        ABSTRACTMechanical alloying of Al-8wt%Ti and Al-12wt%Ti alloy powders has been studies to understand the mechanical alloying process.Various analytical techniques including microvickers hardness test, SEM, optical microscopy, and X-ray diffraction were used to find optimum conditious for mechanical alloying.Homogeneity of Al-8wt%Ti composite metal powders was obtained at the following process condition: addition of 1.5wt% stearic acid as PCA, the milling velocity of 300rpm and the milling time more that 10hours.At the above conditions it was also found that the mechanical alloyed powders reached steady state.At the steady state the particles have a uniform size distribution, a saturation hardness, an equiaxed shape, a composite structure and a chemical homogeneity.       • 기계적 합금화에 의한 Nb-35at%AI혼합분말의 상변화

        To investigate the phase transformation behaviors of mechanically alloyed Nb-35at%AI powders, the mixed Nb-35at%AI powders were mechanically alloyed in SPEX 8000 Mixer/Mill. Mechanical alloying(MA) time was varied between 1 hour and 20 hours. The phase formation behaviors of these mechanically alloyed powders were examined using X-ray diffraction(XRD), scanning electron microscopy(SEM) and differential thermal analysis(DTA), Appreciable amorphization started from 10 hours of MA. The powders mechanically alloyed for 10 hours were in almost amorphous phase. DTA results showed that the powders mechanically alloyed for 10 hours had a strong exothermic peak about 600℃.

        Dummy-Bar Head is used for continuous casting equipment. It is heated up at high temperature and cooled down repeatedly in using. Then the thermal extension by thermal stress and high temperature oxidation occur to it. This easily results in heating damage because Head, contact with melting metal, appears in micro-crack and tension force. In other words, it is material that gives the greatest influence in endurance life. The method of study is to compare and evaluate present material and improved material by the new alloy design. In conclusion, the results are as follows : 1) I think that the following range is suitable to chemical component. ◁표 삽입▷(원문을 참조하세요) 2) Micro-crack occurs in repeated heating and cooling owing to decarburization and thermal extension. Adding carbide stability elements-Cr, Mn, and V-shows martensite and less micro-crack. 3) Dummy-Bar Head is a large cast steel, An effective heat treatment condition is to heat step by step and to cool by force at 880℃. After that, It is effective to cool at 600∼680℃. 4) Improved product maintains the even hardness value. And impact value is over three times higher than present product for repeated heating and cooling. 5) The transition range over the fracture toughness value has smaller in improved product than present product for repeated heating and cooling. Through the fracture toughness evaluation, the characteristics of change is evaluated in materials. And we can make the predictable criteria of life with Refrica method. According to the analysis and investigation of the real condition is use, the improved product is accomplished in optimal alloying design and product processing, and satifies the required condition. I think that it is widely used as proper materials to a higher temperature concerning the economy.

        In other to investigate α-1, 3 glucanase from microorganisms for hydrolysis of dental caries, S. muatns was immobilized by cubes of 28% polycrylamide and coated with same polyacrylamide. The insoluble glucan obtained 1.13g per one liter in TH medium with 5% sucrose by PA immobilization of S. muatns, which was very pure by IR and TLC analysis. α-1, 3 glucanase produced by of the Strptomyces SW-522, was developed by treatment with 5% EMS. The new Sw-522 M1 strain obtained, which improved activity of α-1, 3 glucanase.

        Background : This study was development of moisture tolerance and high-yielding Rehmannia glutinosa cultivar. Methods and Results : Segang is developed by the medicinal crop breeding team of National Institute of Horticulture and Herbal Science(NIHHS), Rural Development Administration(RDA), during the period from 2005 to 2015. The reproduction of Rehmannia glutinosa has been accomplished mainly by vegetative propagation with its seedlings have many variants. The cultivar was selected from seedling of Jihwang 1(check variety). The plant type of Segang is some rising from ground. Regional yield trials conducted at three site from 2014 to 2015. The root yield of Segang was 21.1ton per hectare, which was increased 12% compared with Jihwang 1. Also, Segang has higher catalpol content and dried root ratio compared with Jihwang 1. Conclusion : Segang is a moisture tolerance and high-yielding Rehmannia glutinosa cultivar.

