        Isochaetomium A2, a new bis(naphthodihydropyran-4-one) with antimicrobial and immunological activities from fungus Chaetomium microcephalum

        Isochaetomium A2 (1), a new bis(naphthodihydropyran-4-one), along with chaetochromins A (2) and B(3), was isolated from the solid-state fermented rice cultureof Chaetomium microcephalum. The structure of compound1 was elucidated on the basis of 1D and 2D NMRspectral data, and the relative configuration was confirmedby CD spectrum. Compounds 1–3 possessed significantantimicrobial activity against Escherichia coli 1.044,Staphylococcus aureus 1.252, and Bacillus subtilis 1.079. Moreover, compounds 1–3 showed obvious inhibitoryeffects on mouse spleen cell proliferation with successiveIC50 values of 0.52, 0.19, and 0.24 lM.

        A novel method for identifying rotor-stator rubbing positions using the cepstrum analysis technique

        A novel method of aero-engine rubbing positions identification based on cepstrum analysis is proposed, and the transfer path characteristicswhich reflect the transfer characteristics information from rubbing points to casing measuring points are separated from the vibrationacceleration signals of casing by means of cepstrum analysis. Therefore, there is different transfer characteristics information atdifferent rubbing positions, and in view of this, twenty rubbing positions identification features from the cepstrum are extracted. A largenumber of rubbing experiments of different positions are simulated with the rotor experiment rig of aero-engine, and the characteristicanalysis of experimental samples at different rubbing positions is carried out, and the results indicate the consistency of features to thesame rubbing position and the difference of the features to the different rubbing positions. Finally, the aero-engine rubbing positionsidentification is carried out using the nearest neighbor classification method, the recognition rate reaches 100%, and the effectiveness ofthe method is full verified.

        中国清末の政治改革と明治維新

        메이지유신은 세계사의 대사건이다. 일본은 메이지유신을 통해 통일된 근대국가를 구축하여 아시아에서 일찍이 근대화를 이룩하였다. 메이지유신은 단순히 일본을 ‘一新’ 시켰을 뿐만 아니라 동아시아, 특히 중국 청말의 개혁에 지대한 영향을 미쳤다. 특히 무술변법운동의 三傑이 요시다 쇼인의 사상에 매료되었고, 이들은 무술변법운동에 메이지유신을 모델로 한 정치 개혁을 시도하였다. 본 논문에서는 이러한 시각으로 청말의 개혁자들이 중국의 정치 개혁을 구상할 때, 메이지 유신의 어떤 곳에 관심을 가지고, 또 어떻게 메이지 유신을 보고 있었는지에 대해 살펴보았다. 나아가 그들은 메이지 유신으로부터 무엇을 배워, 중국 청말의 정치 개혁에 어떻게 담았는지, 그들이 고안한 메이지 유신 모델은 중국 청말의 개혁에 어떤 의미가 있었는지에 대해 고찰하였다. (明治維新は世界史の大事件である。日本は明治維新によって統一した近代国家を構築し、アジアでいち早く近代化を成し遂げた。明治維新は単に日本を「一新」させたのみならず、東アジア、とりわけ中国清末の改革にも多大な影響を及ぼした。さて、清末の改革者たちは中国の政治改革を構想する際、明治維新のどういうところに関心を持ち、またどのように明治維新を見ていたのかを検討した。さらに、彼らは明治維新から何を学び、それを中国清末の政治改革にどのように生かしたのかなどが問題関心である。具体的には、清末の外交家、改革者黄遵憲、戊戌の変法の指導者、思想家康有為、彼の弟子、改革運動家梁啓超を中心に、彼らが「尊王攘夷」運動や幕末志士から何を学び、また、「戊戌の変法」期前後の改革の実践とどう関わったかを考察することによって、清末の政治改革と明治維新との関わりを明らかにする。) The Meiji Restoration is a major event in world history.Japan established a unified modern nation under the Meiji Restoration and was the first country to modernize in Asia.The year 2018 marks the 150th anniversary of the Meiji Restoration, which provides an opportunity to examine the significance of the Meiji Restoration from various angles. The Meiji Restoration not only “renewed” Japan, but also greatly influenced reforms in East Asia, especially in the late Qing Dynasty. In 1898, at the end of China's Qing Dynasty, the Bogu Incident was an attempt at political reform modeled after the Meiji Restoration. In other words, the reformers of the late Qing Dynasty were interested in what the Meiji Restoration was and how they learned from it and how it was used in the political reform of the late Qing Dynasty. This chapter focuses on the reform of the late Qing Dynasty, the reformer Huang Joon-hun, the leader of Bogu, the thinker Kang Yu-yu, his disciples and reformist Liang Qichao.

