It has been reported that tight sportswear could have complicated influence on physiological function of human body. The purpose of this present study was to investigate the effect of wearing gradient compression tights (GCT) on muscle strength and EMG activity during repeated isokinetic muscle contractions. Four healthy male undergraduate students performed maximal voluntary isokinetic concentric muscle contractions on biomechanical test and training systems with GCT and loose pants as control (Cont) respectively. During each test, the peak torque of extensor and flexor contractions and the surface electromyography (sEMG) of the recrus femoris and medial gastrocnemius was recorded simultaneously, the peak torque was recorded as an indicator of muscle strength, and the average amplitude and mean power frequency of sEMO were calculated as indicators of EMG activity. The results showed that: the peak torque decreased gradually during continuous muscle contractions both when the Cont and GCT were worn. average sEMG and mean power frequency declined along with the repetitions of muscle contractions for both wearing conditions, and the change tendency was consistence with that of peak torque. There was no obvious, difference between the peak torque recorded wearing the Cont or wearing GCT, but when GCT were worn, average sEMG was lower and mean power frequency was higher than the Cont condition. In 24 samples obtained from four subjects, 80% of results showed the same trend. So we could make a conclusion that wearing ocr had no obvious effect on the improvement of muscle strength, but it would affect the EMO activity posiuvly.
ZNF424, a novel human KRAB/C2H2 zinc finger protein, suppresses NFAT and p21 pathway

Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]

Detection of the number of vegetative cells and endospores is necessary for quality control during the production of orally administered probiotic Bacillus licheniformis-containing tablets (BCT). However, there is no standard method for the rapid detection of vegetative cells and endospores in China. In this study, a simple flow cytometry (FCM) method was used to monitor the population dynamics of BCT. Using a specific fluorescent stain, SYBR green I, flow cytometric analysis could easily differentiate two morphological states of B. licheniformis. Compared with plate count assay (PCA) for determining the number of vegetative cells and endospores, the percentage of endospores determined by FCM was ~10% higher than that by PCA. Advantages of the FCM method over conventional methods include lower labor work, shorter detection time, and higher accuracy. Therefore, this simple FCM method could be a practical tool for monitoring quality control during the production of probiotic BCT

Two kinds of colossal electroresistance (CER) cells, Ag/Pr$_{0.7}$Ca$_{0.3}$MnO$_{3}$/Pt and In/Pr$_{0.7}$ Ca$_{0.3}$MnO$_{3}$/Pt, were prepared to study the memory function. Different reversible-resistance-switching phenomena were observed, depending on the applied electrodes. The I-V characteristics of the two cells have opposite hysteresis properties. The power consumptions and the threshold voltages were also investigated. In particular, the Ag/Pr$_{0.7}$Ca$_{0.3}$MnO$_{3}$/Pt cell with an Ohmic contact interface exhibited better resistance switching retention and small power consumption. In contrast, the In/Pr$_{0.7}$Ca$_{0.3}$MnO$_{3}$/Pt cell with a Schottky contact interface exhibited poor resistance switching retention and large power consumption. The results suggest that the CER cell with an Ohmic contact is better for the resistance memory function.

A fibrinolytic enzyme was produced by an edible mushroom of Pleurotus ostreatus using submerged culture fermentation. The enzyme was purified from the culture supernatant by applying a combination of freeze-thaw treatment, ammonium sulfate precipitation, hydrophobic interaction, and gel filtration chromatographies. The enzyme was purified by a 147-fold, with a yield of 7.54%. The molecular masses of the enzyme an determined by gel filtration and SDSPAGE were 13.6 and 18.2 kDa, respectively. The isoelectric point of the enzyme was 8.52. It hydrolyzed fibrinogen by cleaving the α and β chains of fibrinogen followed by the γ chains, and also activated plasminogen into plasmin. The enzyme was optimally active at 45°C and pH 7.4. The enzyme activity was completely inhibited by EDTA, whereas protease inhibitors of TPCK, SBTI, PMSF, aprotinin and pepstatin showed no inhibition on its activity. The partial amino acid sequences of the enzyme as determined by Q-TOF2 were ATFVGCSATR, GGTLIHESSHFTR, and YTTWFGTFVTSR. These sequences showed a high degree of homology with those of metallo-endopeptidases from P. ostreatus and Armillaria mellea. The purified enzyme can also be applied as a natural agent for oral fibrinolytic therapy or prevention of thrombosis.

In order to study the damage mechanism of surrounding rocks under water-rock coupling, an experimental study on the microscopic law of water absorption and mechanics of sandy mudstone was carried out in the sandy mudstone stratum of Wanfu coal mine in Shandong, China. The results indicate that the strength of sandy mudstone exhibits three stages of relative softening, rapid softening, and decelerated softening. The speed of water molecules entering the microscopic pore structure was analyzed based on the percentage of water molecules in the pore structure. The acoustic emission energy release process is divided into energy accumulation stage, stable release stage and intense release stage. With the increase of water content saturation, (Spectral width of the multifractal spectrum) tends to decrease and the distribution of events is gradually uniform. The action of water gradually expands and penetrates the cracks and pores inside the sandy mudstone, which makes f (Frequency of occurrence of the signal) change from negative to positive values.
Characterization of the fluoridating process of hydroxyapatite

F-substituted hydroxyapatites were synthesized by chemical precipitation method using Ca(CH3COO)2·H2O and KH2PO4 asreactants and KF·2H2O as fluorination agent. Fourier transform infrared spectrometer (FTIR), X-ray diffraction (XRD) andX-ray fluorescence spectrometer (XRF) were used to characterize samples. The results showed that the more fluorine wasadded the more OH ions replaced by F. Moreover, OH-F bond was formed between crystalline, and the bond was strongerthan that of original OH-OH, so the grains of hydroxyapatite grew larger and the crystallinity increased as F content increased.
