        Abnormal differentiation of regulatory T cells and Th17 cells induced by perinatal bisphenol A exposure in female offspring mice

        Background Bisphenol A (BPA) is an environmental estrogen widely exposed to human beings, and there are more studies on its reproductive toxicity, endocrine disruption and neurobehavioral disorders. Recent few studies have found that BPA has immunotoxicity, and its mechanism is not clear. Therefore, the effects of BPA on immune system have attracted extensive attention. The aim of this study was to investigate the effect and mechanism of perinatal exposure to BPA on regulatory T cells (Treg) and Th17 cells in female offspring mice. Methods Twenty-one pregnant C57BL/6 mice were randomly divided into three groups: a control group, low-dose BPA (0.2 μg/mL) and high-dose BPA (2.0 μg/mL) exposure group. All received BPA exposure via drinking water from gestational day 6 to the end of lactation. Female offspring were fed a normal diet and drinking water for 1 month. The percentages of Treg and Th17 cells, the levels of Foxp3 and RORγt protein and IL-17 and TGF-β from spleen tissue or blood were measured in female offspring. Results The percentage of Treg cells and levels of Foxp3 protein decreased, while the percentage of Th17 cells and levels of RORγt protein increased, which showed a dose–effect relationship. The levels of serum TGF-β were significantly lower and the levels of serum IL-17 were statistically higher in BPA-exposed female offspring compared with controls (P < 0.05 or P < 0.01). But there were no statistical difference in the levels of serum TGF-β and IL-17 between 0.2 μg/mL and 2.0 μg/ mL BPA groups (P > 0.05). Conclusion BPA exposure during pregnancy and lactation could cause abnormal differentiation and function of Treg and Th17 cells in female offspring mice, which was associated with down-regulated Foxp3 and up-regulated RORγt protein, respectively. Our findings indicated that BPA exposure during early development may play an important role in the development of autoimmune diseases later.

        Improvements of Metabolites Tolerance in Clostridium acetobutylicum by Micronutrient Zinc Supplementation

        Micronutrient zinc is of great importance for acetone-butanol-ethanol (ABE) fermentation by Clostridium acetobutylicum. The effect of zinc supplementation on toxic metabolites (formic, acetic, butyric acid and butanol) tolerance during ABE fermentation was investigated under various stress-shock conditions without pH control. Great improvements on cell growth, glucose utilization and butanol production were achieved. In the presence of 0.45 g/L formic acid, zinc contributed to 11.28 g/L butanol produced from 55.24 g/L glucose compared to only 5.27 g/L butanol from 29.49 g/L glucose in the control without zinc supplementation. More importantly, relatively higher levels of 7.5 g/L acetic acid, 5.5 g/L butyric acid and 18 g/L butanol could be tolerated by C. acetobutylicum with zinc supplementation while no fermentation was observed under the same stress-shock condition respectively, suggesting that the acids and butanol tolerance in C. acetobutylicum could be significantly facilitated by pleiotropic regulation of micronutrient zinc. Thus, this paper provides an efficient bioprocess engineering strategy for improving stress tolerance in Clostridium species.

        CHMP1A suppresses the growth of renal cell carcinoma cells via regulation of the PI3K/mTOR/p53 signaling pathway

        Background: CHMP1A, a member of the ESCRT-III complex family, has been indicated as a brand-new inhibitor gene of tumors. Our previous research has revealed that CHMP1A plays a vital role in the development and progression of renal cell carcinoma (RCC). Objective: To investigate the potential target pathway of the regulation of the tumor cell growth by CHMP1A. Methods: The effect of CHMP1A on mTOR pathway was elucidated by western blotting. The effect of CHMP1A on the expression of p53 was evaluated, and A498 cell growth was assessed by colony formation and MTT assays. The expression of p53 was knocked down by shRNA-p53, and the effect of CHMP1A on mTOR after knockdown of p53 was evaluated. The effect of CHMP1A on apoptosis and its relationship with MDM2 pathway were detected by western blotting and FCM. Finally, the relationship between the regulation of p53 by CHMP1A and the PI3K/mTOR pathway was detected. Results: This study showed that the mTOR pathway was suppressed significantly in CHMP1A-overexpressing A498 and 786-0 cells; moreover, the enhanced expression of p53 and the reduced proliferation were shown in CHMP1A-overexpressing A498 cells. Furthermore, CHMP1A was able to regulate the PI3K/PTEN/mTOR and MDM2/p53 pathways in order to suppress RCC. In addition, CHMP1A regulated Bax and Bcl-2 via MDM2/p53 to induce the apoptosis of tumor cells and upregulated the expression of p53 via the PI3K/mTOR pathway. Conclusions: The results convey that CHMP1A-related suppression of RCC is closely related to the PI3K/mTOR/p53 pathway.

