      • V. vulnificus의 cyolysin에 의해 형성된 Pored의 이온 투과성에 관한 연구

        V. vulnificus가 분비하는 cytolysin의 pore 형성 효과를 전기생리학적으로 확인하고 그 pore의 이온선택성을 밝히고자, V. vulnificus cytolysin의 표적세포로 예산되는 폐내피세포인 CPAE 세포에서 여러 bath용액에서의 cytolysin pore의 이온투과성을 비교 관찰하여 다음과 같은 결과를 얻었다. 1. Cytolysin 1 HU/ml을 세포내외에 투여시 2-7분의 lag time 후 sensitivity 차이없이 115.6 ± 5.1 pS의 단일 전도도를 갖는 pore를 시간 의존적으로 형성하였으며, 이런 pore의 이온투과성은 세척 후에도 지속적으로 유지되었다. 2. Bath용액의 140 mM KCI을 같은 농도의 NaC1 및 CsC1로 치환하여도 cytolysin pore 의 이온투과성에는 유의한 변동이 없었다. 3. Bath용액의 140 mM KCI을 같은 농도의 CaC1(2) 및 BaCl2로 치환하면 cytolysin pore의 이온투과성이 각각 1.8배와 2.3배로 현저하게 증가하였다. 4. Bath용액의 C1 농도를 동일하게 하고자, 각각 140 mM KCI과 70 mM MgC1(2)로 치환시 cytolysin pore의 이온투과성에는 유의한 차이가 없었다. 5. Bath용액의 KCI농도를 140 mM에서 10mM과 1 Mm로 감소시 cytolysin pore의 역전전압이 0 mV에서 각각 -31.1 ± 1.5mV와 -42.3 ± 1.8 mV로, C1 이온의 역전전압에 근접하게 이동하였다. 이상의 실험결과를 종합하변, V. vulnificus cytolysin은 세포막내외의 차이없이 시간의존 적인 pore를 형성하는데, 이 pore의 투과성은 막전위에 비의존적이며 주로 C1 이온에 선택성이 높음을 시사한다. Cytolysin produced by V. vulnificus has been incriminated as one of the important virulence determinants in V. vulnificus infection. Ion selectivity of cytolysin-induced pres has been examined in CPAE cells, a cell line of pulmonary endothelial cells, using inside-out patch clamp techniques. In symmetrical KCI concentration (140 mM). intracellular and extracellular cytolysin formed the ion-permeable pores with single channel conductance of 115.6±5.1 pS (n=6) in a time-dependent manner and a voltage-independent manner. The pore currents were consistently maintained after washout of cytolysin. Replacement of intracellular 140 mM KCI with 140 mM NaCl, 140 mM CsCl and 70 mM MgCl2 did not affect the pore currents, but replacement of it with equimolar CaCl2 and BaCl2 increased the pore current by 1.8 and 2.4 times of control current (140 mM KCL), respectively. When the intracellular KCI concentration was lowered from 140 to 10 and 1 mM, zero-current membrane potentials shifted from 0 mV to -31.1 ± 1.5 and -42.3 ± 1.8 mV, respectively. These results indicate that cytolysin produced by V. vulnificus formed the CI(-) -selective pores in CPAE cells.

        Identification of interacting proteins of retinoid-related orphan nuclear receptor gamma in HepG2 cells

        The retinoid-related orphan nuclear receptor gamma (RORγ) plays critical roles in regulation of development, immunity and metabolism. As transcription factor usually forms a protein complex to function, thus capturing and dissecting of the RORγ protein complex will be helpful for exploring the mechanisms underlying those functions. After construction of the recombinant tandem affinity purification (TAP) plasmid, pMSCVpuro RORγ-CTAP(SG), the nuclear localization of RORγ-CTAP(SG) fusion protein was verified. Following isolation of RORγ protein complex by TAP strategy, seven candidate interacting proteins were identified. Finally, the heat shock protein 90 (HSP90) and receptor-interacting protein 140 (RIP140) were confirmed to interplay with RORγ by co-immunoprecipitation. Interference of HSP90 or/and RIP140 genes resulted in dramatically decreased expression of CYP2C8 gene, the RORγ target gene. Data from this study demonstrate that HSP90 and RIP140 proteins interact with RORγ protein in a complex format and function as co-activators in the RORγ-mediated regulatory processes of HepG2 cells.         Finding Blue Tracks in Gephyrocharax melanocheir Fish Similar to the Locations of Acupuncture Meridians after Injecting Alcian Blue

        This study investigated whether a meridian-like distribution of Alcian blue (AB) existed after it was injected into a fish's body and suggested a new animal model for meridian study. Twenty Gephyrocharax melanocheir fish with translucent bodies were injected with AB at a point near the spinal column or the dorsal fin. Distribution of AB was observed using a digital camera and a stereomicroscope. Three or more obvious blue tracks were found: one along the spinal column, another along the posterior margin of the abdomen extending to the superior margin of the anal fin, and a third along both sides of the dorsal fin. They were similar to the locations of the governor, conceptual vessel, and urinary bladder meridians, respectively, on the human body according to the classic theory of traditional Chinese medicine. A few other blue tracks were also found, which apparently did not correspond to any known meridians. The results show that the tracks of AB share important similarities with the locations of classically described meridians and that they are mainly distributed in the interstitial space around bones and blood vessels and inside muscular interstices. This study may provide a new experimental animal model for exploring acupuncture meridians.

