        Characterization and validation of an alternative reference bacterium Korean Pharmacopoeia Staphylococcus aureus strain

        The National Culture Collection of Pathogens (NCCP) is a microbial resource bank in Korea that collects pathogen resources causing infectious disease in human and distributes them for research and education. The NCCP bank attempts to discover strains with various characteristics and specific purposes to provide diverse resources to researchers. Staphylococcus aureus American Type Culture Collection (ATCC) 6538P is used as a reference strain in the microbial assay for antibiotics in the Korean and in the United States Pharmacopoeias. We aimed to analyze domestically isolated microbial resources from the NCCP to replace the S. aureus reference strain. Staphylococcus aureus strains were identified using matrix- assisted laser desorption/ionization time-of-flight mass spectrometry and the VITEK-2 system and characterized by multilocus sequence typing, 16S rRNA sequencing, and antibiotic susceptibility testing. Several candidate strains had similar characteristics as the reference strain. Among them, the nucleotide sequence of the 16S rRNA region of NCCP 16830 was 100% identical to that of the reference strain; it was sensitive to six types of antibiotics and showed results most similar to the reference strain. A validity evaluation was conducted using the cylinder-plate method. NCCP 16830 presented valid results and had the same performance as ATCC 6538P; therefore, it was selected as an alternative candidate strain.

        Transient Labor: Past and Present of Young Female White-collar Work Force in South Korea

        The paper aims to investigate transient labor pattern of young South Korean women by employing qualitative method, interwoven with the analysis of theoretical texts from Judith Butler and Cho Han Haejoang, and historiographic texts that show how female workers integrated into Korea`s patriarchal labor market. Women`s transient labor is analyzed though different historical figurations of `working` women, who constantly `do` gender, conforming to the State`s nation-family trope. With 1988 Equal Employment Law, accompanied by Seoul Olympics, the changes seemed to appear, however, contemporary corporate culture continues to marginalize female workers through taboo and sanctions, which shaped the performativity of idealized working women. In turn, office girls reenact the traditional feminine ideal, or become `flowers` in the offices. Some young Korean women enjoy relative freedom from being bread-winner and are allowed to pursue alternative career. Yet, the alleged female empowerment under recent regime of neoliberal self-portfolioization and so-called `spec` accumulation only helps the reproduction of female subalternity, which leads to the ques-tion of female education and its unfulfilled promises.

      • Influence on the performance of the transparent front surface layer by thermal annealing for SHJ solar cells

        We carried out the thermal annealing to improve the efficiency of SHJ solar cells. To find the optimal temperature, we have investigated for temperature and then ambient gas, respectively. For 150°C and Ar atmosphere, we can be obtained the constant values compared to as deposited sample. However, over 150°C, it was decreased to Hall mobility because of re-crystallinity. Therefore, thermal annealing of ITO and µc-SiO:H layers was contributed to improving FF for SHJ solar cells.

      • Rapid quantitative assessment of Salmonella contamination using real-time PCR assay

        Introduction: Salmonella infection poses a zoonotic threat in one health, and expeditious quantification of Salmonella cells in food and products may provide a useful toolkit for public health and animal industry. In this work, we utilized quantitative real time-PCR to estimate the number of Salmonella cells in a given sample, and attempted to validate the relationship between Cq value and DNA concentration with the number of existing bacterial cells. Materials and Methods: We used Salmonella Typhimurium ATCC 14028 to set a standard curve. After incubating overnight in BPW, cells were serially diluted in 1 ml saline. All diluents were plated on XLD agar plates, and subjected to genomic DNA extraction. DNA concentration for the sample from each diluent was measured using nano-drop. Quantitative real time-PCR was performed using DNA samples from all diluents and primers targeting invA gene. We then compared the Cq values with genomic DNA concentration and the cell number obtained by direct plating. Results: The Cq values increased reciprocally to the gDNA concentration and the number of ATCC 14028 cells in the diluents. Conclusions: Our results suggest reciprocal relationship between Salmonella gDNA concentration and the Cq value. Further investigations may include quantitative real time-PCR with diluted DNA samples.

