Maintenance of stem cell characteristics after long-term cryopreservation

The human eyelid adipose-derived stem cells (HEACs) are known as a candidate source for stem cell-based therapy. HEACs possess the ability to proliferate in vitro and multipotency to differentiate into adipogenic, osteogenic and chondrogenic cells. To be used later than the time of collection, a long-term storage is needed. In this study, we investigated stem cell characteristics after cryopreservation of HEACs for 6 months and 1 year in liquid nitrogen. Frozen-thawed stem cells have shown that cumulative cell and doubling numbers were similar to those of fresh HEACs. After thawing, HEACs expressed stem cell-related genes of SCF, NANOG, OCT4, and TERT, ectoderm-related genes of NCAM and FGF5, mesoderm/endoderm-related genes of CK18 and VIM. They also consistently expressed transcripts of the immune-related genes of HLA-ABC and β2M. To induce mesodermal differentiation, cell were cultivated in adipogenic, osteogenic or chondrogenic medium for 2~3 weeks. After each differentiation culture, HEACs expressed adipocyte-, osteocyte- and chondrocytespecific genes. They were also stained with Oil red O, von Kossa, or alcian blue, revealing adipogenic, osteogenic, or chondrogenic character, respectively. The results suggest that long-term storage up to 1 year do not affect their biological properties, HEACs may be suitable for clinical application on cell-based therapies.
Complete genome sequence of soybean dwarf virus infecting soybean (Glycine max L.)

We found a soybean plant displaying severe viral disease symptoms, such as stunting, rugosity, and yellowing. To reveal the viral agent, we extracted total RNA from the leaf sample and prepared a ribosome-deleted library followed by RNAsequencing. Raw data were de novo assembled, and the assembled contigs were used in a BLASTX search against a viral protein database. We identified four contigs associated with Soybean dwarf virus (SbDV) belonging to the genus Luteovirus in the family Luteoviridae. We obtained the complete genome of SbDV isolate HS with 5,686 nucleotides (nt), encoding five open reading frames (ORFs). The phylogenetic analysis showed that SbDV isolate HS belongs to Group A containing isolates from the USA and the Czech Republic. Here, we report the complete genome sequence of SbDV isolate HS infecting soybean in Korea obtained by RNA-sequencing.
Korean EFL Learners’ Use of Formulaic Language

This study aims to investigate the extent to which Korean EFL learners make use of formulaic language in comparison with that of English L1 speakers. With reference to the PHRASE List proposed by Martinez and Schmitt (2012), the corpora of speaking and writing samples of English L1 speakers and Korean EFL learners from the International Corpus Network of Asian Learners of English were analysed. The results show that Korean students use a comparatively small number of formulaic expressions, and use them far less frequently than L1 speakers in all the three categories examined: verb groups, modalization, and connectives. They also tend to use a few items overly frequently for an extended range of functions. These imply that formulaic language is an important factor in the variation and development of L2 communicative competence and therefore raising students’ awareness of its forms and functions should be an integral part of instruction.
Bioactivities and Potential Mechanisms of Action for Conjugated Fatty Acids

Since conjugated linoleic acid (CLA) was identified as a principal anticancer component from ground beef in the 1980s, CLA research has discovered that CLA has a wide range of biologically beneficial effects. Clinical studies with CLA are on the rise, and it is apparent that CLA may not be as effective in humans as in rodents, in particular its anti-obesity aspect. In addition, research with regard to other conjugated fatty acids as well as CLA metabolites is still in its infancy. Investigation of bioactivities for other conjugated fatty acids and CLA metabolites may help to extend the understanding of CLA and its mechanisms of actions. This may pose an opportunity to use CLA more efficiently and expand the future use of other conjugated fatty acids as pharmacological agents to assist current treatments.
Antioxidant Activity of Water Extract from Black Raspberry on C2C12 Muscle Cells

Black raspberry (Rubus occidentalis) is black-fruited raspberry native to north america and known to have various activities. This study investigated the antioxidant activities and intracellular ROS level of water extract from black raspberry (ROW) on C2C12 cells. Antioxidant activity was evaluated by 2,2"-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTs) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities. Total phenolic and flavonoid contents also measured. Intracellular ROS level was evaluated using DCF-DA method at concentrations of 200-500 μg/mL on H₂O₂-induced C2C12 myoblast cell. Intracellular ROS production showed higher level in the H₂O₂ treated group than control group and dose-dependent decrease in the sample group. These results suggest black raspberry water extract have antioxidant activity and effect against oxidative stress.
Surfactant-free Palladium Nanoparticles on Protein Particle Scaffold as High Performance Catalysts

Nano-biocomposite which is a combined material of metal nanoparticles and biomaterials has the great potential to be used as a good catalyst in various reactions because of its improved catalytic properties including high selectivity, durability, and possibility for tandom reactions. Here we report a synthesis and catalytic studies of palladium nanoparticles dispersed protein particles (PdNP-PPs). PdNP-PPs were easily synthesized by reducing Pd precursor in the presence of the protein particles. Due to the amine functional groups regularly expressed on the surface of the protein particles, the synthesized PdNP-PPs have well dispersed small and surfactant-free Pd nanoparticles on their surface. The PdNP-PPs exhibited high catalytic activity and good recyclability compared with polyvinylpyrrolidone (PVP)-coated Pd nanoparticles in the hydrogenation of p-nitrophenol into p-aminophenol because of clean and large surface area of Pd nanparticles and stable surface of charge of the protein particles.
