        Comparative analysis of liver transcriptome reveals adaptive responses to hypoxia environmental condition in Tibetan chicken

        Objective: Tibetan chickens, which have unique adaptations to extreme high-altitude environments, exhibit phenotypic and physiological characteristics that are distinct from those of lowland chickens. However, the mechanisms underlying hypoxic adaptation in the liver of chickens remain unknown. Methods: RNA-sequencing (RNA-Seq) technology was used to assess the differentially expressed genes (DEGs) involved in hypoxia adaptation in highland chickens (native Tibetan chicken [HT]) and lowland chickens (Langshan chicken [LS], Beijing You chicken [BJ], Qingyuan Partridge chicken [QY], and Chahua chicken [CH]). Results: A total of 352 co-DEGs were specifically screened between HT and four native lowland chicken breeds. Gene ontology and Kyoto encyclopedia of genes and genomes enrichment analyses indicated that these co-DEGs were widely involved in lipid metabolism processes, such as the peroxisome proliferator-activated receptors (PPAR) signaling pathway, fatty acid degradation, fatty acid metabolism and fatty acid biosynthesis. To further determine the relationship from the 352 co-DEGs, protein-protein interaction network was carried out and identified eight genes (ACSL1, CPT1A, ACOX1, PPARC1A, SCD, ACSBG2, ACACA, and FASN) as the potential regulating genes that are responsible for the altitude difference between the HT and other four lowland chicken breeds. Conclusion: This study provides novel insights into the molecular mechanisms regulating hypoxia adaptation via lipid metabolism in Tibetan chickens and other highland animals.

        An optimization approach for black-and-white and hinge-removal topology designs

        An optimization approach for black-and-white and hinge-removal topology designs is studied. To achieve this motive, an optimal topologyallowing grey boundaries is found firstly. When a suitable design has been obtained, this solution is then used as a starting pointfor the follow-up optimization with the goal to free unfavorable intermediate elements. For this purpose, an updated optimality criterionin which a threshold factor is introduced to gradually suppress elements with low density is proposed. The typical optimality method andnew technique proposed are applied to the design procedure sequentially. Besides, to circumvent the one-point hinge connection problemproducing in the process of freeing intermediate elements, a hinge-removal strategy is also proposed. During the optimization, the binaryconstraints on design variables are relaxed based on the scheme of solid isotropic material with penalization. Meanwhile, the meshindependencyfilter is employed to ensure the existence of a solution and remove well-known checkerboards. In this way, a solution thathas few intermediate elements and is free of one-point hinge connections is obtained. Finally, different numerical examples including thecompliance minimization, compliant mechanisms and vibration problems demonstrate the validity of the proposed approach.

        Synthesis of Carbon Nanotube-CuxO(x = 8, 64) Nanocrystal Composites

        A simple, rapid and eco-friendly method has been introduced to prepare suboxide CuxO(x = 8, 64)nanocrystals on multiwall carbon nanotube (MWCNT) composites. The structural characterization by using high-resolution transmission electron microscopy and X-ray diffraction methods indicates that CuxO(x = 8, 64) can be uniformly grown on the sidewalls of carbon nanotube under direct copper deposition at room temperature. Our results suggest that the growth method of nano-CuxO (x = 8, 64) crystals could be used to synthesize other nano-metal suboxides, such as MxO [M=metal;x = 1, 2, 4, 8 and 64]

        Research on static and dynamic behaviors of PC track beam for straddle monorail transit system

        In this study, in-situ static and dynamic tests of four pre-stressed concrete (PC) track beams with different span lengths and curvatures in a straddle monorail transit system were reported. In the static load tests, the strain and deflection at critical sections of the PC track beams were measured to determine the load bearing capacity and stiffness. The dynamic responses of strain, deflection, acceleration, and displacement at key positions of the PC track beams were measured under different train speeds and train loads to systematically study the dynamic behaviors of the PC track beams. A three-dimensional finite element model of the track beam-vehicle coupled vibration system was established to help understand the dynamic behavior of the system, and the model was verified using the test results. The research results show that the curvature, span length, train speed, and train loads have significant influence on the dynamic responses of the PC track beams. The dynamic performance of the PC track beams in the curve section is susceptible to dynamic loads. Appropriate train loads can effectively reduce the impact of the train on the PC track beam. The PC track beams allow good riding comfort.

