        Assessment of nephrotoxicity of herbal medicine containing aristolochic acid in mice

        Background/Aims: It is undetermined if herbal medicines (HM) containing aristolochic acid (AA)-containing have similar nephrotoxicity to AA itself. Methods: We administered HM containing a high concentration of AA for 5 days (short-term study) or a low concentration of AA for 30 days (long-term study) to C57BL/6 mice; for comparison, same dose of AA compound was used as controls. Results: The nephrotoxicity in the HM- and AA-treated mice was compared in terms of renal function, histopathology, oxidative stress, apoptotic cell death, and mitochondrial damage. Short-term HM treatment resulted in acute kidney injury (marked renal dysfunction, acute tubular necrosis, and neutrophil gelatinase-associated lipocalin [NGAL] expression) in which the severity of renal dysfunction and histopathology was comparable with that induced by the administration of AA alone. Long-term HM treatment resulted in features of chronic kidney disease (CKD, mild renal dysfunction and tubular atrophy and dilatation). No significant differences in these parameters were observed between the HM- and AA-treated mice. HM-induced oxidative stress (8-hydroxy-2’-deoxyguanosine and manganese- dependent superoxide dismutase expression) and apoptotic cell death (terminal deoxynucleotidyl transferase dUTP nick end labelling [TUNEL]-positive cells and active caspase-3 expression) were similar in HM- and AA-treated mice in the short-term and long-term studies. Mitochondrial injury, evaluated by electron microscopy, was also similar in HM- and AA-treated mice in the short-term and long-term studies. Conclusions: The nephrotoxic potential of HM containing AA was similar to that of AA itself.

        한국인 및 중국 한족 정신분열병 환자의 5-HT2A 수용체 유전자 -1438A/G 다형성

        Objectives : The purpose of the present study was to investigate the association between -1438A/G polymorphism of 5-HT2A receptor gene and schizophrenia in Korean and Han Chinese population. Methods : A sample of 184 Korean patients with schizophrenia and 96 Korean healthy normal controls and 96 Han Chinese patients with schizophrenia and 96 Han-Chinese healthy normal controls were genotyped for a single nucleotide polymorphism with in 5-HT2A receptor gene (promoter region, A-1438G) by Msp I Resthction Fragment Length Polymorphism (RFLP). Results : There was no difference in allelic frequencies and genotype frequencies of -1438A/G polymorphism between Korean schizophrenics and controls (p=0.13) and Han Chinese schizophrenics and controls (p=0.40). Also, -1438A/G Poly-morphism did not show ethnical difference between Korean and Han Chinese controls. The Scale for the Assessment of Negative Symptoms (SANS) scores showed no significant differences between genotypes of -1438A/G polymorphism in both of Korean and Han Chinese schizophrenics. Conclusion : These results suggest that -1438A/G polymorphism of the 5-HT2A receptor gene is not causally related to the development of schizophrenia in Korean and Han Chinese population, and there no ethnic difference between Korean and Han Chinese population.

        한국산과 중국산 '사과배' 과실의 생태적 특성

        Postharvest characteristics of 'Pingguoli' fruit produced between China and Korea were compared to find an ecological difference. There was a difference in fresh weight per fruit, thus, fruit weight produced in Korea was much higher than those in China. However, no significant differences were confirmed in the level of soluble solid content and flesh firmness. The acidity of Korean 'Pingguoli' was higher than Chinese ones. The external appearance of Korean 'Pingguoli' was not attractive due to the rough and rugged surface. Thus, the prospect of 'Pingguoli' cultivation in Korea seems not to b e promised. The general trend of respiration and ethylene evolution during storage did not significantly differed but the amount of CO_2 and ethylene production was higher in fruits produced in China. There was a strong similarity in respiration and ethylene evolution pattern to typical climacteric fruits. Thus, 'Pingguoli' is considered as a climacteric fruit. Even though the rate of ethylene production of 'Pingguoli' was relatively high, the storability of 'Pingguoli' showed a strong potential. The further studies in postharvest physiology of pear using 'Pingguoli' and in its breeding potentials as a source of long storage life are needed.^2)

        MH(Maleic hydrazide)처리가 포도 부초 생장억제 및 품질에 미치는 영향

        Spray of maleic hydrazide (MH) was confirmed to be highly effective on the inhibition of lateral shoot growth regardless of grape cultivars and application concentration examined in his experiment. The optimal spray times were 30 days after full bloom in 'Kyoho' and 35 days in 'Campbell Early'. However, the optimum season of application should be adjusted depending on the vine vigor. The apical buds of lateral shoot was significantly affected at the concentration of 3,900 ㎎·L^-1 resulting in the death and/or abscission. Thus the optimal concentration of MH seemed to be 1,950 ㎎·L^-1. The inhibition effect by MH application was derived from the decrease of node number and length. No adverse effect on fruit quality was found by MH application. In contrast, there was a tendency of anthocyanin increase in 'Campbell Early' grapes. Results indicated that MH could be an effective agent to reduce the labor charge through the inhibition of later shoot growth.