        Silencing of long noncoding RNA HOXA11-AS inhibits the Wnt signaling pathway via the upregulation of HOXA11 and thereby inhibits the proliferation, invasion, and self-renewal of hepatocellular carcinoma stem cells

        Hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths, but its molecular mechanisms are not yet well characterized. Long noncoding RNAs (lncRNAs) play crucial roles in tumorigenesis, including that of HCC. However, the role of homeobox A11 antisense (HOXA11-AS) in determining HCC stem cell characteristics remains to be explained; hence, this study aimed to investigate the effects of HOXA11-AS on HCC stem cell characteristics. Initially, the expression patterns of HOXA11-AS and HOXA11 in HCC tissues, cells, and stem cells were determined. HCC stem cells, successfully sorted from Hep3B and Huh7 cells, were transfected with short hairpin or overexpression plasmids for HOXA11-AS or HOXA11 overexpression and depletion, with an aim to study the influences of these mediators on the self-renewal, proliferation, migration, and tumorigenicity of HCC stem cells in vivo. Additionally, the potential relationship and the regulatory mechanisms that link HOXA11-AS, HOXA11, and the Wnt signaling pathway were explored through treatment with Dickkopf-1 (a Wnt signaling pathway inhibitor). HCC stem cells showed high expression of HOXA11-AS and low expression of HOXA11. Both HOXA11-AS silencing and HOXA11 overexpression suppressed the self-renewal, proliferation, migration, and tumorigenicity of HCC stem cells in vivo, as evidenced by the decreased expression of cancer stem cell surface markers (CD133 and CD44) and stemness-related transcription factors (Nanog, Sox2, and Oct4). Moreover, silencing HOXA11-AS inactivated the Wnt signaling pathway by decreasing the methylation level of the HOXA11 promoter, thereby inhibiting HCC stem cell characteristics. Collectively, this study suggested that HOXA11-AS silencing exerts an antitumor effect, suppressing HCC development via Wnt signaling pathway inactivation by decreasing the methylation level of the HOXA11 promoter.

      • Luciferase Assay to Screen Tumour-specific Promoters in Lung Cancer

        Objective: Specific promoters could improve efficiency and ensure the safety of gene therapy. The aim of our study was to screen examples for lung cancer. Methods: The firefly luciferase gene was used as a reporter, and promoters based on serum markers of lung cancer were cloned. The activity and specificity of seven promoters, comprising CEACAM5 (carcinoembryonic antigen, CEA), GRP (Gastrin-Releasing Peptide), KRT19 (cytokeratin 19, KRT), SFTPB (surfactant protein B, SP-B), SERPINB3 (Squamous Cell Carcinoma Antigen, SCCA), SELP (Selectin P, Granule Membrane Protein 140kDa, Antigen CD62, GMP) and DKK1 (Dickkopf-1) promoters were compared in lung cancer cells to obtain cancer-specific examples with strong activity. Results: The CEACAM5, DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB promoters were cloned. Furthermore, we successfully constructed recombinant vector pGL-CEACAM5 (DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB) contained the target gene. After cells were transfectedwith recombinant plasmids, we found that the order of promoter activity from high to low was SERPINB3, DKK1, SFTPB, KRT19, CEACAM5, SELP and GRP and the order for promoters regarding specificity and high potential were SERPINB3, DKK1, SELP, SFTPB, CEACAM5, KRT19 and GRP. Conclusion: The approach adopted is feasible to screen for new tumour specific promoters with biomarkers. In addition, the screened lung-specific promoters might have potential for use in lung cancer targeted gene therapy research.

      • Naphthazarin Derivatives : Synthesis, Cytotoxic Mechanism and Evaluation of Antitumor Activity

        The rate of the GSH conjugate formation, the inhibition of DNA topoisomerse-Ⅰ and the cytotoxic activity against L1210 cels of the naphtoquinones showed the same order; 5,8-dimethoxy-1,4-naphthoquinone (DMNQ)>6-(1-hudroxyethyl)-DMNQ>2-(1-hydroxyethyl)-DMNQ; the steric hindrance of the substituents, particularly 2-substutent, in reacting with cellular nucleophiles must be the main cause for lowering the bioactivities. Acetylation of 2-(1-hydroxyethyl)-DMNQ producing 2-(acetyloxyethyl)-DMNQ potentiated the bioactivities; 2-(1-hydroxyethyl)-DMNQ did not react with GSH and the enzyme, and showed ED_50 of 0.680 ㎍/㎖, whereas the values of 2-(1-acetyloxyethyl)-DMNQ were the conjugate formation of 0.14μM, IC_50 value of 81 μM for the enzyme inhibition and ED_50 of 0.146 ㎍/㎖ for the cytotoxcity. Furthemore, the acetylation 2-(1-hydroxyethyl)-DMNQ (T/C, 119%) enhaced the T/C values for the mice bearing S-180 tumor (T/C of 2-(1-acetyloxyethyl)-DMNQ, 276%]. It was assumed that the difference in bbioactivities ensued by acetylation was based on the mechanism of the so-called bioreductive alkylation.
