The unreduced (2n) gametes have long been used in triploid breeding of citrus. In lemon, the previously reported mechanisms for 2n megagametophyte formation were controversial, whereas that for 2n pollen production is still unknown. Herein, the frequency of and mechanism underlying 2n megagametophyte and 2n pollen formation in ‘Eureka’ lemon were investigated based on cytological observation and genotyping of the triploid hybrids between ‘Eureka’ lemon and ‘Early gold’ sweet orange. As a result, 4.79% of the viable pollens of ‘Eureka’ lemon were identified as the 2n pollen with a larger diameter (70.16 ± 3.92 μm). The 2n pollen might be resulted from the formation of parallel spindles at meiosis stage II. Among the 204 plantlets regenerated from embryo rescue following the sexual cross, 12 were triploids as identified by flow cytometry. According to the analysis of heterozygosity transmission using 13 pericentromeric single nucleotide polymorphism (SNP) markers and 20 randomly distributed simple sequence repeat (SSR) markers, 11 triploids were identified to be originated from the fertilization of 2n megagametophytes of ‘Eureka’ lemon, with a frequency of 5.39%. Among them, nine 2n megagametophytes were supposed to be arisen from the second division restitution (SDR), whereas the other two were from postmeiotic genome doubling (PMD). These results to understand the mechanism underlying 2n gamete formation in lemon are valuable for its efficient polyploid breeding.
Comparison of stomach microRNA transcriptomes of Tibetan and Yorkshire pigs by deep sequencing

MiRNAs regulate the expression of target genes in diverse cellular processes and hence play important roles in different physiological processes, yet little is known about the stomach microRNAome (miRNAome) of the Tibetan pig. The objective of this experiment was to investigate differentially expressed stomach miRNAs participating in digestion. Firstly, we isolated total RNA by Trizol reagent from three Tibetan and three Yorkshire purebred pigs stomach samples at 90-day-old. Secondly, a comprehensive analysis of Tibetan and Yorkshire pig stomach miRNAomes was performed by small RNA sequencing in the Illumina HiSeq 2000 system. Finally, SYBR Green Real-time RT-PCR was performed to validate the differentially expressed miRNAs. We identified 318 unique miRNAs, 260 were co-expressed in both libraries, 17 and 31 miRNAs were specifically expressed in Tibetan and Yorkshire pigs respectively. Fifty six differentially expressed miRNAs were identified by the identifying differentially expressed genes 6 (IDEG6). Kyoto encyclopedia of genes and genomes analysis revealed that some of the differentially expressed miRNAs were associated with protein and fat digestion. Two differentially expressed miRNAs (miR-214-3p and ssc-un39) participating in the digestion of lipid were identified. Additionally, qRT-PCR results suggested that a higher expression of miR-214-3p in the Tibetan pig stomach could lead to relatively lower expression of calcium-dependent phospholipase A2, which is an enzyme important for the digestion of glycerol phospholipid. This study has delineated the different stomach miRNAs expression patterns of Tibetan and Yorkshire pigs, which would help explain the regulatory mechanisms of miRNAs in digestion of Tibetan pigs, and contribute to utilize a the unique digestion merits of Tibetan pig in future porcine hybridization breeding.

Cold water paddy field soils are relatively unproductive, but can be ameliorated by supplementing with inorganic fertilizer from animal waste-based composts. The yield of two rice cultivars was significantly raised by providing either chicken manure or cow dung-based compost. The application of these composts raised the soil pH as well as both the total nitrogen and ammonium nitrogen content, which improved the soil’s fertility and raised its nitrification potential. The composts had a measurable effect on the abundance of nitrogencycling- related soil microbes, as measured by estimating the copy number of various bacterial and archaeal genes using quantitative real-time PCR. The abundance of ammonia oxidizing archaea and bacteria was markedly encouraged by the application of chicken manure-based compost. Supplementation with the composts helped promote the availability of soil nitrogen in the cold water paddy field, thereby improving the soil’s productivity and increasing the yield of the rice crop.

In the present work, the infl uence of various retrogression treatments on hardness, electrical conductivity and mechanicalproperties of a high Zn-containing Al–Zn–Mg–Cu alloy is investigated and several retrogression regimes subjected to a samestrength level are proposed. The precipitates are qualitatively investigated by means of transmission electron microscopy(TEM) and high-resolution transmission electron microscopy techniques. Based on the matrix precipitate observations, thedistributions of precipitate size and nearest inter-precipitate distance are extracted from bright-fi eld TEM images projectedalong ⟨110⟩ Al orientation with the aid of an imaging analysis and an arithmetic method. The results show that GP zonesand η′ precipitates are the major precipitates and the precipitate size and its distribution range continuously enlarge with theretrogression regime expands to an extent of high temperature. The nearest inter-precipitate distance ranges obtained arequite the same and the average distance of nearest inter-precipitates show a slight increase. The infl uence of precipitates onmechanical properties is discussed through the interaction relationship between precipitates and dislocations.
Self‐assembly of covalent porphyrin compound and its enhanced electrochemiluminescence performance

