        Iron Determination in Rat Plasma Samples by Inductively Coupled Plasma Emission Spectrometry and Application to Pharmacokinetic Studies

        This paper describes an inductively coupled plasma emission spectrometric method for the analysis of Fe in rat plasma. Calibration curves were obtained in the range of 0.125-1.50 ${\mu}g{\cdot}mL^{-1}$. The relative standard deviation ranges from 5.93% to 6.80%, and accuracy was between 87.6 and 102.0%. Dilution with water had no influence on the performance of the method, which could then be used to quantify Fe concentration in plasma up to 0.50 ${\mu}g{\cdot}mL^{-1}$. The limit of quantification was 0.10 ${\mu}g{\cdot}mL^{-1}$. At this level, the average relative standard deviation was 6.8%. The results indicate that the method meets the accuracy and precision requirements for the pharmacokinetic studies. The Fe concentration in rat plasma was measured and the main pharmacokinetic parameters were calculated by Topfit 2.0 (GmbH. Shering AG, Godecke AG, Germany).

        Subcellular Characterization of Porcine Oocytes with Different Glucose-6-phosphate Dehydrogenase Activities

        The in vitro maturation (IVM) efficiency of porcine embryos is still low because of poor oocyte quality. Although brilliant cresyl blue positive (BCB+) oocytes with low glucose-6-phosphate dehydrogenase (G6PDH) activity have shown superior quality than BCB negative (-) oocytes with high G6PDH activity, the use of a BCB staining test before IVM is still controversial. This study aimed to shed more light on the subcellular characteristics of porcine oocytes after selection using BCB staining. We assessed germinal vesicle chromatin configuration, cortical granule (CG) migration, mitochondrial distribution, the levels of acetylated lysine 9 of histone H3 (AcH3K9) and nuclear apoptosis features to investigate the correlation between G6PDH activity and these developmentally related features. A pattern of chromatin surrounding the nucleoli was seen in 53.0% of BCB+ oocytes and 77.6% of BCB+ oocytes showed peripherally distributed CGs. After IVM, 48.7% of BCB+ oocytes had a diffused mitochondrial distribution pattern. However, there were no significant differences in the levels of AcH3K9 in the nuclei of blastocysts derived from BCB+ and BCB- oocytes; at the same time, we observed a similar incidence of apoptosis in the BCB+ and control groups. Although this study indicated that G6PDH activity in porcine oocytes was correlated with several subcellular characteristics such as germinal vesicle chromatin configuration, CG migration and mitochondrial distribution, other features such as AcH3K9 level and nuclear apoptotic features were not associated with G6PDH activity and did not validate the BCB staining test. In using this test for selecting porcine oocytes, subcellular characteristics such as the AcH3K9 level and apoptotic nuclear features should also be considered. Adding histone deacetylase inhibitors or apoptosis inhibitors into the culture medium used might improve the efficiency of IVM of BCB+ oocytes.

        Effects of Sn and Mn Addition on the Microstructure and Mechanical Properties of As-Extruded Mg–2Al–1Zn Alloys

        The effects of low Sn (1.0 wt%) and Mn (0.5 wt%) content on the microstructures and mechanical properties of as-extrudedMg–2Al–1Zn (AZ21) alloys were systematically investigated. The combined addition of Sn and Mn significantly refinedthe grains of the AZ21 alloy, reducing the average grain size from 25.77 to 2.17 μm. The volume fraction of the dynamicallyrecrystallized (DRXed) grains decreased from 78.49 to 43.81% with Sn (1.0 wt%) and Mn (0.5 wt%) addition. The Al8Mn5,Al11Mn4,and Al9Mn4phases were formed in Mg–2Al–1Zn–1Sn–0.5Mn (AZTM2110) alloy, and the Mg17Al12phase disappeared. The orientation relationship between Al9Mn4and Al11Mn4was (002)Al11Mn4//(̄ 302)Al9Mn4, [ ̄ 100]Al11Mn4//[223]Al9Mn4,and the orientation relationship between Al8Mn5and α-Mg was ( ̄ 252)Al8Mn5//(̄ 12̄ 13)α−Mg. The Al9Mn4and Al11Mn4phasesgrew in the same direction. The intensity of basal fiber texture of AZTM2110 alloy decreased by 21.7%. The ultimatetensile strength, yield strength, and elongation of the AZ21 alloy increased by 29.03%, 65.05%, and 61.02%, respectively. Grain-refinement strengthening was the main mechanism underlying the improvement in AZTM2110 alloy strength. Grainrefinement and the weakened basal fiber texture improved the plasticity of the AZTM2110 alloy.

        Investigation of the Phenomenon of Propagated Sensation along the Channels in the Upper Limb Following Administration of Acupuncture and Mock Laser

        Background: Similar to De Qi psychophysical responses, propagated sensation along the channels (PSC) is considered an important phenomenon in traditional Chinese acupuncture. In acupuncture clinical trials, different acupuncture manipulation techniques are used to enhance the propagation of sensation along the channels to facilitate an optimum therapeutic result. Aim: To examine and compare the PSC reported by participants in a clinical trial following the administration of acupuncture and inactive mock laser. Methods: The study was embedded in a two-arm parallel design multicenter, randomized clinical trial, the Tennis Elbow AcupuncturedInternational StudydChina, Hong Kong, Australia, Italy (TEA IS CHAI). Needle sensations were measured using a validated instrument, the Massachusetts General Hospital Acupuncture Sensation Spreading Scale. Ninety-six participants with lateral elbow pain were randomly allocated into two groups in a 1:1 ratio; the acupuncture treatment group (n Z 47) and the mock laser control group (n Z 49). Participants in both groups received the intervention at two acupoints, LI10 and LI11, consisting of 2 minutes of either standardized needle manipulation or mock laser at each acupoint with a rest period between each intervention period. Data were collected immediately following the interventions at the first and the ninth session within the clinical trial. Results: Although participants in both groups perceived PSC radiating to similar sites along the upper limb, the frequency of the reported radiation sites among the two intervention groups for both radiation up the limb (p < 0.05) and radiation down the limb (p < 0.001) were statistically significantly different. Among the radiating sensation sites recorded within the two study groups, the sensations were reported as radiating a greater distance down the forearm to the wrist compared to up the arm. Evaluation of PSC across the four study sites revealed a statistically significant difference in frequency of the reported radiation down the limb sites in each study group and radiation up the limb sites only in control group only (p < 0.001). Conclusion: The findings of the study demonstrated that the PSC phenomenon is not just associated with needling but can be perceived when using a mock laser.
