maturation of dendritic cells are critical in immunotherapy, and the potency of DCs isassociated with high levels of antigen presentation on major histocompatibility complex (MHC) class Iand II and the expression of costimulatory signals. Graphene oxide (GO) and its derivatives have manyexcellent physicochemical properties, including large and hydrophobic surface available for interactingwith hydrophobic or aromatic drugs via p–p stacking forces,flexibility of the chemical modification onthe surfaces, and capacity of DC activation. In this study, we designed a simple strategy to achieve the codeliveryof a DNA vaccine and hydrophobic immune adjuvant (R848) and to enhance the adjuvanticity ofR848 via the synergistic effect of GO and R848. Thiolated low-molecular-weight polyethylenimine(TPEI1.8) was crosslinked with 4-aminothiophenol-modified GO (TGO) via the formation of disulfidebonds. Thus, TGO with its assembled TPEI1.8 could not only load R848 but also electrostatically interactwith the DNA vaccine. Owing to the reducibility of the disulfide bond in the cellular environment, theDNA vaccine could be readily released. This system can significantly enhance the DNA transfection, theexpression of the costimulatory signal, and the level of antigen presentation to MHC class I DCs for theiractivation and maturation.

        A New Micromechanical Criterion for Ductile Fracture of G20Mn5QT Cast Steels Under Shear Stress

        A new micromechanical criterion was proposed for ductile fracture caused by shear in low stress triaxiality regions. The infl uences of both the stress triaxiality and the Lode parameter were reasonably considered. The formula of the critical plastic strain for ductile fracture by shear was deduced based on the idea that the evolution of microvoids in materials under shear stresses is a successive procedure of nucleation, growth, elongation and collapse. Six shear specimens of G20Mn5QT cast steels were tested under monotonic tension. Three confi gurations of these specimens provided diff erent combinations of the stress triaxiality and the Lode parameter. The critical void compression ratio for the void collapse was determined based on unit cell analysis and the shape of the dimples on the fracture surfaces of the test specimens. Parameters in the proposed criterion were then calibrated for G20Mn5QT cast steels based on tests and complementary fi nite element analysis of the shear specimens. Finally, diff erent fracture mechanisms of G20Mn5QT cast steels were discussed.

        Ductile Fracture Analysis of Welded Hollow Spherical Joints Subjecting Axial Forces with Micromechanical Fracture Models

        Two micromechanical fracture models, void growth model (VGM) and stress modifi ed critical strain (SMCS) model, were adopted to distinguish the failure mechanism of welded hollow spherical (WHS) joints under axial load based on FE analysis. Ductile fracture was successfully predicted for WHS joints under axial tension. The predicted fracture location is at the weld toe between WHS joints and circular hollow section members, which is consistent with corresponding test results. The predicted fracture load is lower than the peak load on the load–displacement curve, which indicates that the failure mechanism of WHS joints under axial tension is fracture due to inadequate strength and the fracture load should be taken as the ultimate load bearing capacity of the joints. A simplifi ed SMCS model was proposed and verifi ed for ductile fracture prediction of WHS joints under axial tension. Micromechanical fracture analysis was also conducted on WHS joints under axial compression. It was obtained by both VGM and SMCS model that no fracture would occur before the load reached its peak value, the reason of which was discussed by tracing the variation of the equivalent plastic stain and stress triaxiality at the potential location of fracture. Therefore, the failure mechanism of WHS joints under axial compression is losing stability with the depression of the sphere cap and the peak load on the load–displacement curve should be taken as the ultimate load bearing capacity of WHS joints.

        RBM24 exacerbates bladder cancer progression by forming a Runx1t1/TCF4/miR-625-5p feedback loop

        RNA–binding motif protein 24 (RBM24) acts as a multifunctional determinant of cell fate, proliferation, apoptosis, and differentiation during development by regulating premRNA splicing and mRNA stability. It is also implicated in carcinogenesis, but the functions of RBM24 in bladder cancer (BC) remain unclear. In the present study, we revealed that RBM24 was upregulated in BC tissues. Importantly, we found that a higher level of RBM24 was correlated with poor prognosis in BC patients. Overexpression of RBM24 promoted BC cell proliferation, while depletion of RBM24 inhibited BC cell proliferation in vivo and in vitro. Mechanistically, RBM24 positively regulated Runx1t1 expression in BC cells by binding to and enhancing Runx1t1 mRNA stability. Furthermore, Runx1t1 in turn promoted RBM24 expression by interacting with the transcription factor TCF4 and suppressing the transcription of miR-625-5p, which directly targets RBM24 and suppresses RBM24 expression. RBM24-regulated BC cell proliferation was moderated via the Runx1t1/TCF4/miR-625-5p feedback loop. These results indicate that the RBM24/Runx1t1/TCF4/miR-625-5p positive feedback loop participates in BC progression. Disruption of this pathway may be a potential therapeutic strategy for BC treatment.

