        Effects of Chronic Mild Stress in Female Bax Inhibitor-1-Gene Knockout Mice

        Objective: The anti-apoptotic protein Bax inhibitor-1 (BI-1) is a regulator of apoptosis linked to endoplasmic reticulum (ER)stress, and BI-1-/- mice exhibit increased sensitivity to tissue damage. The purpose of this study was to investigate the role of BI-1 in the pathogenesis of chronic mild stress (CMS)-induced depression-like behaviors in BI-1-/- mice. Methods: We delivered CMS for 2 or 6 weeks in BI-1-knockout and wild-type mice. Control groups of BI-1-knockout and wild-type mice were left undisturbed. The measured parameters were sucrose consumption at weeks 1, 2, 3, 4, 5, and 6, spontaneous locomotion, and a forced swimming test (FST) at weeks 2 and 6. Results: Significant decreases in sucrose consumption and increases in immobility time in the FST were observed in both stress groups compared with the non-stress groups. Interestingly, at week 2, but not at week 6, BI-1-/--stress mice showed less sucrose intake and greater immobility time than did BI-1+/+-stress mice. Conclusion: These results suggest that BI-1 may play role in protecting against the depressogenic effects of CMS in the short term, but not in the long term. Further study is required to deepen understanding of the role of BI-1 in protecting against depression.

      • Design of Tool Automatic Identification and Database Management System Based on RFID

        RFID (Radio Frequency Identification) is a non-contact automatic identification technology, without human intervention. It is convenient to apply and it can identify fast moving objects. Around the tool and the tool information, the problem of low operational efficiency and low degree of tool management information are still unsolved. In this paper, RFID technology is applied in the tool automatic identification and management system. The tool automatic identification and database management system based on RFID is built. The application model, tool information database design and Upper Computer management software of the RFID in tool identification and management are analyzed. The overall architecture and the software function module of the system are constructed. The database management system is developed by the LabVIEW software. The interface design between LabVIEW and Access database is completed by using ActiveX, as a result, the information of Access database can be written-in and read-out by the management system. Meanwhile, the communication protocol between the Upper Computer and Lower Computer is programed that based on VISA. The RS232 is applied to make the communication between the Upper Computer and Lower Computer come true. Finally, take the SECO's Solid Carbide Ball-end Milling Cutter as an example to have an experiment. Through the experiment, the system proved to be with high reliability and efficiency.

      • Anti-metastasis Activity of Black Rice Anthocyanins Against Breast Cancer: Analyses Using an ErbB2 Positive Breast Cancer Cell Line and Tumoral Xenograft Model

        Background: Increasing evidence from animal, epidemiological and clinical investigations suggest that dietary anthocyanins have potential to prevent chronic diseases, including cancers. It is also noteworthy that human epidermal growth factor receptor 2 (ErbB2) protein overexpression or ErbB2 gene amplification has been included as an indicator for metastasis and higher risk of recurrence for breast cancer. Materials and Methods: The present experiments investigated the anti-metastasis effects of black rice anthocyanins (BRACs) on ErbB2 positive breast cancer cells in vivo and in vitro. Results: Oral administration of BRACs (150 mg/kg/day) reduced transplanted tumor growth, inhibited pulmonary metastasis, and decreased lung tumor nodules in BALB/c nude mice bearing ErbB2 positive breast cancer cell MDA-MB-453 xenografts. The capacity for migration, adhesion, motility and invasion was also inhibited by BRACs in MDA-MB-453 cells in a concentration dependent manner, accompanied by decreased activity of a transfer promoting factor, urokinase-type plasminogen activator (u-PA). Conclusions: Together, our results indicated that BRACs possess anti-metastasis potential against ErbB2 positive human breast cancer cells in vivo and in vitro through inhibition of metastasis promoting molecules.

