        Induction and Purification of a Thermophilic Chitinase Produced by Aeromonas sp. DYU-Too7 Using Glucosamine

        In the presence of chitin, Aeromonas sp. DYU-Too7 can produce extra-cellular, chitin-degrading enzymes. Chitin analogues and other carbon sources can be used to cultivate this bacterial strain. The chitinases produced by the strain were higher in the GIcN (glucosamine) medium than those in other media. The maximal chitinase activity occurred in the medium containing 0.1% GlcN. Cultivation of Aeromonas sp. DYU-Too7 in the GlcN medium sped up the chitinase production; however, the same result did not appear when it was cultivated in the (Chitin + GlcN) medium. This result may indicate that GlcN can be utilized by Aeromonas sp. DYU-Too7 as a carbon source and an inducer to produce chitinases. A chitinase with a molecular mass of 36 kDa was further purified and characterized to have an optimal reacting pH of 5.0 and an optimal reacting temperature of $50^{\circ}C$. This chitinase showed high stability in the proximity of $30^{\circ}C$ and also high stability in the proximity of pH 7.0. The hydrolysates of colloidal chitin, with the aid of the 36-kDa chitinase, were analyzed by an HPLC and found to be chitobiose.

      • 추상적 환경창조를 위한 De Stijl건축의 협력작업에 관한 연구

        세계 1차 대전을 전후해서 거의 동시대에 출현한 다양한 Avant-Garde예술운동들은 더 나은 사회의 건설과 그 배경요소로서의 이상적 환경창조를 목표로 다양한 예술분야 사이의 협력작업을 수행했다. 본 연구는 이러한 운동들 증에서 추상적 환경을 이상적 배경으로 간주하고 이의 창조를 위한 건축가와 화가 사이의 협력작업을 하나의 이상으로 추구한 De Stijl의 건축을 중심으로 전개과정에서 나타난 추상적 환경창조를 위한 협력작업의 시기별 양상과 이에 따른 작품변화를 통해 양자의 상관관계를 규명하였다. 추상적 환경창조를 위한 De Stijl건축의 협력작업은 양상과 건축적 결과를 기준으로 제1(1917~21), 2(1922~23), 3기(1924~31)의 3시기로 구분할 수 있다. 제1기에 화가는 공간적 효과의 창출에 있어 건축가에게 종속되었다. 제2기는 화가가 건축가의 권한을 초월하는 권리를 확보한 시기로서 추상적 환경형성을 위한 기반이 조성되었다. 제3기는 모든 협력작업의 중단 후에 건축가와 화가가 추상적 환경창조를 위한 독자적인 탐구를 모색한 시기이다. Before and after the First World War various Avant-Garde movements appeared simultaneously and it accomplished collaborative works between various parts of art which aimed the establishement of better society and the creation of ideal environment as a supporting element. Among these movements, De Stijl regared the abstact environment as an ideal background. In order to create the abstract environment, De Stijl continously pursued collaborative works between architects and painters as an ideal. The purpose of this study is to examine the architectural characteristics of De Stijl focused on the changes of collaborative aspects and buildings appeared in development process of collaborative works for creation of abstract environment.

      • Oud의 집합주택에서 나타나는 공동체의식 제고를 위한 계획기법에 관한 연구

        요약문본 연구는 공동체의 구성요소로서의 집합주택의 의미와 20세기 초엽의 시대적 상황 과 사회문제 해결에의 요구를 분석하고, 그에 대한 건축적 대응으로 간주되는 당시의 많은 공동체 중에서 가장 현실적인 사례인 네델란드 건축가 Oud의 집합주택으로 동동체적 성격 의 규명과 함께 거주자들의 공동체의식 제고를 위한 다양한 계획기법을 고찰하므로써 현재 의 상황에 적용가능한 하나의 선례적 모델로 제시하고자 하였다. Oud는 자신의 집합주택단 지에서 외부와의 접촉을 단절하는 내향적·폐?거 단지배치와 공동생활을 지원하는 공동시 설의 부여, 구분의 폐지와 생활양식을 동질화 시키는 단위주호의 반복, 차별적인 외형의 modernity를 통한 정체성의 부여와 같은 계획기법을 통하여 집합주택 거주자들의 공동체의 식을 제고하고 있으며, 이러한 계획기법들은 시대적·공간적 간극에 대한 보충연구를 통해 현재의 상황에 적용 할 수 있는 선례로 고려할 만한 가치를 지닌 것으로 사료된다.AbstractThe purpose of this study is to consider the planning technique for uplifting of sense of the community in social housings of J. J. P. Oud, and to present the result as a precedential model which is able to apply in the current situation. Oud uplifted the sense of community of residents through the means of introversive and closed block arrangement, endowment of communal facilities, repetition of unit and abolition of discrimination, and endowment of identity through the modernity of facade.

