        A Generalized Hidden Markov Model and Its Applications in Recognition of Cutting States

        In the traditional hidden Markov model (HMM) for statistical learning and classification, aleatory uncertainty because of randomness and epistemic uncertainty that is due to the lack of knowledge are not differentiated. In this paper, a generalized hidden Markov model (GHMM) in the context of generalized interval probability theory is proposed. The parameters of GHMM are in the form of generalized interval probability, which provides a concise representation for the two kinds of uncertainty simultaneously. The generalized versions of the forward-backward, Viterbi, and Baum-Welch algorithms in GHMM are developed. The proposed algorithms take advantage of the algebraic property in the generalized interval probability. These algorithms provide an efficient approach to train the GHMM. The trained GHMM is used for state recognition with the criterion of maximum log-likelihood. A case study on recognizing the cutting states in manufacturing processes is provided to demonstrate the application of GHMM. The interval forms of the cutting signal are considered as the inputting of GHMM. The cutting states are recognized based on the learning algorithm of GHMM. With the two uncertainty components quantified, the reliability of GHMM in state recognition is superior to that of HMM.

      • An Efficient and Robust Data Integrity Verification Algorithm Based on Context Sensitive

        There exist two key problems about data aggregation that should be thoroughly explored - algorithm design in networking layer, and algorithm design in application layer. Those two problems should be subtlety tackled in termers of high efficiency and robustness. Therefore, the former one requires the survivability and highly reliable design at networking layer, the latter one usually asks for high efficiency and robustness at application layer. Moreover, the optimization of algorithms is also considered for further enhancement. The integrity check is a key requirement for optimization. The context-aware and cross-layer design is applied in the optimization. A dynamic fragment odd-even parity checking code is proposed, and a context-aware aggregative integrity check code is proposed.

        IL-33 promotes IL-10 production in macrophages: a role for IL-33 in macrophage foam cell formation

        We evaluated the role of IL-10- in IL-33-mediated cholesterol reduction in macrophage-derived foam cells (MFCs) and the mechanism by which IL-33 upregulates IL-10. Serum IL-33 and IL-10 levels in coronary artery disease patients were measured. The effects of IL-33 on intra-MFC cholesterol level, IL-10, ABCA1 and CD36 expression, ERK 1/2, Sp1, STAT3 and STAT4 activation, and IL-10 promoter activity were determined. Core sequences were identified using bioinformatic analysis and sitespecific mutagenesis. The serum IL-33 levels positively correlated with those of IL-10. IL-33 decreased cellular cholesterol level and upregulated IL-10 and ABCA1 but had no effect on CD36 expression. siRNA-IL-10 partially abolished cellular cholesterol reduction and ABCA1 elevation by IL-33 but did not reverse the decreased CD36 levels. IL-33 increased IL-10 mRNA production but had little effect on its stability. IL-33 induced ERK 1/2 phosphorylation and increased the luciferase expression driven by the IL-10 promoter, with the highest extent within the − 2000 to − 1752 bp segment of the 5′-flank of the transcription start site; these effects were counteracted by U0126. IL-33 activated Sp1, STAT3 and STAT4, but only the STAT3 binding site was predicted in the above segment. Site-directed mutagenesis of the predicted STAT3-binding sites (CTGCTTCCTGGCAGCAGAA→CTGCCTGGCAGCAGAA) reduced luciferase activity, and a STAT3 inhibitor blocked the regulatory effects of IL-33 on IL-10 expression. Chromatin immunoprecipitation (CHIP) confirmed the STAT3-binding sequences within the − 1997 to − 1700 and − 1091 to − 811 bp locus regions. IL-33 increased IL-10 expression in MFCs via activating ERK 1/2 and STAT3, which subsequently promoted IL-10 transcription and thus contributed to the beneficial effects of IL-33 on MFCs.

