To identify novel virulence genes, a mutant library of Xanthomonas citri subsp. citri 29-1 was produced using EZ-Tn5 transposon and the mutants were inoculated into susceptible grapefruit. Forty mutants with altered virulence phenotypes were identified. Nine of the mutants showed a complete loss of citrus canker induction, and the other 31 mutants resulted in attenuated canker symptoms. Southern blot analysis revealed that each of the mutants carried a single copy of Tn5. The flanking sequence was identified by plasmid rescue and 18 different ORFs were identified in the genome sequence. Of these 18 ORFs, seven had not been previously associated with the virulence of X. citri subsp. citri and were therefore confirmed by complementation analysis. Real-time PCR analysis showed that the seven genes were upregulated when the bacteria were grown in citrus plants, suggesting that the expression of these genes was essential for canker development.

The indefinite linear quadratic (ILQ) optimal control problem for continuous-time descriptor Markov jump systems (DMJSs) is discussed in this work. Firstly, by using elementary linear algebra approach, the ILQ problem of DMJSs can be equivalent to standard LQ problem of Markov jump systems (MJSs) with some inequality and rank conditions. Then sufficient condition of the ILQ problem for DMJSs being solvable is obtained according to the LQ theory of MJSs, the optimal state feedback control is given, and the optimal cost value can be guaranteed to be nonnegative, moreover, the obtained optimal closed-loop system is stochastically admissible. Lastly, a numerical example is provided to verify the validity of the presented methods.

E-commerce represents the acquisition and sale, or the transmission of funds or data through an electronic platform. E-commerce is a paradigm shift that influences marketers and customers to improve current market processes. The significant challenges in e-commerce are the accuracy and performance factors during a business transaction, which has been substantially enhanced using Cloud Computing Techniques (CCT). The growth of e-commerce management has been increased due to massive internet penetration, and particularly small and emerging companies are increasingly using this alternative as a differentiated business model. E-commerce has significant environmental impacts and highly utilized in today’s market scenario. Further, the replacement has not been thoroughly explored. Current research has been carried out to describe the e-commerce scenario to analyze market trends. This study further discusses the essential variables to the performance of market models for e-commerce. For example, e-procurement of products/services, electronic supply chain management, e-distribution and selling support (supplier connections, e-fulfilment) and online e-auctions (transactional) can represent important e-commerce capabilities, which can contribute to marketing strategy implementation effectiveness, resulting in higher export performance.

Utilizing a computer-aided simulation approach, this work studies the impacts of different inter-stop transport distances on the transport efficiency evaluated by the transport time cost intensity of a typically composed Chinese freight train hauled by representative types of locomotives in view of different target speeds. It is found that the decelerated decrease of the transport time cost intensity with improving the target speed is much traded off by decreasing the stop-spacing especially below approximately 20.00 km. Moreover, the decrease of the inter-stop transport distance especially shorter than about 20.00 km obviously increases the transport time cost intensity in an accelerated way for the same target speed. Such a trend is more apparent when the target speed becomes relatively high. Therefore, it is suggested that the inter-stop transport distance of a freight train ought to be over 20.00 km for relatively high transport efficiency.

Background PTX is widely used in cancer treatments. Objective In this paper, we explored the role and potential molecular mechanism of propofol in regulating PTX sensitivity in PC cells. Methods Prostatic cancer cell line PC3 was treated using diferent concentrations of PTX (10 nM, 50 nM), propofol (150 μM, 300 μM) or transfected with overexpressed HOTAIR plasmid. HOTAIR expression was analyzed by RT-qPCR. Apoptosis of PC3 cells was observed by fow cytometry method while cell viability was evaluated by CCK-8. Moreover, apoptosis-related genes, Bcl-2 and Bax were detected by Western blot methods. E-cadherin, N-cadherin and Vimentin protein concentrations were monitored by ELISA. Results PTX signifcantly increased apoptosis of PC3 cells and reduced cell viability in a dose-dependent manner. Moreover, Protein expression of Bcl-2 was obviously inhibited while Bax protein expression level was provoked. Furthermore, E-cadherin protein concentration increased while N-cadherin and Vimentin decreased due to increasing PTX treatments. HOTAIR expression dropped due to PTX treatment while overexpression of HOTAIR induced cell viability, EMT and deterred apoptosis. Propofol ignited the PTX function while upregulation of HOTAIR partially reversed this. Conclusion Propofol enhanced paclitaxel sensitivity in prostatic cancer cells through modulation of HOTAIR in vitro.
Security Model of Stadium Evacuation Combined with Multi-agent and Cellular Automata

In order to design a reasonable pedestrian evacuation and exclude the security risks in large stadiums, this thesis proposes the research of security model of evacuation in stadiums combined with multi-agent and cellular automata. The research is based on cellular automata model and the process makes extended analysis to the cell’s behavior of autologous, and then it makes simulation experiments of the process of simulating the evacuation in large stadiums. Simulation results finally show that Agent-CA combines the advantages of multi-agent and cellular automata, which fully considers the individual internal factors. Compared to traditional cell cellular automata, it is more close evacuation situation of the realistic major sports stadium, and it shortens the evacuation time and improves the safety.

The performances of Constant Weights (CW) method (including Direct Feedback (DF) method as a special case), Method of Successive Averages (MSA) and Reversed MSA are comparatively studied in this research by utilizing a new four-step feedback model with the application of the person-trip data collected in Jabodetabek of Indonesia in 2002. It is found that both constant and changeable weights of these feedback solutions on new matrices should be at least no smaller than 0.50 to make the feedback procedure converge quickly and the application of the new feedback model based on the DF method converges most rapidly. Moreover, it is numerically confirmed that trip matrices from the applications of different feedback solutions to the feedback process will lead to a unique convergence in different efficiencies and the weights’ values over 0.50 on new matrices will also perform the unique convergence efficiently.
