      • Common promoter polymorphism in monocyte differentiation antigen CD14 is associated with serum triglyceride levels and body mass index in non-diabetic individuals

        <P>Abstract</P><P>Aims </P><P>Growing evidence supports the hypothesis that chronic low-grade inflammation related to innate immunity may play an important role in the pathophysiology of Type 2 diabetes mellitus (T2DM). The monocyte differentiation antigen CD14 gene (<I>CD14</I>) acts as the receptor for lipopolysaccharide (LPS) and augments monocyte/macrophage inflammatory responses.</P><P>Methods </P><P>We have sequenced the gene, including all exons, exon/intron boundaries, and the −1.5 kb of the 5′ flanking region. Two common loci (minor allele frequency > 0.05) were genotyped in 775 T2DM patients and 316 control subjects recruited in the Korean T2DM Study.</P><P>Results </P><P>Eight polymorphisms, including four non-synonymous forms, were identified in <I>CD14</I>. No polymorphisms were found in association with T2DM. However, one common promoter SNP (<I>−260T>C</I>) was significantly associated with both the serum triglyceride level (TG) and body mass index (BMI) in non-diabetic control subjects. Individuals who carried the minor allele (C) had higher TG levels (1.65 ± 0.81 vs. 1.46 ± 0.80 mmol/l; <I>P</I> = 0.0007) and BMI (23.96 ± 3.00 vs. 23.28 ± 3.22 kg/m<SUP>2</SUP>; <I>P</I> = 0.04) as compared with subjects carrying T/T genotypes.</P><P>Conclusion </P><P>Our data suggest that lipid metabolism and obesity, important pathophysiological elements of T2DM and the metabolic syndrome, are regulated by complex mechanisms that include the CD14 gene polymorphism-mediated genetic propensity to non-specific inflammatory responses.</P>

      • Differentiation of Recurrent Tumor and Posttreatment Changes in Head and Neck Squamous Cell Carcinoma: Application of High b-Value Diffusion-Weighted Imaging

        <P><B>BACKGROUND AND PURPOSE:</B></P><P>High b-value DWI has been expected to have an additional diagnostic role and demonstrated some promising results in head and neck cancer. The aim of this study was to evaluate the diagnostic performance of DWI at a high b-value (<I>b</I>=2000 s/mm<SUP>2</SUP>) compared with a standard b-value (<I>b</I>=1000 s/mm<SUP>2</SUP>) and the ratio of ADC values of high and standard b-values for their ability to differentiate between recurrent tumor and posttreatment changes after the treatment of head and neck squamous cell carcinoma.</P><P><B>MATERIALS AND METHODS:</B></P><P>A total of 33 patients diagnosed with head and neck squamous cell carcinoma were enrolled in the present study; all had contrast-enhancing lesions on follow-up MR imaging. All patients underwent single-shot echo-planar DWI at <I>b</I>=1000 s/mm<SUP>2</SUP> and <I>b</I>=2000 s/mm<SUP>2</SUP>, and corresponding ADC maps were generated (ADC<SUB>1000</SUB> and ADC<SUB>2000</SUB>, respectively). The mean ADC<SUB>1000</SUB>, ADC<SUB>2000</SUB>, and ADC<SUB>ratio</SUB> (ADC<SUB>ratio</SUB> = ADC<SUB>2000</SUB>/ADC<SUB>1000</SUB> × 100) values were evaluated within a manually placed ROI with contrast-enhanced T1-weighted images as references. For the statistical analysis, we performed a Student <I>t</I> test and multivariate logistic regression.</P><P><B>RESULTS:</B></P><P>The mean ADC<SUB>1000</SUB> in recurrent tumor was significantly lower than that in posttreatment changes (<I>P</I> < .001), whereas the mean ADC<SUB>2000</SUB> resulted in no significant difference (<I>P</I> = .365). The mean ADC<SUB>ratio</SUB> was significantly higher in recurrent tumor than that in posttreatment changes (73.5 ± 7.2% vs 56.9 ± 8.8%, respectively; <I>P</I> < .001). Multivariate logistic regression analysis revealed that the ADC<SUB>ratio</SUB> was the only independently differentiating variable (<I>P</I> = .024). The sensitivity, specificity, and accuracy of ADC<SUB>ratio</SUB> were 95.0%, 69.2%, and 84.8%, respectively, by use of the optimal cutoff value of 62.6%.</P><P><B>CONCLUSIONS:</B></P><P>We suggest that the ADC<SUB>ratio</SUB> calculated from the ADC<SUB>1000</SUB> and ADC<SUB>2000</SUB> is a promising value for the differentiation of recurrent tumor and posttreatment changes in head and neck squamous cell carcinoma.</P>

        임신 및 각종 갑상선질환에서 갑상선 기능 판정에 관한 연구 : 혈청유리 T4의 진단적 의의에 관한 고찰 The diagnostic value of free thyroxine by RIA

