Purpose: Our previous studies have shown that kinesin family member 11 (KIF11) is markedly overexpressed in human breast cancer cells or tissues and positively correlated with distant metastasis and prognosis in patients with breast cancer, suggesting an important role in the regulation of cancer stem cells. Herein, we examined the role of KIF11 in breast cancer stem cells. Methods: In the current study, we validated our previous findings through analysis of data collected in The Cancer Genome Atlas. Endogenous KIF11 was stably silenced in MCF-7 and SKBR-3 cells. Flow cytometry was used to measure the proportion of side-population (SP) cells. Mammosphere culture and tumor implantation experiments in immunodeficient mice were used to assess the self-renewal ability of breast cancer cells. Real-time polymerase chain reaction, western blot, immunofluorescence staining, luciferase reporter assays and Wnt agonist treatment were conducted to investigate the signaling pathways regulated by KIF11. Results: We found that the expression level of KIF11 was positively correlated with stem cell-enrichment genes. The proportion of SP cells was significantly reduced in KIF11-silenced cells. Silencing endogenous KIF11 not only reduced the size and number of mammospheres in vitro, but also reduced the ability of breast cancer cells to form tumors in mice. Simultaneously, we found that KIF11 was involved in regulating the activation of the Wnt/β-catenin signaling pathway. Conclusion: Endogenous KIF11 enhances the self-renewal of breast cancer cells by activating the Wnt/β-catenin signaling pathway, thereby enhancing the characteristics of breast cancer stem cells.

This paper presents a nontraditional laser power system in which an active compensated pulsed alternator (ACPA) drives a flashlamp directly without the use of capacitor groups. As a result, the volume of the laser system is decreased because of the high energy density of the ACPA. However, the difficulty in matching the output of the alternator with the laser flashlamp is a significant issue and needs to be well analyzed. In order to solve this problem, based on the theory for ACPA, the authors propose a simplified model for the system of ACPA with flashlamp load by the way of circuit simulation. The simulation results preliminarily illuminate how the performance of the ACPA laser power system is affected. Meanwhile, the simulation results can also supply a consultation for future ACPA laser power system design and control.
Enhanced Morris for the extraction of significant parameters in high-dimensional design optimization

The traditional Morris method cannot ensure the consistent extraction of significant parameters because its random sampling strategy often leads to an improper coverage of the input space, especially when the number of sample points is small. To overcome the drawbacks, we developed an improved sampling strategy to enhance the performance of Morris method based on Latin hypercube sampling (LHS) with the idea of Central composite design (CCD) and a fluctuant step to generate a more uniform points set. A comparison on the results of a numerical example obtained by the enhanced Morris, the traditional methods and CCS-Morris demonstrated that the former had a much better performance. The application of the enhanced Morris method in the high-dimensional optimization of a compressor shell with 15 design parameters aimed at increasing its natural frequencies and keeping its mass relatively light demonstrates that the method is effective and applicable in engineering design.

A partitioned fluid-structure interaction (FSI) solving strategy that depends on problem characteristics is applied to quantitatively obtain the coupling effects of a fluid-structure system in a single-blade centrifugal pump on the unsteady flow. A two-way coupling method is employed to realize strong FSI effects in the calculation procedure. The successful impeller oscillation measurement using two proximity sensors validated the FSI simulation accuracy in a complicated and practical fluid-structure system having a rotating component. The results show that the hydrodynamic force deviation can be observed in the results for the coupled versus uncoupled cases. Additionally,the coupled unsteady pressure is larger than the uncoupled value for every monitoring point at every impeller rotation position. Comparison results for different monitoring points under an overload condition and partial-load condition display the same regularities. To some extent, this interaction mechanism would affect the accuracy and reliability of the unsteady flow and rotor deflection analysis.

