        Hypoxia-induced PLOD2 regulates invasion and epithelial-mesenchymal transition in endometrial carcinoma cells

        Background Procollagen-lysine, 2-oxoglutarate 5-dioxygenase 2 (PLOD2) was induced in hypoxia and participated in cancer development. However, the role of PLOD2 in endometrial carcinoma remains unclear. Objective To explore the influences and regulation mechanism of PLOD2 in endometrial carcinoma under hypoxic condition. Methods The small interfering RNA (siRNA) targeting to PLOD2 and pcDNA3.1-PLPD2 were transfected to endometrial carcinoma cells to alter PLOD2 expression. Cell proliferation ability was determined by colony formation assay. Wound healing assay used to detect cell migration ability. Transwell invasion assay was used to detect cell invasion ability. Results PLOD2 and Hypoxia-inducible factor-1α (HIF-1α) were induced by hypoxia. Down-regulation of PLOD2 did not affect endometrial carcinoma cell proliferation ability, while inhibited cell migration, invasion under hypoxic condition. Besides, down-regulation of PLOD2 increased the levels of γ-catenin and E-cadherin and decreased levels of Fibronectin and Snail under hypoxic condition. Down-regulation of PLOD2 also inactivated Src and phosphoinositide 3-kinase (PI3K)/ protein kinase B (Akt) signaling under hypoxic condition. The promoting effects of PLOD2 overexpression on migration, invasion and epithelial-mesenchymal transition (EMT) of endometrial carcinoma cells were reversed by Akt inhibitor (MK2206) under hypoxic condition. Conclusion PLOD2 expression was increased in endometrial carcinoma cells under hypoxic condition. PLOD2 modulated migration, invasion, and EMT of endometrial carcinoma cells via PI3K/Akt signaling. PLOD2 may be a potential therapeutic target for endometrial carcinoma.

        Temporal Variations in Seaweed Biomass and Coverage in Korean Coasts: Ongdo, Chungnam

        Temporal variations of seaweed biomass and coverage were seasonally examined at Ongdo in the Yellow Sea, Korea from August 2006 to April 2008. Average seaweed biomass was 245.79 g/$m^2$ in wet weight and coverage was 16.49% with seasonal variations from 13.97% in spring to 18.55% in autumn. Seaweeds were distributed across the shore gradient from the high intertidal to 10m depth in the subtidal zone. Biomass was always higher in the subtidal zone (310.24 g/$m^2$) than in the intertidal zone (181.35 g/$m^2$). Of total seaweed biomass, 76.52% (first year) and 80.32% (second year) occurred from the low intertidal zone, down to depth of 1 to 5m. Gelidium amansii had the highest importance value and biomass, and sub dominant species were Chondrus ocellatus and Chondria crassicaulis. Coarsely-branched seaweeds comprised the highest proportion of biomass (214.84 g/$m^2$, or 87.41% of the total biomss). Seasonal variations in algal biomass were largely explained by fluctuations in the biomass of coarsely-branched and thick-leathery forms. In conclusion, seaweed biomass of Ongdo shore was very low because of perennial G. amansii showing low biomass as compared to kelp or Sargassum spp. However, these results indicate Ongdo is good place to grow seaweeds because coarsely-branched form seaweeds including G. amansii are dominant at unpolluted and clean environment.

        Flow performance of cryomodules in C-ADS InjectorⅡ

        Two β=0.10 cryomodules are required for the China Accelerator Driven Subcritical System (C-ADS) injector II accelerator. Flow design is of great importance in the performance of cryomodules, including thermal design, flow distribution, pressure dropand so on. This paper will study convection heat transfer of helium and relation among the pipe diameter, mass flow rate andReynolds number. Furthermore, the influence of flow geometries on pressure drop and flow distribution will also be done. It wasfound that the theoretical flow distribution were in good agreement with the experimental data.

      • Structural Conservation and Food Habit-related Liver Expression of Uncoupling Protein 2 Gene in Five Major Chinese Carps