        To evaluate the diagnostic accuracy of the measurement of free thyroxine(FT4) by radioimmunoassay, we measured free T4 and T4, T3, T3RU, TSH and TBG serum levels by radioimmunoassay in 18 healthy persons and 52 patients with various thyroid diseases and 11 normal pregnant women. The results are as follows. 1) In 19 cases of overt hyperthyroidism, T3, free T4 and FTI, T4/TBG ratio reflect hyperfunction in all cases. T4 is increased in 94%(18/19) and TBG and TSH are decreased in 79%(15/19). 2) In 8 patients with overt hypothyroidism, TSH is increased in all cases and free T4 and FTI is decreased in all cases. T4 is decreased in 87.5%(7/8), T3 is decreased in 75%(6/8) and T4/TBG ratio is decreased in 62.5%(5/8). 3) In 5 patients who are clinically in euthyroid state after treatment of hyperthyroidism, T4 free T4, FTI and TSH are in the normal range in all cases and T3 is normal in 60%(3/5) and slightly increased in 40%(2/5). 4) In 10 patients who showed clinically borderline hypothyroidism after treatment of hyperthyroidism, TSH is increased in all cases and free T4 and FTI are decreased in all cases, but T4 and T3, T4/TBG ratio are in the normal limit in all cases. So after treatment of hyperthyroidism, TSH, free T4 or FTI are recommended as optimal function test. 5) In normal pregnancy, free T4, FTI and T4/TBG ratio reflect normal function, but the other parameters unreliable due to the influence of increased TBG. Also TBG and TSH level in pregnancy is increased significantly compared with normal healthy control group. 6) The coefficients of correlation between T4 and FTI were 0.862(p〈0.001) and 0.685(p〈0.001) between free T4 and T4/TBG ratio. In most patients, diagnostic value of free T4 was comparable and even superior to FTI, so free T4 measurement can be used routinely with thyrotropin assay in the diagnosis of hypothyrodism or with T3 for the diagnosis of hyperthyroidism.

        Effect of Dietary Copper Sources (Cupric Sulfate and Cupric Methionate) and Concentrations on Performance and Fecal Characteristics in Growing Pigs

        This study was conducted to assess the effects of organic and inorganic copper on performance in growing pigs. A total of 100 pigs, average age 63 d and initial body weight 21.46${\pm}$1.13 kg, were assigned to five treatment groups. Dietary treatments included i) CON (basal diet, 0 ppm Cu), ii) T1 (basal diet with 67 ppm Cu as cupric sulfate, $CuSO_4$), iii) T2 (basal diet with 134 ppm Cu as $CuSO_4$), iv) T3 (basal diet with 67 ppm Cu as cupric methionate, CuMet) and v) T4 (basal diet with 134 ppm Cu as CuMet). Throughout the entire experimental period, ADG (average daily gain), ADFI (average daily feed intake) and G/F (gain: feed) ratios showed no significant differences. The dry matter digestibility was improved in the T1, T2, T3, and T4 treatments (p<0.05), as compared with CON. Nitrogen digestibility was improved in the T3 treatment group as compared with CON (p<0.05). As compared with the T1 treatment group, fecal pH values were improved in the CON, T3, and T4 treatment groups (p<0.05). Fecal Cu concentrations were significantly lower in the CON, T3, and T4 treatment groups than in T1 and T2 (p<0.05). The incidence of diarrhea was reduced when the pigs were fed on the T2, T3, and T4 diets as compared with CON. In conclusion, diets supplemented with 67 or 134 ppm Cu as CuMet may prove effective in improving nutrient digestibility and fecal pH value in growing pigs, and fecal Cu concentrations may be reduced by CuMet supplementation.

        This study was conducted to investigate the physico-chemical characteristics of various bone extracts in Hanwoo cattle. The experimental materials are as follows; T1(Shank bone extract), T2(Tail extract), T3(Beef feet extract), and T4(The other bones extract). In the proximate composition, the T1 and T2 showed higher moisture and fat contents than those of T3 and T4(p<0.05), and the T3 had the highest protein content among the bone extracts(p<0.05). The quality properties including salinity, sugar content, turbidity and yellowness(b*) values were significantly higher in the T3 than the other bone extracts(p<0.05). However, the pH value of T3 was significantly lower than the other bone extracts. In the mineral contents, calcium contents of bone extracts were 36.33-48.57mg/kg, and the content of phosphorus was the lowest in T1(4.62mg/kg) and T2(19.88mg/kg) was the highest(p<0.05). Contents of sodium and potassium were the highest in T3(215.83mg/kg) and T2(54.59mg/kg), while the content of magnesium was the lowest in T3(12.28mg/kg). In conclusion, these results could be suggested to be the basis information for processed meat extract products industry in korea using Hanwoo bone extract.

