        Group B Streptococcus Vaccines: Progress and Potential

        Group B Streptococcus (GBS, Streptococcus agalactiae) is a pathogen that causes sepsis and meningitis, particularly in newborns, as well as severe infections in the elderly and those at high risk. For many years, the administration of intrapartum antimicrobial prophylaxis (IAP) has been a standard method to prevent neonatal GBS infection. However, IAP may be unsuitable in low-income settings due to its high cost and difficult accessibility to medical institutions. Additionally, IAP may lead to the emergence of antimicrobial-resistant bacteria. Hence, an alternative method for the control of GBS, such as a vaccine, is needed. An effective vaccine will likely prevent the further spread of GBS and be cost-effective compared with IAP. GBS vaccines have been under development for the past two decades, and several candidates have shown potential. In this review, we discuss the current development of GBS vaccines, including types and their implementation in different target populations.

        Engineering an auto-activated R protein that is in vivo activated by a viral protease

        <P>Autonomous hypersensitive responses (self-HRs) are caused by constitutively active R proteins. In this study, we identified an auto-activated form of the R gene Pvr9 (autoPvr9); the auto-activation results from an amino acid substitution between its NBS and LRR domains. Self-HR was strongly reduced or completely inhibited by fusion of an extra peptide to the autoPvr9 N-terminal or C-terminal, respectively. When an NIa recognition site was placed between autoPvr9 and the extra peptide, the fusion construct could trigger an NIa-dependent HR. Several C-terminal fusions were tested, but only those that maintained detectable protein expression were capable of an NIa-dependent HR. Our results suggest the potential for transforming malfunctioning and auto activated R proteins into a new construct targeting potyviral NIa proteases.</P>

      • The Role of Obeticholic Acid (Farnesoid X receptor agonist) on Improving Liver Biochemistry Parameters in Primary Biliary Cholangitis: A Systematic Review and Meta-Analysis

        Aims: Primary biliary cholangitis (PBC) is a chronic, progressive, autoimmune cholestatic liver disease that leading to end stage liver disease and death. Higher levels of Y-glutamiltransferase (GGT), liver enzyme and bilirubin levels correlated with disease progression and predictive value of survival outcomes. Obeticholic acid (OCA) has been reported to have great potency in activating FXR which will protect hepatocyte against bile acid toxicity. The aim of this study was to assess the effect of obeticholic acid (OCA) on liver biochemistry parameters in PBC based on randomized control trials. Methods: Literature search was conducted using PubMed database until April 2020 to find randomized control trial (RCTs), which assessed 10 mg obeticholic acid administration on liver enzyme, Y-glutamiltransferase (GGT) and conjugated bilirubin in PBC patients. Treatment effect were considered as Mean difference and standard deviation (SD) change from the baseline. We performed data analysis using RevMan 5.3. Results: A total of 2 trials (211 participants) were included in the meta-analysis. The result suggested that obeticholic acid has significant effect on improving ALT(-15.2U/L,95%CI:-19.58 to -10.66; P<0.00001), ALT(-9.12U/L,95%CI:-12.23 to -6.02; P<0.00001), GGT(-99.70U/L, 95%CI: -125.68 to -73.71; P<0.00001) and Conjungated bilirubin (-0.08U/L, 95%CI: -0.13 to -0.03; P<0.002). Conclusions: Administered of 10 mg Obeticholic acid improved liver biochemistry parameters in PBC. However, further study with large-scale and better design are needed to confirm the results and eliminated the bias.

      • Efficacy and Safety of Dacomitinib in Advance Non Small Cell Lung Cancer (NSCLC): Systematic Review and Meta Analysis

        Background EGFR activating mutations are common oncogenic driver in patients with non-small-cell lung cancer (NSCLC). Dacomitinib is a pan-HER inhibitor that irreversibly binds to EGFR, HER2 and HER4 tyrosine kinase. The Aim of this study was to assess the efficacy and safety dacomitinib in NSCLC patients. Methods Literature search was conducted using PubMed, Cochrane and scopus database until August 2020 to find randomized control trial (RCTs), which assessed Dacomitinib administration on NSCLC patients. The outcomes of dacomitinib efficacy included overall survival and median progression free survival (hazard ratio 95%CI). The most common adverse event were measured by Risk Ratio (RR) and 95%CI. We performed data analysis using RevMan 5.3. Results A total of 4 RCTs involved 1240 intervention and 998 control participants. The Result showed that dacomitinib group improved progression free survival compare to control group significantly (HR 0.71, 95%CI: 0.56 to 0.90; P=0.004 I2=79%). There is no significant effect dacomitinib gorup on overall survival (0.83, 95%CI, 0.59 to 1.18; P=0.30 I2=87%). The most common grade >=3 Adverse event were Diarrhea (RR 6.01 95%CI: 2.36 to 15.34; P=0.0002 I2=55%) and paronychia (RR 4.20 95%CI: 1.88 to 9.36; P=0.0005 I2=0%) Conclusions Dacomitinib improve progression free survival but not overall survival in NSCLC patients. However, further studies with larger scale and better designs are needed to confirm the Results and eliminate bias.

