A novel therapeutic approach of Hachimi-jio-gan to diabetes and its complications

Great efforts have been made to improve both the quality of life and life expectancy of diabetes by treating problems associated with chronic complications such as neuropathy, retinopathy and nephropathy. In particular, diabetes is an increased risk of developing several types of kidney disease, and the predominant cause of end-stage renal disease in patients with this disorder is diabetic nephropathy. Therefore, prevention of the occurrence and progression of diabetes and its complications has become a very important issue. The scientific observations of an animal model of streptozotocin-induced diabetes, spontaneously occurring diabetes and diabetic nephropathy in this study suggest that one of the Kampo prescriptions, Hachimi-jio-gan comprising eight constituents, is a novel therapeutic agent.
Effectiveness of Green Tea Tannin on Rats with Chronic Renal Failure

The effects of green tea tannin on nephrectomized rats were examined. There were increases in blood urea nitrogen, serum creatinine, and urinary protein, and a decrease in creatinine clearance in the nephrectomized control rats, whereas better results for these parameters were obtained in rats given green tea tannin after nephrectomy, demonstrating a suppressed progression of the renal failure. When the renal parenchyma was partially resected, the remnant kidney showed a decrease in the activity of radical scavenger enzymes. Green tea tannin, however, was found to lighten the kidney under such oxidative stress. Mesangial proliferation and glomerular sclerotic lesions, which were conspicuous in the rats that were not given green tea tannin after nephrectomy, were also relieved.
Persimmon Oligomeric Proanthocyanidins Extend Life Span of Senescence-Accelerated Mice

The anti-aging activities of persimmon oligomeric proanthocyanidins (POPs), reported to improve life span and behavioral characteristics associated with the aging process, were investigated using the senescence-accelerated mouse (SAM) P8, which is a good model for studies on aging-related behavioral changes as well as life span. We demonstrated that the administration of POPs extended the life span of SAMP8. In addition, POPs elevated Sirt1 expression, which is recognized as an essential factor for life span extension in the brain. On the other hand, the administration of POPs did not induce stereotypical behaviors such as rearing, jumping, and hanging from the lid of a cage, whereas food restriction increased these frequencies without a significant change in motor function. The present study suggests a promising role of POPs as anti-aging agents to extend life span, although further studies elucidating their anti-aging mechanisms acting are needed.
당뇨 쥐에서 리치로부터 분리된 저분자 폴리페놀인 Oligonol의 NADPH Oxidase 의존형 활성산소종 생산을 통한 세포사 및 췌장보호 효과

Background : We have previously reported that Oligonol, a low-molecular polyphenol derived from lychee fruit, has protective effect on the liver and kidney of diabetic animal model. In this study, we examined whether Oligonol has any beneficial effects on pancreas of diabetic rats. Methods and Results : Oligonol was orally administered at a dose of 10 and 20 mg/kg body weight for 10 days to STZ-induced diabetic rats, and the effects were compared with those of vehicle-treated diabetic control and non-diabetic control rats. The administration of Oligonol reduced hyperglycemia in diabetic rats through an improvement of serum and pancreatic insulin levels. The increased reactive oxygen species levels in pancreas of diabetic control rats was attenuated by the Oligonol administration through inhibiting the expression of NADPH oxidase-related proteins. The enhanced expression of pro-apoptotic proteins in pancreas of diabetic control rats was significantly reduced by Oligonol administration through down-regulation of phosphor-c-Jun N-terminal kinases protein in pancreas. Furthermore, the expressions of cell proliferation-related protein were also augmented in Oligonol treated-diabetic rats. However, Oligonol treatment led to improved histological changes in the pancreas. Conclusion : These pancreatoprotective effects of Oligonol were achieved through attenuation of oxidative stress and its sensitive protein expression associated with apoptosis and cell proliferation in diabetic rats.
