Hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths, but its molecular mechanisms are not yet well characterized. Long noncoding RNAs (lncRNAs) play crucial roles in tumorigenesis, including that of HCC. However, the role of homeobox A11 antisense (HOXA11-AS) in determining HCC stem cell characteristics remains to be explained; hence, this study aimed to investigate the effects of HOXA11-AS on HCC stem cell characteristics. Initially, the expression patterns of HOXA11-AS and HOXA11 in HCC tissues, cells, and stem cells were determined. HCC stem cells, successfully sorted from Hep3B and Huh7 cells, were transfected with short hairpin or overexpression plasmids for HOXA11-AS or HOXA11 overexpression and depletion, with an aim to study the influences of these mediators on the self-renewal, proliferation, migration, and tumorigenicity of HCC stem cells in vivo. Additionally, the potential relationship and the regulatory mechanisms that link HOXA11-AS, HOXA11, and the Wnt signaling pathway were explored through treatment with Dickkopf-1 (a Wnt signaling pathway inhibitor). HCC stem cells showed high expression of HOXA11-AS and low expression of HOXA11. Both HOXA11-AS silencing and HOXA11 overexpression suppressed the self-renewal, proliferation, migration, and tumorigenicity of HCC stem cells in vivo, as evidenced by the decreased expression of cancer stem cell surface markers (CD133 and CD44) and stemness-related transcription factors (Nanog, Sox2, and Oct4). Moreover, silencing HOXA11-AS inactivated the Wnt signaling pathway by decreasing the methylation level of the HOXA11 promoter, thereby inhibiting HCC stem cell characteristics. Collectively, this study suggested that HOXA11-AS silencing exerts an antitumor effect, suppressing HCC development via Wnt signaling pathway inactivation by decreasing the methylation level of the HOXA11 promoter.
Vibration attenuation in periodic composite Timoshenko beams on Pasternak foundation

Periodic and quasi-periodic Timoshenko beams on Pasternak foundation are investigated using the differential quadrature method. Not only band gaps in the beams but also the dynamic response of them is analyzed. Numerical results show that vibration in periodic beams can be dramatically attenuated when the exciting frequency falls into band gaps. Different from the band structures of periodic beams without foundation, the so-called critical frequency was found because of the Pasternak foundation. Its physical meaning was explained in detail and a useful formula was given to calculate the critical frequency. Additionally, a comprehensive parameter study is conducted to highlight the influence of foundation modulus on the band gaps.
Molecular and Biochemical Characterization of a Novel Intracellular Low-Temperature-Active Xylanase

A 990 bp full-length gene (xynAHJ2) encoding a 329- residue polypeptide (XynAHJ2) with a calculated mass of 38.4 kDa was cloned from Bacillus sp. HJ2 harbored in a saline soil. XynAHJ2 showed no signal peptide, distinct amino acid stretches of glycoside hydrolase (GH) family 10 intracellular endoxylanases, and the highest amino acid sequence identity of 65.3% with the identified GH 10 intracellular mesophilic endoxylanase iM-KRICT PX1-Ps from Paenibacillus sp. HPL-001 (ACJ06666). The recombinant enzyme (rXynAHJ2) was expressed in Escherichia coli and displayed the typical characteristics of low-temperatureactive enzyme (exhibiting optimum activity at 35 o C, 62% at 20 o C, and 38% at 10 o C; thermolability at ≥45 o C). Compared with the reported GH 10 low-temperature-active endoxylanases, which are all extracellular, rXynAHJ2 showed low amino acid sequence identities (<45%), low homology (different phylogenetic cluster), and difference of structure (decreased amount of total accessible surface area and exposed nonpolar accessible surface area). Compared with the reported GH 10 intracellular endoxylanases, which are all mesophilic and thermophilic, rXynAHJ2 has decreased numbers of arginine residues and salt bridges, and showed resistance to Ni 2+ , Ca 2+ , or EDTA at 10 mM final concentration. The above mechanism of structural adaptation for low-temperature activity of intracellular endoxylanase rXynAHJ2 is different from that of GH 10 extracellular low-temperature-active endoxylanases. This is the first report of the molecular and biochemical characterizations of a novel intracellular low-temperatureactive xylanase.

Statistical experimental designs combined with solvent engineering for optimization of enzymatic synthesis of L-ascorbyl palmitate were developed. First, the composition of the solvent for co-dissolving polar and apolar substrates was determined. The co-solvent mixture of tert-pentanol:DMSO at a ratio of 9:1 (v/v) and the optimal biocatalyst were obtained. Then, the Plackett-Burman design was implemented to screen the variables that significantly influence the conversion. The method of steepest ascent was used to approach the proximity of optimum. After determining the Plackett-Burman and steepest ascent designs, the optimum values were determined by central composite design under response surface methodology. The statistical analysis showed that the optimum reaction conditions (temperature 50oC, enzyme concentration 5.8 g/L,and substrate molar ratio 11:1, stirring rate 160 rpm, amount of molecular sieve 50 g/L, time 18 h) led to the maximum conversion (66.44%) and production concentration (20.63 g/L). A very satisfactory conversion (64.74%) and production concentration (20.13 g/L) could be achieved in short time (6 h).
Deep Local Multi-level Feature Aggregation Based High-speed Train Image Matching

At present, the main method of high-speed train chassis detection is using computer vision technology to extract keypoints from two related chassis images firstly, then matching these keypoints to find the pixel-level correspondence between these two images, finally, detection and other steps are performed. The quality and accuracy of image matching are very important for subsequent defect detection. Current traditional matching methods are difficult to meet the actual requirements for the generalization of complex scenes such as weather, illumination, and seasonal changes. Therefore, it is of great significance to study the high-speed train image matching method based on deep learning. This paper establishes a high-speed train chassis image matching dataset, including random perspective changes and optical distortion, to simulate the changes in the actual working environment of the high-speed rail system as much as possible. This work designs a convolutional neural network to intensively extract keypoints, so as to alleviate the problems of current methods. With multi-level features, on the one hand, the network restores low-level details, thereby improving the localization accuracy of keypoints, on the other hand, the network can generate robust keypoint descriptors. Detailed experiments show the huge improvement of the proposed network over traditional methods.

The importance of information technology (IT) has been emphasized in developing countries recently. Despite the importance of the topic area of the productivity of IT in developing countries, the literature to date is relatively sparse. The findings of almost all these studies are based on data collected in developed countries. Few studies have been conducted to validate these results and to see if they are still applicable in developing countries. This study tries to investigate the effects of IT investment on productivity in the electronics industry of China, which is a representative developing country, with production function model. The results show that there is a positive impact of IT investment on productivity in China similar to previous studies in developed countries.