        Position and Force Control of Bilateral Teleoperation Systems With Time-varying Delays Based on Force Estimation

        A position and force tracking control based on force estimation is proposed for bilateral teleoperation systems with time-varying delays. A time-delay state observer is employed to estimate the system state variables affected by the delays. To estimate the interaction forces effectively, a force estimation algorithm with adaptive law is designed. Based on the estimated states and forces, a P+D controller is designed to simultaneously guarantee the position and force tracking of the system. The stability and tracking performance of the closed-loop system are proved via Lyapunov functions, and the feasibility of the proposed control is verified by both simulations and experiments. The proposed control can improve the position and force tracking performance of bilateral teleoperation systems under time-varying delays. Meanwhile, it neither requires force measurement nor the bound of the derivative of the time-varying delays to be within one.

      • Polymorphisms in Genes of the De Novo Lipogenesis Pathway and Overall Survival of Hepatocellular Carcinoma Patients Undergoing Transarterial Chemoembolization

        Aberrant expression of genes in de novo lipogenesis (DNL) pathway were associated with various cancers, including hepatocellular carcinoma (HCC). Single nucleotide polymorphisms (SNPs) of DNL genes have been reported to be associated with prognosis of some malignancies. However, the effects of SNPs in DNL genes on overall survival of HCC patients receiving transarterial chemoembolization (TACE) treatment are still unknown. In present study, nine SNPs in three genes (ACLY, ACACA and FASN) in DNL pathway were genotyped using the Sequenom iPLEX genotyping system in a hospital-based cohort with 419 HCC patients treated with TACE, and their associations with HCC overall survival were evaluated by Cox proportional hazard regression analysis under three genetic models (additive, dominant and recessive). Although we did not find any significant results in total analysis (all p>0.05), our stratified data showed that SNP rs9912300 in ACLY gene was significantly associated with overall survival of HCC patients with lower AFP level and SNP rs11871275 in ACACA gene was significantly associated with overall survival of HCC patients with higher AFP level. We further identified the significant interactions between AFP level and SNP rs9912300 or rs11871275 in the joint analysis. Conclusively, our data suggest that genetic variations in genes of DNL pathway may be a potential biomarker for predicting clinical outcome of HCC patients treated with TACE.

        Thermodynamic performance analysis of a coupled transcritical and subcritical organic Rankine cycle system for waste heat recovery

        We present a novel coupled organic Rankine cycle (CORC) system driven by the low-grade waste heat, which couples a transcriticalorganic Rankine cycle with a subcritical organic Rankine cycle. Based on classical thermodynamic theory, a detailed performance analysison the novel CORC system was performed. The results show that the pressure ratio of the expander is decreased in the CORC and theselection of the working fluids becomes more flexible and abundant. With the increase of the pinch point temperature difference of theinternal heat exchanger, the net power output and thermal efficiency of the CORC all decrease. With the increase of the critical temperatureof the working fluid, the system performance of the CORC is improved. The net power output and thermal efficiency of the CORCwith isentropic working fluids are higher than those with dry working fluids.

        연구논문 : 국내외기업의 인적자원관리에 따른 경영성과 및 노사관계 분석

        본 연구는 국내 진출 외국기업과 국내기업의 인적자원관리가 기업경영성과 및 노사관계에 미치는 영향을 분석한다. 2005-2009년간의 사업체 패널자료를 사용하였고 확률효과모형(Radom Effect Model)을 활용하고 있다. 주요 분석결과는 다음과 같다. 기업의 근로자 수가 많을수록 당기매출액과 영업이익이 높 으며 외국기업의 경우 기업규모가 클수록 이직률이 낮은 것으로 나타났다. 한편, 국내기업의 경우 임금수준이 높더라도 이직률이 높게 나타나고 있는데 이는 국내기업에서의 이직률은 단순한 임금수준에 의해 결정되는 것이 아니라 업무강도나 환경 등 다양한 요소에 의해 결정되기 때문으로 파악된다. 인력계획을 실시하면 국내기업의 경우 당기매출액과 영업이익이 높아지는 것으로 분석되었다. 근로자 중 여성비율이 높을수록 당기매출액과 영업이익에는 부정적인 영향을 미친 것으로 분석되었다. 다면평가 제를 실시할 경우 대체로 매출액, 영업이익은 물론이고 노사관계에도 긍정적인 것으로 나타났다. 마지막 으로 근참법에 근거한 노사협의회가 실질적으로 유지될수록 노사관계가 원만하며 근로자들의 이직률도 낮출 수 있는 것으로 분석되었다. This study is designed to empirically analyze of the effects of human resource management on performance and labor relation in Korea. For the analysis, it uses 2005-2009 Workplace Panel Survey (WPS) data-set. Empirical methods of this thesis is Random Effect Model. The findings of this study were as follows. The company`s number of employees on the basis of company size, the more the higher current proceeds and operating profit. In the case of foreign companies, the larger company size, the lower turnover rate. On the other hand, in the case of domestic companies, even if the wage level is high turnover rate is higher. The turnover rate of domestic companies is not determined by a simple wage level, but it is assumed to be determined by the strength or environment. Working manpower system has been shown to enhance the current proceeds and operating profit in the case of domestic companies. The higher the ratio of female workers, the negative impact of the current proceeds and operating profit. Multi dimensional evaluation has been shown to enhance the current proceeds and operating profit in addition to labor relation. If the labor-management council in accordance with the LMC Act remains substantially, labor relations are amicable, and turnover rates are lower.

