        Tip60 and p300 function antagonistically in the epigenetic regulation of HPV18 E6/E7 genes in cervical cancer HeLa cells

        Background High-risk HPV is a causative factor of cervical cancer. HPV DNA fragments integrate into host genome resulting in the constitutive expression of HPV genes E6 and E7 under the regulation of transcription factors, such as p300 and Tip60. Interestingly, Tip60, a factor with HAT (histone acetyl transferase) activity, represses HPV18 E6/E7 genes while another HAT p300 activates the transcription of HPV18 E6/E7. Objective To explore the mechanism for the opposite roles of Tip60 and p300 in the virus gene regulation, and the influence of Tip60 and p300 in histone modifications in the regulatory sequence of HPV18 genes. Methods Tip60 or p300 was either knocked down or overexpressed in HeLa cells. The effects on HPV E6E7 expression were determined with RT-qPCR. The association of RNA polymerase II and the enrichment of acetylated or methylated histones in HPV promoter region were measured by ChIP assays with specific antibodies. Results ChIP results showed that Tip60 and p300 differently affected the modifications of histone H3K9 and the deposition of nucleosomes in HPV18 long control region (LCR). HPV18 LCR in HeLa cells is bivalent chromatin carrying both the active histone H3K9 acetylation mark and the repressive histone H3K9 trimethylation mark, the balance is maintained by Tip60 and p300. Conclusion(s) Based on the roles of Tip60 and p300 in HPV gene regulation, chemical compounds targeting Tip60 or p300 are potential anti-cervical cancer drugs.

        Object Classification based on Weakly Supervised E2LSH and Saliency map Weighting

        The most popular approach in object classification is based on the bag of visual-words model, which has several fundamental problems that restricting the performance of this method, such as low time efficiency, the synonym and polysemy of visual words, and the lack of spatial information between visual words. In view of this, an object classification based on weakly supervised E2LSH and saliency map weighting is proposed. Firstly, E2LSH (Exact Euclidean Locality Sensitive Hashing) is employed to generate a group of weakly randomized visual dictionary by clustering SIFT features of the training dataset, and the selecting process of hash functions is effectively supervised inspired by the random forest ideas to reduce the randomcity of E2LSH. Secondly, graph-based visual saliency (GBVS) algorithm is applied to detect the saliency map of different images and weight the visual words according to the saliency prior. Finally, saliency map weighted visual language model is carried out to accomplish object classification. Experimental results datasets of Pascal 2007 and Caltech-256 indicate that the distinguishability of objects is effectively improved and our method is superior to the state-of-the-art object classification methods.

        Measurement of unsteady transition on a pitching airfoil using dynamic pressure sensors

        Unsteady boundary layer transition on a pitching OA209 airfoil in a wind tunnel was detected by using pressure fluctuation measurement at different oscillation frequency. Thirty Kulite dynamic pressure transducers flush-mounted on the airfoil surface recorded pressure signatures, and root mean square of pressure signatures were calculated. Results indicated that the criterion of transition for static airfoil defined as the peak of root mean square of pressure fluctuation was still suitable for detection of transition on a pitching airfoil. Fixed transition experiment for pitching airfoil was performed to validate the conclusion. Effect of oscillation frequency on transition was investigated. For small reduced frequency, the hysteresis loop is larger near leading edge. With increasing in the oscillation frequencies, the transition was promoted and relaminarization was enhanced.

        Beta-defensin gene (DEFB1) polymorphisms are associated with the susceptibility to chronic respiratory diseases

        In the present study, we aim to investigate the correlations between polymorphisms in the DEFB1 gene and the susceptibility to chronic respiratory diseases (CRDs). Electronic searches in multiple scientific literature databases (PubMed, EBSCO, Ovid, Springerlink, Wiley, Web of Science, Wanfang database, China National Knowledge Infrastructure database and VIP database) were made to retrieve studies on the associations between DEFB1 gene polymorphisms and CRDs. We used strict inclusion criteria in the present meta-analysis. Data analyses were performed with STATA software (version 12.0; Stata Corp, College Station, TX, USA). Of 76 initiallyretrieved articles, 11 were finally incorporated into our meta-analysis, enrolling 1343 patients with CRDs (477 with chronic obstructive pulmonary disease (COPD), 305 with asthma, 286 with cystic fibrosis (CF), 130 with ventilator- associated pneumonia (VAP), 145 with pulmonary tuberculosis, and 1261 healthy controls into our metaanalysis. Our study showed that -44 C/G (rs1800972) and 1654 G/A (rs2738047) in DEFB1 are strongly associated with the increased susceptibility to CRDs. The subgroup analysis on disease types showed that the -44 C/G (rs1800972) in DEFB1 is associated with the susceptibility to COPD, VAP and CF, and the 1654 G/A (rs2738047) associated with the susceptibility to COPD and asthma. Subgroup analyses based on ethnicity indicated that -44 C/G (rs1800972) may confer increased susceptibility to CRDs in Asians but not in Caucasians. Taken together, -44 C/G (rs1800972) and 1654 G/A (rs2738047) are strongly associated with CRD susceptibility, while associations of SNPs -52 G/A (rs1799946) and -20 G/A (rs11362) with CRDs needs further investigation.