      • THE IMPACT OF ENDORSER ETHNICITY AND PORTRAYAL ON CHINESE WOMEN’S ATTITUDE TOWARD LUXURY ADVERTISING

        This study aims to examine the impact of endorser ethnicity and portrayal on consumers’ attitude toward luxury ad and brand. In addition, the moderating role of individual’s appearance self-esteem is examined. In the ads of luxury brands in China, it is common to have two types of endorser ethnicity (i.e., international vs. local) and two types of endorser portrayal (i.e., sexy vs. smart). Endorser ethnicity in an ad may influence consumers’ self-referencing behavior which refers to an individual’s tendency to encode communication information differently depending on the level on which the self is implicated in the information (Rogers et al., 1997). Asian consumers’ self-reference level is found to be higher when they are exposed to ads featuring an Asian model than a Western model (Martin et al., 2004). In other words, if an international luxury brand adopts a localization advertising strategy in China by using a Chinese endorser, the local consumers are more likely to generate a higher level of self-reference.

      • WORD-OF-MOUTH AND LUXURY BRAND CONSUMPTION: THE ROLE OF ATTITUDE FUNCTIONS

        This study aims to investigate the effect of word-of-mouth on the purchase of genuine and counterfeit luxury brands. It also examines the roles of attitude functions and social norm in the relationship between word-of-mouth communication and consumers’ luxury brand evaluation. A 2 (information source: advertising vs. word-of-mouth) × 2 (luxury brand: genunie vs. counterfeit) between-subjects experimental design was used to collect the data. A total of 153 adult consumers from Shanghai, China were recruited and randomly assigned to one of the four experimental conditions. Prior to the conduction of the actual experiment, a pretest of 30 respondents was conducted to determine the brands and scenarios selected for the experiments. Attitude function (social-adjustive function or value-expressive function) is measured by a self-monitoring scale. Social norm, luxury brand evaluations on genuine and counterfeit luxury brands were measured. MANOVA and ANOVA were performed to examine the proposed hypotheses. Results indicated that purchase intention for counterfeit luxury brand was moderated by subjective norm. Word-of-mouth increased the subjective norm related to the disapproval of counterfeits, compared to traditional advertising. Subjective norm was found to strongly influence consumers’ counterfeit luxury brand evaluation. The relationship between subjective norm and counterfeit consumption was positively significant. The moderating effects of self-monitoring and subjective norm on consumers’ evaluations and consumption for counterfeit luxury brands were found to significant. In short, the findings support the proposed hypotheses and showed that positive word-of-mouth was an efficient way to enhance consumers’ purchase intentions for genuine luxury brands, and also an effective means to decrease purchase intentions for counterfeit luxury brands. The results reveal that social-adjustive function and value-expressive function served by luxury brand consumption can be increased by positive word-of-mouth. Furthermore, higher levels of social-adjustive function and value-expressive function served by publicly consumed product (vs. privately consumed product) are found, indicating that product categories also affect the attitude functions served by luxury brand consumption. Managerial recommendations are provided to the marketing managers for luxury brands.

      • The effect of thiobarbituric acid on tyrosinase: inhibition kinetics and computational simulation.

        <P>Tyrosinase plays various roles in organisms and much research has focused on the regulation of tyrosinase activity. We studied the inhibitory effect of thiobarbituric acid (TBA) on tyrosinase. Our kinetic study showed that TBA inhibited tyrosinase in a reversible noncompetitive manner (K(i) 5 14.0 ± 8.5 mM and IC?????? 5 8.0 ± 1.0 mM). Intrinsic and ANS-binding fluorescences studies were also performed to gain more information regarding the binding mechanism. The results showed that no tertiary structural changes were obviously observed. For further insight, we predicted the 3D structure of tyrosinase and simulated the docking between tyrosinase and TBA. The docking simulation was successful with significant scores (binding energy for AutoDock4: -5.52 kcal/mol) and suggested that TBA was located in the active site. The 11 ns molecular dynamics simulation convinced that the four HIS residues (residue numbers: 57, 90, 250, and 282) were commonly responsible for the interaction with TBA. Our results provide a new inhibition strategy that works using an antioxidant rather than targeting the copper ions within the tyrosinase active site.</P>