A novel Zn-coordination covalent porphyrin assembly (TCPP-BZA-Zn) is designed. The assembly structure is synthesized through the amidation reac- tion between the porphyrin terminal carboxyl group and the amino group of benzylamine (BZA), and further assembled through π–π stacking. In particular, the inherently ordered structure of TCPP-BZA-Zn with Zn as the catalytic active center endows the porphyrin assembly structure with several obvious advantages, such as high ion transport properties and high electrocatalytic per- formance. In the presence of hydrogen peroxide as a co-reaction reagent, TCPP-BZA-Zn/GCE showed excellent ECL behavior. The amplification phenome- non of ECL was further studied by cyclic voltammetry and the corresponding mechanism was proposed. Based on TCPP-BZA-Zn, an electrochemilumines- cence sensor was constructed for copper ion detection. The ECL intensity of the sensor shows a good linear relationship with the concentration of copper ion in the range of 10 nM–1 mM, and the detection limit is 1.3 nM.
Research and Implementation of Android Embedded Code Generation Method based on Rule Model

Based on the in-depth study of the Android embedded application program frame and project file, this paper proposes a code generation method based on model rule, the method discusses the problem of the layout of the Android embedded application interface, defines the constraint rules between the components and component object model and event model. And code generation rules on the Android embedded applications are defined and used throughout the code generation process, it solves the visualization development of Android embedded application program and the code automatic generation. Finally, a simple examples is given to verify the process of automatic generation of Android embedded code.

Precipitation characteristics influencing fatigue crack propagation contained matrix precipitate, grain boundary precipitateand precipitate free zone for Al–Zn–Mg–Cu alloys. Over-aging treatment could effectively regulate precipitation and then tobe able to change fatigue crack propagation behavior compared with the peak aging state. In the current work, typical T651and T7651 aging tempers of the alloy were extracted via hardness, electrical conductivity and mechanical properties underone-step and two-step aging treatments. Fatigue crack propagation (FCP) rate under them was tested and correspondingprecipitation characteristics and fracture morphology were observed. The results indicated that fatigue crack propagationresistance for the T7651 temper possessed an obvious improvement on the side of that for the T651 temper, which was alsosupported by fracture appearance, including tearing ridge, tearing dimple and fatigue striation. The precipitation observationshowed that the T651 alloy contained GPI zone, GPII zone and ηʹ phase while the T7651 alloy possessed ηʹ phase and η phase.Compared with the T651 temper, matrix precipitate for the T7651 temper distinctly owed an expanding of size distributionand an enlargement of average size while cuttable phase still remained the dominance for both tempers. Grain boundaryprecipitate and precipitate free zone manifested no obvious difference between the two aging tempers. Cut and bypass mechanismsof dislocation–precipitate interactions were used for explanation and it revealed the reinforced cuttable phase was infavor of enhancing fatigue crack propagation resistance. A theoretical model which directly correlated FCP rate with matrixprecipitate characteristics was employed to calculate FCP rate by substituting quantitative precipitate characteristics and thecalculation results were vaguely consistent with the experimental measurement, which proved its reliability and feasibility.

Background: Synaptic plasticity contributes to nociceptive signal transmission and modulation, with calcium/ calmodulin-dependent protein kinase II (CaMK II) playing a fundamental role in neural plasticity. This research was conducted to investigate the role of CaMK II in the transmission and regulation of nociceptive information within the nucleus accumbens (NAc) of naïve and morphine-tolerant rats. Methods: Randall Selitto and hot-plate tests were utilized to measure the hindpaw withdrawal latencies (HWLs) in response to noxious mechanical and thermal stimuli. To induce chronic morphine tolerance, rats received intraperitoneal morphine injection twice per day for seven days. CaMK II expression and activity were assessed using western blotting. Results: Intra-NAc microinjection of autocamtide-2-related inhibitory peptide (AIP) induced an increase in HWLs in naïve rats in response to noxious thermal and mechanical stimuli. Moreover, the expression of the phosphorylated CaMK II (p-CaMK II) was significantly decreased as determined by western blotting. Chronic intraperitoneal injection of morphine resulted in significant morphine tolerance in rats on Day 7, and an increase of p-CaMK II expression in NAc in morphine-tolerant rats was observed. Furthermore, intra-NAc administration of AIP elicited significant antinociceptive responses in morphine-tolerant rats. In addition, compared with naïve rats, AIP induced stronger thermal antinociceptive effects of the same dose in rats exhibiting morphine tolerance. Conclusions: This study shows that CaMK II in the NAc is involved in the transmission and regulation of nociception in naïve and morphine-tolerant rats.