        수출 전 저장기간과 모의 수출에 따른 '후지' 사과의 상품성 변화

        관행적인 수확기인 11월 6일에 수확한 '후지' 사과는 80% 이상이 밀증상을 나타내었으나 에틸렌 발생량을 기준으로 볼 때 climacteric stage에 도달하지는 않았다. 모의 수출 전 2개월 저장기간 중 밀증상은 점차 감소하였다. 저장기간 중 가용성 고형물 수준은 일정하지 않았으나 경도와 생체중 감소가 저장기간일 길수록 증가하였다. 저장기간에 따라 모의 수출과 소매과정을 경과시킨 과실의 품질은 전반적으로 낮아졌지만 무처리에 비하여 30 um 플라스틱 필름 포장, 에틸렌제거제 처리는 전반적으로 과실의 품질을 소매단계까지 우수하게 유지시키는 결과를 보였다. 반면에 왁스처리는 외관품질을 우수하게 유지시켰지만 갈변장해를 일으켜 바람직하지 않았다. 따라서 수출용 과실은 건조한 환경에서 장기간 수송되고 판매되므로 품질저하가 심하여 이를 방지하기 위해서는 플라스틱 필름과 같은 moisture barrier 및 에틸렌 제거환경으로 선적하는 것이 바람직할 것으로 판단된다. The percentage of watercore occurrence harvested on Nov. 6 (commercial harvest season) was over 80% but fruits seemed not to reach climacteric stage on the basis of ethylene evolution. The occurrence ratio and severity of watercore gradually decreased with the increase of preshipment storage for 2 months. The level of soluble solids did not show a clear tendency during preshipment storage but flesh firmness, fresh weight, and acidity gradually decreased. Although the overall quality of fruit reduced along with the increase of preshipment storage duration after exposure to retail condition at ambient temperature for 1 week, plastic film wrapping (PE 30 um) as well as plastic film wrapping plus ethylene scrubbing had a benefit on keeping freshness of fruit even after retail condition. Waxed fruit showed excellent appearance but browning disorder was developed. Results indicated that moisture barrier such as plastic film wrapping and ethylene removal have a great potential on keeping freshness of fruit during export which required long-term transport under low humidity environment.

        Vertical equipment isolation using piezoelectric inertial-type isolation system

        Among anti-seismic technologies, base isolation is a very effective means of mitigating damage to structural and nonstructural components, such as equipment. However, most seismic isolation systems are designed for mitigating only horizontal seismic responses because the realization of a vertical isolation system (VIS) is difficult. The difficulty is primarily due to conflicting isolation stiffness demands in the static and dynamic states for a VIS, which requires sufficient rigidity to support the self-weight of the isolated object in the static state, but sufficient flexibility to lengthen the isolation period and uncouple the ground motion in the dynamic state. To overcome this problem, a semi-active VIS, called the piezoelectric inertia-type vertical isolation system (PIVIS), is proposed in this study. PIVIS is composed of a piezoelectric friction damper (PFD) and a leverage mechanism with a counterweight. The counterweight provides an uplifting force in the static state and an extra inertial force in the dynamic state; therefore, the effective vertical stiffness of PIVIS is higher in the static state and lower in the dynamic state. The PFD provides a controllable friction force for PIVIS to further prevent its excessive displacement. For experimental verification, a shaking table test was conducted on a prototype PIVIS controlled by a simple controller. The experimental results well agree with the theoretical results. To further investigate the isolation performance of PIVIS, the seismic responses of PIVIS were simulated numerically by considering 14 vertical ground motions with different characteristics. The responses of PIVIS were compared with those of a traditional VIS and a passive system (PIVIS without control). The numerical results demonstrate that compared with the traditional and passive systems, PIVIS can effectively suppress isolation displacement in all kinds of earthquake with various peak ground accelerations and frequency content while maintaining its isolation efficiency. The proposed system is particularly effective for nearfault earthquakes with long-period components, for which it prevents resonant-like motion.

