Lactate as a major epigenetic carbon source for histone acetylation via nuclear LDH metabolism

Histone acetylation involves the transfer of two-carbon units to the nucleus that are embedded in low-concentration metabolites. We found that lactate, a high-concentration metabolic byproduct, can be a major carbon source for histone acetylation through oxidation-dependent metabolism. Both in cells and in purified nuclei, 13C3-lactate carbons are incorporated into histone H4 (maximum incorporation: ~60%). In the purified nucleus, this process depends on nucleus-localized lactate dehydrogenase (LDHA), knockout (KO) of which abrogates incorporation. Heterologous expression of nucleus-localized LDHA reverses the KO effect. Lactate itself increases histone acetylation, whereas inhibition of LDHA reduces acetylation. In vitro and in vivo settings exhibit different lactate incorporation patterns, suggesting an influence on the microenvironment. Higher nuclear LDHA localization is observed in pancreatic cancer than in normal tissues, showing disease relevance. Overall, lactate and nuclear LDHA can be major structural and regulatory players in the metabolism–epigenetics axis controlled by the cell’s own status or the environmental status.

소통의 정치윤리: 필리프 6세 치세(1328~1350)의 정치 담론

본 논문은 정치적 정당성이 취약했던 필리프 6세 치세(1328-1350)에 나타난 정치적 담론들을 살펴보기 위해 당대에 간행된 네 편의 정치윤리서의 특징들을 소개하고 그 의미를 당대의 맥락에서 조망해보고자 한다. 이 저작들은 필리프 드 비트리가 저술한 『세 송이 백합관』, 장 드 비녜가 번역한 『테오도로스 팔라이올로고스의 교훈』과 『장기로 보는 귀족과 평민의 도덕에 대한 책』, 장 드 마리니가 저술한 『국가와 통치』이다. 그리고 앞의 두 저작은 필리프 6세에게, 뒤의 두 저작은 당시 세자인 장(장2세)에게 헌정되었다. 이 네 편의 정치윤리서는 다음과 같은 세 가지 공통점, 즉 1) 교회의 영향력에 대해 점점 더 자율적으로 되어 가는 정치공동체에 대한 관심 증대, 2) 정치적 의사소통을 바탕으로 한 정치공동체의 내재적 구성, 3) 왕권 신성화와는 무관한 정치적 의사소통에 있어서의 국왕 주도권이라는 성격을 지닌다. 물론 정치공동체의 자율성에 대한 이러한 관심은 필리프 4세 치세의 정치담론들이 보여주는 왕의 신체에 각인된 정치체의 신성화, 또는 ‘선택받은 민족’이라는 표현으로 나아가지 않는다. 오히려 1330-40년대 정치적 위기들로 짓눌린 프랑스 왕궁에서는 정치공동체의 한 구성원에 불과한 국왕과 정치사회 구성원들 사이의 실질적인 의사소통이 이루어져야 한다고 강조하고 있다. 즉 이 저작들은 적어도 1350년대 이전까지 프랑스 왕정이 절대주의적 경향에서는 멀리 떨어져 있었다는 점을 보여준다. 결국 1350년에 즉위한 장 2세는 왕국의 개혁을 주도하기 시작한다. 그러나 중요한 점은 왕권이 어떠한 주장이나 담론에 찬성이나 반대를 했다기보다 의사소통의 장이나 텍스트 생산의 영역에서 주도권을 잡아 나가기 시작했다는 사실이다. This paper introduces the characteristics of four texts on political ethics, published in the reign of Philip VI (1328-1350) whose political legitimacy was weak, to examine political discourses at that time, and looks at their meanings in the pol itical context. These four texts are Le Chapel des trois fleurs de lis written by Philippe de Vitry, Les Enseignements de Theodore Paleologue and Le livre de la moralite des nobles hommes et des gens de peuple sus le gieu des esches translated by Jean de Vignay, and L’Estat et le gouvernement, written by Jean de Marigny. The previous two works were dedicated to Philip VI, and the latter two, to the crown prince, Jean(Jean II). These have three things in common: 1) Growing interest in the political community, which was becoming increasingly autonomous from the influence of the church ; 2) Immanent formation of polity based on political communication ; 3) Royal intiative in political communication, far from sanctification of kingship. Of course, this interest in the autonomy of the politiy does not go toward the sanctification of the political body incarnated in the king's body, or the expression “chosen people”, as demonstrated by political discourses under the reign of Philip IV the Fair. Rather, in the royal court weighed by the political crises of the 1330s-40s, they stress that actual communication between members of the political society and the king, also as its member, should take place. In other words, these works show that the French monarchy was far from absolutist tendencies until at least the 1350s. Eventually, King Jean II, who ascended the throne in 1350, began to lead the reform of the kingdom. What is important here is that the kingship, rather than for or against any argument or discourse, began to take the initiative in the field of communication or text production.