The aim of this study was to compare the functional characteristics of mesenchymal stromal cells (MSCs) from a sheep model of traumatic temporomandibular joint (TMJ) fibrous and bony ankylosis. A sheep model of bilateral TMJ trauma-induced fibrous ankylosis on one side and bony ankylosis on the contralateral side was used. MSCs from fibrous ankylosed callus (FAMSCs) or bony ankylosed callus (BA-MSCs) at weeks 1, 2, 4, and 8 after surgery were isolated and cultured. MSCs derived from the bone marrow of the mandibular condyle (BM-MSCs) were used as controls. The MSCs from the different sources were characterized morphologically, phenotypically, and functionally. Adherence and trilineage differentiation potential were presented in the ovine MSCs. These cell populations highly positively expressed MSC-associated specific markers, namely CD29, CD44, and CD166, but lacked CD31 and CD45 expressions. The BA-MSCs had higher clonogenic and proliferative potentials than the FA-MSCs. The BA-MSCs also showed higher osteogenic and chondrogenic potentials, but lower adipogenic capacity than the FA-MSCs. In addition, the BA-MSCs demonstrated higher chondrogenic, but lower osteogenic capacity than the BM-MSCs. Our study suggests that inhibition of the osteogenic and chondrogenic differentiations of MSCs might be a promising strategy for preventing bony ankylosis in the future.

The core of Confuianism is Inhak(An Exposition of Benevolence 仁學). This paper seeks to trace Tasan`s view of Inhak by comparing the views of Inhak of the Song-Ming confuianists. Through restoration of Confuianism, the Inhak of Confucius is re-established by Tasan in the moral-based confucianism from the Song-Ming confuianist`s nature-based confucianism. Tasan`s view of Inhak is not a plain theoretical criticism on the view of Inhak of Song Confucianism, but it is a active criticism which stems from a strong critical mind to solve the issues of the time. And the attitude towards the Song-Ming confuianists are an open criticism which is succeeding and at the same time criticizing their theoretical results. However, Tasan`s view of Inhak is connected to his theory of human nature and mandate(性命論). And the theory is based on Tasan`s religious view on heaven. His view of Inhak may be explained in reality on the ground of morality and but also on the background of transcendental, external and absolute `heaven as a supreme ruler`(上帝天). Tasan`s view of Inhak speaks for the strong spirit of the time and the regional characteristics of 18-19 century of the Chosun Dynasty(朝鮮). It is also a notice of the fusion of east and west thoughts of Confucianism and Christianity. The reasons for Tasan to break from the world view and the view of man of Neo-confuciaunism of Song Dynasty and reinterpret the Six Chinese Classics(六經) and the Four Books of Ancient China(四書) based on new world views and the view of man are: first, the social, economical, political realities of Chosun Dynasty at the time; second, the doctrinal and ideological trend of Neo-confucianism of Song Dynasty; third, the weakness of the theory of Neo-confucianism of Song Dynasty itself on the social and economical issues; fourth, the effect of Catholics which he got very much interested in, in his late teens and the effect of the newly imported western scientific thoughts.
Enhancing the Anaerobic Digestion of Corn Stalks Using Composite Microbial Pretreatment

A composite microbial system (XDC-2) was used to pretreat and hydrolyze corn stalk to enhance anaerobic digestion. The results of pretreatment indicated that sCOD concentrations of hydrolysate were highest (8,233 mg/l) at the fifth day. XDC-2 efficiently degraded the corn stalk by nearly 45%, decreasing the cellulose content by 22.7% and the hemicellulose content by 74.1%. Total levels of volatile products peaked on the fifth day. The six major compounds present were ethanol (0.29 g/l), acetic acid (0.55 g/l), 1,2-ethanediol (0.49 g/l), propionic acid (0.15 g/l), butyric acid (0.22 g/l), and glycerine (2.48 g/l). The results of anaerobic digestion showed that corn stalks treated by XDC-2 produced 68.3% more total biogas and 87.9% more total methane than untreated controls. The technical digestion time for the treated corn stalks was 35.7% shorter than without treatment. The composite microbial system pretreatment could be a cost-effective and environmentally friendly microbial method for efficient biological conversion of corn stalk into bioenergy.