        Graphene Oxide Nanosheet-Composited Poly(N-isopropylacrylamide) Hydrogel for Cell Sheet Recovery

        Cell sheet engineering technique has been applied to treat various tissues without the use of a traditional scaffold. To date, methods for the cell sheet harvesting depend mostly on grafted poly(N-isopropylacrylamide) (pNIPAAm) thin layers, while the native pNIPAAm hydrogel, which possibly presents the easiest way to prepare thermo-responsive materials, is not suitable for the cell sheet harvesting due to its low cell attachment. In this study, the graphene oxide (GO) nanosheet was utilized as an additive to enhance the bio-compatibility of the pNIPAAm hydrogel. Different concentrations of GO nanosheets were added to prepare GO/pNIPAAm composite hydrogels through the in-situ free radical polymerization with polyethylene glycol dimethacrylate (PEGDA) as a cross-linker. The results indicated that the physical properties of the composite hydrogels had little difference with that of the native pNIPAAm hydrogel. However, the cell attachment, proliferation and detachment behaviors on the composite hydrogel surface were greatly enhanced. Monkey fibroblast COS7 cells attached and proliferated better on the GO/pNIPAAm composite hydrogel, while intact COS7 cell sheets could be harvested from the composite hydrogels by simply lowering the temperature. In contrast, the cells appeared as clusters on the native pNIPAAm hydrogel. Furthermore, when HeLa and COS7 cells were seeded successively onto the micropatterned GO/pNIPAAm hydrogel, there could be the formation of a patterned HeLa/COS7 cell layer. The geometrically patterned GO/pNIPAAm hydrogel may provide an easy-to-prepare material for releasing patterned cell sheets compared to the specific cell-adhesive proteins reported to make patterned cell layers.

        Effects of a multi-strain probiotic on growth, health, and fecal bacterial flora of neonatal dairy calves

        Objective: The aim of this study was to investigate the effects of dietary supplementation with a multi-strain probiotic (MSP) product containing of Bifidobacterium animalis, Lactobacillus casei, Streptococcus faecalis, and Bacillus cerevisiae on growth, health, and fecal bacterial composition of dairy calves during the first month of life. Methods: Forty Holstein calves (24 female and 16 male) at 2 d of age were grouped by sex and date of birth then randomly assigned to 1 of 4 treatments: milk replacer supplementation with 0 g (0MSP), 2 g (2MSP), 4 g (4MSP), and 6 g (6MSP) MSP per calf per day. Results: Supplementation of MSP did not result in any significant differences in parameters of body measurements of calves during the 30 d period. As the dosage of MSP increased, the average daily gain (p = 0.025) and total dry matter intake (p = 0.020) of calves showed a linear increase. The fecal consistency index of the 2MSP, 4MSP, and 6MSP group calves were lower than that of the 0MSP group calves (p = 0.003). As the dosage of MSP increased, the concentrations of lactate dehydrogenase (p = 0.068) and aspartate aminotransferase (p = 0.081) in serum tended to decrease, whereas the concentration of total cholesterol increased quadratically (p = 0.021). The relative abundance of Dorea in feces was lower (p = 0.011) in the 2MSP, 4MSP, and 6MSP group calves than that in the 0MSP group calves. The relative abundance of Dorea (p = 0.001), Faecalibacterium (p = 0.050), and Mitsuokella (p = 0.030) decreased linearly, whereas the relative abundance of Prevotella tended to increase linearly as the dosage of MSP increased (p = 0.058). Conclusion: The MSP product can be used to reduce the diarrhea, improve the performance, and alter the composition of the fecal bacteria in neonatal dairy calves under the commercial conditions.       • The Important Anti-Apoptotic Role and Its Regulation Mechanism of PTTG1 in UV-Induced Apoptosis

        Pituitary tumor transforming gene (PTTG1) is widely detected in many tumors. Increasing evidence reveals that PTTG1 is associated with cell proliferation, cellular transformation and apoptosis. However, the functions of PTTG1, especially its role in DNA damage-induced apoptosis, remain largely unclear. In this report, we used UV irradiation to induce apoptosis in HeLa cells to examine the role of PTTG1 in UV-induced apoptosis by RNAi-mediated knockdown and overexpression of PTTG1. RNAi-mediated knockdown of PTTG1 expression increased and overexpression of PTTG1 decreased the UV-induced apoptosis. Furthermore, UV irradiation decreased PTTG1 mRNA and protein expression. These effects were found to be mediated by JNK pathway. Therefore, PTTG1 had an important anti-apoptotic role in UV-induced apoptosis and this role was mediated by JNK pathway. These results may provide important information for understanding the exact role and the regulation mechanism of PTTG1 in UV-induced apoptosis.

      • The Influence Analysis of Hamilton Chaos Phase Trajectory in Additive White Noise Channel

        Previously a novel chaos M-ary modulation and demodulation method based on spatiotemporal chaos Hamilton oscillator was proposed and is applied in M-ary communication. Without chaos synchronization circumstance, the performance of communication has been improved in bandwidth efficiency, transmission efficiency and anti-white- noise performance compared with traditional communication method. Due to the working performance of zone partition demodulator is strictly connected with channel noise, it is necessary to study the influence of Hamilton phase trajectory deeply by the channel noise. In this paper, two methods for designing of the boundary of zone partition demodulator in additive white Gaussian noise are discussed, which are using statistics and curve fitting ways. Therefore, the problem about how to determine the boundary of zone partition demodulator in white noise is solved. Finally, the chaos M-ary communication system based on Hamilton oscillator is constructed and simulated in white noise channel. The result shows that the method proposed in this paper can improve the anti-white-noise performance of the whole communication system.