        The full-length cDNA of grass carp (Ctenopharyngodon idellus) and silver carp (Hypophthalmichthys molitrix) uncoupling protein 2 (UCP2) was obtained from liver. The grass carp UCP2 cDNA was determined to be 1152 bp in length with an open reading frame that encodes 310 amino acids. Five introns (Intron 3, 4, 5, 6 and 7) in the translated region, and partial sequence of Intron 2 in the untranslated region of grass carp UCP2 gene were also obtained. Gene structure comparison between grass carp and mammalian (human and mouse) UCP2 gene shows that, the UCP2 gene structure of grass carp is much similar to that of human and mouse. Partial UCP2 cDNA sequences of bighead carp (Aristichthys nobilis) and mud carp (Cirrhinus molitorella), were further determined. Together with the common carp (Cyprinus carpio) UCP2 sequence from GenBank (AJ243486), multiple alignment result shows that the nucleotide and amino acid sequences of the UCP2 gene, were highly conserved among the five major Chinese carps that belong to four subfamilies. Using beta-actin as control, the ratio UCP2/beta-actin mRNA (%) was determined to be $149.4{\pm}15.6$ (common carp), $127.4{\pm}22.1$ (mud carp), $96.7{\pm}12.7$ (silver carp), $94.1{\pm}26.8$ (bighead carp) and $63.7{\pm}16.2$ (grass carp). The relative liver UCP2 expression of the five major Chinese carps, shows a close relationship with their food habit: benthos and detrituseating fish (common carp and mud carp) > planktivorious fish (silver carp and bighead carp) > herbivorious fish (grass carp). We suggest that liver UCP2 might be important for Chinese carps to detoxify cyanotoxins and bacteria in debris and plankton food.

      • Sleep Duration and Cancer Risk: a Systematic Review and Meta-analysis of Prospective Studies

        To assess the risk of cancers associated with sleep duration using meta-analysis of published cohort studies, we performed a comprehensive search using PubMed, Embase and Web of Science through October 2013. We combined hazard ratios (HRs) from individual studies using meta-analysis approaches. A random effect dose-response analysis was used to evaluate the relationship between sleep duration and cancer risk. Subgroup analyses and sensitivity analyses were also performed. Publication bias was evaluated using Funnel plots and Begg's test. A total of 13 cohorts from 12 studies were included in this meta-analysis, which included 723, 337 participants with 15, 156 reported cancer outcomes during a follow-up period ranging from 7.5 to 22 years. The pooled adjusted HRs were 1.06 (95% CI: 0.92, 1.23; P for heterogeneity =0.003) for short sleep duration, 0.91 (95% CI: 0.78, 1.07; P for heterogeneity <0.0001) for long sleep duration. In subgroup analyses stratified by cancer type, long duration of sleep showed an inverse relation with hormone-related cancer (HR=0.79; 95% CI: 0.65, 0.97; P for heterogeneity =0.009) and a greater risk of colorectal cancer (HR=1.29; 95% CI: 1.09, 1.52; P for heterogeneity =0.346). Further meta-analysis on dose-response relationships showed that the relative risks of cancer were 1.00 (95% CI: 0.99, 1.01; P for linear trend=0.9151) for one hour of sleep increment per day, and 1.00 (95% CI: 0.98, 1.01; P for linear trend=0.7749) for one hour of sleep increment per night. No significant dose-response relationship between sleep duration and cancer was found on non-linearity testing (P=0.5053). Our meta-analysis suggests a positive association between long sleep duration and colorectal cancer, and an inverse association with incidence of hormone related cancers like those in the breast. Studies with larger sample size, longer follow-up times, more cancer types and detailed measure of sleep duration are warranted to confirm these results.

        Amphiphilically modified chitosan copolymer for enhanced oil recovery in harsh reservoir condition

        A novel amphiphilically grafting natural chitosan copolymer (PAMCS) was prepared by using one stepwater-free radical polymerization strategy and the basic parameters for PAMCS were systematicallycharacterized. The rigid D-glucosamine unit, intermolecular association and hydrogen bondingsynergistically endowed PAMCS solution with rheological properties, super-high salt resistance andtemperature tolerance in harsh reservoir condition. PAMCS solution exhibited viscoelastic behaviorand formulated the unique displacement mechanism in comparison to that for partially hydrolyzedpolyacrylamide. Moreover, the mass concentration, injective rate and polymer solution slug related to oilrecovery efficiency for PAMCS were investigated from the economic point.

        Polyaniline-modified graphitic carbon nitride as electrode materials for high-performance supercapacitors

        Graphitic carbon nitride (g-C3N4) has attracted extensive attention in energy storage due to its suitable and tunable bandgap, high chemical/thermal stability, earth abundance and environmental friendliness. However, its conductivity should be improved to work as the electrode materials in supercapacitors. In this report, we have prepared a two-dimensional composite (CN-PANI) based on g-C3N4 and polyaniline (PANI) by in-situ polymerization, which can be efficiently applied as electrode material for supercapacitors. The introduction of PANI can increase the conductivity of the electrode, and the porous structure of g-C3N4 can provide enough channels for the transport of electrolyte ions and improve the electrode stability. As a result, the obtained CN-PANI demonstrates excellent specific capacitance (234.0 F g−1 at 5 mV/s), good rate performance and high cycling stability (86.2% after 10,000 cycles at 50 mV/s), showing great potential for high-rate supercapacitors.
