Rapid screening of roundup ready soybean in food samples by a hand-held PCR device

Insulated isothermal PCR (iiPCR) method was recently available for rapid on-site detection ofroundup ready soybean (RRS; event GTS40-3-2) in food materials and products. Performance of thismethod was evaluated in this study. The 100% detection endpoint for the RRS by iiPCR was found insamples containing 0.1% RRS, equivalent to the results of the reference real-time PCR (rtPCR). Analysisof nucleic acids of soybean-based processed food products indicated 95% agreement between the iiPCRand rtPCR for RRS detection. By testing soybean milk and tofu samples using simple pretreatmentmethods, we found that the agreements between iiPCR and rtPCR methods of the aforementionedsamples were 80% and 90%, respectively. Replicated tests of all discrepant samples implied that thesesamples had trace amounts of RRS, suggesting that the iiPCR system is more sensitive than the rtPCRmethod. In conclusion, the iiPCR technology can be a useful point-of-need tool to help make a timelydecision in the consumption of genetically modified organisms.

A structure-controlled model for hot spring exploration in Taiwan by remote sensing

Hot Spring Law of Taiwan was passed in legislativeassembly on 3 June 2003. Hot springs would become one of themost important natural resources for recreation purposes. Bothpublic and private sectors wil invest large amount of capital inthis area in the near future. The value of remote sensing technol-ogy is to give a critical tool for observing the landscape to find outmega-scaled geological structures, which may not be able to befound by conventional approaches. The occurrences of the hotsprings in Taiwan are mostly in metamorphic and sedimentaryrocks, other than in volcanic environments. Local geothermalanomaly or heat of springs transfer by liquid convection otherthan conduction or radiation. The deeply-seated fractures of hardrocks are the conduit of the convection of hot water, which couldbe as deep as 3000 meters in a hypothetical model of Taiwan.Clues to find outcrops of hot spring can be obtained by a struc-ture-controlled model deduced by geological lineaments observedby satellite images and stereoscopic interpretation of aerial pho-tographs. A case study conducted in Eastern Taiwan wil be dem-onstrated.

Ulcerative colitis (UC) is an inflammatory bowel disease characterized by chronic mucosal inflammation of the colon, and the prevalence and incidence of UC have been steadily increasing in Taiwan. A steering committee was established by the Taiwan Society of Inflammatory Bowel Disease to formulate statements on the diagnosis and management of UC taking into account currently available evidence and the expert opinion of the committee. Accurate diagnosis of UC requires thorough clinical, endoscopic, and histological assessment and careful exclusion of differential diagnoses, particularly infectious colitis. The goals of UC therapy are to induce and maintain remission, reduce the risk of complications, and improve quality of life. As outlined in the recommended treatment algorithm, choice of treatment is dictated by severity, extent, and course of disease. Patients should be evaluated for hepatitis B virus and tuberculosis infection prior to immunosuppressive treatment, especially with steroids and biologic agents, and should be regularly monitored for reactivation of latent infection. These consensus statements are also based on current local evidence with consideration of factors, and could be serve as concise and practical guidelines for supporting clinicians in the management of UC in Taiwan. Crohn`s disease (CD) is a chronic relapsing and remitting inflammatory disease of the gastrointestinal tract. CD is rare in Taiwan and other Asian countries, but its prevalence and incidence have been steadily increasing. A steering committee was established by the Taiwan Society of Inflammatory Bowel Disease to formulate statements on the diagnosis and management of CD taking into account currently available evidence and the expert opinion of the committee. Thorough clinical, endoscopic, and histological assessments are required for accurate diagnosis of CD. Computed tomography and magnetic resonance imaging are complementary to endoscopic evaluation for disease staging and detecting complications. The goals of CD management are to induce and maintain remission, reduce the risk of complications, and improve quality of life. Corticosteroids are the mainstay for inducing re-mission. Immunomodulating and biologic therapies should be used to maintain remission. Patients should be evaluated for hepatitis B virus and tuberculosis infection prior to treatment and receive regular surveillance for cancer. These consensus statements are based on current local evidence with consideration of factors, and could be serve as concise and practical guidelines for supporting clinicians in the management of patients with CD in Taiwan. Cis-3-O-p-hydroxycinnamoyl ursolic acid (HCUA), a triterpenoid compound, was purified from Elaeagnus oldhamii Maxim. This traditional medicinal plant has been used for treating rheumatoid arthritis and lung disorders as well as for its anti-inflammation and anticancer activities. This study aimed to investigate the anti-proliferative and apoptotic-inducing activities of HCUA in oral cancer cells. HCUA exhibited anti-proliferative activity in oral cancer cell lines (Ca9-22 and SAS cells), but not in normal oral fibroblasts. The inhibitory concentration of HCUA that resulted in 50% viability was 24.0 μM and 17.8 μM for Ca9-22 and SAS cells, respectively. Moreover, HCUA increased the number of cells in the sub-G1 arrest phase and apoptosis in a concentrationdependent manner in both oral cancer cell lines, but not in normal oral fibroblasts. Importantly, HCUA induced p53-mediated transcriptional regulation of pro-apoptotic proteins (Bax, Bak, Bim, Noxa, and PUMA), which are associated with mitochondrial apoptosis in oral cancer cells via the loss of mitochondrial membrane potential. HCUA triggered the production of intracellular reactive oxygen species (ROS) that was ascertained to be involved in HCUA-induced apoptosis by the ROS inhibitors YCG063 and N-acetyl-L-cysteine. As a result, HCUA had potential antitumor activity to oral cancer cells through eliciting ROS-dependent and p53-mediated mitochondrial apoptosis. Overall, HCUA could be applicable for the development of anticancer agents against human oral cancer.
A Simple Partial Discharge Detector for Low-Voltage Rotating Electrical Machines

A new, simple partial discharge (PD) detector for low-voltage rotating electrical machines using acoustical emission (AE) technique is dseveloped in this study. Common electric components were used in the detector, which reduces the cost of inspection, comparing with the traditional PD detection methods that use expensive equipment costing from tens of thousands to several millions dollars. Experimental results by resonant type AE sensors (150 ㎑) which utilize power line-cycle in a microcontroller unit (MCU) as reference to measure the PDs generated in a low-voltage motor are presented. The AE signals are then amplified by a pre-amplifier (30 ㎑ ~ 300 ㎑, 34 ㏈). Since the resonant frequency of the sensor is much lower than that of the electromagnetic (EM) interferences around the motor, the effects of noise is substantially reduced by this method. In the mean time, the use of 150-㎑ resonant type AE sensor also avoids the disturbance of mechanical vibration noise. According to the experiment results, the measurement system developed in this study can be used to detect the PDs’ AE signals correctly. The AE measurement scheme proposed in this study provides an effective, low-cost method for PD measurements.
