        Solid-Phase Extraction of Caffeine and Catechin Compounds from Green Tea by Caffeine Molecular Imprinted Polymer

        In this work, caffeine and some catechin compounds + C, EC, EGC, and EGCG were extracted from green tea by using molecular imprinted polymers (MIP) as sorbent materials in a solid-phase extraction (SPE) process known as MISPE (molecular imprinted solid-phase extraction). For synthesis of MIP, caffeine was employed as the template, MAA as the monomer, EGDMA as the crosslinker, and AIBN as the initiator. A solution of caffeine (0.2 mg/mL in methanol) was utilized in the solid extraction cartridges following loading, washing, and elution procedures with acetonitrile, methanol, and methanol-acetic acid (90/10, %v/v) as the solvents, respectively. This solid-phase extraction protocol was applied for the extraction of caffeine and some catechin compounds from green tea. A comparison was made between the results obtained with the MIP cartridges and a traditional C18 reversed-phase cartridge. It was thereupon found that the recovery of caffeine by the MIP-based sorbent used in this work was almost two and four times greater than that by a commercially available C18 material. A quantitative analysis was conducted by high performance liquid chromatography (HPLC) using a C18 column (5 m, 250 4.6 mm) with methanol/water (40/60, %v/v) as the mobile phase at a flow rate of 0.5 mL/min.

        To study the effects of ethanol extract of the Rhodiola Sachalinensis on the anti-oxidation and blood pressure and skeletal muscles contractility in rats. The ethanol extracts and fractions of Rhodiola Sachalinensis were determinated the anti-oxidation effects by DPPH (1,1-dipenyl- 2-picrylhydrazyl) method and comparing with the BHA (Butylated hydroxyanisole) and AA (Ascorbic acid). The gastrocnemius and soleus muscle contractility were observed by stmulating the sciatic nerve with electricity after affusing stomach with ethanol extract of the 200 mg/kg dosage of Rhodiola Sachalinensis for 4 weeks. The ethanol extract of the Rhodiola Sachalinensis could shorten latent period, increase maximal contractive extent and time and relaxative time at the twitch and complete tetanus of gastrocnemius and soleus muscles. The ethanol extract of the Rhodiola Sachalinensis shows high anti-oxidation effect and can enhance skeletal muscles contractility in rats.

        Adsorption Isotherms of Catechin Compounds on (+)Catechin-MIP

        A molecular imprinted polymer (MIP) using (+)catechin ((+)C) as a template and acrylamide (AM) as a functional monomer was prepared. Acetonitrile was used as the porogen with ethylene glycol dimethacrylate (EGDMA) as the crosslinker and 2,2'-azobis(isobutyronitrile) (AIBN) as the initiator. The adsorption isotherms in the MIP were measured and the parameters of the equilibrium isotherms were estimated by linear and nonlinear regression analyses. The linear equation for original concentration and adsorpted concentrations was then expressed, and the adsorption equilibrium data were correlated into Langmuir, Freundlich, quadratic, and Langmuir Extension isotherm models. The mixture compounds of (+)C and epicatechin (EC) show competitive adsorption on specific binding sites of the (+)catechin-MIP. The adsorption concentrations of (+)C, epicatechin (EC), epicatechin gallate (ECG), and epigallocatechin gallate (EGCG), on the (+)catechin-molecular imprinted polymer were compared. Through the analysis, the (+)catechin-molecular imprinted polymer showed higher adsorption ability than blank polymer which was synthesized molecular imprinted polymer without (+)catechin. Furthermore, the competitive Langmuir isotherms were applied to the mixture compounds of (+)C and EC.

        Separation of Catechin Compounds by Retention Theory in RP-HPLC

        <P> An efficient optimization method was used to separate catechin compounds by RP-HPLC. In this work, the binary mobile phase of water and acetonitrile was used with the buffer of acetic acid (AA). The elution profiles were calculated by the plate theory based on the linear and quadratic equations of retention factor, lnk=Inkw+SF, lnk=L+MF+NF2, k=A+B/F, where F was the vol.% of acetonitrile. We modified the retention theory to calculate the elution profile in both isocratic and gradient modes. The final calculated results showed that the first mobile phase composition was water in 0.1% AA/acetonitrile in 0.1% AA, 90/10 vol.%, followed 30 min later by the second composition of mobile phase, which was linearly changed to 70/30 vol.%. In the experimental conditions, the agreement between the experimental data and the calculated values was relatively good. Catechin compounds from Korean and Chinese green tea as potential powers of anticancer and antioxidant components were target materials in this work. The full content of the catechin compounds of EGC, (+) C, EC, EGCG, and ECG in Chinese green tea extracted was 249.92 mg/g, so it was 2.15 times of that from Korean tea.</P>

Acetonitrile was used as the porogen with ethylene glycol dimethacrylate (EGDMA) as the crosslinker and 2,2'-azobis(isobutyronitrile) (AIBN) as the initiator. By a linear and nonlinear regression analysis, the experimental parameters in the equilibrium isotherms were estimated. Then, the linear and quadratic equations for concentration and sorbents to adsorption amounts were expressed, and the adsorption equilibrium data were also correlated into the Freundlich isotherm model. Comparisons of caffeine adsorption isotherm on C18 particles as well as the molecular imprinted polymer were made. The results showed that the caffeine-imprinted polymer showed extraordinarily higher adsorption ability than C18 particles

        Primary study of volatiles composition of Rhodiola sachalinensis by using gas chromatography and mass spectrometry (GC/MS)

        To determine the volatile compounds in Rhodiola sachalinensis, hydro-distillation (HD) and headspace liquid-phase micro-extraction (HS-LPME) were used to extract 75 and 68 volatiles, respectively. Geraniol (24.73%),n-octanol (15.56%), and linalool (14.51%) were the most abundant essential oils detected in the HD samples, while geraniol (24.17%) and n-octanol (15.81%) were also detected at high levels in the HS-LPME samples. The main chemical classes of the essential oils were monoterpene alcohols in both the HD and HS-LPME samples at 59.02% and 37.64%,respectively. The O-heterocyclic (8.52%) and aromatic (5.92%) compounds were more abundant in the HS-LPME samples than in the HD samples.
