        Development of seven SNP molecular markers based on the key genes related to red colour of fruit skin in Japanese apricot (Prunus mume)

        Fruit color is an important factor that aff ects its quality. The red-skinned cultivars of Japanese apricot ( Prunus mume Sieb. et Zucc.) have high medicinal economic benefi ts and are mainly used for deep-processed products because of their eye-catching red pigmentation. In contrast, molecular markers related to the red color trait in Japanese apricot fruit haven’t been reported. This study identifi ed twenty-eight SNPs markers based on genome resequencing data. The average number of alleles per SNP marker site in red and green-skinned groups of Japanese apricot cultivars was 1.9643, with heterozygosity ranging from 0.02 to 0.75, and an average Shannon index of 0.517 and 0.4420, respectively, indicating high dispersion and diversity. The total germplasm was divided into two (K = 2) clusters, including 10 and 34. Most red-skinned Japanese apricot cultivars were classifi ed as cluster I, showing the signifi cant genetic diff erence between the two cultivar groups ( Fst = 0.54). According to PIC values, a total of seven SNPs markers showed high polymorphism ( PIC > 0.5), including PmSNP_1 ( 4-CL ), PmSNP_4 ( WD40 ), PmSNP_5 ( MYB29 ), PmSNP_18 ( UFGT6 ) and PmSNP_27 ( UFGT3 ) that were highly correlated with red color trait. Among them, PmUFGT3 exhibited the highest polymorphism, which is signifi cant for future research on developing and utilizing germplasm resources related to red fruit skin.

        Treatment of oily wastewater from cold-rolling mill through coagulation and integrated membrane processes

        The feasibility of applying coagulation-integrated microfiltration (MF) as a pretreatment for an ultrafiltration (UF) feed in oily wastewater treatment was investigated. The effects of different coagulants on oil removal rates from wastewater were studied. The maximum oil removal rate of 82% was obtained after coagulation with 130 mg/L of polyaluminium chloride (PAC). UF flux reached 95 L/(㎡·h) with coagulation-integrated MF as pretreatment. This value was 2.5 times higher than that flux obtained without pretreatment. The value of UF flux increased as the transmembrane pressure (TMP) and cross-flow velocity (CFV) of the UF module increased. UF flux gradually increased when TMP and CFV exceeded 0.4 MPa and 3 m/s, respectively, because of concentration polarization and membrane fouling stabilization. Chemical oxygen demand reduction and oil removal rate reached 95.2% and 98.5%, respectively, during integrated membrane processing with a PAC concentration of 130 mg/L, TMP of 0.4 MPa, and CFV of 3 m/s for UF. In addition, sequentially cleaning the fouling membrane with NaOH and HNO₃ aqueous solutions caused UF flux to recover to 90%. These encouraging results suggested that the hybrid integrated membrane process-based coagulation and MF + UF are effective approaches for oily wastewater treatment.

      • Lentivirus-mediated Silencing of Rhomboid Domain Containing 1 Suppresses Tumor Growth and Induces Apoptosis in Hepatoma HepG2 Cells

        Rhomboids were identified as the first intramembrane serine proteases about 10 years ago. Since then, the study of the rhomboid protease family has blossomed. Rhomboid domain containing 1 (RHBDD1), highly-expressed in human testis, contains a rhomboid domain with unknown function. In the present study, we tested the hypothesis that RHBDD1 was associated with proliferation and apoptosis in hepatocellular carcinoma using recombinant lentivirus-mediated silencing of RHBDD1 in HepG2 cells. Our results showed that down-regulation of RHBDD1 mRNA levels markedly suppressed proliferation and colony formation capacity of HepG2 human hepatoma cancer cells in vitro, and induced cell cycle arrest. We also found that RHBDD1 silencing could obviously trigger HepG2 cell apoptosis. In summary, it was demonstrated that RHBDD1 might be a positive regulator for proliferative and apoptotic characteristics of hepatocellular carcinoma.