        Cross-immunizing potential of tumor MAGE-A epitopes recognized by HLA-A*02:01-restricted cytotoxic T Lymphocytes

        Almost all melanoma cells express at least one member of the MAGE-A antigen family, making the cytotoxic T cells (CTLs) epitopes with cross-immunizing potential in this family attractive candidates for the broad spectrum of anti-melanoma immunotherapy. In this study, four highly homologous peptides (P264: FLWGPRALA, P264I9: FLWGPRALI, P264V9: FLWGPRALV, and P264H8: FLWGPRAHA) from the MAGE-A antigens were selected by homologous alignment. All four peptides showed high binding affinity and stability to HLA-A*02:01 molecules, and could prime CTL immune responses in human PBMCs and in HLA-A*02:01/Kb transgenic mice. CTLs elicited by the four epitope peptides could cross-lyse tumor cells expressing the mutual target antigens, except MAGE-A11 which was not tested. However, CTLs induced by P264V9 and P264I9 showed the strongest target cell lysis capabilities, suggesting both peptides may represent the common CTL epitopes shared by the eight MAGE-A antigens, which could induce more potent and broad-spectrum antitumor responses in immunotherapy.         Key challenges, recent advances and future perspectives of rechargeable lithium-sulfur batteries

        Lithium-sulfur (Li-S) battery, which releases energy by coupling high abundant sulfur with lithium metal,is considered as a potential substitute for the current lithium-ion battery. Thanks to the lightweight andmulti-electron reaction of sulfur cathode, the Li-S battery can achieve a high theoretical specific capacityof 1675 mAh g1 and specific energy of 2600 Wh kg1. However, some key scientific issues limit its practicalapplication, such as the ‘‘shuttle effect” of lithium polysulfides, large volume change, poor conductivityof sulfur and its solid-state products, and self-discharge phenomenon. Of these, the ‘‘shuttle effect”is recognized as the most critical challenge affecting on electrochemical performance. Hence, this reviewfirst systematically introduces the development of Li-S batteries and the corresponding ‘‘shuttle effect”. Then, the latest work on anode, cathode, separator and electrolyte are summarized. Finally, some promisingviews on the future research direction of this battery system are put forward.

        Driver circuit to eliminate bridge leg crosstalk in SiC MOSFETs

        This study involves the crosstalk caused by the high-frequency parasitic parameters in the driver circuits of SiC MOSFETs, which are high-speed switching devices. A lumped parameter model for the high-frequency crosstalk of a driver circuit was established. Then, the relationships between the parameters and crosstalk of the driver circuit were analyzed. Finally, an improved SiC MOSFET driver circuit was proposed. The optimized driver circuit parameters can reduce the driver signal oscillation and minimize the crosstalk of the driver circuit. Verification was performed on a test platform of a full-bridge LLC converter. Test results show that the improved driver circuit can remarkably suppress crosstalk, increase the turn-off speed of SiC MOSFETs and reduce the switching loss. This demonstrates the effectiveness of the driver circuit.

      • Using Frequency Scaling on Virtualized Memory in Cloud Datacenters

        As the increasing of IT-infrastructure in cloud platforms, rapidly growth of energy consumption becomes a critical problem in many cloud datacenters. Conventionally, most of studies on energy-efficiency optimization concentrate on CPU related energy costs instead of memory subsystem, since CPU often dominates the total energy consumption in modern servers. However, such a situation is gradually changing as more and more cloud datacenters are equipped with larger and larger memory systems for dealing with data-intensive applications. In this paper, we present a novel mechanism, namely frequency scaling on virtualized memory (FSVM), which applies DVFS technology on memory subsystem based on the characteristics of active VM instances. Comparing with previous studies, our approach provides a fine-grained memory energy consumption conservation mechanism for virtualized servers. Extensive experiments are conducted to investigate the effectiveness and performance of our FSVM, and the results indicate that it can significantly improve the energy-efficiency of memory subsystem in virtualized servers.

        Is There Volume Transmission Along Extracellular Fluid Pathways Corresponding to the Acupuncture Meridians?

        Volume transmission is a new major communication signaling via extracellular fluid (interstitial fluid) pathways. It was proposed by the current authors that such pathways can explain the meridian phenomena and acupuncture effects. To investigate whether meridian-like structures exist in fish body and operate via volume transmission in extracellular fluid pathways, we injected alcian blue (AB) under anesthesia into Gephyrocharax melanocheir, which has a translucent body. The migration of AB could be seen directly and was recorded by a digital camera. The fish was then embedded and cut transversely to observe the position of tracks in three dimensions. Eight longitudinal threadlike blue tracks were recognized on the fish. The positions of these threadlike tracks were similar to meridians on the human body. Transverse sections showed that these tracks distributed to different layers of distinct subcutaneous loose connective tissues and intermuscular septa. Lymphatic vessels were sometimes associated with the extracellular blue tracks where the migration of AB occurred. Extracellular fluid pathways were found on fish through their transport of AB. These pathways operating via volume transmission appeared to be similar in positions and functions to the acupuncture meridians in Chinese medicine.