        정상 임신에서 임신주기에 따른 갑상선기능의 변화

        It is well known that normal pregnancy is accompanied by a rise in serum concentrations of thyroxine-binding globulin(TBG) and human chorionic gonadotropin (hCG). Alterations of biochemical parameters of thyroid function are recognized during gestation and sensitive tests to evaluate the alterations easily are required. Therefore, a cross-sectional study was undertaken in 140 healthy pregnant women to evaluate the efficacy of free T_4 measured by 2-step RIA compared to other thyroid function tests and to confirm the changes of thyroid function according to the stages of normal pregnancy. The sensitivities of free T_4 index, free T_4(by 2-step RIA), T_3 and TSH were realtively high(99.3%, 93.6%, 92.9%, 83.6%, respectively) compared to those of T_4 and T_3 bead upgake(49.3%, 21.4%) during all stages of pregnancy. There were positive correlations between free T_4 index and free T_4 or total T_4(r=0.68, r=0.72; p$lt;0.001). The values of free T_4 index sharply decreased from 3.22+-0.10(meam +-SEM) during 6th-12th week to an plateau after 16th-20th week of gestation(p$lt;0.01). The serum concentrations free T_4 and T_3 bead uptake also significantly decreased from 1.65+-0.05 ng/dl, 24.7+- 0.7% during 6th-12th week to an plateau after 16th-20th week of gestation, respectively(p$lt;0.001), No differences were found in the changes of serum concentrations of T_3, T_4 and TSH according to the stages of pregnancy. In conclusion, it is adequate to measure some tests including free T_4 index and free T_4 to evaluate thyroid function during pregnancy. The thyroid physiology and changes of thyroid function according to the stages of pregnancy should be considered in the interpretation of thyroid function status during pregnancy(J Kor Soc Endocrinol 9: 183-189, 1994).

        Sorghum × Sudangrass hybrid 의 입모저장에 관한 연구 ; 1 . 입모저장기간이 생육특성 및 기호성 변화에 미치는 영향

        Growth characteristics and palatability by different harvesting dates were measured in order to determine its availability and effective storage period when sorghum × sudangrass hybrid were stored as foggage. Experiment was carried out with six treatments as follows; T1, T2 and T3 were foggaged at 120, 160 and 200㎝ in plant length and T4, T5 and T6 were foggaged at boot, milk and dough stage on 7 october after regrowth respectively. Rice straw was offered to examine the relative palatability a, control. 1. Growth characteristics such as plant length, leaf length, leaf width, leaf number and stem hardness were decreased with prolongation of foggage period at all treatments. Especially, decrease and falling ratio were a heigher at T5 and T6 than the other treatments. 2. Greeness grade of leaf showed fast decoloration at TI and T2 treatments, but stem showed fast decoloration at TS and T6 treatments. Leaf and stem decolorized in mid-October and mid-november in all treatments. 3. Leaf ratio was deceased with prolongation of foggage period, but stem showed gradually increase. Grain ratio was stable increase at T6 treatments, but not at T5 treatment, 4. Dry matter contents of leaf and grain were stable increased at all treatment but stem was multifarious according to weather conditions. 5. Changes of palatability were high at T1, T2 and T3 treatments untill mid-October, but T4 treatment and rice straw showed high in early December. 6. According to the results obtained in this study, it was suggested that when sorghum x sudangrass hybrid is used as foggage, foggaging period would be suitable until mid-november and growth stage for foggaging would be recommendable from T2 to boot stage for maximum availability.

        톱밥 발효사료 급여 및 분말어유의 첨가사료가 돈육의 이화학적 성질에 미치는 영향

        This study was conducted to investigate the influence of pork quality pigs which were fed with diets supplemented with fermented sawdust and sardin powder oil. The pigs were randomly assigned to one of the four treatments: a), control (commercial feed); b), T1 (30% fermented sawdust); c), T2 (10% sardin powder oil) d), T3 (30% fermented sawdust, l0% sardin powder oil and 30% limited amino acid) from 30 kg to 110 kg B. Wt. and slaughtered. The samples were stored at 0±1 C. The physico-chemical properties were determined at the dust of 2, 4, 6, 8 and 15. For the loin the pH's of all the treatments on the 15th day were higher than those of the 2nd day, and that of TI was higher than those of the others. The total moisture contents of all treatments decreased with the storage (P$lt;0.05), and there were no significant differences among treatments on the 15th day. The water soluble proteins of all treatments increased with time and were highest on the 8th day and decreased gianibicanth thereater. T1 had significantly higher water soluble protein than others(P$lt;0.05). The salt soluble proteins of all treatments increased with lure the days(P$lt;0.05). T1 had higher salt soluble protein than others during the storage period(P$lt;0.05). The brittleness, hardness, cohesiveness, gumminess and chewiness of all the treatments were not significantly different among during the period, but elasticity of them increased after from the 2nd day. The brittleness of control was higher than that of the others on the 15th day, and the hardness and chewiness of control were higher than those of the others on the 6th day(P$lt;0.05). Forthebelly the pH's of all the treatments on the 15th day were higher than those of the at 2nd day(P$lt;0.05). T1 showed higher pH than the others during the period(P$lt;0.05). The total moisture contents of control and T1 were significantly decreased with time and there were no significant differences between T2 and T3. T1 showed the highest value. The water soluble proteins of all the treatments decreased with time and was lower on the 15th day(P$lt;0.05). That of control and T1 was significantly higher than that of others on the 2nd day, but there were no significant differences among the treatments on the 15th day. The salt soluble proteins of all treatments increased with time (P$lt;0.05), and they were highest on the 15th day. T1 had significantly higher salt soluble protein than the others on the 8th and 15th days.