      • Nonsurgical correction of a severe anterior deep overbite accompanied by a gummy smile and posterior scissor bite using a miniscrew-assisted straight-wire technique in an adult high-angle case

        In the present report, we describe the successful use of miniscrews to achieve vertical control in combination with the conventional sliding MBT<SUP>TM</SUP> straightwire technique for the treatment of a 26-year-old Chinese woman with a very high mandibular plane angle, deep overbite, retrognathic mandible with backward rotation, prognathic maxilla, and gummy smile. The patient exhibited skeletal Class II malocclusion. Orthodontic miniscrews were placed in the maxillary anterior and posterior segments to provide rigid anchorage and vertical control through intrusion of the incisors and molars. Intrusion and torque control of the maxillary incisors relieved the deep overbite and corrected the gummy smile, while intrusion of the maxillary molars aided in counterclockwise rotation of the mandibular plane, which consequently resulted in an improved facial profile. After 3.5 years of retention, we observed a stable, well-aligned dentition with ideal intercuspation and more harmonious facial contours. Thus, we were able to achieve a satisfactory occlusion, a significantly improved facial profile, and an attractive smile for this patient. The findings from this case suggest that nonsurgical correction using miniscrew anchorage is an effective approach for camouflage treatment of high-angle cases with skeletal Class II malocclusion.

      • Association of Genetic Polymorphisms at 1q22 but not 10q23 with Gastric Cancer in a Southern Chinese Population

        Objective: Data from a recent genome-wide association studiesy of gastric cancer (GC) and oesophageal squamous cell carcinoma in Chinese living in the Taihang Mountains of north-central China suggest that 1q22 and 10q23 are susceptibility-associated regions for GC. However, this has not been confirmed in southern Chinese populations. The aim of this study was to investigate whether these polymorphisms at 1q22 and 10q23 are associated with the risk of GC in a southern Chinese population. Methods: We selected seven top significant associated single nucleotide polymorphisms (SNPs) at 1q22 and 10q23 and conducted a population-based case-control study in a southern Chinese population. Genotypes were determined using MassARRAYTM system (Sequenome, San Diego, CA). Results: Two SNPs at 1q22, rs4072037 and rs4460629, were significantly associated with a reduced risk of GC, best fitting the dominant genetic model. Logistic regression models adjusted for age and sex showed that rs4072037 AG and GG (OR=0.64, P=0.017, compared with AA) and rs4460629 CT and TT (OR=0.54, P=0.0016, compared with TT) significantly reduced the risk of GC. However, no significant results for the five SNPs at 10q23 were obtained in this study. Conclusion: These outcomes indicate that 1q22 is associated with GC susceptibility in this southern Chinese population, while an association for the locus at 10q23 was not confirmed.

      • Research on non-destructive testing technology for existing bridge pile foundations

        Pile foundations of existing bridges lie in soil and water environment for long term and endure relatively heavy vertical loads, thus prone to damages, especially after stricken by external forces, such as earthquake, collision, soil heap load and etc., and the piles may be injured to certain degrees as well. There is a relatively complete technical system for quality inspection of new bridge pile foundations without structures on the top. However, there is no mature technical standard in the engineering community for the non-destructive testing technology specific to the existing bridge pile foundations. The quality of bridge pile foundations has always been a major problem that plagues bridge maintenance. On the basis of many years' experiences in test engineering and theoretical studies, this study developed a new type of detection technology and equipment for the existing bridge piles.

        ASTROD-I is a mission concept under study to realize the general concept of ASTROD (Astrodynamical Space Test of Relativity using Optical Devices). This mission concept has one spacecraft carrying a payload of a telescope, ve lasers, and a clock together with ground stations (ODSN: Optical Deep Space Network) to test the optical scheme of interferometric and pulse ranging and yet give important scientic results. The scientic goals include a better measurement of the relativistic parameters, a better sensitivity in using optical Doppler tracking method for detecting gravitational waves, and measurement of many solar system parameters more precisely. The weight of this spacecraft is estimated to be about 300-350 kg with a payload of about 100-120 kg. The spacecraft is to be launched with initial period about 290 days and to pass by Venus twice to receive gravity-assistance for achieving shorter periods. With good orbit design, after about 370 days from launch, the spacecraft will arrive at the other side of the Sun for the determination of relativistic parameters.