        시토키닌 활성물질 Thidiazuron 처리가 포도 '거봉'(Vitis labruscana)의 果粒肥大 및 品質에 미치는 影響

        본 실험은 새로운 비 punne계 사이토카이닌 활성 물질인 thidiazuron(TDZ)이 포도 '거봉'의 과립 중 및 과실 품질에 미치는 영향을 조사하기 위하여 실시하였으며 대조 약제로 forchlorfenuron(CPPU)을 이용하였다. 생리적 낙화 후 5일의 TDZ를 처리는 과방의 무핵과립의 착립수를 증가시켜 과방중은 유의하게 증가하였으나 과립중은 증가하지 않았다. 그러나 낙화후 10 또는 15일 처리는 무핵과립의 착립수를 감소시켜 과립중이 유의하게 증가하였다. 따라서 TDZ처리는 생리적 낙과가 어느 정도 진행된 만개 10~15일 후에 처리하는 것이 유리하였다. GA 25mg·L^(-1)처리도 과립중 증가에 효과적이었으나 TDZ와 GA혼용 처리는 각 약제를 단용 처리할 때보다 과립중 증대 효과가 더욱 컸다. 두 약제간에는 TDZ가 대조약제인 CPPU가보다 과립비대 증진 효과가 더욱 컸으며 TDZ처리 농도가 20㎎·L^(-1)에서도 약해는 발생하지 않았으나 착색과 성숙은 더욱 지연되는 것으로 나타나 적정 처리농도는 5~10㎎·l^(-1)이었다. This experiment was conducted to find the effect of a non-purine cytokinin, thidiazuron(TDZ), on the increase of berry size and fruit quality in 'Kyoho' grapes. For the comparison, the effect of forchlorfenuron(CPPU) was also examined The number of shot berries per cluster was significantly increased by the application of TDZ at 5 days after calyptra fall, resulting in the increase of cluster weight but no increase of each berry weight. The increase of berry weight was found when TDZ was applied at 10 to 15 days after calyptra fall. Gibberellin(GA 25㎎·L^(-1)) was also effective on the increase berry weight and the combined application of GA and TDZ further increased the berry weight. TDZ showed stronger effect on the increase of berry weight than CPPU at the same concentration. Berry ripening was more delayed at higher concentration of TDZ but no chemical injury was observed even at 20㎎·L^(-1). Results indicated that TDZ was effective on the quality improvement through berry weight increase and the recommended concentration TDZ for 'Kyoho' gapes was 5 to 10㎎·L^(-1).

        A Novel Self-Healing Polyurethane with High Transparency and Strength: Effects of Multiple Supermolecular Forces

        It is a considerable challenge for expanding the applications of self-healing polyurethane because of the dilemma in balancing its mechanical and self-healing properties. Herein, a series of self-healing polyurethane systems (PUT-x) with short-range ordered rigid aromatic structures were synthesized by simply adjusting two kinds of diisocyanates, and by ingenious addition of aromatic chain extender. We hope to regulate the π-π interactions and further to manipulate the properties of obtained polyurethanes via adjusting the amount of the benzene rings in the molecular chains of the PUT-x systems. The PUT-5 with decent self-healing ability, optimal mechanical properties, acceptable thermal stability and favorable transparency was selected to incorporate liquid metal and fluorescent powder to fabricate conductive wire and anti-counterfeiting film. Significantly, the liquid metal could be recycled from the conductive wire conveniently without using strong acids or strong bases. Moreover, PUT-5 exemplarily withstood long-term soaking in normal saline, and no deformation could be identified.

        Five new anthraquinones from the seed of Cassia obtusifolia

        A phytochemical investigation on the seeds ofCassia obtusifolia led to the isolation of five new anthraquinones,including one aglycon and four glycosides. Thestructures were elucidated by analysis of extensive spectroscopicdata and the results of acid hydrolysis. All theseisolates were evaluated for their inhibitory effects againsta-glucosidase, and 1 showed potent activity with IC50value of 185 ± 15 lM.

      • An Adaptive Sliding Mode-Like P-type Iterative Learning Control for Robot Manipulators

        Iterative learning control(ILC) is used to track the trajectory of repetitive motion, especially for industrial robot. The proposed control scheme is based upon the feedback of tracking error and corresponding change rate, which construct a switching surface, with the use of a proportional structure, for which it is called sliding mode-like P-type ILC. In addition, an adaptive item was added to not only cope with the unknown parameters and disturbances but also. remove chatter phenomenon happening in control process. To verify the proposed control law, a conventional PD ILC and an adaptive PD ILC are added as comparison with all the same setting conditions. We use root mean square error (RSME) each iteration trial as performance evaluation index. Finally, simulation results are provided to illustrate the effectiveness of the proposed controller. As a result, it can achieve better performance.