        Bag of Visual Words Method based on PLSA and Chi-Square Model for Object Category

        The problem of visual words` synonymy and ambiguity always exist in the conventional bag of visual words (BoVW) model based object category methods. Besides, the noisy visual words, so-called “visual stop-words” will degrade the semantic resolution of visual dictionary. In view of this, a novel bag of visual words method based on PLSA and chi-square model for object category is proposed. Firstly, Probabilistic Latent Semantic Analysis (PLSA) is used to analyze the semantic co-occurrence probability of visual words, infer the latent semantic topics in images, and get the latent topic distributions induced by the words. Secondly, the KL divergence is adopt to measure the semantic distance between visual words, which can get semantically related homoionym. Then, adaptive soft-assignment strategy is combined to realize the soft mapping between SIFT features and some homoionym. Finally, the chi-square model is introduced to eliminate the “visual stop-words” and reconstruct the visual vocabulary histograms. Moreover, SVM (Support Vector Machine) is applied to accomplish object classification. Experimental results indicated that the synonymy and ambiguity problems of visual words can be overcome effectively. The distinguish ability of visual semantic resolution as well as the object classification performance are substantially boosted compared with the traditional methods.

        Construction of Strontium Titanate/Binary Metal Sulfide Heterojunction Photocatalysts for Enhanced Visible-Light-Driven Photocatalytic Activity

        A novel strontium titanate/binary metal sulfide (SrTiO3/SnCoS4) heterostructure was synthesized by a simple two-step hydrothermal method. The visible-light-driven photocatalytic performance of SrTiO3/SnCoS4 composites was evaluated in the degradation of methyl orange (MO) under visible light irradiation. The photocatalytic performance of SrTiO3/SnCoS4-5% is much higher than that of pure SrTiO3, SnCoS4, SrTiO3/SnS2 and SrTiO3/CoS2. The SrTiO3/SnCoS4 composite material with 5 wt.% of SnCoS4 showed the highest photocatalytic efficiency for MO degradation, and the degradation rate could reach 95% after 140 min irradiation time. The enhanced photocatalytic activity was ascribed to not only the improvement of visible light absorption efficiency, but also the construction of a heterostructure which make it possible to effectively separate photoexcited electrons and holes in the two-phase interface.

        Effects of Loading Rates on the Hysteretic Response of Resilient Variable Friction Dampers

        A novel resilient variable friction damper (RVFD) with a simple construction was proposed. To analyze the eff ects of loading rate on the stability and energy dissipation performance of the RVFD, two RVFDs were tested under diff erent loading rates and the hysteretic behaviors of the compared with that of the traditional constant friction dampers. Then the peak force, energy dissipation behavior, uneven gap opening of the grooved outer plates (GOPs), and performance of the disc springs under diff erent loading rates were studied. The results show that although no self-centering system was provided, only the RVFDs without a self-centering system still can exhibit stable self-centering performance and eff ective energy dissipation behaviors, especially with due to the using e of athe grooved steel plates with greater groove angle. When the loading rate is less than 10 mm/s, the peak forces of the RVFD slightly decrease with the increase in loading rates, while when the loading rate is greater than 10 mm/s, its eff ects could be neglected. The uneven gap opening of the GOPs infl uences the clamping forces of the disc springs in the bolts under diff erent loading amplitudes. The disc springs in the RVFDs not only provide the increasing clamping forces for energy dissipation and the restoring forces for self-centering behavior, but also dissipate the energy, which accounts for about 10% of the total energy dissipation of the RVFD.

        A Novel Therapeutic Effect of a New Variant of CTLA4-Ig with Four Antennas That Are Terminally Capped with Sialic Acid in the CTLA4 Region

        Rheumatoid arthritis (RA) is a multifactorial immune-mediated disease, the pathogenesis of which involves different cell types. T-cell activation plays an important role in RA. Therefore, inhibiting T-cell activation is one of the current therapeutic strategies. Cytotoxic T-lymphocyte antigen 4-immunoglobulin (CTLA4-Ig), also known as abatacept, reduces cytokine secretion by inhibiting T-cell activation. To achieve a homeostatic therapeutic effect, CTLA4-Ig has to be administered repeatedly over several weeks, which limits its applicability in RA treatment. To overcome this limitation, we increased the number of sialic acid-capped antennas by genetically engineering the CTLA4 region to increase the therapeutic effect of CTLA4-Ig. N-acetylglucosaminyltransferase (GnT) and α2,6-sialyltransferase (α2,6-ST) were co-overexpressed in Chinese hamster ovary (CHO) cells to generate a highly sialylated CTLA4-Ig fusion protein, named ST6. The therapeutic and immunogenic effects of ST6 and CTLA4-Ig were compared. ST6 dose-dependently decreased paw edema in a mouse model of collagen-induced arthritis and reduced cytokine levels in a co-culture cell assay in a similar manner to CTLA4-Ig. ST6- and CTLA4-Ig-induced T cell-derived cytokines were examined in CD4 T cells isolated from peripheral blood mononuclear cells after cell killing through irradiation followed by flow- and magnetic-beadassisted separation. Interestingly, compared to CTLA4-Ig, ST6 was substantially less immunogenic and more stable and durable. Our data suggest that ST6 can serve as a novel, less immunogenic therapeutic strategy for patients with RA.

      • Pricing Strategy of Dual Channel Supply Chain under Asymmetric Information

        Aiming at the dual channel supply chain which sells a single product, and composed of the retailer as the leader and the manufacturer as the follower, firstly, by establishing the Stackelberg game model, we study the manufacturer and the retailer’s optimal pricing strategy under the condition of retailer’s innovation cost coefficient is common knowledge, then further analyze the manufacturer and the retailer’s optimal pricing strategy under the retailer’s innovation cost coefficient is his own private information, and discuss the existence condition of perfect Bayesian Nash equilibrium pricing. The research result showed that under the condition of asymmetric information, even the manufacturer has belief revision, his optimal pricing is always consistent with the type of retailer’s optimal pricing, but there are several pricing strategy choices to retailer. Under different parameters’ values, the dynamic game model with asymmetric information possibly exist separating equilibrium, pooling equilibrium, and semi-separating equilibrium.