        Overexpression of the potato StEPF2 gene confers enhanced drought tolerance in Arabidopsis

        Epidermal patterning factor 2 (EPF2) is a negative regulator of stomatal development, and is essential for plant growth, development, and environmental stress responses. However, the role of EPF2 in potato (Solanum tuberosum) has not been investigated to date. Here, we cloned and characterized the potato EPF2-like gene (StEPF2). StEPF2 is predominantly intercellular space localized and its transcripts were rhythmically expressed, and showed the highest expression in apical unexpanded leaves. Expression of StEPF2 was markedly down-regulated in response to abscisic acid and sodium chloride treatments; however, upon the application of polyethylene glycol, the expression of StEPF2 peaked at 4 h and then decreased gradually. Overexpression of StEPF2 in Arabidopsis (OE) substantially reduced stomatal density and photosynthetic rate, but had little effects on plant growth. Under drought stress, OE lines maintained higher photosynthetic rates, photosystem II efficiency, and instantaneous water use efficiency than wild-type (WT) plants. Moreover, OE lines showed less water loss and hydrogen peroxide accumulation in detached leaves compared with WT plants. Thus, our results suggest that StEPF2 acts as a negative regulator of stomatal development in potato, indicating that the role of EPF2 is conserved across plant species. Overall, StEPF2 represents an important target for the development of drought-tolerant potato cultivars via genetic engineering.

      • Impact of IL-2 and IL-2R SNPs on Proliferation and Tumor-killing Activity of Lymphokine-Activated Killer Cells from Healthy Chinese Blood Donors

        One of the goals of tumor immunotherapy is to generate immune cells with potent anti-tumor activity through in vitro techniques using peripheral blood collected from patients. However, cancer patients generally have poor immunological function. Thus using patient T cells, which have reduced in vitro proliferative capabilities and less tumor cell killing activity to generate lymphokine-activated killer (LAK) cells, fails to achieve optimal clinical efficacy. Interleukin-2 (IL-2) is a potent activating cytokine for both T cells and natural killer cells. Thus, this study aimed to identify optimal donors for allogeneic LAK cell immunotherapy based on single nucleotide polymorphisms (SNP) in the IL-2 and IL-2R genes. IL-2 and IL-2R SNPs were analyzed using HRM-PCR. LAK cells were derived from peripheral blood mononuclear cells by culturing with IL-2. The frequency and tumor-killing activity of LAK cells in each group were analyzed by flow cytometry and tumor cell killing assays, respectively. Regarding polymorphisms at IL-2-330 (rs2069762) T/G, LAK cells from GG donors had significantly greater proliferation, tumor-killing activity, and IFN-${\gamma}$ production than LAK cells from TT donors (P<0.05). Regarding polymorphisms at IL-2R rs2104286 A/G, LAK cell proliferation and tumor cell killing were significantly greater in LAK cells from AA donors than GG donors (P<0.05). These data suggest that either IL-2-330(rs2069762)T/G GG donors or IL-2R rs2104286 A/G AA donors are excellent candidates for allogeneic LAK cell immunotherapy.

        Modification of hollow BiOCl/TiO2 nanotubes with phosphoric acid to enhance their photocatalytic performance

        Organic pollutants in dyed wastewater are hazardous to human health and the environment. The photocatalyticdegradation of pollutants is considered a green treatment with significant social and environmental benefits. Inthis study, hollow BiOCl/TiO2 nanotubes with open pores at both ends, prepared by hydrothermal synthesis, weremodified using phosphoric acid to promote photoelectron transfer. The hollow nanotubes offer an increased numberof electroactive sites, which enhances the photoelectric transfer efficiency and reduces the electron-hole recombinationrate. The modification by H3PO4 significantly optimized the photocatalytic activity of the nanotubes. The results showthat the H3PO4 modified hollow BiOCl/TiO2 (molar ratio of BiOCl to TiO2 is 0.8) nanotubes exhibited the most efficientphotocatalytic performance toward rhodamine B (RhB) with a photodegradation efficiency of up to 98.8% undervisible-light illumination. These nanotubes have broad application prospects for the effective removal of organic pollutantsfrom wastewater and can potentially inform future research on photocatalysts.