        Impact of wilting and additives on fermentation quality and carbohydrate composition of mulberry silage

        Objective: The objective of this study was to investigate the effects of wilting and additives on the fermentation quality, structural and non-structural carbohydrate composition of mulberry silages. Methods: The selected lactic acid bacteria strains Lactobacillus plantarum 'LC279063' (L1), commercial inoculant Gaofuji (GF), and Trichoderma viride cellulase (CE) were used as additives for silage preparation. Silage treatments were designed as control (CK), L1, GF, or CE under three wilting rates, that is wilting for 0, 2, or 4 hours (h). After ensiling for 30 days, the silages were analyzed for the chemical and fermentation characteristics. Results: The results showed that wilting had superior effects on increasing the non-structural carbohydrate concentration and degrading the structural carbohydrate. After ensiling for 30 days, L1 generally had a higher fermentation quality than other treatments, indicated by the lower pH value, acetic acid, propionic acid and ammonia nitrogen (NH<sub>3</sub>-N) content, and the higher lactic acid, water soluble carbohydrate, glucose, galactose, sucrose, and cellobiose concentration (p<0.05) at any wilting rate. Wilting could increase the ratio of lactic acid/acetic acid and decrease the content of NH<sub>3</sub>-N. Conclusion: The results confirmed that wilting degraded the structural carbohydrate and increased the non-structural carbohydrate; and L1 exhibited better properties in improving fermentation quality and maintaining a high non-structural carbohydrates composition compared with the other treatments.

        On Deploying Relays for Connected Indoor Sensor Networks

        This paper considers the crucial problem of deployingwireless relays for achieving a connected wireless sensor networkin indoor environments, an important aspect related to the managementof the sensor network. Several algorithms have been proposedfor ensuring full sensing coverage and network connectivity. These algorithms are not applicable to indoor environments becauseof the complexity of indoor environments, in which a radiosignal can be dramatically degraded by obstacles such as walls.Wefirst prove theoretically that the indoor relay placement problem isNP-hard. We then predict the radio coverage of a given relay deploymentin indoor environments. We consider two practical scenarios;wire-connected relays and radio-connected relays. For thenetwork with wire-connected relays, we propose an efficient greedyalgorithmto compute the deployment locations of relays for achievingthe required coverage percentage. This algorithm is proved toprovide a Hn factor approximation to the theoretical optimum,whereHn = 1+ 12 +· · ·+ 1n = ln(n)+1, and n is the number ofall grid points. In the network with radio-connected relays, relayshave to be connected in an ad hoc mode. We then propose an algorithmbased on the previous algorithm for ensuring the connectivityof relays. Experimental results demonstrate that the proposedalgorithms achieve better performance than baseline algorithms.

        Reports : Aurora-A kinase-inactive mutants disrupt the interaction with Ajuba and cause defects in mitotic spindle formation and G2/M phase arrest in HeLa cells

        Aurora-A is a centrosome-localized serine/threonine kinase that is overexpressed in multiple human cancers. We previously reported an intramolecular inhibitory regulation of Aurora-A between its N-terminal regulatory domain (Nt, amino acids [aa] 1-128) and the C-terminal catalytic domain (Cd, aa 129-403). Here, we demonstrate that although both Aurora-A mutants (AurA-K250G and AurA-D294G/Y295G) lacked interactions between the Nt and Cd, they also failed to interact with Ajuba, an essential activator of Aurora-A, leading to loss of kinase activity. Additionally, overexpression of either of the mutants resulted in centrosome amplification and mitotic spindle formation defects. Both mutants were also able to cause G2/M arrest and apoptosis. These results indicate that both K250 and D294/Y295 are critical for direct interaction between Aurora-A and Ajuba and the function of the Aurora-A complex in cell cycle progression.