        Evaluation of food effects on the pharmacokinetics of Pelargonium sidoides and Coptis with each bioactive compound berberine and epicatechin after a single oral dose of an expectorant and antitussive agent UI026 in healthy subjects

        UI026 is an expectorant and antitussive agent which is a new combination of Pelargonium sidoides extract and Coptis extract. The bioactive compounds of Pelargonium sidoides and Coptisextracts were identified as epicatechin and berberine, respectively. This study evaluated the effect of food on the pharmacokinetics (PKs) and safety of UI026. A randomized, openlabel, single-dose, 2-treatment, parallel study in 12 healthy male subjects was performed. Subjects received a single oral dose of UI026 (27 mL of syrup) under a fed or fasted condition according to their randomly assigned treatment. Blood samples for the PK analysis were obtained up to 24 hours post-dose for berberine and 12 hours post-dose for epicatechin. The PK parameters were calculated by non-compartmental analysis. In the fed condition, the mean maximum plasma concentration (C max ) and mean area under the plasma concentrationtime curve from time zero to the last observed time point (AUC last ) for berberine were approximately 33% and 67% lower, respectively, compared with the fasted condition, both showing statistically significant difference. For epicatechin, the mean C max and mean AUC lastwere about 29% and 45% lower, respectively, compared to the fasting condition, neither of which showed a statistically significant difference. There were no drug-related adverse events. This finding suggests that food affects the systemic exposure and bioavailability of berberine and epicatechin.

        Monitoring of Red Pepper Powder and Seasoned Red-Pepper Sauce using Species-Specific PCR in Conjunction with Whole Genome Amplification

        고추는 한국에서 매우 중요한 양념 중 하나이다. 하지만 수입 고춧가루와 다진 양념(다대기)에 부과되는 관세율(45%/270%)의 차이로 인해, 다진 양념이 수입된 후, 건 조 및 분쇄 과정을 거쳐 고춧가루로 제작되고 있는 실정 이다. 본 연구에서는 종 특이 PCR 기술과 whole-genome amplification 방법을 접목하여 고춧가루(N=45) 및 다진 양 념(N=5) 제품의 사용원료(고추, 마늘, 양파, 파, 생강)를 분 석하였다. 모니터링 결과, 39개 고춧가루 제품은 표시사항 을 준수하였으며, 6개 고춧가루 및 5개 다진 양념 제품은 제조 기준을 충족시키지 못했다. 따라서 분석 제품의 22% 가 표시사항을 준수하지 못한 것으로 밝혀졌으며, 본 연구에 사용한 분석 방법은 고춧가루 제품에 사용된 원료 분석에 적합한 방법임을 입증하였다. Red pepper is one of the most important spices popularly utilized in Korea. Because of the differences in tariff rates between red pepper powder and seasoned red-pepper sauce, seasoned red-pepper sauce is often therefore imported by consumers, then dried, ground, and added to red pepper powder for cost effective purposed to use the product the most effectively. In this study, we combined species-specific polymerase chain reaction (PCR) assays (for red pepper, garlic, onion, spring onion, and ginger) with whole-genome amplification (WGA). Thirty-nine red pepper powders were well in accordance with their labels. However, six red pepper powder and five seasoned redpepper sauce products failed to meet their compliance requirements. As a consequence, our monitoring results revealed that the overall mislabeling rate detected in this study was identified at 22%. Thus, our findings showed that the species-specific PCR in conjunction with WGA was an ideal method to identify raw materials that are used in the manufacturing of red pepper powder and seasoned red-pepper sauce.

      • Improving Hall mobility mechanism of sputtered AZO film for thin-film silicon solar cell application

        In this work, we carried out to reduce the optical absorption and to enhance Hall mobility in TCO film for improving optoelectrical properties. In terms of mean free path, we used for the varying distance between the target and substrate. To confirm improving crystallinity in AZO film, we have used the substrate temperature and then analyzed the XRD, film stress, EDAX, XPS, and PL, respectively.

        Utilization of Preventive Therapy in Korean Migraine Patients

        Background: Migraine is a common neurological disorder that affects the quality of life and causes several health problems. Preventive migraine treatment can reduce migraine frequency, headache severity, and health care costs. This study aimed to estimate the utilization of migraine preventive therapy and associated factors in eligible patients. Methods: We studied 534 patients with migraine who were eligible for migraine preventive therapy using 2017 National Patient Sample (NPS) data from the Health Insurance Review and Assessment Service (HIRA). We estimated the migraine days by calculating the monthly average number of defined daily dose (DDD) of migraine-specific acute drug. Patients with a monthly average number of DDD of 4 or more were considered as subjects for preventive treatment. Chi-square test and multiple logistic regression analysis were used to determine the association between the preventive therapy and the influencing variables. Results: Less than half of the eligible patients for prophylaxis (n=234, 43.8%) were prescribed preventive therapy. Multiple logistic regression results show that migraine preventive therapy was influenced by age, the type of migraine, and some comorbidities. Patients over the age of 50 tend to receive less prophylactic treatment than under the age of 40. On the other hand, migraine patients with epilepsy or depression were more likely to receive preventive therapy. Sumatriptan was the most preferred medication for acute treatment, and propranolol was the most commonly prescribed drug for prevention. Conclusions: More than half of the patients who were candidates for migraine prophylaxis were not receiving suitable preventive treatment. Positive factors affecting the use of migraine prevention were the presence of comorbidities such as epilepsy and depression.