        The purpose of this study is to analyze Frank Lloyd Wright's architectural characteristics and constitution of Plan in his Houses. Frank Llod Wright was unswerving in his architectural idea through his long career and exercised his influence over many other Architects. He discovered the essential elements of architectural consitution in American tradition and developed the elements into his houses. Through this study, We could interprete the characteristics of his Organic Architecture and wanted to seek interrelationship between his Organic Architecture and elements of Architectural constitution expressed in his building plans.

        Through quantitative analysis of vertical circulation space as the subject of study, This paper; first; examine the characteristics of human behavior in terms of proxemics second; select the prototypes of action by analyzing human activity through Ergonomics third; classify human body dimensions according to action concerned with the basis of anthropometrics By synthesizing above considerations, element of spatial form, which has physical fitness to human behavior in vertical circulation space, could be analogized such as follows; * Decision of optimal width and handrail form according to space conditions (Fig; 9, 10, 11) * Decison of the slope and form decreasing energy consumption and stress of a pedestrian, that is, the relation between riser and tread (Fig.12, Table 4) Finally, I think that the comprehensive study on vertical circulation space, including qualitative analysis excluded in this paper is needed.

      • 機能主義 理論에 따른 建物 內·外部 構成에 關한 硏究

        This study is to re-interpret the concepts of functionalism in process of dialectical development, and to present the possibilities in the way that functionalism can be accommodated in contemporary architecture. In the course of analyzing functionalist theory functionalism could be classified into the rationalistic and geometric functionalism based on logos and the romantic and organic functionalism based on pathos, and architectural characteristics in the state of modern and present situations be presented. Particularly, functionalist principles in the internal and external compositions of building through examinations on two typical Architect's works could be established. In the result, functionalist approach in contemporary architecture are developed in harmony with geometric and organic approaches, and then the adaptable approaches founded on the morality of geometric functionalism and the reconstructed approaches of organic functionalism on the basis of currently functional needs are thought to be the sources that can solve architectural problems at this time.

        Analysing all the problems with which Gyongsan campus of Y. N. University is confronted in the academic and physical aspects, Authors can set a core of each academic zone and introduce a Campus System that includes the concept of multiversity. This thesis deals with site planning as a basic study for the construction of multi-purpose typed 'Convention Center' which may be the most critical of the cores. Informations that is acquired from this thesis would be a useful guideline for the possible building design.

        This study is concerned with the transmission method of architectural meanings on the basis of semiotics. Architectural meanings changed by various causes during their transmitting through the communication channel shown in fig 3 are arrived at user's brain in various situations of receiving information. For perceiving those relationships further more in details it is necessary to analysis following items. 1. Architectural signs: Index, Icon, Iconic symbol, Irrelative symbol. 2. Causes of meaning change: Signal, Social, Historical, Psychological, Foreign, Creative cause. 3. Contents of architectural meaning: Referential, Informative, Expressive, Aesthetical, Intentional content. 4. Situations of receiving information: Direct, Creative, Intentionaly metaphorical, Extentional, Metaphorical, Paradoxical, Reductive situation. We can find out that above several elements are ordered in architecture according to human needs.

      • 現代建築의 콘텍스츄얼리즘的 傾向에 관한 硏究

        The pluralism of the contemporary architecture is an environment to solve the problems presented the criticism of the Modern Movement and a mechanism to get a new standard of value with the transformation of the Modern Architecture's ethical concept into the problem of meaning. Contextualism, A methodology of architectural conditions, is represented in two ways such as the philosophical and the historical movement in architecture. It's methodology of building solution has been found from the urbanic field. The main problems in architecture are understood as urban structures in which cities have their own inner logics. The asthetic characteristics of the elements in the urban space are considered as physical structures. Contextualists concern the special building, architecture synthetically rather than analytically about modern cities and architecture, and develop the theme of modern cities broadly. European rationalists whose profects the ornamental properties are found from, have synthesized organic architecture with 1920s' rationalism and represented modern rationality.

        The logic of Modern Architecture in the early part of the 20th century was begun to be criticized intensely, and functionalism, idealism, philosophy of university brought about the loss of identity. The attempt to retrieve the humanity by a methodology for solutions of given problems leads contemporary architecture to the pluralism. Consequently, contemporary architecture is to harmonize architecture with environment, and that is possible when we introduce the concept of place into architecture. The concept of place is not new, but rediscovery. Place is every-where human exists. For a long time a place has been understood in Genius Loci and a guardian deity. It is parted into natural and man-made place, and they are subdivided into romantic, classical, cosmic, complex landscape/architecture. Place is not abstract or geometrical but concrete. By definition, combination and concentration of space and control of the direction, place is where man orients, shelters and feels enclosure, intimacy, security and position. A method to express a sense of place based on a mental image of human who experiences a place is embodied through visualization, complementation and symbolization and when a sense of place is expressed. It is possible that man orients and is individualized.