        Isolation and validation of a candidate <i>Rsv3</i> gene from a soybean genotype that confers strain-specific resistance to soybean mosaic virus

        <P>Soybean mosaic virus (SMV), a member of the genus Potyvirus, significantly reduces soybean production worldwide. Rsv3, which confers strain-specific resistance to SMV, was previously mapped between the markers A519F/R and M3Satt in chromosome 14 of the soybean [Glycine max (L.) Mem] genotype L29. Analysis of the soybean genome database revealed that five different NBS-LRR sequences exist between the flanking markers. Among these candidate Rsv3 genes, the full-length cDNA of the Glyma.14g204700 was successfully cloned from L29. Over-expression of Glyma.14g204700 in leaves inoculated with SMV inhibited viral infection in a soybean genotype lacking Rsv3. In addition, the transient silencing of the candidate gene caused a high accumulation of an avirulent strain in L29 carrying Rsv3. Our results therefore provide additional line of evidence to support that Glyma.14g204700 is likely Rsv3 gene that confers strain-specific resistance to SMV.</P>

        The Effect of Trust and Customer Value on Loyalty: An Empirical Study in Indonesia

        The world of tourism is a tangible form of a global business trip, which is very promising because it will be estimated to be a sector capable of driving world economic growth resulting from tourists’ ever-increasing movement. Currently, in Indonesia, marketing activities in this sector are an integrated process, where the main target is providing customer satisfaction. Thus, the company’s marketing activities are expected to be in accordance with customer expectations. The essential purpose of this study was to analyze the mediating role of the effect of trust and customer value on loyalty. The population of this study was users of the Lentera Wisata service in Semarang City, Indonesia. The sampling used a purposive sampling method, with the criteria of male and female customers and customers who have used the Lentera Wisata Semarang service bureau more than once. The test employed multiple regression analyses and the Sobel test. The test results revealed that trust and customer value positively and significantly affected satisfaction. However, trust did not affect customer loyalty. Meanwhile, customer value and satisfaction significantly and positively influenced loyalty. Additionally, the Sobel test results revealed that satisfaction mediated the effect of trust and customer value on customer loyalty.

        A plant intron enhances the performance of an infectious clone <i>in planta</i>

        <P><B>Abstract</B></P> <P>Although infectious clones are fundamental tools in virology and plant pathology, their efficacy is often reduced by the instability of viral sequences in <I>Escherichia coli</I>. In this study, we constructed an infectious clone of PepMoV (pPepMoV) in a bacterial binary vector (pSNU1); the clone induces symptoms of PepMoV in agroinfiltrated plants. During its modification and maintenance in <I>E. coli</I>, however, the pPepMoV infectious clone was instable in the bacteria. Manipulation of this unstable clone in the bacterial strain DH10B led to the spontaneous formation of a recombined clone with high stability in the bacteria but with reduced infectivity due to an unwanted insertion of an <I>E. coli</I> sequence in the NIa-protease coding region. Replacement of this sequence with a plant intron restored infectivity and maintained plasmid stability. In addition to restoring plasmid growth in both <I>E. coli</I> and <I>Agrobacterium</I>, the presence of the intron in the PepMoV sequence enhanced the accumulation of PepMoV in agroinfiltrated leaves and resulted in symptom induction in upper systemic leaves that was nearly as strong as with PepMoV sap-inoculation. Plant introns have been previously used to stabilize plasmids in <I>E. coli</I> without any effect or with an unexpected lag in symptom development. In contrast, the current results demonstrated the in vivo enhancement of an infectious clone by a plant intron.</P> <P><B>Highlights</B></P> <P> <UL> <LI> An infectious clone of PepMoV in a bacterial binary vector was constructed. </LI> <LI> The pPepMoV infectious clone was instable and lost infectivity during manipulation. </LI> <LI> During its propagation in <I>E. coli</I>, the pPepMoV infectious clone spontaneously acquired a bacterial transposon. </LI> <LI> A plant intron inserted in the pPepMoV restored infectivity and maintained plasmid stability. </LI> <LI> We have demonstrated the in vivo enhancement of an infectious clone by a plant intron. </LI> </UL> </P>