        Kinetics of a Cloned Special Ginsenosidase Hydrolyzing 3-O-Glucoside of Multi-Protopanaxadiol-Type Ginsenosides, Named Ginsenosidase Type 3

        In this paper, the kinetics of a cloned special glucosidase, named ginsenosidase type III hydrolyzing 3-O-glucoside of multi-protopanaxadiol (PPD)-type ginsenosides, were investigated. The gene (bgpA) encoding this enzyme was cloned from a Terrabacter ginsenosidimutans strain and then expressed in E. coli cells. Ginsenosidase type III was able to hydrolyze 3-O-glucoside of multi-PPD-type ginsenosides. For instance, it was able to hydrolyze the 3- O-β-D-(1→2)-glucopyranosyl of Rb1 to gypenoside XVII, and then to further hydrolyze the 3-O-β-D-glucopyranosyl of gypenoside XVII to gypenoside LXXV. Similarly, the enzyme could hydrolyze the glucopyranosyls linked to the 3-O- position of Rb2, Rc, Rd, Rb3, and Rg3. With a larger enzyme reaction Km value, there was a slower enzyme reaction speed; and the larger the enzyme reaction Vmax value, the faster the enzyme reaction speed was. The Km values from small to large were 3.85 mM for Rc, 4.08 mM for Rb1, 8.85 mM for Rb3, 9.09 mM for Rb2, 9.70 mM for Rg3(S), 11.4 mM for Rd and 12.9 mM for F2; and Vmax value from large to small was 23.2 mM/h for Rc, 16.6 mM/h for Rb1, 14.6 mM/h for Rb3, 14.3 mM/h for Rb2, 1.81mM/h for Rg3(S), 1.40 mM/h for Rd, and 0.41 mM/h for F2. According to the Vmax and Km values of the ginsenosidase type III, the hydrolysis speed of these substrates by the enzyme was Rc>Rb1>Rb3>Rb2>Rg3(S)>Rd>F2 in order.

      • Spatial Orientation of Indoor Fire Ignition Based on Monocular Vision and Virtual Scene

        The spatial orientation of fire ignition can support an automatic fire suppression system (AFSS). However, due to the ambiguity of 2D images, most of the existing methods tend to study fire detection and cannot carry out spatial orientation. To solve this problem, we propose a spatial orientation method of indoor fire ignition based on monocular vision and virtual scenes. First, a hierarchical virtual scene construction method is proposed to realize the rapid construction of indoor scenes. Second, the characteristics of the fire are analyzed to obtain fire ignition in a 2D image. Finally, the spatial orientation of indoor fire ignition is calculated by analyzing the characteristics of fire attachment and the principle of three-dimensional imaging. The experimental results show that the absolute error of the spatial orientation of our method is 10.27 cm, and the relative error is 4.08%, which can meet the needs of AFSS.

      • Prognostic Role of Nucleophosmin in Colorectal Carcinomas

        Aim: Recent research suggests that nucleophosmin (NPM) may be a prognostic marker in colorectal carcinomas (CRC). We here tested its use to predict the survival of CRC patients. Methods: We investigated NPM expression by immunohistochemistry in histologically normal to malignant colorectal tissues and evaluated its association with clinicopathological variables. Overall and disease-free survival after tumor removal were calculated by the Kaplan-Meier method, and differences in survival curves were analyzed by the log-rank test. The Cox proportional hazards model was used for multivariate analysis of prognostic factors. Results: NPM expression was found significantly upregulated in CRC compared to adjacent colorectal tissue, villous adenoma, tubular adenoma and normal colorectal mucosa (p<0.05 for all). NPM expression was statistically linked to cancer embolus, lymph node metastasis, differentiation grade, and recurrence of CRC. Overall and disease-free survival of NPM-negative CRC patients tended to be better than those for patients with NPM-positive lesions (log-rank statistic, p<0.05 for all). Multivariate analysis indicated NPM expression as an independent prognostic indicator for CRC patients (p<0.05 ). Conclusion: Our results suggest that NPM expression can predict the survival of CRC patients. Prognosis of CRC is determined by not only many known prognostic factors but also by NPM expression.