        Platform Technologies for Research on the G Protein Coupled Receptor: Applications to Drug Discovery Research

        G-protein coupled receptors (GPCRs) constitute an important class of drug targets and are involved in every aspect of human physiology including sleep regulation, blood pressure, mood, food intake, perception of pain, control of cancer growth, and immune response. Radiometric assays have been the classic method used during the search for potential therapeutics acting at various GPCRs for most GPCR-based drug discovery research programs. An increasing number of diverse small molecules, together with novel GPCR targets identified from genomics efforts, necessitates the use of high-throughput assays with a good sensitivity and specificity. Currently, a wide array of high-throughput tools for research on GPCRs is available and can be used to study receptor-ligand interaction, receptor driven functional response, receptor-receptor interaction, and receptor internalization. Many of the assay technologies are based on luminescence or fluorescence and can be easily applied in cell based models toreduce gaps between in vitro and in vivo studies for drug discovery processes. Especially, cell based models for GPCR can be efficiently employed to deconvolute the integrated information concerning the ligand-receptor-function axis obtained from label free detection technology. This review covers various platform technologies used for the research of GPCRs, concentrating on the principal, non-radiometric homogeneous assay technologies. As current technology is rapidly advancing, the combination of probe chemistry, optical instruments, and GPCR biology will provide us with many new technologies to apply in the future.

      • Detection of calcium signaling in MCH1 receptor

        Melanin concentrating hormone(MCH)은 동물에서 외측 시상하부와 zona incerta에서 발현되는데 에너지 균형과 체중을 조절하는 중요한 매개체로 작용한다. 사람에서 MCH는 MCH receptor subtype 1 (MCH1)과 subtype 2 두 가지의 수용체를 통해 전달되는데 그 중 MCH1의 신호전달 과정을 통해 음식섭취와 에너지 소비를 조절하기 때문에 MCH1 antagonist로 비만, 대사 증후군의 치료제로 연구되고 있다. MCH1는 두 가지의 G protein-coupled receptors로써 신호 전달되는데 Gi- or Gq-dependent 경로를 다중 활성화한다. 본 실험에서는 Gq 경로를 통해 변화하는 세포 내 칼슘 이온을 형광염료를 이용해 확인하기 위하여 MCH1이 발현되어있고, Gq pathway를 활성화하는 Gα16 단백질이 동시에 발현되어 있는 세포를 사용하였다. MCH 가 결합했을 때 MCH1의 Gq 경로를 통해 소포체에서 세포질로 방출되는 칼슘 이온이 형광 염료와 결합하여 형광을 띄는 adherent 세포를 현미경을 통해 관찰하여 MCH를 첨가하였을 때 세포에서 형광의 변화를 동영상으로 얻을 수 있었다. 그리고 다기능 플레이트 신호측정장비인 EnVison을 사용하여 세포 내 칼슘 변화를 형광 신호 값으로 측정 함으로써 서로 다른 농도의 MCH 첨가에 따른 농도 의존성 반응 곡선을 구할 수 있었다.

        Microbial transformation of ginsenoside Rg3 to ginsenoside Rh2 by Esteya vermicola CNU 120806.

        <P>In the present investigation, we successfully employed a cell-free extract of Esteya vermicola CNU 120806 to convert ginsenoside Rg3 to Rh2. Three important factors including pH, temperature and substrate concentration were optimized for the preparation of Rh(2). The optimal condition was obtained as follows: 50C, pH 5.0 and substrate concentration of 3 mg ml(-1). The yield of conversion was up to 90.7%. In order to identify the specificity of the β-glucosidase activity of Esteya vermicola CNU 120806, ginsenoside Re (protopanaxatriol saponins) was treated under the same reaction system. Interestingly, no new metabolite was generated, which elucidated that the enzymatic process only occurred by hydrolysis of the terminal glucopyranosyl moieties at the C-3 carbon of ginsenoside Rg(3). The crude enzyme extract can be used for commercial ginsenoside Rh(2) preparation.</P>