멀티미디어 시대로의 급격한 변화에 따라 영상시대로 접어든 현재 가독성 위주의 합리적인 방식에서 탈피한 실험적 타이포그래피는 시간의 개념을 내포한 동적인 표현을 통하여 무빙타입을 감성적 이미지로서 보이게 한다. 본 논문은 레터링 디자인에 근거하여 문자의 구조적 특징을 문자의 움직임에 반영한 동적 문자(Animated Type)에 관한 연구이다. 이론적 배경으로 시간 공간 움직임의 개념을 알아보고 동적 문자에 대하여 정의하였다. 동적 문자의 형태 변화를 그리기 회전 형성 변형 치환 결합과 같은 유형별로 구분하여 특징과 사례를 살펴보았다. 이것은 동적 문자의 구조적 변화과정에 초점을 둔 세분화된 구분으로서 향후 동적 문자가 표현하는 어휘의 우연적 장식적 특징과 표현 주제를 동반한 시각 아이덴티티의 특징을 예상가능하게 한다는 결론을 얻었다. 또한 변형의 시작점과 종료점의 설정은 동적문자가 인터렉티브 미디어를 이용한 정보의 시각적 표기로 쓰일 수 있게 하는 중요한 특징임을 보여준다. 이러한 연구는 문자가 움직임뿐만 아니라 사운드 프로그래밍의 요소와 결합하고 새로운 미디어를 통해 다양하게 전개되는 모션 타이포그래피의 실험을 위한 선행연구로의 기여할 수 있을 것으로 기대 된다. We are experiencing a rapid development of multimedia leading us into a era of visual language in motion. As a result experimental typography assigns less value on the readability of type and makes it easier for us to accept the moving type as an expressional image rather than type to read meaning. This research is about animated type sets based on the sequential lettering design reflecting upon the structural characteristics of each letter. As a supporting theory it also explores the concepts of time space motion and defines the meaning of animated type. The examples of animated type in this research can be classified by its characteristics of structural changes under the concept of drawing rotation forming transforming replacing and assembling. These classifications are more focused on the sequence of change. They allow us to draw the conclusion that we can predict the style of words consisting animated type that is characterized by coincidential decorative motion demonstrated through the visual identity of assigned theme in motion. Setting starting and ending points of motion in animated type is the main characteristic of visual images representing the change of data as a response to interaction in media. This research can be meaningful as a pre-study for the experimentation of moving types in combination with not only motion but also sound and computer programing with the emergence of new media in the future.

Purpose: Accurate monitoring of soil strength is a key technology applicable to various precision agricultural practices. Soil strength has been traditionally measured using a cone penetrometer, which is time-consuming and expensive, making it difficult to obtain the spatial data required for precision agriculture. To improve the current, inefficient method of measuring soil strength, our objective was to develop and evaluate an in-situ system that could measure horizontal soil strength in real-time, while moving across a soil bin. Methods: Multiple cone-shape penetrometers were horizontally assembled at the front of a vertical plow blade at intervals of 5 cm. Each penetrometer was directly connected to a load cell, which measured loads of 0-2.54 kN. In order to process the digital signals from every individual transducer concurrently, a microcontroller was embedded into the measurement system. Wireless data communication was used between a data storage device and this real-time horizontal soil strength (RHSS) measurement system travelling at 0.5 m/s through an indoor experimental soil bin. The horizontal soil strength index (HSSI) measured by the developed system was compared with the cone index (CI) measured by a traditional cone penetrometer. Results: The coefficient of determination between the CI and the HSSI at depths of 5 cm and 10 cm (r 2 = 0.67 and 0.88, respectively) were relatively less than those measured below 20 cm (r 2 ≥ 0.93). Additionally, the measured HSSIs were typically greater than the CIs for a given numbers of compactor operations. For an all-depth regression, the coefficient of determination was 0.94, with a RMSE of 0.23. Conclusions: A HSSI measurement system was evaluated in comparison with the conventional soil strength measurement system, CI. Further study is needed, in the form of field tests, on this real-time measurement and control system, which would be applied to precision agriculture.

This study examines how a country`s cultural factors impact the whistleblowing system in a country by analyzing Hofstede`s cultural dimensions (power distance, individualism, uncertainty avoidance, and masculinity) and Global Integrity`s whistleblowing measures. The results of Pearson correlations show that power, distance and uncertainty avoidance have statistically significant relations to the whistleblowing scores. After multivariate regression analyses considering democratization, GDP per capita, primary school enrollment, and government accountability of each country with panel data of 37 countries from 2004 to 2008 with gaps, this study confirms that individualism and masculinity are positively related to the whistleblowing scores and power, distance, and uncertainty avoidance negatively affect the whistleblowing scores.