      • A Systematic Review of Studies on Cyber Physical System Security

        Cyber-Physical System (CPS) is a system of systems which integrates physical system with cyber capability in order to improve the physical performance [1]. So far, it is being widely applied in areas closely related to national economy and people’s daily lives. Therefore, CPS security problems have drawn a global attention and an appropriate risk assessment for CPS is in urgent need. According to the researches and discussions in recent years, we believe that most researchers have already established a comprehensive understanding about CPS. This paper systematically introduced CPS’s conception, development and applications assisted. In addition, concerning about its aspects of safety and security, we also analyzed CPS’s risks and new requirements as an up-to-date technique brings. We elaborate the existing work and propose a research focus that has not been paid enough attention to, and proposed a security framework for CPS. At last, after providing a classic modeling and simulation method of CPS, we bring forward a new idea for accessing the experimental results into existing systems.

        Polymorphisms of melatonin receptor genes and their associations with egg production traits in Shaoxing duck

        Objective: In birds, three types of melatonin receptors (MTNR1A, MTNR1B, and MTNR1C) have been cloned. Previous researches have showed that three melatonin receptors played an essential role in reproduction and ovarian physiology. However, the association of polymorphisms of the three receptors with duck reproduction traits and egg quality traits is still unknown. In this test, we chose MTNR1A, MTNR1B, and MTNR1C as candidate genes to detect novel sequence polymorphism and analyze their association with egg production traits in Shaoxing duck, and detected their mRNA expression level in ovaries. Methods: In this study, a total of 785 duck blood samples were collected to investigate the association of melatonin receptor genes with egg production traits and egg quality traits using a direct sequencing method. And 6 ducks representing two groups (3 of each) according to the age at first eggs (at 128 days of age or after 150 days of age) were carefully selected for quantitative real-time polymerase chain reaction. Results: Seven novel polymorphisms (MTNR1A: g. 268C>T, MTNR1B: g. 41C>T, and g. 161T>C, MTNR1C: g. 10C>T, g. 24A>G, g. 108C>T, g. 363 T>C) were detected. The single nucleotide polymorphism (SNP) of MTNR1A (g. 268C>T) was significantly linked with the age at first egg (p<0.05). And a statistically significant association (p<0.05) was found between MTNR1C g.108 C>T and egg production traits: total egg numbers at 34 weeks old of age and age at first egg. In addition, the mRNA expression level of MTNR1A in ovary was significantly higher in late-mature group than in early-mature group, while MTNR1C showed a contrary tendency (p<0.05). Conclusion: These results suggest that identified SNPs in MTNR1A and MTNR1C may influence the age at first egg and could be considered as the candidate molecular marker for identify early maturely traits in duck selection and improvement.

        Single-cell RNA sequencing analysis of human bone-marrow-derived mesenchymal stem cells and functional subpopulation identification

        Mesenchymal stem cells (MSCs) are a common kind of multipotent cell in vivo, but their heterogeneity limits their further applications. To identify MSC subpopulations and clarify their relationships, we performed cell mapping of bone-marrow-derived MSCs through single-cell RNA (scRNA) sequencing. In our study, three main subpopulations, namely, the stemness subpopulation, functional subpopulation, and proliferative subpopulation, were identified using marker genes and further bioinformatic analyses. Developmental trajectory analysis showed that the stemness subpopulation was the root and then became either the functional subpopulation or the proliferative subpopulation. The functional subpopulation showed stronger immunoregulatory and osteogenic differentiation abilities but lower proliferation and adipogenic differentiation. MSCs at different passages or isolated from different donors exhibited distinct cell mapping profiles, which accounted for their corresponding different functions. This study provides new insight into the biological features and clinical use of MSCs at the single-cell level, which may contribute to expanding their application in the clinic.
