Cytological, genetic, and proteomic analysis of a sesame (Sesamum indicum L.) mutant Siyl‑1 with yellow–green leaf color

Background Both photosynthetic pigments and chloroplasts in plant leaf cells play an important role in deciding on the photosynthetic capacity and efficiency in plants. Systematical investigating the regulatory mechanism of chloroplast development and chlorophyll (Chl) content variation is necessary for clarifying the photosynthesis mechanism for crops. Objective This study aims to explore the critical regulatory mechanism of leaf color mutation in a yellow–green leaf sesame mutant Siyl-1. Methods We performed the genetic analysis of the yellow-green leaf color mutation using the F2 population of the mutant Siyl-1. We compared the morphological structure of the chloroplasts, chlorophyll content of the three genotypes of the mutant F2 progeny. We performed the two-dimensional gel electrophoresis (2-DE) and compared the protein expression variation between the mutant progeny and the wild type. Results Genetic analysis indicated that there were 3 phenotypes of the F2 population of the mutant Siyl-1, i.e., YY type with light-yellow leaf color (lethal); Yy type with yellow-green leaf color, and yy type with normal green leaf color. The yellowgreen mutation was controlled by an incompletely dominant nuclear gene, Siyl-1. Compared with the wild genotype, the chloroplast number and the morphological structure in YY and Yy mutant lines varied evidently. The chlorophyll content also significantly decreased (P < 0.05). The 2-DE comparison showed that there were 98 differentially expressed proteins (DEPs) among YY, Yy, and yy lines. All the 98 DEPs were classified into 5 functional groups. Of which 82.7% DEPs proteins belonged to the photosynthesis and energy metabolism group. Conclusion The results revealed the genetic character of yellow-green leaf color mutant Siyl-1. 98 DEPs were found in YY and Yy mutant compared with the wild genotype. The regulation pathway related with the yellow leaf trait mutation in sesame was analyzed for the first time. The findings supplied the basic theoretical and gene basis for leaf color and chloroplast development mechanism in sesame.

광고의 대업인 정음반포가 단지 세종의 예단에서 나왔다고 하더라도 소중화로 자처하는 그 당시 사회에서 그대로 준수되었다고는 할 수 없을 것이다. 그 당시 최만리의 상소에 나타난 견해는 수구에 물들어 있는 일반적인 견해라고 하겠고 굳이 최만리 한사람만의 문화상 반역도 아닐 것이다. 이러한 일반적인 분위기속에서 세종은 왕자적 권위에 입각하여 이를 추진하여야만 하였으나, 가장 신임하던 집현전학사 가운데서 마저 완강한 반대에 봉착하게 되었으므로 집현전과는 별개의 기관인 정음청같은 것은 다시 금내에 설치하였음도 이해할 수 있다. 그러나 이 정음청이 무엇을 한 기관이었던가 하는 의문은 아직 의문 그대로 남아있고, 그 당시의 분위기를 이해하는데 정음청의 연구는 불가결의 여건인 것만도 사실이다. 하긴 정음청에 관하여도 이제가지 문헌적으로 제서에서 언급한 바 있고, 또 가장 상세히 기록한 홍모(기문)의 「정음발달사」가 있어, 다시 사족을 붙인다는 것은 쑥스러운 일이지만 관점을 달리하여 정음 실용의 배경을 살피는데 중요한 관약이 되는 당청의 시말을 더듬어 새로운 소견을 제시해 보고자 한다.

This paper argues on Deuteronomy in which Moses repeatedly urged the Israel people to take possession of the Promised Land, and in which Moses was preaching the Laws to Israel to impress God's Word on their hearts. In Deuteronomy the Israel people were to remember what God had already done for them, and to obey Him, fear Him, love Him, and hold fast to Him. In this respect, education was made much of in Deuteronomy. And this paper aims at examining the thought of education in Deuteronomy, centering around educational languages. For abundant educational terminology can be found to be involved in it. On the basis of educational arguments, it can be found that linguistic commentary, admonitions, educational community, historical theology, the Shema found in 6: 4-9, festivals, and brotherly have been dealt with in serious respects. 우슬약침(牛膝藥鍼)이 Rat의 Adjuvant 관절염(關節炎)에 미치는 영향(影響)

To investigate effects of Achyranthis Radix herbal-acupuncture on adjuvant arthritis in rats, the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum and histological test of the muscular tissue were measured in the arthritis part. 1. After elicitating arthritis of Sprague dawley(SD) rats by injection of Freund's complete adjuvant for 2 weeks, normal saline was injected for the Exp. I group and Achyranthis Radix herbal-acupuncture was injected for the Exp. II group during 30days. Selected point was $D\acute{u}b\acute{i}(ST_{35})$ in both the groups. And then the volume of the paw were checked. The volume of the paw was $0.84{\pm}0.14mm$ in the Exp. I group and $0.47{\pm}0.11mm$ in the Exp.II group, the swelling of the paw was restricted significantly in the Exp. II group(p<0.05). 2. The number of WBC was $10.34{\pm}0.14(10^3/ml)$ in the normal group and $37.47{\pm}5.46(10^3/ml)$ in the Exp. I group. It was $21.24{\pm}2.58(10^3/ml)$ in the Exp. II group. This fact showed that the group Exp. II with Achyranthis Radix herbal-acupuncture was more effective than the Exp. II group in the treatment of arthritis(p<0.05). 3. The content of the total protein in the blood serum was $6.14{\pm}0.43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.41{\pm}0.68g/dl$ in the Exp. II group. There was no significance in total protein between the Exp. II group and the Exp. I group from the statistical analysis. 4. The content of albumin in the blood serum was $2.94{\pm}0.13g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in Chang Tong-young the Exp. I group and $3.15{\pm}0.27g/dl$ in the Exp. II group. This fact showed that the Exp. II group had significant increase in the serum albumin from the statistical analysis compared with the Exp. I group. 5. The content of the globulin in the blood serum was $3.19{\pm}0.48g/dl$ in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group and $3.26{\pm}0.57g/dl$ in the Exp. II group. There was no significance in the serum globulin between the Exp. II group and Exp. I group from the statistical analysis. 6. In histological finding, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed small amount of inflammatory cells, the refrained inflammatory state and even recovering state.
