Oxygen tank for synergistic hypoxia relief to enhance mitochondria-targeted photodynamic therapy

Background: Mitochondria play an essential role in cellular redox homeostasis maintenance and meanwhile serve as an important target for organelle targeted therapy. Photodynamic therapy (PDT) is a promising strategy for organelle targeted therapy with noninvasive nature and highly spatiotemporal selectivity. However, the efficacy of PDT is not fully achieved due to tumor hypoxia. Moreover, aerobic respiration constantly consumes oxygen and leads to a lower oxygen concentration in mitochondria, which continuously limited the therapeutic effects of PDT. The lack of organelle specific oxygen delivery method remains a main challenge. Methods: Herein, an Oxygen Tank is developed to achieve the organelle targeted synergistic hypoxia reversal strategy, which not only act as an oxygen storage tank to open sources and reduce expenditure, but also coated with red blood cell membrane like the tank with stealth coating. Within the oxygen tank, a mitochondrion targeted photosensitizer (IR780) and a mitochondria respiration inhibitor (atovaquone, ATO) are co-loaded in the RBC membrane (RBCm) coated perfluorocarbon (PFC) liposome core. Results: Inside these bio-mimic nanoparticles, ATO effectively inhibits mitochondrial respiration and economized endogenous oxygen consumption, while PFC supplied high-capacity exogenous oxygen. These Oxygen modulators reverse the hypoxia status in vitro and in vivo, and exhibited a superior anti-tumor activity by mitochondria targeted PDT via IR780. Ultimately, the anti-tumor effects towards gastric cancer and colon cancer are elicited in vivo. Conclusions: This oxygen tank both increases exogeneous oxygen supply and decreases endogenous oxygen consumption, may offer a novel solution for organelle targeted therapies.
SIRT3-Mediated Dimerization of IDH2 Directs Cancer Cell Metabolism and Tumor Growth

<P>These findings identify SIRT3 as a potential tumor suppressor, mediating its effects by limiting the ability of IDH2 to drive cancer cell metabolism and malignant progression.</P><P>The isocitrate dehydrogenase IDH2 produces α-ketoglutarate by oxidizing isocitrate, linking glucose metabolism to oxidative phosphorylation. In this study, we report that loss of SIRT3 increases acetylation of IDH2 at lysine 413 (IDH2-K413-Ac), thereby decreasing its enzymatic activity by reducing IDH2 dimer formation. Expressing a genetic acetylation mimetic IDH2 mutant (IDH2<SUP>K413Q</SUP>) in cancer cells decreased IDH2 dimerization and enzymatic activity and increased cellular reactive oxygen species and glycolysis, suggesting a shift in mitochondrial metabolism. Concurrently, overexpression of IDH2<SUP>K413Q</SUP> promoted cell transformation and tumorigenesis in nude mice, resulting in a tumor-permissive phenotype. IHC staining showed that IDH2 acetylation was elevated in high-risk luminal B patients relative to low-risk luminal A patients. Research on Grey Predictive Control of PMSM Based on Reduced-order Luenberger Observer

To further improve the dynamic and anti-interference performance of permanent magnet synchronous motor (PMSM) control system, a dual closed loop model predictive control (MPC) strategy combining with grey prediction and reduced-order Luenberger observer is proposed in this paper for speed and current control. To be specifi c, fi rstly, the dual closed loop MPC strategy is introduced based on the mathematical model of PMSM, that is, the model predictive controller is used for the outer loop of speed, and the deadbeat predictive controller for the inner loop of current. Secondly, considering the grey prediction method can establish grey model and whiten the system grey quantity to some extent, it is used in the PMSM control system to improve the dynamic performance. Finally, to solve the problem that the motor is easy to be aff ected by disturbances, a reduced-order Luenberger observer is designed to combine with the model predictive controller to improve the system anti-interference performance. The simulation results show that the dual MPC strategy based on grey prediction makes the system more stable and dynamic response faster, while the dual MPC strategy combining with the grey prediction and the reduced-order Luenberger observer makes the system more resistant to load disturbance.

As a degenerative joint disease, osteoarthritis (OA) constitutes a major cause of disability that seriously affects the quality of life of a large population of people worldwide. However, effective treatment that can successfully reverse OA progression is lacking until now. The present study aimed to determine whether two small non-coding RNAs miR-29a and miR-140, which are significantly down-regulated in OA, can be applied together as potential therapeutic targets for OA treatment. MiRNA synergy score was used to screen the miRNA pairs that potentially synergistically regulate OA. An in vitro model of OA was established by treating murine chondrocytes with IL-$1{\beta}$. Transfection of miR-29a and miR-140 via plasmids was investigated on chondrocyte proliferation and expression of nine genes such as ADAMTS4, ADAMTS5, ACAN, COL2A1, COL10A1, MMP1, MMP3, MMP13 and TIMP metallopeptidase inhibitor 1 (TIMP1). Western blotting was used to determine the protein expression level of MMP13 and TIMP1, and ELISA was used to detect the content of type II collagen. Combined use of miR-29a and miR-140 successfully reversed the destructive effect of IL-$1{\beta}$ on chondrocyte proliferation, and notably affected the MMP13 and TIMP1 gene expression that regulates extracellular matrix. Although co-transfection of miR-29a and miR-140 did not show a synergistic effect on MMP13 protein expression and type II collagen release, but both of them can significantly suppress the protein abundance of MMP13 and restore the type II collagen release in IL-$1{\beta}$ treated chondrocytes. Compared with single miRNA transfection, cotransfection of both miRNAs exceedingly abrogated the suppressed the protein production of TIMP1 caused by IL-$1{\beta}$, thereby suggesting potent synergistic action. These results provided1novel insights into the important function of miRNAs' collaboration in OA pathological development. The reduced MMP13, and enhanced TIMP1 protein production and type II collagen release also implies that miR-29a and miR-140 combination treatment may be a possible treatment for OA.

We introduce the fundamental theorem of upper and lower solutions for a class of singular ($p_1,\;p_2$)-Laplacian systems and give the proof by using the Schauder fixed point theorem. It will play an important role to study the existence of solutions.
Multiplicity results of positive solutions for singular generalized Laplacian systems

We study the homogeneous Dirichlet boundary value problem of generalized Laplacian systems with a singular weight which may not be in $L^{1}$. Using the well-known fixed point theorem on cones, we obtain the multiplicity results of positive solutions under two different asymptotic behaviors of the nonlinearities at $0$ and $\infty$. Furthermore, a global result of positive solutions for one special case with respect to a parameter is also obtained.