        Nurse Led Smartphone Electrographic Monitoring for Atrial Fibrillation after Ischemic Stroke: SPOT-AF

        Background and Purpose Paroxysmal atrial fibrillation (PAF) underlying acute stroke frequently evades detection by standard practice, considered to be a combination of routine electrocardiogram (ECG) monitoring, and 24-hour Holter recordings. We hypothesized that nurse-led in-hospital intermittent monitoring approach would increase PAF detection rate. Methods We recruited patients hospitalised for stroke/transient ischemic attack, without history of atrial fibrillation (AF), in a prospective multi-centre observational study. Patients were monitored using a smartphone-enabled handheld ECG (iECG) during routine nursing observations, and underwent 24-hour Holter monitoring according to local practice. The primary outcome was comparison of AF detection by nurse-led iECG versus Holter monitoring in patients who received both tests: secondary outcome was oral anticoagulant commencement at 3-month following PAF detection. Results One thousand and seventy-nine patients underwent iECG monitoring: 294 had iECG and Holter monitoring. AF was detected in 25/294 (8.5%) by iECG, and 8/294 (2.8%) by 24-hour Holter recordings (P<0.001). Median duration from stroke onset to AF detection for iECG was 3 days (interquartile range [IQR], 2 to 6) compared with 7 days (IQR, 6 to 10) for Holter recordings (P=0.02). Of 25 patients with AF detected by iECG, 11 were commenced on oral anticoagulant, compared to 5/8 for Holter. AF was detected in 8.8% (69/785 patients) who underwent iECG recordings only (P=0.8 vs. those who had both iECG and 24-hour Holter). Conclusions Nurse-led in-hospital iECG surveillance after stroke is feasible and effective and detects more PAF earlier and more frequently than routine 24-hour Holter recordings. Screening with iECG could be incorporated into routine post-stroke nursing observations to increase diagnosis of PAF, and facilitate institution of guideline-recommended anticoagulation.

        The influence factors on CeSn0.8W0.6Ox/TiO2 for catalytic removals of NO, CO and C3H8

        Series of CeSn0.8W0.6Ox/TiO2 catalysts were tested for selective catalytic reduction of NO and forsynergistic catalytic removals of CO and C3H8 from diesel engine exhaust. Results revealed that catalyst12%-CeSn0.8W0.6Ox/TiO2 calcined at 500 C exhibited the optimal catalytic performance for NH3-SCR ofNO. The catalyst obtained more than 90% NO conversion at a wide temperature range of 252–456 C. BothCO and C3H8 could be oxidized into CO2 by the optimized catalyst. Moreover, excellent redox property,rich surface acidity and big specific surface area were the promotional factors for good catalyticperformance in catalytic removals of NO, CO and C3H8.

        Mechanical and Thermal Properties of Epoxy Resins Modified by a Novel Thermoplastic-polyimide

        In this paper, a novel polyimide with trifluoromethyl group (PIS) was synthesized successfully and used toimprove the toughness of epoxy resins. Five modified epoxy resins (5, 7.5, 10, 12.5 and 15 wt%) were prepared andinvestigated, respectively. The structure, thermal properties, tensile properties, viscoelastic properties, toughness andmorphology of modified epoxy resins were studied by Fourier Transform Infrared Spectroscopy (FT-IR), thermogravimetricanalysis (TGA), Instron 5967, Dynamic Mechanical Analysis (DMA), and Scanning Electron Microscopy (SEM). Theresults showed that the epoxy resins modified with PIS exhibited good thermal stability which the glass transitiontemperatures increased from 135.9 oC to 146.4 oC. Tensile strength of modified epoxy resins increased from 40.4 MPa to92.3 MPa. The KIC values increased from 0.86 MPa·m1/2 to 1.56 MPa·m1/2 indicated that the toughness enhancement wasmarkedly. The fracture modes also were examined by SEM images and exhibited the fracture mode changed from the brittleone to ductile fracture with varied weight of the novel trifluoromethyl-polyimide.