        말뚝 인발모형실험에 대한 유한요소해석의 적용성 평가

        본 연구에서는 말뚝의 인발저항에 대한 합리적 평가를 위하여, 다양한 지반 조건(상대밀도, 세립분 함유율)에서 말뚝의 인발거동 모사에 대한 유한요소해석의 적용성을 평가하였다. 즉, 기존의 실내모형실험 결과(You et al., 2018)를 대상으로 동일한 조건에서의 유한요소해석을 실시하였고, 유한요소해석에 이용된 해석모델의 신뢰성을 검증하였다. 또한 수치해석을 활용한 말뚝의 인발거동 모사에 대한 적정성을 평가하였다. 그 결과, 가상지반이 적용된 축대칭 해석을 이용하여 말뚝의 주면마찰력을 평가할 수 있는 것으로 확인되었다. 또한 다양한 지반 조건에 대하여 말뚝-지반 경계면의 전단저항 특성을 반영할 수 있는 축대칭 해석은 말뚝의 합리적인 인발거동 모사가 가능한 해석방법으로 활용이 가능한 것으로 분석되었다. 따라서 본 연구를 통하여 제안된 해석모델은 말뚝의 응력-변형 관계를 통한 인발거동을 적절하게 모사할 수 있는 것으로 판단되었다. This paper describes the applicability of FEA(Finite Element Analysis) to the simulation of pile pullout behavior under various soil conditions (relative density and fines content), in order to evaluate reasonably the pullout resistance of pile. That is, the results of previous research (You et al., 2018) were analyzed by FEA under the same conditions. The FEA results showed that axisymmetric analysis using virtual ground was able to evaluate the skin friction of the pile. Also, axisymmetric analysis, which can apply the shear resistance characteristics of the pile-soil interface in various soil conditions, could be used as an analytical method that can simulate a reasonable pile pullout behavior. Therefore, the analytical model proposed in this study was able to simulate appropriately the pullout behavior based on the stress-strain relationship of the pile-soil interface.

        지반조건에 따른 말뚝의 인발저항 평가를 위한 말뚝-지반 경계면 강도감소계수 고찰

        본 연구에서는 원지반의 상대밀도와 세립분 함유율의 조건에 따른 말뚝의 인발거동 특성 규명을 위하여, 유한요소해석을 수행하였다. 수치해석에서는 말뚝과 지반 경계에서의 전단거동을 원활히 모사하기 위해서 경계요소와 경계면 강도감소계수(Rinter)를 부여하였으며, 그 결과를 기존의 실험적 연구결과(You et al., 2018)와 비교함으로써 본 수치해석 방법의 신뢰성을 검증하였고, 말뚝-지반 경계면에서의 변형 특성과 함께 경계요소에 대한 Rinter값의 결정방법을 제시하였다. 해석 결과, 본 연구에서 적용된 해석모델을 이용한 수치해석은 말뚝의 인발모형실험에 의한 말뚝과 지반의 경계면 특성을 적절하게 모사하였다. 또한 제시된 Rinter의 적용에 있어서, 반드시 지반의 상대밀도와 세립분 함유율 조건을 고려해야 할 필요가 있음을 확인 하였다. This paper describes the results of finite element analysis (FEA), in order to investigate a characteristics of pile pullout behavior according to the conditions of the relative density and fines content in original ground. In the FEA, a boundary elements and strength reduction factors (Rinter) on pile-soil interface were applied to simulate appropriately the shear behavior at the pile-soil interface, and then the reliability of numerical analysis method was verified by comparison of FEA results and previous experimental research(You et al., 2018). In addition, a the deformation characteristics at the pile-soil interface and determination method of Rinter value was laid out. The results showed that the FEA, based on the analytical model applied in this study simulates appropriately the characteristics of the pile-soil interface by pullout model test of pile. In order to apply the suggested Rinter value, it is necessary to consider the condition of the relative density and the fines content in ground.
